目的:探讨miR-496是否通过靶向mTOR影响宫颈癌HeLa细胞的周期、增殖、迁移、侵袭以及裸鼠移植瘤的生长.方法:选取2020年12月至2021年12月期间于河北医科大学第四医院接受手术的74例宫颈癌患者的肿瘤标本和癌旁组织标本,qPCR、WB法和免疫荧光法检测宫颈癌组织中miR-496和mTOR在mRNA和蛋白水平的表达.利用TargetScan数据库预测miR-496的靶基因并用双荧光素酶报告基因实验进行验证.将HeLa细胞按转染物不同分为对照组、miR-496 mimic组和miR-496 mimic+pMIR-mTOR组,CCK-8法、流式细胞术和Transwell实验分别检测miR-496和mTOR对HeLa细胞增殖、周期、迁移和侵袭的影响.将对照组、miR-496 mimic组HeLa细胞接种至BALB/c裸鼠皮下构建宫颈癌裸鼠移植瘤模型,21d后处死小鼠,剥离小鼠肿瘤组织并称取瘤质量,免疫荧光法和WB法检测miR-496过表达对移植瘤组织中mTOR和Ki67表达的影响.结果:在宫颈癌组织中,miR-496呈低表达,而mTORmRNA和蛋白呈高表达.miR-496能够靶向结合mTORmRNA的3'-UTR.与对照组和miR-496 mimic+pMIR-mTOR组相比,miR-496 mimic组HeLa细胞的miR-496水平和S期细胞比例均显著升高,而增殖水平、迁移和侵袭细胞数均显著降低(均P<0.01).成功构建宫颈癌裸鼠移植瘤模型,接种21d后,miR-496 mimic组移植瘤质量、移植瘤组织中mTOR和Ki67的表达水平均显著低于对照组(均P<0.01).结论:miR-496在宫颈癌组织中呈低表达,miR-496过表达可通过靶向调控mTOR抑制HeLa细胞的恶性生物学行为和裸鼠移植瘤的生长.
Background and purpose: MicroRNA (miRNA) has been proved to be related to the pathogenesis of cervical cancer. The study aimed to analyze the mutual relation between miR-496 and SFMBT1 and the mechanism of miR-496 in cervical cancer. Methods: Target gene of miR-496 was predicted and confirmed. The effect of miR-496 mimics on SFMBT1 expression was detected by real-time fluorescence quantitative polymerase chain reaction (RTFQ-PCR) and Western blot. Changes of SFMBT1 expression in cervical cancer tissues were analyzed using immunofluorescence and Western blot. Cell counting kit-8 (CCK-8) and transwell assays were used to detect the effects of both miR-496 and SFMBT1 on HeLa cell proliferation, migration and invasion, respectively. The impact of miR-496 on tumor growth was also explored in nude mice model. Results: Compared with para-cancerous tissues, mRNA (1.69±0.23 vs 1.00±0.12) and protein (1.73±0.28 vs 1.00±0.15) of SFMBT1 were highly expressed in tissues of cervical cancer; miR-496 could inhibit mRNA (0.81±0.07 vs 1.00±0.15) and protein (0.26±0.02 vs 1.00±0.14) expression of SFMBT1 via specifically binding to the 3’-UTR of SFMBT1. In cell experiments, HeLa cell proliferation (1.39±0.10 vs 2.01±0.09), migration (40.50±3.17± vs 28.42±1.21) and invasion (15.03±1.67 vs 25.71±2.56) abilities were suppressed in miR-496 mimics group compared with control group. Abilities of cell migration (33.21±2.66 vs 19.28±1.50) and invasion (30.11±2.73 vs 15.03±1.67) were enhanced in miR-496+SFMBT1 group compared with miR-496 mimics group. In nude mice xenograft model of cervical cancer, tumor volume and weight were obviously reduced in miR-496 group compared with control group. The degree of lesion and SFMBT1 expression and Ki-67 proliferative index were obviously decreased in miR-496 group (P<0.01). Conclusion: MiR-496 inhibits biological behaviors of cervical cancer cells and transplanted tumor growth of nude mice via targeted regulation of SFMBT1.
N2O是影响全球气候变化的主要温室气体之一,对臭氧层的消耗起着催化作用.农田土壤作为温室气体第二大排放源,如何降低其N2O的排放已成为不断探索的重大课题.生物炭是在缺氧条件下由含碳量丰富的生物质热解而成的一种高比表面积、多孔性材料,具有提高土壤肥力,促进作物生长等诸多优点,已成为研究热点.对农田N2O排放的影响以及作用机制已有较多研究,但尚未形成统一定论.综述了N2O排放机制,以及生物炭特性、生物炭与环境因子的相互作用对农田N2O排放的影响,并对未来生物炭对温室气体排放的影响研究提出方向性建议.
为探讨2种赋分制烤烟外观质量数字化评价方法与感官质量是否相关,选用3个产地中部正组相邻4个等级烟叶进行数字化打分及相关分析.结果表明,2种外观质量评价方法均能有效区分不同等级烤烟外观质量,但2种方法对部分外观质量指标敏感度差异明显.方法一(10分制)对外观质量的识别灵敏度较高且与感官质量呈极显著相关.方法二(100分制)在部分近相邻等级外观质量识别中易于出现较大误差,且未能反映出颜色显著差异.
As an alternative to fossil fuels, biomass burning represents a feasible way of greening the tobacco flue-curing-drying process, especially when it is self-produced. In this work, a biomass briquette fuel (BBF) burner used for flue-curing tobacco was designed and assembled using modern and mature electromechanical technologies, while also making use of its own structure to prevent high-temperature damage to the photoelectric elements during fuel combustion, and deploying an intelligent tobacco controller (ITC) to ensure orderly heating. A comparison between this new burner and a traditional coal-fired heating furnace revealed that when ITC was in operation, the BBF could meet the heating needs for tobacco curing, and the temperature difference between the target and the actual dry-bulb temperature (DBT) in the bulk curing barn was well controlled within a range of ±0.5oC. Additionally, the BBF burner fully used the biomass briquette and reduced the cost per kilogram of dry leaf curing. Furthermore, non-manual management heating modes could be realized during firing. Considering that the structure of the new device is similar to gas or oil burners, in terms of heating via connecting sockets, it can also potentially be used in small-scale boilers if the ITC programming chip is changed.
烟草长柄分泌型腺毛的分泌物对烟叶的品质有重要影响,为了明确长柄腺毛的遗传特性,以纯合长柄分泌型腺毛烟草材料T.I.1068为父本、非分泌型腺毛烟草材料T.I.1112为母本,构建6个世代的群体(P1、P2、F1、F2、BC1、BC2),利用主基因+多基因混合遗传模型分离分析方法,研究烟草长柄分泌型和非分泌型腺毛的遗传规律.结果表明,烟草长柄分泌型腺毛与非分泌型腺毛相对比例的遗传由2对加性-显性-上位性主基因+加性-显性上位性多基因模型控制.BC1、BC2和F2世代主基因的遗传率分别为80.05%、97.88%和96.50%,主基因遗传率很高,可以在早代进行腺毛种类的选择;BC1、BC2和F2世代多基因的遗传率分别为16.15%、1.55%和2.75%,BC1代应考虑多基因的影响,BC2和F2代多基因的影响较小.可见,长柄分泌型与非分泌型腺毛相对比例的遗传受2对主基因控制,以主基因效应为主,受基因间的互作的影响.
为探讨产地之间烤烟颜色色差及其对感官质量的影响,运用图像处理技术和感官评吸技术对铜仁、驻马店、吉安烟叶颜色正、反面色差进行了分析.结果表明,铜仁烟叶颜色正、反面RGB(红、绿、蓝三色)色值最大,吉安正面RGB色值最小,驻马店反面RGB色值最小,不同产地同等级烟叶颜色RGB色值叶片反面差异较小;不同产地烟叶颜色正、反面色差差异明显,其中吉安烟叶正、反面色差最大,极显著或显著大于铜仁和驻马店烟叶,铜仁和驻马店烟叶差异不明显.等级之间C2F与C3L之间色差有显著差异,而C2F、C3F、C4F等级差异不显著;铜仁和驻马店烟叶正、反面色差对感官评吸质量影响未达到显著水平,吉安烟叶正、反面色差对感官评吸质量影响较大,且随着色差的降低感官评吸质量也随之下降.
In order to improve the quality and industrial usability of upper leaves and to study the impact of different leaf structure, leaf blade structure of the top six leaves of tobacco plants in Xuchang was divided into groups of loose, slightly loose, slightly dense, dense, very dense.. The relationships between leaf structure of different grades and appearance quality, physical characteristics, chemical composition and sensory quality were studied. The results showed that, when the leaf structure changed from tight toward loose, the appearance quality became better, and the loose leaf was the best, followed by the slightly loose leaf. And the physical property tended to be suitable, and the blade density of loose and slightly loose leaves reduced significantly. The chemical composition tended to be suitable, and the protein content of loose and slightly loose leaves reduced significantly, and the total and reducing sugar content increased. The loose leaf’s starch content reduced significantly and sugar/alkali ratio rose gradually. The sensory quality was among the lower middle to good level, and the loose leaf was the best, followed by the slightly loose leaf. The industrial usability rose gradually, and the ratio of loose and slight loose leaf in class I cigarette formula was 100% and 85.71% respectively. The tobacco quality with slight dense structure was medium, which was usually used in class II and III cigarette formula. The tobacco quality with dense and very dense structure was poor, which were only used in class Ⅳ and Ⅴ cigarette formula. The tobacco samples with different leaf structure can be used in training, professional grade guidance, tobacco purchasing, receiving, and the leaf structure is available characteristic index of appearance quality in selecting usable tobacco in redrying and processing.
In order to study the genetic characteristics of flue-cured tobacco,5 flue-cured tobacco varieties (lines) were used as materials,and the Griffing hybrids were combined with 10 groups.The total carotenoids degradation products of flue-cured tobacco the combining ability of 3 substances,beta demascenone,giant bean three ketene,and farnesyl acetone,and the genetic parameters were analyzed.The results showed that the genetic characteristics of the carotenoid degradation products were mainly controlled by non-additive effects.664-01 and Yunyan 87 had not only a higher content of carotenoid degradation products,but also a high general combining ability (GCA) value,which was the 2 excellent parents to cultivate high aroma varieties.T NC 89 × 664-01,Zhongyan 100 × Yunyan 87,Qinyan 96 × Yunyan 87 had a high specific combining ability (SCA) effect value,which had important application value in improving the breeding practice of the breeding objectives of aroma.The genetic determinant of traits was high,which indicated that the reliability of selection by phenotype was high.
为了实现烟叶烘烤环节减工降本,利用生物质颗粒燃料代替煤炭作燃料,采用智能生物颗粒燃料燃烧机,对密集式烤房进行简单改造,与普通密集式烤房做烟叶烘烤对比试验.试验点选取了同一田块、相同品种、田间长势正常、叶片着生部位相同且成熟度一致的鲜烟进行对比,同时选择同一烘烤工艺;以烘烤时间、能耗成本、烟叶外观质量为研究指标,评价烟叶烘烤效果.结果表明:采用新型生物质燃料热风炉供热烤房在烘烤管理时间上节省21.7 h,燃料成本比煤多出188.72元/烤,耗电成本节省了16.06元/烤,人工成本节省216元/烤,上中等烟比例提高了3.01个百分点,均价提高了1.54 元/kg.智能生物颗粒燃料燃烧机供热点火简单,自动进燃料,燃烧完全彻底,省去了烘烤加煤、出渣等繁琐费力劳动,管理方便,易清理维护,一定程度上提高烟叶外观质量和内在品质,同时降低了对环境的污染.
The concept and characteristics of modern tobacco aigriculture were introduced,the status of tobacco production and the problems of developing modern tobacoo agriculture in china were analyzed,and some suggestions for the quickening development were put forward in this article.
In order to breed new tobacco varieties with medicable function,the total DNA of medicinal plant mentha(Mentha haplocalyx Briq.) was introduced into oriental tobacco cv.Basma via pollen-tubeway,the descendants variated comprehensively.A hybrid descendant assuming better traits,designated BD804,was obtained after screening for 3 consecutive years.Routine chemical component analysis indicated that there was no noticeable difference between BD804 and tobacco,and GC/MS test showed that the content of menthol in BD804 was far higher than that in tobacco.78 primers were used to detect the genomes of tobacco,mentha and BD804 with RAPD.The results indicated that 668 bands were produced in tobacco,mentha and BD804,among them,14 bands of BD804 were discovered only in mentha,and the similarity rates of BD804 with tobacco and mentha were 92.80% and 5.84%,respectively.It indicated that some DNA fragments of mentha were integrated into tobacco and tobacco genome variation resulted.
