Из водных растворов декстрана, полигексаметиленгуанидин гидрохлорида и Совидона, а также их смесей с таурином, сформированы устойчивые ультратонкие защитные пленки офтальмологического применения. Введение таурина в растворы декстрана и полигексаметиленгуанидин гидрохлорида вызывает ассоциацию макромолекул полимера в двумерной пленке и понижение поверхностной активности раствора. Введение таурина в растворы Совидона изменяет конформацию макромолекулярных глобул и увеличивает поверхностную активность раствора. Применение исследованных растворов в качестве основы глазных капель не повреждает липидный слой слезной пленки, так что при восстановлении функции слезных желез экстренно сформированная полимерная пленка может быть успешно заменена на природную липидную за счет естественных процессов.
A new approach to formulation of eye drops containing the active substances, taurine and adjuvant polymers such as dextran, poly(hexamethyleneguanidine)hydrochloride and Sovidon is proposed. According to this, the eye drops formulation must be started with studying the physicochemical properties of the polymer and taurine solutions, particularly from evaluation of their surface tension and wettability of the substrate similar to mucous layer of lacrimal film. The optimal values of solution concentrations providing effective delivery of drugs into intracellular space of epithelial cells were determined.
Oligo(hexamethyleneguanidine) (OHMG) hydrochloride samples with various molecular weights synthesized from hexamethylenediamine and guanidine hydrochloride were characterized by 13C NMR, ultracentrifugation, and capillary viscosimetry techniques. It was found that OHMG hydrochloride has 0.16 - 1.08 branching points per molecule. Number-average molecular weights Mn and weight-average molecular weights Mw were calculated from 13C NMR and ultracentrifugation data. A comparative analysis of these characteristics shows that the synthesized oligomers have a wide molecular-weight distribution (Mw/Mn = 5.4 - 9.3).
