目的 通过观察脑缺血再灌注后脑内12/15-脂氧合酶代谢产物15-羟基二十碳四烯酸(HETE)含量和过氧化物酶体增殖物激活受体(PPAR)γ 表达的变化,探究12/15脂氧合酶与脑缺血再灌注损伤的相关性.方法 将SD大鼠90只随机分为15-HETE治疗组(干预组,30只)、假手术组(30只)和缺血再灌注模型组(模型组,30只).应用线栓法制备缺血再灌注损伤模型,干预组在手术前30 min脑内注射15μl 15-HETE,模型组脑内注射等量的磷酸盐缓冲液(PBS).应用Western印迹实验考察脑组织中12/15-脂氧合酶和PPARγ 在蛋白水平的变化.应用Real Time-PCR考察脑组织中12/15-脂氧合酶和PPARγ 在基因水平的变化.应用酶联免疫吸附试验(ELISA)试剂盒考察脑内15-HETE含量变化.结果 模型组12/15-脂氧合酶和PPARγ 在基因和蛋白水平表达均明显增加.与模型组相比,给予15-HETE治疗后PPARγ 基因和蛋白水平明显增加.结论 外源性给予15-HETE能够有效激活PPARγ,降低脑梗死面积,为脑缺血再灌注损伤治疗提供根据.
Objective To investigate the efficacy of salicylazosulfapyridine combined with herbal coloclysis in the treatment of ulcerative colitis and its effect on serum interleukin-2(IL-2) and C-reactive protein(CRP).Methods The clinical data of 118 patients with ulcerative colitis admitted to our hospital from January 2008 to December 2010 were retrospectively analyzed.They were randomly divided into two groups:control group(n=58),in which patients were given oral salicylazosulfapyridine treatment(6.0 g,tid;reduced to 4.5 g,tid,after 15 days);and treatment group(n=60),in which patients were given herbal coloclysis qd in addition to the oral salicylazosulfapyridine treatment.Treatment in each group lasted both 20 days.The changes of IL-2 and CRP levels were compared before and after treatment.Results The efficay was significantly superior in the treatment group than in the control group(u=-3.464,P<0.01).In both groups,the IL-2 levels were increased significantly after treatment,and the CRP levels were decreased significantly after treatment(P<0.05).The IL-2 and CRP levels of the treatment group were significantly better than those of the control group(P<0.05).Conclusion The salicylazosulfapyridine combined with herbal coloclysis shows good efficacy in treating ulcerative colitis,which may be achieved by influencing the change of serum IL-2 and CRP.
