The purpose of the work was to assess the influence of a phenotype of oxidative metabolism on intensity of pharmacodynamic effect of the multistage hypotensive therapy administered to patients with arterial hypertension of high and very high risk. The research included 120 patients with arterial hypertension of 2-3 degrees. The high risk of developing cardiovascular complications was detected in 58 people (48.3%), very high - in 62 patients (51.7%). The results of the conducted research showed the dependence of the degree of lowering arterial pressure on phenotypical option of oxidative metabolism estimated after 14-week treatment of patients with arterial hypertension with various schemes of multistage hypotensive therapy including amlodipin as a starting drug. This fact enables to make the individualized choice of initial option of pharmacotherapy for patients with arterial hypertension.
The correction of endothelial dysfunction is currently the most important component of cardiovascular therapy. The drugs used for pathogenetic treatment have a wide range of protective effects that can be enhanced by combining with the well-known drug Magnesium Orotate. We assessed the impact of 0.33 mg/kg/day and 0.66 mg/kg/day Rosuvastatin (Rozart) and 100 mg/kg/day Magnesium Orotate and their combined administration on lipid peroxidation level in rats’ serum in experimental endothelial dysfunction. The study revealed that the combined use of low-dose Rosuvastatin and Magnesium can decrease lipid peroxidation level in rats’ serum.
Determination of the phenotype of oxidizing metabolism allows to personalize the dosage of the drugs in treatment of the patients with peptic ulcer. This approach allows to reach maximum efficiency and economic profitability of the treatment that leads to stop pain and dyspepsia, to change рН, to eradicate Helicobacter pylori, thus, making essential impact on the forecast of the disease and quality of life of patients.
