Objective To compare the depiction of brain lesions by T2-weighted image, fluid-attenuated inversion-recovery sequency and three-dimensional double inversion recovery sequence in patients with multiple sclerosis(MS).Method 23 patients with MS underwent magnetic resonance imaging(MRI)examination at 3.0T,including T2WI,Flair and 3D-DIR sequences,to compare the relevance ratio of lesions in different areas of the brain at these three sequences.Results In total lesions ,there were statistical differences among T2WI,Flair and 3D-DIR sequences in detecting intracortical lesions and proximai cortical lesions, Compared with T2WI and Flair, 3D-DIR was more sensitive to detect intracortical lesions and proximai cortical lesions.Conclusion MRI 3D-DIR can improve the detection of intracortical and proximai cortical lesions in patients with MS.
micro RNA(miRNA)是一类长度约为19~25个核苷酸的非编码单链RNA分子,通常在转录后水平调控靶基因的降解或抑制翻译。越来越多的证据表明,miRNA可通过靶向免疫系统中关键信号转导分子的表达,从而在多个环节上参与调控机体的固有免疫反应和适应性免疫反应。多发性硬化(multiple sclerosis ,MS)是一种累及中枢神经系统的自身免疫性疾病,最近研究发现,多种miRNA在介导MS致病过程中起关键作用。本文就miRNA 在T细胞分化和发育以及在MS中的作用进行综述。
目的:探讨阿托伐他汀对老年自身免疫性脑脊髓膜炎(EAE)大鼠脑白质髓鞘碱性蛋白(MBP)和少突胶质细胞转录因子(OLIG)1蛋白表达的影响。方法取老年 Wistar 雄性大鼠80只,随机分为对照组、模型组、阿托伐他汀大剂量组(8 mg/kg 组)和阿托伐他汀小剂量组(2 mg/kg组),每组20只。采用新鲜豚鼠50%全脊髓匀浆液和完全弗氏佐剂(CFA)免疫制备EAE模型,免疫后第2天开始,模型组给予生理盐水灌胃,阿托伐他汀组给予阿托伐他汀不同剂量灌胃,持续灌胃21 d,于第22天处死,取脑干和脊髓待测。采用实时荧光定量和免疫组织化学染色检测各组脑干、脊髓中MBP和OLIG1的mRNA水平和蛋白阳性表达强度。结果与对照组比较,模型组 MBP mRNA水平和蛋白阳性表达强度明显升高、OLIG1明显降低(P<0.01);阿托伐他汀小剂量组MBP mRNA水平和蛋白阳性表达强度升高、OLIG1降低(P<0.01);阿托伐他汀大剂量组 MBP mRNA水平和蛋白阳性表达强度略升高、OLIG1略降低,无显著差异(P>0.05),模型组、阿托伐他汀大、小剂量组差异显著(P<0.01)。结论阿托伐他汀能够通过下调MBP、上调OLIG1 mRNA水平和蛋白阳性表达强度,对EAE大鼠的脑白质髓鞘损伤具有保护作用,且大剂量作用优于小剂量。
