口腔扁平苔藓(oral lichen planus,OLP)是一种慢性炎症性黏膜病,已有文献表明精神因素是其重要的致病因素之一.同时,OLP又是一种免疫失调引起的自身免疫性疾病,笔者怀疑精神因素可能通过某些因子作用于局部免疫,引起免疫失调,从而在OLP的发病机制中发挥作用.5-羟色胺(5-hydroxytryptamine,5-HT)是一种单胺类神经递质,可作为神经系统和免疫系统双边接触的介质.5-HT可能通过与多种受体结合,激活不同信号通路,对情绪调节以及免疫细胞的增殖、迁移、凋亡等过程均起到重要的调节作用.近年来的研究表明5-HT可能参与OLP的发生、发展过程.该文针对5-HT及其受体的功能、它们在自身免疫性疾病中的特异性表达以及在OLP中可能发挥的作用作一综述.
氧化锆陶瓷的优良特性使其逐步成为口腔修复领域的理想材料,其应用范围和应用形式近年来发展迅速.氧化锆目前已经在种植体、基台、冠修复、全牙弓种植修复等方面应用于临床,其临床效果受到广泛关注,长期临床研究报道不断涌现.本文就近年来氧化锆在口腔种植修复中的实际临床效果进行综述,所做结论均基于至少有1年以上随访的研究,重点关注氧化锆与传统材料修复体在力学性能、生物相容性和美观性方面的优劣,为口腔种植修复中氧化锆材料的选用提供参考.
背景:骨髓间充质干细胞在体外培养传代过程中存在细胞老化现象,但其机制尚不明确.组蛋白去乙酰化酶可以使组蛋白脱乙酰化而使染色质处于浓缩状态,进一步关闭基因.目的:探讨骨髓间充质干细胞衰老过程中组蛋白去乙酰化酶9的表达模式.方法:通过生物信息学挖掘技术分析组蛋白去乙酰化酶在骨髓间充质干细胞中的表达;通过体外传代培养骨髓间充质干细胞,β-半乳糖甘酶染色和qRT-PCR方法检测其衰老状态,qRT-PCR、Western blot及免疫荧光技术检测组蛋白去乙酰化酶9的表达模式.结果 与结论:①数据挖掘、生物信息学分析显示在骨髓间充质干细胞衰老过程中,所有组蛋白去乙酰化酶亚型中,只有组蛋白去乙酰化酶9呈现高表达;②骨髓间充质干细胞在传至10代时可表现明显的衰老状态,如β-半乳糖甘酶染色阳性细胞最为明显,衰老相关基因表达上调;③组蛋白去乙酰化酶9的表达与数据挖掘结果一致,呈现上调;组蛋白去乙酰化酶9在年轻骨髓间充质干细胞的胞核与胞浆中均有表达,但其在胞核中的表达较低,衰老骨髓间充质干细胞胞浆中几乎不表达组蛋白去乙酰化酶9,而其在胞核中的表达增加;④结果表明,组蛋白去乙酰化酶9在骨髓间充质干细胞衰老过程中具有重要的作用,但其衰老表型与组蛋白去乙酰化酶9发挥作用的分子机制尚待进一步研究.
神经细胞粘附分子1(neural cell adhesion molecule l,NCAM1)是一种非钙依赖性粘附分子,能介导细胞与细胞、细胞与细胞外基质间的相互作用.在神经系统的发育、免疫细胞功能的调节、细胞的识别及转移、肿瘤的浸润与生长等方面发挥重要作用.近年来的研究表明NCAM1可能参与了口腔扁平苔藓(oral lichen planus,OLP)的发生、发展及癌变过程.本文将从NCAM1的结构、功能及调节,NCAM1在OLP中的异常表达,NCAM1在OLP发生、发展中的可能作用及NCAM1与OLP癌变等方面作一综述.
目的 分析口腔鳞状细胞癌(OSCC)与慢性牙周炎(CP)患者血清、唾液中白介素1β(IL-1β)表达水平,探讨IL-1β与两者间的相关关系.方法 收集16例健康志愿者(H组)、18例口腔鳞状细胞癌患者(O组)、20例慢性牙周炎患者(CP组)、26例口腔鳞状细胞癌伴牙周炎患者(O+CP组)空腹血清及唾液样本,采用酶联免疫实验法(ELISA)检测样本中IL-1β的浓度,并记录四组牙周临床指标.结果 唾液中IL-1β浓度O+CP组显著高于O组、CP组、H组,且O+CP组与另外三组间差异显著(P<0.05);血清中IL-1β表达水平在O+CP组、O组、CP组和H组依次降低.结论 血清及唾液中IL-1β水平可能在口腔鳞状细胞癌及牙周炎发展过程中起重要作用.
目的:评估基质金属蛋白酶-13(MMP-13)在慢性牙周炎患者经过牙周基础治疗前后的非刺激性全唾液、龈沟液及血清中的表达水平.方法:纳入20名慢性牙周炎患者和20名牙周健康者,采用ELISA分别检测慢性牙周炎患者治疗前、后及健康人非刺激性全唾液、龈沟液及血清中MMP-13的水平,检测患者牙周临床指标BI、PD和CAL,并进行组间比较.结果:患者除非刺激性唾液外,患者治疗后的龈沟液和血清中MMP-13水平显著降低(P<0.05),并且MMP-13水平与健康对照组无显著差异(P>0.05).患者治疗后龈沟液中BI和PD也明显降低,并与MMP-13水平相关(P<0.05).结论:龈沟液中MMP-13的表达水平可能成为慢性牙周炎的检测指标之一.
目的 研究发现CD147可能参与调节牙周疾病的进展过程,因而评估牙周炎患者经牙周基础治疗前后和健康者的非刺激性全唾液、龈沟液及血清中CD147水平,探讨其与牙周炎的相关性以及作为牙周炎诊断及预后标志物的可能性.方法 酶联免疫吸附试验(ELISA)检测20例牙周炎患者治疗前、后及20名健康人非刺激性全唾液、龈沟液及血清中CD147的水平,并记录牙周袋探诊深度(PD)、附着丧失(AL)和出血指数(BI).结果 经牙周基础治疗6周后,牙周炎患者的临床指标除AL外BI及PD均低于治疗前(P<0.05);治疗后非刺激性全唾液、龈沟液及血清中CD147水平明显降低,与治疗前相比差异具有统计学意义(P<0.05);除血清外,牙周炎患者治疗前、后的非刺激性全唾液和龈沟液中CD147水平仍高于健康对照组,其差异有统计学意义(P<0.05).结论 牙周炎患者、健康者的非刺激性全唾液及龈沟液中均有CD147表达,并随牙周炎症减轻CD147表达降低.
Family with sequence similarity 20-member C (FAM20C), a recently characterized Golgi kinase, performs numerous biological functions by phosphorylating more than 100 secreted proteins. However, the role of FAM20C in the salivary glands remains undefined. The present study demonstrated that FAM20C is mainly located in the cytoplasm of duct epithelial cells in the salivary glands. Fam20c(f/f); Mmtv-Cre mice were created in which Fam20c was inactivated in the salivary gland cells and observed that the number of ducts and the ductal cross-sectional area increased significantly, while the number of acinar cells was reduced. The granular convoluted tubules (GCTs) exhibited an accumulation of aberrant secretory granules, along with a reduced expression and altered distribution patterns of beta nerve growth factor, alpha-amylase and bone morphogenetic protein (BMP) 4. This abnormality suggested that the GCT cells were immature and exhibited defects in developmental and secretory functions. In accordance with the morphological alterations and the reduced number of acinar cells, FAM20C deficiency in the salivary glands significantly decreased the salivary flow rate. The Na+, Cl- and K+ concentrations in the saliva were all significantly increased due to dysfunction of the ducts. Furthermore, Fam20c deficiency significantly increased BMP2 and BMP7 expression, decreased BMP4 expression, and attenuated the canonical and noncanonical BMP signaling pathways in the salivary glands. Collectively, the results of the present study demonstrate that FAM20C is a key regulator of acinar and duct structure and duct maturation and provide a novel avenue for investigating novel therapeutic targets for oral diseases including xerostomia.

目的 研究基质金属蛋白酶-3(matrix metalloproteinase-3,MMP-3)在舍格伦综合征患者唇腺和唾液中的表达水平,以及明确其表达水平与病理分级的关系.方法 采集35例舍格伦综合征患者(实验组)和20例黏液腺囊肿患者(对照组)的唇腺和唾液,应用酶联免疫吸附试验法(ELISA)检测唾液中MMP-3的表达水平,应用免疫组化方法观察唇腺中MMP-3的表达,使用图像分析软件(IPP 6.0)定量分析两组唇腺中MMP-3的表达强度,并将舍格伦综合征患者按照其病理分级分为低的1、2级和高的3、4级两组.结果 MMP-3在实验组唾液和唇腺中的表达高于对照组,差异有统计学意义(P<0.05);MMP-3在病理分级高的患者唾液和唇腺中表达水平高于病理分级低的患者,差异有统计学意义(P<0.05).结论 舍格伦综合征患者唇腺中的MMP-3可能参与了腺泡以及腺体基质的破坏,导致疾病的发生;唾液以及唇腺中MMP-3的表达水平可能成为舍格伦综合征的一项检测指标,并为临床诊治提供参考依据.
目的:比较口腔扁平苔藓(OLP)(网纹型和糜烂型)和口腔鳞状细胞癌(OSCC)患者血清和唾液中的sCD44表达水平.方法:收集23例健康对照者、46例OLP患者(23例网纹型和23例糜烂型)及23例OSCC患者的血清和唾液进行研究.采用酶联免疫吸附试验(ELISA)法检测血清和唾液中的sCD44,使用MATLAB软件进行统计分析.结果:健康对照组、OLP组(网纹型和糜烂型)和OSCC组的血清中sCD44的表达水平比较差异无统计学意义(P>0.05).唾液中sCD44表达水平由低到高依次是:健康对照组,网纹型OLP组,糜烂型OLP组,OSCC组,且其差异具有统计学意义(P<0.05).结论:唾液中sCD44的表达水平,OSCC患者高于糜烂型OLP;糜烂型OLP高于网纹型OLP,其均高于健康人表达水平.血清中sCD44的表达水平,OLP和OSCC患者与健康人无差异.唾液中sCD44有望在口腔黏膜癌前病变的相关诊断中具有一定价值.
Aspirin has positive effects on bone marrow mesenchymal stem cells (BMSCs) osteogenic differentiation. However, researchers did not give much thought to its effect on BMSCs adipogenic differentiation. Here, we analyzed the effect of aspirin on the BMSCs adipogenic differentiation. To detect whether the effect of aspirin on the adipogenic differentiation of BMSCs is associated with the disturbed epigenetic modification, the expression of histone deacetylases (HDACs), activity of HDACs and HAT, global histone H3 acetylation and H3k9 acetylation alterations were investigated. Moreover, to further explore and understand the binding mode between aspirin and HDACs, an attempt was made to identify the interaction between aspirin and the HDACs with the aid of in silico docking study. The results showed that aspirin could induce inhibition of BMSCs adipogenesis. The level of HDAC activity, global histone H3 acetylation, and H3k9 acetylation were all down regulated during adipogenic differentiation, and aspirin can reverse these decreases. Furthermore, the HDAC isoforms have different expression patterns in those progresses. The expression of HDAC9 was increased in a does-dependent manner when aspirin was introduced during BMSCs adipogenic differentiation. Docking study showed that high affinity of HDAC9 to aspirin was existed, suggesting that HDAC9 may has an important role in the process of aspirin-induced suppression of adipogenesis. Further studies are needed to define the intricate mechanisms of the HDAC isoforms, and all of these enable us to understand aspirin and its efficacy of inhibition of adipogenic differentiation and pave the way to aspirin clinical using for the tissue regenerating.
Objective:To research the expression of matrix metalloprotase 9 (MMP-9) in the saliva and the relationship with histopathologic grading of patients with Sjogren Syndrome.Methods:The expressions of MMP-9 in the saliva of 27 Sjogren Syndrome patients (experimental group) and 29 underlip mucocele patients (control group) were detected with ELISA and the results of Sjogren Syndrome patients were pathological graded with low level (1 and 2) and high level (3 and 4).Results:In contrast to control group,the MMP-9 levels in Sjogren Syndrome patients were significantly increased (P<0.05),and the level of MMP-9 in the high pathologic grade was higher than that of low grade (P<0.05).Conclusion:The level of MMP-9 in the saliva may be a test indicator for the Sjogren Syndrome,which might provide reference for clinical diagnosis and treatment.
Hypoxia regulates a number of cell biological processes, including cell survival, development and differentiation. Deferoxamine (DFO), an oral chelator for blood transfusion patients, has been demonstrated to induce hypoxia and is frequently used as a hypoxia‑mimicking agent. The purpose of the present study was to investigate the influence of DFO on the proliferation, migration and osteogenic differentiation of human periodontal ligament cells (hPDLCs). The effects of DFO on hPDLC viability and migration were measured using an MTT and wound healing assay. To characterize the hypoxia microenvironment, the expression of hypoxia‑inducible factor‑1α (HIF‑1α) in hPDLCs treated with DFO was quantified using the reverse transcription‑quantitative polymerase chain reaction (RT‑qPCR). Subsequently, the osteogenic differentiation potential of DFO was determined by RT‑qPCR of the mRNA of osteogenic markers (runt‑related transcription factor 2 [Runx‑2], osteopontin [OPN] and collagen type I [Col‑1]). The alkaline phosphatase activity and mineral deposition were analyzed using alizarin red S staining. The MTT and wound healing assays demonstrated that low‑concentrations of DFO had little impact on hPDLC viability and migration 48 h into the treatment. DFO upregulated the expression of hPDLC genes specific for osteogenic differentiation: HIF‑1α, Runx‑2, OPN and Col‑1. Furthermore, formation of mineralized nodules was enhanced by DFO. The present study suggests that DFO provided favorable culture conditions to promote the osteogenic differentiation and mineralization of hPDLCs. The mechanism underlying these alterations remains to be elucidated.
GGsTOP is a novel and selective inhibitor of gamma-glutamyl transferase (GGT), a cell-surface enzyme that has a key role in glutathione homeostasis and the maintenance of cellular reactive oxygen species (ROS). ROS are essential for wound healing. However, little is known about the molecular mechanisms underlying the inhibition of GGT by GGsTOP in human periodontal ligament cells (hPLCs). The present study assessed GGT expression in mouse periodontal ligament tissues, GGT activity in hPLCs, and the potential physiological effect of GGsTOP on hPLC migration. Immunohistochemical analysis confirmed that GGT was widely expressed in mouse periodontal ligament tissue. Treatment with GGsTOP was associated with greater proliferation and migration of hPLCs, and higher levels of cellular ROS compared with untreated hPLCs. However, the increase in intracellular ROS was attenuated in hPLCs co-cultured with the anti-oxidant N-acetylcysteine (NAC), a precursor of glutathione. The higher ROS levels associated with GGsTOP treatment were in parallel with increases in the levels of type I collagen and alpha smooth muscle actin, which was inhibited in hPLCs co-cultured with NAC. Thus, GGsTOP may promote hPLC migration and participate in the maintenance of the periodontal ligament apparatus via the ROS pathway.
Objective To explore the relationship between rheumatoid arthritis ( RA) and chronic periodontitis ( CP) by observing the role of IL-18 in serum and gingival crevicular fluid ( GCF) of patients with CP and RA. Methods 83 subjects were selected and divided into four groups:16 healthy participants as the control group, 20 patients with CP, 21 patients with RA, and 26 patients with CP and RA. The serum and GCF levels of IL-18 were measured by enzyme-linked immunosorbent assay ( ELISA) , and clinical perio-dontal parameters including bleeding index ( BI) , probing depth( PD) , and clinical attachment loss( CAL) were examined. Results The IL-18 levels in GCF of patients with CP and RA [(221. 83±43. 38) pg/mL] were significantly higher than those in the CP group [(198. 00±37. 72) pg/mL], RA group [(138. 10±23. 16) pg/mL] and the control group [(135. 12±27. 11) pg/mL] (P<0. 05). Serum levels of IL-18 in the CP and RA group [(321. 02±260. 57) pg/mL], CP group [(220. 60±228. 37) pg/mL], RA group [(186. 29±253. 03) pg/mL] and HP group [(169. 56±143. 82) pg/mL] showed a decreasing trend. Only in CP and RA group, the serum levels of IL-18 were significantly higher than those in the control group (P<0. 05). Conclusion IL-18 can be one of the bio-logical foundations that correlates chronic periodontitis and rheumatoid arthritis.
Exacerbations of oral lichen planus (OLP) have been linked to the periods of psychological stress, anxiety and depression. The specific mechanism of the interaction is unclear. The aim of this study was to explore the candidate genes or molecules that play important roles in the interaction between OLP and depression. The BITOLA system was used to search all intermediate concepts relevant to the "Gene or Gene Product" for OLP and depression, and the gene expression data and tissue-specific gene data along with manual checking were then employed to filter the intermediate concepts. Finally, two genes (NCAM1, neural cell adhesion molecule 1; CD4, CD4 molecule) passed the follow-up inspection. By using the text mining can formulate a new hypothesis: NCAM1 and CD4 were identified as involved or potentially involved in the interaction between OLP and depression. These results offer a new clue for the experimenters and hold promise for developing innovative therapeutic strategies for these two diseases.
Objective This present study was to detect the role of IL-17A on the correlation between chronic periodontitis(CP) and rheumatoid arthritis(RA).Methods According to the inclusion criteria selected 85 subjects were detected into four groups:①20 patients with chronic periodontitis;②23 patients with the RA;②26 patients with CP and RA;④16 healthy numbers as the control group.The clinical periodontal parameters including probing depth (PD),clinical attachment loss(CA L),and bleeding index(BI) were examined,and the serum and GCF levels of IL-17A were measured by enzyme-linked immunosorbent assay(ELISA).Results The IL-17A levels in serum of patients with CP and RA[(2216.0 ± 1520.87)pg/mL]were significantly higher than the RA group [(851.59 ± 975.81) pg/mL],CP group [(1039.7 ± 1004.83) pg/mL] and the control group[(209.48 ± 50.02)pg/mL].Serum levels of IL-17A in CP group and RA group were significantly higher than the healthy control group(P <0.05).The IL-17A levels in GCF of in the HP group[(356.75 ±69.07)pg/mL],RA group[(381.08 ± 79.48) pg/mL]/CP group [(397.82 ± 108.53) pg/mL] and patients with CP and RA[(414.71 ± 103.09)pg/mL] showed a trend of increase,but no statistical difference (P >0.05).Conclusions IL-17A may have some significance in the study of the correlation between chronic periodontitis and rheumatoid arthritis.
Objective To evaluate the MMP-9 levels in the unstimulated whole saliva (UWS),gingival crevicular fluid (GCF) and serum of patients with chronic periodontitis before and after the initial phase of periodontal treatment and to determine whether they are possible diagnostic and prognostic biomarkers for periodontitis.Methods Twenty patients with periodontitis and 20 healthy controls were included,and periodontal conditions (BI,PD,CAL) were assessed.The levels of MMP-9 in UWS,GCF and serum were measured by enzyme-linked immunosorbent assay before and after the treatment (BT and AT).Results ① In addition to CAL,the conventional periodontal treatment resulted in significant improvement in clinical parameters compared with before treatment.[CAL BT(5.50 ± 1.97) mm,AT (5.50 ± 1.97) mm,P =1.000 ; PD BT (7.05 ± 1.81) mm,AT(4.23 ± 1.06)mm,P =0.000;BI BT 3.75 ±0.44,AT 0.20 ±0.41,P =0.000]②MMP-9 levels in UWS,GCF and serum were significantly decreased six weeks after treatment[UWS BT(22.89 ± 5.28)ng/mL,AT(18.11 ±7.19) ng/mL,P =0.000;GCF BT(41.80 ±2.90) ng/mL,AT(35.71 ±4.49) ng/mL,P =0.000;serum BT(6.67 ± 5.318) ng/mL,AT (2.47 ± 2.713) ng/mL,P =0.004] ③In addition to serum,MMP-9 levels in UWS and GCF before and after the initial phase of periodontal treatment revealed significant increases compared with the control group[UWS of control group (6.78 ± 3.02) ng/mL,GCF (30.20 ± 3.64) ng/mL,P =0.000 ; serum of control group (1.18 ± 0.88) ng/mL,compared with BT and AT P =0.004,P =0.055].Conclusions The high expression of MMP-9 in unstimulated whole saliva is likely to become indicators in chronic periodontitis,and may provide a reference for clinical diagnosis and treatment.
目的:通过调查该院牙周病患者的疾病构成及其变化趋势,从而满足牙周病患者需求,并为口腔健康和牙周病防治提供临床依据.方法:收集2009~2013年哈尔滨医科大学附属第二临床医学院牙周黏膜科的牙周病患者资料,并按1999年国际牙周病分类诊断,采用Excel、SPSS软件进行数据整理分析.结果:主要诊断为牙周病的患者共8058人次,按1999年国际牙周病分类诊断共7874人次,占97.72%.共24种疾病,其中所占比例较高者(构成比≥3.00%)依次为:慢性牙周炎(35.62%)、慢性龈炎(14.94%)、反应全身疾病的牙周炎(6.73%)、扁平苔藓(累及牙龈)(5.47%)、伴牙髓病变的牙周炎(4.71%)、牙周脓肿(4.43%)、侵袭性牙周炎(3.67%)、牙龈瘤(3.56%)、青春期龈炎(3.00%).复诊1557人次,不同就诊原因致使其接受治疗和复诊情况具有显著差异.结论:本次调查在一定程度上反应出哈尔滨市综合医院牙周病患者的疾病构成及患者对该疾病的认知程度,且患者就诊原因与其治疗和复诊情况具有相关性.
