The correlation between the total activity of the complement system (TACS) and the content of modified LDL (mLDL) in blood serum when modeling microgravity in Earth conditions was evaluated. Healthy volunteers were placed in a state of antiorthostatic hypokinesia for 21 days. Microgravity in ICR mice was modeled by tail suspension for 14 days. TACS, activity of the alternative pathway (in humans), and the content of mLDL in serum from volunteers and mice were measured by turbidimetry. On day 7 of antiorthostatic hypokinesia, volunteers had a significant increase in TACS, which persisted until day 21. The content of mLDL in blood serum increased significantly on day 7. In experiments with mice, TACS increased significantly on day 14, and mLDL content decreased significantly. The limitation of mobility, which is an integral part of microgravity modeling, causes an increase in the overall hemolytic activity of the complement system in both humans and laboratory animals, indicating a response to significant physiological stress.
Endogenous multiple modified LDL (mLDL) and the renin-angiotensin system play a significant role in the development of atherosclerosis. It has been found that by behavioral and hemodynamic parameters the physiological activity of angiotensin II (Ang II) in combination with mLDL is considerably modified due to weakening of its diuretic effect and the inversion of hypertensive and tachyarrhythmic effects. Atherosclerosis is a long-term pathological process, so a single administration of artificially synthesized Ang I-mLDL complexes can be considered a model of the first contact of the body with pathogenic factors. We believe that angiotensin complexes with mLDL participate in adaptive and compensatory mechanisms at the initial stages of atherosclerosis and later contribute to its progression.
We have previously demonstrated multidirectional changes in trypsin activity and some serum biochemical parameters in rats under conditions of water and food deprivation with following satiation. In the present study, administration of soybean trypsin inhibitor (STI) to animals subjected to water and food deprivation caused a moderate decrease in trypsin activity and an increase in the serum concentration of total protein, which attested to deceleration of protein metabolism. Significant changes in the content of cholesterol and triglycerides against the background of STI administration were also found indicating intensification of lipid and energy metabolism. These findings suggest that STI prevents disturbances of metabolic processes in the body under conditions of water and food deprivation. However, the peculiarities of STI influence on metabolic processes require further study.
The parameters of the cytokine profile and functional activity of the complement system in the blood of rats were studied during different time periods of chronic unpredictable mild stress using a model of sequentially alternating low-intensity stress effects for 1, 2, 3, and 4 weeks. In the dynamics of observation, a general tendency towards multidirectional fluctuations in the concentration of cytokines was revealed: an increase in IL-10, but a decrease in IL-4 in comparison with the control. Statistically significant changes in the level of IL-10 were noted after 2, 3, and 4 weeks, IL-4 — after 2 and 4 weeks of stress loads. The percentage of lysis of the C3 component in rats gradually increased by the 2nd week of chronic stress, but then decreased and practically did not differ from the control values (intact animals) by the end of the study. These results illustrate the specificity of changes in the indicators of the C component of the complement system and the cytokine profile of the blood reflecting activity of the cellular and humoral components of the immune response in rats exposed to repeated stress factors of different origins and duration.
Длительные стрессорные воздействия вызывают дизрегуляцию большинства компонентов иммунной системы. Большой интерес исследователей к изучению системы комплемента и цитокинов связан с широким спектром их функций. Цель исследования – изучение показателей концентрации цитокинов и функциональной активности системы комплемента в крови крыс на модели хронического непредсказуемого мягкого стресса разной длительности. Методы. Исследование проведено на 42 крысах-самцах Wistar. Выделены 5 экспериментальных групп: контрольная группа; четыре опытные группы, которых подвергали многократным стрессорным нагрузкам на модели хронического непредсказуемого мягкого стресса в течение 1, 2, 3 и 4 недель соответственно. Концентрацию ИЛ-4 и ИЛ-10 в сыворотке крови крыс определяли методом твердофазного ИФА, функциональную активность системы комплемента – с помощью скрининг-теста в реакции комплемент-зависимого лизиса эритроцитов человека. Результаты. Выявлено статистически значимое повышение уровня ИЛ-10 через 2, 3 и 4 недели, но снижение концентрации ИЛ-4 в крови крыс через 2 и 4 недели стрессорных воздействий. Функциональная активность системы комплемента увеличивалась через 2 недели хронического стресса, но не отличалась от контрольных значений в последующие периоды. Заключение. Выявленные изменения иллюстрируют специфику активности клеточного и гуморального звеньев иммунного ответа в условиях многократных непредсказуемых стрессорных воздействий разной длительности. Long-term stress causes dysregulation of most components of the immune system. The great interest of researchers in studying the complement system and cytokines is associated with a wide range of their functions. The purpose of the study was to study the concentrations of cytokines and the functional activity of the complement system in the blood of rats using a model of chronic unpredictable mild stress of different durations. Methods. The study was conducted on 42 male Wistar rats. There were 5 experimental groups: control group; four experimental groups that were subjected to repeated stress loads in a chronic unpredictable mild stress model for 1, 2, 3 and 4 weeks, respectively. The concentration of IL-4 and IL-10 in the blood serum of rats was determined by solid-phase enzyme immunoassay, the functional activity of the complement system was determined using a screening test in the reaction of complementdependent lysis of human erythrocytes. Results. A statistically significant increase in the level of IL-10 was revealed after 2, 3 and 4 weeks, but a decrease in the concentration of IL-4 in the blood of rats after 2 and 4 weeks of stress exposure. The functional activity of the complement system increased after 2 weeks of chronic stress, but did not differ from control values in subsequent periods. Conclusion. The identified changes illustrate the specificity of the activity of the cellular and humoral components of the immune response under conditions of repeated unpredictable stressors of varying duration.
Childhood adversity can induce maladaptive behaviors and increase risk for affective disorders, post-traumatic stress disorder, personality disorders, and vulnerability to stress in adulthood. Deprivation of maternal care interrupts brain development through the disturbance of various neurotransmitters, however, the details remain unclear. The features of the symptoms of disorders are largely determined by early stress protocol, genetic characteristics (line), and the sex of the animals. The purpose of current study was (1) to assess behavioral changes in adult Wistar rats of both sexes after early life stress; (2) to determine the levels of monoamines in brain structures involved in the motor, emotional, and social reactions in rats aged 1 and 2 months; and (3) to determine the level of monoamines after physical or emotional stress in adult rats. The rat pups were separated from their dams and isolated from siblings in tight boxes at a temperature of 22–23 °C for 6 h during postnatal days 2–18. The data were processed predominantly using two-way analysis of variance and the Newman–Keys test as the post hoc analysis. The adult rats demonstrated an increase in motor activity and aggressiveness and a decrease in levels of anxiety and sociability. Behavioral disturbances were accompanied by region-, sex-, and age-dependent changes in the levels of monoamines and their metabolites. The dopaminergic and noradrenergic systems were found to be sensitive to psycho-emotional stress.
Background and Aims : To determine the quality characteristics of nutrition in individuals with low cardiovascular risk (SCORE 2 < 2,5%) and to assess the relationship of nutritional characteristics with metabolic disorders and body composition.Methods: The study included 90 patients: 43.8 ± 3.2 years, 37 men (47%) and 41 womеn (53%). 62 (69%) participants with central obesity (CO), low cardiovascular risk (SCORE ≤1%, SCORE 2 < 2,5%, CMDS 0-1). All patients were provided with a nutritional questionnaire consisting of 11 questions.Results: Inappropriate nutrition was registered in all patients with CO. A number of diet factors had a correlation with TG, LDL, TC, WC, body fat mass, hepatic steatosis and epicardial fat. Among them: processed meat (r = 0.3; r = 0.4; r = 0.3; r = 0.4; r = 0.4; r = 0.4; r = 0.48, respectively; p = 0.001), the absence of nuts (r = 0.4; r = 0.4; r = 0.3; r = 0.4; r = 0.4; r = 0.4; r = 0.3, respectively; p = 0.001), insufficient quota of fruits and vegetables (r = 0.7; r = 0.7; r = 0.6; r = 0.7; r = 0.7; r = 0.5; r = 0.7 , respectively; p = 0.001). It is important that according to the nutritional scale the cardiovascular diseases risk became higher in 57% of patients.Conclusions: The low cardiovascular risk group is very heterogeneous due to the high prevalence of central obesity and unbalanced nutrition. The food scale helps to identify problem areas of the diet within 2 minutes. Aim. To study the relationship between the abundance of the genera in the gut microbiota (GM) and levels of serum biomarkers of chronic systemic inflammation and endotoxemia in patients with HFpEF.Material and methods. The composition of GM among 42 patients with HFpEF (men, 57,1%) was assessed by 16S rRNA sequencing. The median age was 67,0 years, interquartile range [64,0; 71,5] years. Correlation and multivariate regression analysis (with adjustments for sex and age) of relationships between the relative abundance of intestinal bacteria and the concentrations of serum biomarkers including high-sensitivity C-reactive protein (hsCRP), interleukins (IL) 1β and 6, the soluble suppressor of tumorigenicity (sST2), and the level of lipopolysaccharide (LPS) was carried out.Results. According to multivariate regression analysis, the relative abundance of Haemophilus was directly related to the concentration of IL-1β (odds ratio (ОR) 32,37, 95% confidence interval (CI) 2,071237,69, p=0,025), Coriobacteriaceae (unclassified) — with IL-6 (ОR 6,27, (1,42-36,74), p=0,024), Porphyromonadaceae (unclassified) — with sST2 (ОR 5,96, (1,33-34,39), p=0,028), and the relative abundance of the genera Pseudomonas (ОR 7,09, (1,45-42,39), p=0,020), Parasutterella (ОR 4,55, (1,07-22,76), p=0,047) and Clostridiaceae (unclassified) (ОR 4,85, (1,06-24,7), p=0,045) was directly associated with LPS levels.Conclusion. In patients with HFpEF, the relative abundance of some GM genera (e.g., Haemophilus, Coriobacteriaceae (unclassified), Porphyromonadaceae (unclassified), Pseudomonas, Parasutterella, Clostridiaceae (unclassified)) is statistically significantly associated with the concentration of biomarkers of chronic systemic inflammation and endotoxemia.

Complement activation is an integral part of the innate immune defence but also influences directly triglyceride synthesis. The study aim was to evaluate a potential association between central obesity, C3a-desArg and atherosclerosis in individuals with low SCORE-2 risk.Methods: A pilot study, including patients at low risk according to SCORE-2, was designed. Anthropometry and bioimpedance analysis were performed in every subjects alongside with laboratory tests for cholesterol profile, multiply modified LDL (mmLDL) and C3a-desArg. Central obesity (CO) was detected according to the IDF criteria. Individuals were consecutively screened for carotid atherosclerosis using ultrasound method. ANOVA test and Pearson’s chi-square test were used to assess the impact of different metabolic parameters on atherosclerosis prevalence.Results: 127 adult subjects were screened (mean age: 43(3) years; BMI 27(5) kg/m2; and 47% male). 80 (63%) participants with central obesity. Prevalence of atherosclerosis was 32% (41).The level of mmLDL was significantly higher in the CO group than that in the control, regardless of gender: 20.8 U (8.5) vs 13.6 U (7), respectively (p‹0.01). cIMT average were correlated with WC, body fat mass, TG, TC, LDL, and mmLDL (r = 0.4; 0.3; 0.3; 0.4; 0.4; 0.5, respectively, p = 0.001) and C3a-desArg (r = - 0.4, p = 0.001). According to multivariate analysis: at a C3a-desArg>10.7 ng/ml, atherosclerosis was not detected.Conclusions: С3a-desArg plays an unexpectedly beneficial role in the atherosclerosis. С3a-desArg is critical for triglyceride synthesis and the maintenance of lipid homeostasis,resulting in increased adiposity and atheroprotection. ЦЕЛЬ ИССЛЕДОВАНИЯ Оценить уровень пентраксина-3 (РТХ-3) у пациентов с хронической сердечной недостаточностью (ХСН) ишемического генеза II—IV функционального класса (ФК) по NYHA и выявить возможные связи с наличием метаболического синдрома (МС) и течением заболевания. МАТЕРИАЛ И МЕТОДЫ Обследованы 180 пациентов с ХСН ишемического генеза II—IV ФК, из них 90 пациентов с МС и 90 — без МС. Возраст пациентов в группе МС на момент включения в исследование составил 56,0 [46; 63] года, в группе без МС — 59,5 [51; 70] года (p=0,498). Все пациенты прошли комплексное обследование, включавшее сбор жалоб, изучение анамнеза, физикальное обследование, антропометрические измерения, лабораторное и инструментальное исследование, в том числе исследование уровня РТХ-3. РЕЗУЛЬТАТЫ В группе пациентов с ХСН и МС по сравнению с группой с ХСН и без МС преобладали больные с отягощенным наследственным анамнезом по сердечно-сосудистым заболеваниям (ССЗ) (50% против 34%), с артериальной гипертонией (38% против 24%), более высоким ИМТ (30,4 [27,9; 35] кг/м2 против 24,5 [23; 27] кг/м2), большей выраженностью одышки (67% при нагрузке и 32% в покое против 64% при нагрузке и 24% в покое) и большей частотой увеличения массы тела за последнюю неделю (44% против 19%), а также более высоким уровнем глюкозы (6,0 [5,4; 6,7] против 5,0 [4,7; 5,2]) и менее высоким креатинином (85 [71; 99] против 87 [74; 121]) (p<0,05). У пациентов с ХСН и МС по сравнению с пациентами с ХСН и без МС отмечалось более выраженное структурное ремоделирование миокарда. Средний уровень PTX-3 у пациентов с ХСН и МС составляет 79 нг/мл, без МС — 98 нг/мл (p=0,924). У пациентов с ХСН и МС уровень PTX-3 показал статистически значимую корреляцию с величиной фракции выброса левого желудочка (ФВ ЛЖ) (r= –0,281) и размером левого предсердия (ЛП) (r=0,198), а также с большим числом баллов по ШОКС (r=0,246) и меньшим показателем теста с 6-минутной ходьбой (r= –0,297; p<0,05). У пациентов с ХСН без МС уровень PTX-3 показал статистически значимую корреляцию с величиной ФВ ЛЖ (r= –0,187) и размером ЛП (r=0,192), а также с бо́льшим числом баллов по ШОКС (r=0,232) и меньшим показателем теста с 6-минутной ходьбой (r= –0,196), с выраженностью одышки (r=0,135), отеков (r=0,175) и одышки лежа (положения в постели) (r=0,126; p<0,05). ЗАКЛЮЧЕНИЕ У пациентов с ХСН ишемического генеза II—IV ФК по NYHA в клинических исследованиях целесообразна оценка концентрации PTX-3 в качестве фактора, ассоциированного с ее неблагоприятным течением. Вероятное влияние МС на уровень PTX-3 целесообразно учитывать при планировании дизайна исследований, направленных на оценку его прогностической значимости в отношении ССЗ.
Актуальность. Определение фибриногена (ФГ) по Клауссу является чувствительным к гипо-, дис- и гипер-фибриногенемии, а также к продуктам деградации фибрина, которые влияют на процесс коагуляции и могут быть причиной занижения или повышения уровня фибриногена. Цель настоящей работы - разработка простого турбидиметрического теста для определения фибриногена и оценка его функциональности. Материалы и методы. В работе определяли фибриноген в цитратных плазмах 96 пациентов по методу Клаусса, модифицированным методом Рутберга и новым турбидиметрическим тестом. Результат. Разработан простой турбидиметрический тест определения фибриногена, основанный на определении коагулированного фибриногена при рекальцификации цитратной плазмы по формуле: ФГ (г / л) = ΔА450 × 5,12, где ΔА450 - изменение оптической плотности в опытной пробе; 5,12 - переводной коэффициент ΔА450 в г/л ФГ. Корреляционный анализ показал высокую степень сходимости результатов (R = 0,843). При разнице ФГ более 10% по Клауссу и по разработанному методу констатируют как нарушенную функциональность ФГ. Заключение. Использование простого, доступного турбидиметрического метода определения фибриногена в рутинных исследованиях позволяет оценивать функциональность фибриногена. Background. The Clauss fibrinogen assay is sensitive to hypo-, dys- and hyper-fibrinogenemia, as well as to fibrin degradation products, which may affect coagulation and cause underestimation of or increase the fibrinogen level. The aim of this study was to develop a simple turbidimetric test to measure fibrinogen and evaluate its functionality. Materials and methods. In this study, fibrinogen was measured in citrate plasma from 96 patients using the Clauss assay, a modified Rutberg method, and a new turbidimetric test. Result. A simple turbidimetric test was developed for measuring fibrinogen. This test is based on measuring coagulated fibrinogen during recalcification of citrate plasma according to the formula, FG (g/l) = ΔА450 × 5.12, where ΔА450 is a change in optical density of the experimental sample; 5.12 is a conversion coefficient ΔА450 in g/l FG. Correlation analysis showed a high degree of agreement between the results (R = 0.843). With a FG difference of more than 10% by the Clauss assay and the developed method, impaired functionality of the FG is stated. Conclusion. Using a simple, affordable turbidimetric method to determine fibrinogen in routine studies allows accessing the fibrinogen functionality.

The findings suggest a pronounced presence of the C1q complement component in various DM areas during inflammation, both in the epithelial layer and in the lamina propria, where C1q is localized in the region of the basolateral membrane of enterocytes and some free cells, presumably neutrophils, macrophages and mast cells. A C1q-specific reaction in the epithelial cells and secretory ducts of the duodenal glands indicates a possible secretion of C1q. Co-localization of C1q, cathepsin G and cell adhesion proteins in the DM epithelium implies their interaction in the regulation of tissue metabolism, modification and maintenance of the barrier properties of the intestinal epithelium in normal and with inflammation.

Разработан скрининг-тест для определения множественно-модифицированных липопротеинов низкой плотности (ммЛПНП) в сыворотке крови для рутинных исследований. Инкубация сыворотки в буфере, содержащем 9,1% поливинилпирролидона 35000 (ПВП-35) приводит к полной агрегации ммЛПНП. Патогенность выделенных ммЛПНП подтверждена в тестах связывания системы комплемента и литической активности in vitro , а также в тесте сосудистой дистонии у крыс in vivo . Предлагаемый способ отличается высокой производительностью и низкой себестоимостью. Метод включает 2 операции - смешивание сыворотки с раствором 10% ПВП-35 в 96-луночных иммунологических планшетах, инкубация в течение 10 мин при комнатной температуре и турбидиметрия при 450 нм на ИФА-ридере. Проведены исследования взаимосвязи концентрации ммЛПНП в сыворотке крови и толщины комплекса интима-медиа по данным ультразвукового дуплексного сканирования сонных артерий у 78 человек с низким сердечно-сосудистым (SCORE <1%) и низким кардио-метаболическим рисками (CMDS 0-1). Из всех фракций липидного спектра крови ммЛПНП показали самый высокий коэффициент корреляции с показателями субклинического атеросклероза, что подтверждает высокий атерогенный потенциал ммЛПНП. The proposed method is characterized by high productivity, low cost, includes 2 operations - mixing serum with a solution of 10% PVP-35 in 96-well immunoassay plates, incubating for 10 min at room temperature and turbidimetry at 450 nm on an ELISA reader. A study was made to determine relationship between concentration of mmLDL in the blood serum and thickness of the intima-media complex from data of ultrasound duplex scanning of carotid arteries with 78 people with low cardiovascular risk (SCORE <1%) and low cardiovascular risk (CMDS 0-1). Of all fractions of the lipid spectrum of the blood, mmLDL showed the highest correlation coefficient with the indices of subclinical atherosclerosis, which confirms high atherogenic potential of mmLDL.

В работе исследовали сыворотки крови 90 относительно здоровых лиц с избыточной массой тела (ИМТ >25) без метаболического синдрома. Иммуноферментный анализ функциональной активности системы комплемента по классическому пути проводили с использованием коммерческого набора. Реакцию лизиса эритроцитов барана, сенсибилизированных антителами (ЕА), оценивали турбидиметрически по снижению оптической плотности суспензии при длине волны 620 нм, степень лизиса определяли по калибровочному графику. Результаты. Разработан простой скрининг-тест для определения функциональной активности классического пути системы комплемента. Корреляционный анализ показал высокую степень сходимости результатов (r = 0,496), полученных разными методами. Заключение. Скрининг-тест для определения функциональной активности классического пути системы комплемента является быстрым, информативным и доступным для рутинных исследований. The aim of this work was to develop an accessible test for screening the functional activity of the complement system classic pathway. Materials and methods. Blood serum from 90 relatively healthy, overweight (BMI>25) individuals without the metabolic syndrome was studied. Enzyme immunoassay of the complement system functional activity following the classic pathway was performed using a commercial kit. The lysis of antibody-sensitized sheep erythrocytes was evaluated turbidimetrically by a decrease in suspension optical density at a wavelength of 620 nm; the lysis intensity was determined by the calibration curve. Results. A simple screening test was developed to determine the functional activity of the complement classic pathway. A high degree of correlation (r = 0.496) was observed between results obtained by different methods. This screening test for evaluating the functional activity of classic complement system pathway is fast, informative, and available for routine research.
