Objective To evaluate the clinical significance of autoimmune antibodies in the diagnosis and therapy of hematologic malignancies. Methods During the period of 2011 to 2016,453 patients with hematologic malignancies in Zhejiang provincial TCM hospital were enrolled in this retrospective analysis. The antibodies profiles were tested for each patients by using immunoblotting and indirect immunofluorescence. Results The positive rates of MDS and CLL were higher than those of other malignant hematologic diseases,accounting for 29.7%(27/91)and 27.5%(19/69) respectively. Among the detected autoimmune antibodies,ANA positive rate was the highest,accounting for 11.7%(53/453),while positive rate of Anti-PR3 was lowest,accounting for 2.6%(12/453). RBC and PLT counts in CLL patients with positive antibodies were lower than in those with negative ones,and WBC count in MDS patients with positive antibodies was lower than in those with negative ones. Conclusions Autoimmunity is more frequently observed in CLL and MDS than other malignant hematologic diseases,associated with hemocytopenia. It's helpful for clinical doctors to deepen the understanding of the pathogenesis of MDS and CLL,and to help clinical treatment according to different patients' immune status to detect the autoimmune antibodies.
目的 研究ELTD1基因在人结直肠正常上皮细胞和肿瘤细胞之间的表达差异,明确其表达蛋白的细胞定位.方法 分别抽提对数生长期的人结直肠癌LoVo细胞株和正常上皮细胞NCM460细胞株的总RNA,RT-PCR半定量技术扩增靶基因ELTD1和内参基因B2M(β2微球蛋白)的mRNA,利用ELTD1/B2M光密度比值来表示细胞ELTD1 mRNA的相对表达量;Western blot法检测2种细胞株靶蛋白ELTD1和内参蛋白GAPDH,利用ELTD1/GAPDH比值来表示ELTD1蛋白的相对表达量;间接免疫荧光技术检测LoVo细胞ELTD1蛋白的细胞定位.结果 LoVo细胞和NCM460细胞的ELTD1/B2M灰度比值分别为0.83±0.07和0.11±0.04,ELTD1/GAPDH灰度比值分别是0.74±0.1和0.12±0.04,差异有统计学意义(P<0.05),荧光显微镜下,可见黄绿色荧光信号定位于细胞膜.结论 ELTD1基因在正常上皮NCM460细胞极低表达,而在人结直肠癌LoVo细胞株高表达,ELTD1蛋白定位于细胞膜,ELTD1是一种有助于结直肠癌的早期诊断、靶向治疗、疗效判断及预后的潜在生物标志分子.
This study was aimed to explore the role of full-automatic blood analyzer Sysmex XE-2100 in early screening and diagnosing the hematologic malignancies. A total of 288 samples of the patients with hematologic malignancies was examined. Then, the scatter plots, alarm information and blood smears were analyzed. The results indicated that 76% of these samples showed abnormal scatter plots. CMML and AML-M3 patients had their own characteristic scatter plots, while others hadnt's. The coincident rate of CMML and AML-M3 determined by scatter plots with practical diseases was 100%; the coincident rate of ALL determined by scatter plots with practical disease was 67%. The coincident rate of alarm information of blast cells was 92.5%, the coincident rate of immature granulocytes was 77.1%, the coincident rate of nucleated red blood cells was 33.3%, the coincident rate of atypical lymphocytes was 31.3%. It is concluded that the abnormal scatter plots and alarm information are very important for finding the patients with hematologic malignancies and determining the disease type. The alarm information has high reliability for blast cells and immature granulocytes, but has only mirror value for nucleated red blood cells and atypical lymphocytes.
Objective: To observe the effects of hovenia dulcis thunb on histological changes of liver and serum TNF - α,IL - 6,and IL - 8 levels in rats with non - alcoholic fatty liver,discuss the mechanism. Methods: SD rats were feeding high fat diet for 90 days to build non - alcoholic fatty liver models,different doses hovenia dulcis thumb liquid were given to intervene the formation of non - alcoholic fatty liver for 90 days at the same time. The histological changes of rats’liver were observed by light microscope,serum TNF - α,IL - 6,and IL - 8 levels wre determined by radioimmunoassay. Results: The degree of fatty degeneration and inflammation activity of hepatic tissue in model group was obvious,serum TNF - α,IL - 6,and IL - 8 levels of model rats were significantly higher than those of normal rats; the degree of inflammation activity of hepatic tissue in high and low dose hovenia dulcis thunb groups was obviously improved,serum TNF - α,IL - 6,and IL - 8 levels in the two dose hovenia dulcis thumb groups were significantly reduced. Conclusion: TNF - α,IL - 6,and IL - 8 were related to the occurrence of non - alcoholic fatty liver induced by high fat diet,decrea- sing serum TNF - α,IL - 6,and IL - 8 contents maybe one of the mechanism that hovenia dulcis thumb can effectively prevent non - alcoholic fatty liver.
AIM: To analyze the feasibility of PCR-high-resolution melting curve analysis(HRM) for detecting the site mutations of C2549 and G2548 in leptin gene promoter from the patients with liver cirrhosis,and to explore the relevance between mutant genotypes and physiological and biochemical indexes in liver cirrhosis patients.METHODS: Compared with the method of PCR-restriction fragment length polymorphism(RFLP),the present research used the method of PCR-HRM to analyze the site mutations of C2549 and G2548 in leptin gene promoter in control group(n=100) and liver cirrhosis group(n=100).The physiological and biochemical indexes of the patients were also detected and compared.RESULTS: Leptin gene promoter polymorphism was detected using PCR-HRM with effectiveness,high flux and accuracy.Preliminary results showed that the main mutation of the patients with liver cirrhosis was in C2549 site,but not found in G2548 site.Leptin,free leptin index(FLI),fasting insulin(FINS) and insulin resistance index estimated by homeostatic model assessment(HOMA-IR) in liver cirrhosis group were higher than those in control group.Insulin sensitivity index(ISI) and soluble leptin receptor(sOB-R) in liver cirrhosis group were lower than those in control group with significant difference except leptin level.Meanwhile,FLI showed positively correlated with FINS and HOMA-IR(r=0.45,r=0.53,P<0.05),and negatively with ISI(r=-0.34,P<0.05).In the patients with liver cirrhosis,C2549A heterozygous mutation was predominant.The indexes of HOMI-IR,leptin,sOB-R and FLI of C2549A homozygotes and heterozygotes were higher than those of the wildtypes,which showed significant difference except leptin and sOB-R levels(P<0.05).CONCLUSION: PCR-HRM can be more accurate for identifying leptin promoter polymorphism.The increase in the frequency of C2549A mutation may be closely related with liver cirrhosis.Existence of hyperinsulinemia and insulin resistance may be correlated with leptin level in the patients with liver cirrhosis.
[Objective] To observe the intrinsic relation between hyperbaric oxygen and different types of cerebral apoplexy, sponsor to use limited hyperbaric oxygen resource by differentiation, greatly develop the essence of treatment based on differentiation in TCM. [Method] Divide 54 cases of cerebral apoplexy into 3 types for testing blood rheology before and after hyperbaric oxygen therapy.[Result] The high shear rate, middle shear rate and low shear rate, red cell gathering index and rigidity index in deficient Yin and active wind type were markedly higher than wind sputum uprising type and deficient Qi and blood stasis type(P<0.01). [Conclusion] From blood rheology, it points out that the fittest sign for hyperbaric oxygen to treat cerebral apoplexy is “deficient Yin and active wind type”, which is the pathological basis in changes of blood rheology of cerebral apoplexy.
