BACKGROUND:Both genetic and environmental factors contribute to the development of periodontitis. Genetic studies identified a variety of candidate genes for periodontitis. The aim of the present study is to identify the most promising candidate genes for periodontitis using an integrative gene ranking method.METHODS:Seed genes that were confirmed to be associated with periodontitis were identified using text mining. Three types of candidate genes were then extracted from different resources (expression profiles, genome-wide association studies). Combining the seed genes, four freely available bioinformatics tools (ToppGene, DIR, Endeavour, and GPEC) were integrated for prioritization of candidate genes. Candidate genes that identified with at least three programs and ranked in the top 20 by each program were considered the most promising.RESULTS:Prioritization analysis resulted in 21 promising genes involved or potentially involved in periodontitis. Among them, IL18 (interleukin 18), CD44 (CD44 molecule), CXCL1 (chemokine [CXC motif] ligand 1), IL6ST (interleukin 6 signal transducer), MMP3 (matrix metallopeptidase 3), MMP7, CCR1 (chemokine [C-C motif] receptor 1), MMP13, and TLR9 (Toll-like receptor 9) had been associated with periodontitis. However, the roles of other genes, such as CSF3 (colony stimulating factor 3 receptor), CD40, TNFSF14 (tumor necrosis factor receptor superfamily, member 14), IFNB1 (interferon-β1), TIRAP (toll-interleukin 1 receptor domain containing adaptor protein), IL2RA (interleukin 2 receptor α), ETS1 (v-ets avian erythroblastosis virus E26 oncogene homolog 1), GADD45B (growth arrest and DNA-damage-inducible 45 β), BIRC3 (baculoviral IAP repeat containing 3), VAV1 (vav 1 guanine nucleotide exchange factor), COL5A1 (collagen, type V, α1), and C3 (complement component 3), have not been investigated thoroughly in the process of periodontitis. These genes are mainly involved in bacterial infection, immune response, and inflammatory reaction, suggesting that further characterizing their roles in periodontitis will be important.CONCLUSIONS:A combination of computational tools will be useful in mining candidate genes for periodontitis. These theoretical results provide new clues for experimental biologists to plan targeted experiments.
BACKGROUND:As one of common oral diseases, periodontitis is the leading cause of tooth loss in adults. <br> OBJECTIVE:To review the research progress on periodontitis treatment. <br> METHODS:A computer-based search was performed on the PubMed database and CNKI database for articles addressing periodontitis treatment. The key words were“periodontitis, treatment, tissue engineering, gene engineering”in English and Chinese, respectively. Final y, a total of 57 articles were included to review. <br> RESULTS AND CONCLUSION:Currently, the majority of scholars are stil focus to improve periodontal tissue, promote the regeneration and repair of periodonttal tissue. With the development of tissue engineering and gene technology in recent years, periodontitis has been treated from multiple perspectives and multiple aspects, to solve the problems of patients with periodontitis. However, some treatment methods are stil in their early stage and need in-depth research.
Objective This present study was to detect the role of IL-17A on the correlation between chronic periodontitis(CP) and rheumatoid arthritis(RA).Methods According to the inclusion criteria selected 85 subjects were detected into four groups:①20 patients with chronic periodontitis;②23 patients with the RA;②26 patients with CP and RA;④16 healthy numbers as the control group.The clinical periodontal parameters including probing depth (PD),clinical attachment loss(CA L),and bleeding index(BI) were examined,and the serum and GCF levels of IL-17A were measured by enzyme-linked immunosorbent assay(ELISA).Results The IL-17A levels in serum of patients with CP and RA[(2216.0 ± 1520.87)pg/mL]were significantly higher than the RA group [(851.59 ± 975.81) pg/mL],CP group [(1039.7 ± 1004.83) pg/mL] and the control group[(209.48 ± 50.02)pg/mL].Serum levels of IL-17A in CP group and RA group were significantly higher than the healthy control group(P <0.05).The IL-17A levels in GCF of in the HP group[(356.75 ±69.07)pg/mL],RA group[(381.08 ± 79.48) pg/mL]/CP group [(397.82 ± 108.53) pg/mL] and patients with CP and RA[(414.71 ± 103.09)pg/mL] showed a trend of increase,but no statistical difference (P >0.05).Conclusions IL-17A may have some significance in the study of the correlation between chronic periodontitis and rheumatoid arthritis.
Objective To evaluate the MMP-9 levels in the unstimulated whole saliva (UWS),gingival crevicular fluid (GCF) and serum of patients with chronic periodontitis before and after the initial phase of periodontal treatment and to determine whether they are possible diagnostic and prognostic biomarkers for periodontitis.Methods Twenty patients with periodontitis and 20 healthy controls were included,and periodontal conditions (BI,PD,CAL) were assessed.The levels of MMP-9 in UWS,GCF and serum were measured by enzyme-linked immunosorbent assay before and after the treatment (BT and AT).Results ① In addition to CAL,the conventional periodontal treatment resulted in significant improvement in clinical parameters compared with before treatment.[CAL BT(5.50 ± 1.97) mm,AT (5.50 ± 1.97) mm,P =1.000 ; PD BT (7.05 ± 1.81) mm,AT(4.23 ± 1.06)mm,P =0.000;BI BT 3.75 ±0.44,AT 0.20 ±0.41,P =0.000]②MMP-9 levels in UWS,GCF and serum were significantly decreased six weeks after treatment[UWS BT(22.89 ± 5.28)ng/mL,AT(18.11 ±7.19) ng/mL,P =0.000;GCF BT(41.80 ±2.90) ng/mL,AT(35.71 ±4.49) ng/mL,P =0.000;serum BT(6.67 ± 5.318) ng/mL,AT (2.47 ± 2.713) ng/mL,P =0.004] ③In addition to serum,MMP-9 levels in UWS and GCF before and after the initial phase of periodontal treatment revealed significant increases compared with the control group[UWS of control group (6.78 ± 3.02) ng/mL,GCF (30.20 ± 3.64) ng/mL,P =0.000 ; serum of control group (1.18 ± 0.88) ng/mL,compared with BT and AT P =0.004,P =0.055].Conclusions The high expression of MMP-9 in unstimulated whole saliva is likely to become indicators in chronic periodontitis,and may provide a reference for clinical diagnosis and treatment.
目的:研究通过观察牙周基础治疗对口腔扁平苔藓伴牙周炎患者血清中肿瘤坏死因子-α(tumour necro-sis factor-α,TNF-α)及白细胞介素-6(interleukin-6,IL-6)浓度的影响,探讨牙周干预对口腔扁平苔藓患者治疗的意义.方法:选取口腔扁平苔藓伴牙周炎患者(30)例,将其分为牙周治疗组(15)例和非牙周治疗组(15)例,牙周治疗组采用牙周基础治疗联合药物治疗,非牙周治疗组仅采用药物治疗.应用酶联免疫吸附法检测两组患者在治疗前及治疗后1个月血清中TNF-α及IL-6的浓度.结果:口腔扁平苔藓伴牙周炎患者牙周临床指标及血清中TNF-α表达水平在牙周治疗组治疗后低于治疗前(P<0.05),且较非牙周治疗组治疗后显著降低,差异具有统计学意义(P<0.05);IL-6表达水平牙周治疗组在治疗后较治疗前有下降的趋势,然而在统计学上无显著差异.结论:牙周基础治疗可有效改善口腔扁平苔藓患者牙周状况、临床表现及降低其血清中TNF-α的表达水平,提示牙周干预对口腔扁平苔藓患者具有积极的治疗作用.
