Objective To study the effects of zinc deficiency and supplements on serum and tissue zinc contents,zinc-dependent enzyme activity,hormone level,and protein synthesis in scalded rats. Methods Among 112 Sprague-Dawley rats,56 rats were fed with 40 μg/g zinc diet and other 56 rats with 4 μg/g zinc diet for one week. Eight rats were respectively selected from each group; those from the group fed with 40 μg/g zinc were regarded as normal control group (NC group), and those from the group fed with 4 μg/g zinc were as deficiency control group (DC group). These 16 rats were sacrificed, and their vein blood and tissue specimen were harvested. The remaining 96 rats were subjected to 15% TBSA. Among them,48 rats from the group fed with 40 μg/g zinc were randomly divided into normal burn group (NB group,fed with 40 μg/g zinc continuously) and NH group (fed with 80 μg/g zinc diet). Other 48 rats of 4 μg/g zinc diet were randomly divided into DB group (fed with 10 μg/g zinc diet) and DH group (80 μg/g zinc diet),with 24 rats in each group. One,3 and 7 days after having been scalded,8 rats from each group were sacrificed. The samples of blood,liver tissues,thigh bone,and scalded skin were harvested,the zinc levels in the serum and tissues were determined,and the content of serum alkaline phosphatase (ALP) activity,growth hormone (GH) level,and liver metallothioneine (MT) content were also assayed. Results The rats were fed with zinc deficient diet for a week,and then zinc deficiency condition was successfully produced. In DC group,serum zinc, bone zinc, and wound tissue zinc levels, and ALP activity declined obviously compared with NC group. After burn the zinc levels in liver and wound tissue,and GH level and liver MT content showed increasing tendencies in NB group and DC group.However, the zinc levels in serum and bone, and ALP activity decreased. After zinc supplementation,the zinc levels in serum,liver,and wound elevated in NH and DH group. Among them,the increase extent of the zinc levels in serum and wound were prominent; ALP activity, GH level in serum, and liver MT also showed increasing tendencies,while most obvious increase occurred in BH group. Conclusion With zinc deficiency condition,the contents of zinc in serum and tissue decrease; meanwhile,the functions and activities of zinc-dependent enzymes,hormones,and proteins are also affected. These problems can be resolved by zinc supplementation.
Objective: To study the effects of zinc supplementation by different ways on zinc level in serum and tissues in scalded rats.Methods:80 SD rats were divided randomly into C(control group,no scald),N(scald control group),W(zinc supplemented by wound) and H group(zinc supplemented by diet).All rats had been fed with diet containing zinc 40μg/g for a week and then the rats of C group were skilled.The other rats were scalded on their dorsal skins covering 15% of the total body surface area by boiling water for 10 seconds and fed continuously with diet containing zinc 40μg/g in N and W groups,80μg/g in H groups.A cream containing zinc 761.1μg/g was applied on the scalded skin in W group daily.The level of zinc in serum,liver,bone and scalded skin of N,W and H groups were measured on day 1,3 and 7 after they were scalded,respectively.Results: The level of zinc in serum,bone and scalded skin decreased in the first day after scalded in N and W groups,but it increased in H group.The zinc level in liver increased in all groups.The zinc level in the scalded skin of W group was higher in the seventh day than that at other days.Conclusion: After scalded,the zinc levels in serum and tissues except liver decreased and there was re-distribution of zinc in the body.Zinc supplementation by diet and wound is necessary and can elevate quickly zinc level in serum and wound tissue,which can promote wound healing
OBJECTIVE:To explore the effects of zinc supplementation on zinc and calcium levels in serum and tissue in burned rats.METHODS:Eighty SD rats were randomly divided into C group (control group without scald, n = 8), and N, W, H groups (each consisting of 24 rats), in which the rats were exposed to scalding resulting in partial thickness burns covering 15% of the total body surface area on the back, and then they were fed with diets containing zinc 40 microg/g in N and W groups, and 80 microg/g in H group. A cream containing zinc 761.1 microg/g was applied on the wound in W group at the same time. Eight rats of each group were sacrificed on day 1, 3 and 7 after scald respectively. Venous blood and samples of liver, femur and scald skin were harvested. Zinc and calcium contents in serum and tissues were determined with atomic absorption spectrophotometer.RESULTS:The serum Zn(2+) levels in N, W groups were lower than C group, however, it was obviously higher in H group (up to 16.2 micromol/L) on day 1 after scald. The liver Zn(2+) showed an increasing tendency in all groups, while Ca(2+) level declined in H group, but increased in N, W group. The bone Zn(2+) and Ca(2+) levels showed a progressive declination in all groups from day 1 to 7 after scald. The changes were more obviously in N group than H group (P < 0.05). The Zn(2+) content of the scalded skin increased obviously in H group on first day after scald and in W group on 7th day after scald. The Ca(2+) contents of scalded skin showed marked increases in all groups, especially in N group, but least in W group.CONCLUSION:There are obvious changes in Zn(2+) and Ca(2+) contents of serum and tissues after scald injury and zinc supplementation. The effects of zinc supplementation on calcium level in the tissue need to be further studied.
严重烧伤患者常因锌的大量丢失而造成锌营养不良,而补锌却未成为临床治疗的一个环节,其主要原因在于担心补锌可能造成锌的蓄积毒性或干扰其它相关元素的代谢.为此,笔者通过给烫伤大鼠进行不同剂量补锌,观察其对各主要脏器的锌及部分二价元素代谢的影响.
Objective To study the effects of supplement zinc on serum and tissue zinc levels, serum ALP activity and serum growth hormone levels in scalded rats with deficiency zinc. Methods Scalded rats were randomly divided into three groups: group L (low dose zinc, diet containing zinc 4 μg/g), group M (margin dose zinc, 10 μg/g), and group H (high dose zinc, 80 μg/g). Rats of each group were killed respectively on 1,3,7 day postburn. Samples were obtained and examined. Results The state of zinc deficiency appeared in rats before being scalded by intake of zine deficiency diets. The serum zinc, bone zinc and activity of ALP continuously decreased in the group L, but zinc levels in serum, liver, bone, skin increased. The activity of ALP and GH level also increased in the M and H group, most prominent in the group H. Conclusions Deficiency zinc in rats appears with intake of low zinc diets and becomes more serious after scalding than before. Supplementary zinc can increase the zinc levels in body, especially in skin, and promote the activity of ALP,and in crease GH level.
目的:严重烧伤后往往存在锌营养不良状态,本实验考察不同剂量补锌是否会造成锌的蓄积或影响铜代谢.方法:利用人工半合成饲料技术制成不同含锌饲料;大鼠进食低锌饲料成为缺锌模型,造成 20% TBSA深Ⅱ度烫伤后饲以不同含锌饲料(含锌量分别为: 1.6 μg/g; 24.7 μg/g; 286.9 μg/g),原子吸收法分析血清、肝、肾、脾和睾丸的锌、铜含量.同时设一组大鼠烫伤前后均进食正常含锌饲料(含锌量: 24.7 μg/g) 做对照组.结果:补锌可显著提高血清和各器官的含锌量,补高锌组未发现器官存在锌的过量蓄积;大剂量补锌使血清、肾脏铜含量也明显提高.结论:在严重烧伤后的锌缺乏状态下,短时期内适当加大补锌量不会导致锌的过量蓄积,对铜代谢无不良影响.
钙营养状况调查显示,我国属于低钙摄入国家[1,2].建国以来4次大规模膳食调查结果一致表明,我国居民每天钙平均摄入量不足500mg,不少省份低于400mg,此值不足营养学会1988年制定的有关营养素膳食推荐量标准(RDA值中规定成人每日800mg)的50%[3].有关资料显示,各国都根据正常人的营养需求制定有每日钙的推荐摄入量标准:美国生长期少年儿童为1500mg/日,成人为800mg/日;日本钙推荐摄入量为成年人600mg/日,世界卫生组织推荐绝经后妇女钙的摄入量为1500mg/日.钙摄入不足,会影响生长发育并可致多种疾病,最普遍发生的则是骨质疏松症.钙营养不良不仅是儿童,也已成为老年群体的重要营养问题.为对国民补钙提供可靠的参考,我们对收集到的临床用钙剂及原料成分进行了测定.
锌的生物学作用已有许多报道,但锌对其他元素的影响研究较少,本实验观察了不同途径补锌对大鼠烫伤皮肤锌(Zn)、铜(CU)、铁(Fe)、钙(Ca)、镁(吨)的影响。一、材料和方法1.动物分组:SD大鼠80只,雄性,体重(odt3)g,随机分成C组(伤前对照)8只和正常锌组、高锌组、创面锌组各四只,大鼠单独置于透明塑料笼内。饲料配制:按实验动物营养推荐量标准配制合h量州、mndg(w/W)饲料。先用含Zn40ndg饲料喂养1周,然后,乙醚麻醉C组,心外穿刺抽血,活杀大鼠留取皮肤。其他3组乙醚麻醉后,质量分数0.l的硫化钠背部脱毛,lboCC水,烫伤10秒,形成15%深11”烫伤。正常锌组继用含ZIW
目的:探讨烧伤后补锌对其他微量元素的影响.方法:SD大鼠随机分为烫伤前对照,L组(低锌组),M组(边缘缺锌组),H组(高锌组),均先用 4μg/g低锌饲料喂养 1 周.活杀对照组,其他 3 组致背部深二度烫伤 15%,补充不同剂量锌,观察血清锌、铁和红细胞脆性的变化.结果:L组血清锌下降,血清铁、红细胞脆性增加,H组血清锌明显增高,血清铁、红细胞脆性下降,M组的变化介于L、M组之间.结论:烫伤后缺锌使血清锌下降,红细胞脆性增加,血清铁上升,补锌降低红细胞脆性,血清铁下降,这与锌具有维持红细胞膜稳定性的作用有关.
