The present study aimed to evaluate the antimicrobial susceptibility profile and molecular characteristics of Escherichia coli isolated from various foods, including ground beef, pork, Minas fresh cheese, lettuce, and chicken. A total of 150 E. coli strains were recovered and tested against conventional antimicrobials and crude extracts of Solanum palinacanthum and Siparuna guianensis. Susceptibility testing was conducted using disk diffusion and broth microdilution methods. Additionally, phylogenetic characterization and pathotype identification were performed via polymerase chain reaction (PCR). Results indicated that all E. coli isolates were resistant to at least two antimicrobial classes, with 55.3
ABSTRACT: Leishmaniasis, classified as neglected diseases, is expandig throughout Brazilian territory and is transmitted by the hemantophagy of sandflies. The municipality of Jataí is classified as silently receptive to sandflies. Therefore, this study determined the species of sandflies present in urban area of Jataí. Collections were made mothly from September 2023 to January 2024 using CDC-type light traps. The screened insects were then sent for taxonomic identification at the René Rachou Research Center, Oswaldo Cruz Foundation . As a result, 15 insects were collected; although, only 46,6% (07) were confirmed as sandflies. After analysis four species were identified: Sciopemyia sordellii, Evandromyia walkeri, Evandromyia carmelinoi, and Nyssomyia intermedia. Our results demonstrated that Jataí has a low number of sandflies, which may had been influenced by precipitation and humidity during the collection periody.
Among the primary diseases affecting the Brazilian dairy herd is bovine mastitis, a complex, multifactorial disease whose primary pathogen is Staphylococcus spp., which is known for its high resistance to beta-lactam antimicrobials and environmental factors. The present study aimed to isolate staphylococci from milk samples collected from cows at a dairy farm in Jataí, GO, Brazil. Isolation was carried out using samples from 90 sick animals collected between November and March. After isolation and identification, antibiotic tests were performed to assess resistance and investigate the presence of associated virulence genes. The method chosen for the antibiogram was disk diffusion, which we used to evaluate two groups of animals with clinical and subclinical mastitis at that property. Of the milk samples analyzed, 58.9% were positive for Staphylococcus sp., with 5.5% coagulase-positive (SPC) and 53.3% coagulase-negative (SCN). The SPC bacteria were associated with clinical and subclinical mastitis, while SCN was often associated with subclinical mastitis (68.75%). The high frequency of Staphylococcus sp. indicates they remain a primary causative agent of mastitis, with SCN having the greatest impact in this herd. Among the antibiotics tested, we found three with prevalent resistance, namely, oxacillin, penicillin, and vancomycin. We also identified coagulase negative staphylococci (CoNS) as the predominant cause of subclinical mastitis in the bovine herd. Methicillin-resistant (MRS) and vancomycin resistant Staphylococcus isolates stood out as causes of mastitis. MRS isolates also underwent a polymerase chain reaction (PCR) test to detect the mecA and mecC genes. However, no isolate exhibited those resistance genes. This shows that antibiotic resistance may occur through other mechanisms and genes.
The current public health worry revolves around infections stemming from the consumption of meat contaminated with Salmonella spp., which has been linked to the growing issue of antimicrobial resistance. We investigated the presence of non-typhoidal Salmonella (NTS) in the retail of spicy chicken meat and its antimicrobial susceptibility profile. Forty establishments (supermarkets and butcheries) in Mid-West Brazil were visited in February and July 2017, totaling 80 samples. Salmonella isolation, molecular serotyping and antimicrobial analyses were performed. Of the total establishments, 18 sold meat NTS-positive, 6 of which were contaminated in the two visits, resulting in 24 contaminated samples (30%). Of these, the prevalent serovars were Heidelberg (58%–14/24), Minnesota (21%–05/24), Muenchen (17%–04/24) and Mbandaka (4%–01/24), but none was positive for Typhimurium and Enteritidis. Still, 55.6% (10/18) of the establishments did not have a veterinary inspection seal, and 50% (09/18) sold products beyond their expiration date. Worse still, 87.5% (21/24) of NTS isolates were resistant to at least one antimicrobial agent, being 66.7% (16/24) multidrug-resistant. The bla gene was detected in 41.6% (10/24), confirming resistance by β-lactamase enzymes. These findings highlighted the great risk of foodborne illness due to the possibility of contaminated seasoned chicken meat and transmission of bla resistant genes through the food chain.
The microbiological quality control, of food, for human consumption is extremely important. SIM is focused on inspecting and verifying the microbiological quality of products of animal origin, routinely carrying out inspections and collecting samples for laboratory analysis. This is one of the alternatives to assess whether meat products available for sale are suitable for consumption. The objective was to investigate the microbiological quality of meat products sold in Jatai and inspected by the SIM. 65 results of microbiological tests of samples collected in the years 2019, 2021 and 2022 were analyzed. Of the tests performed, 05 samples were positive for Salmonella spp (7.9%), 05 positive for Staphylococcus Coagulase Positive (SCP) (12, 2%), 08 positive for Thermotolerant Coliforms(8.7%), 08 positive samples for Escherichia coli(26.6%) and 38(95%) for Viable Aerobic Mesophiles at 30(degrees)C. Of the total of 65 samples, 42 (64%) were positive for some microbiological contaminant. The existence of a Municipal Inspection Service implemented in the municipality and the adoption of Good Food Manufacturing Practices by the establishments is an important measure in the control of hygiene and food safety. However, as detected by this research, microbiological analyzes of marketed meat foods are essential to detect hazards and guide adjustment actions.
Mastitis is one of the main diseases of the dairy herd, and is linked to product quality and viability of the activity. The prevalence of mastitis is influenced by environmental and sanitary conditions, with somatic cell count (SCC) being one of the main forms of diagnosis of subclinical mastitis. The objective of this research is to evaluate the epidemiology of mastitis (bacterial groups and CCS) on a good-production property in the southwest of Goias with cows confined in a Free stall system, evaluating the difference in the case series of mastitis in the dry period. and in the rainy season. During the year, there were 180 cases of clinical mastitis (60% in the rainy season and 40% in the dry season), 245 cases of subclinical mastitis (64.5% in the rainy season and 35.5% in the dry season), and 12 cases of mastitis in the after birth of the calf (58% in the rainy season and 42% in the dry season). The CCS in the rainy season was 442,260 cells/ml while in the dry season it was 326,800 cells/ml. A higher prevalence of clinical, subclinical and postpartum mastitis was observed in the rainy season, whether of contagious or environmental origin, as well as higher CCS values. A higher prevalence of clinical and subclinical mastitis was observed in the rainy season, whether of contagious or environmental origin, as well as higher SCC values in the herd. It was possible to note that the presence of humidity and heat probably facilitate the spread of mesophilic microorganisms, which leads to a greater frequency of mastitis in animals.
The presence of Shiga toxin-producing Escherichia coli (STEC) and resistance to beta-lactams in healthy sheep represents a potential public health risk. This study aimed to characterize STEC isolates in sheep feces for toxin production and resistance to beta lactam antibiotics. In the present study, among the 40 isolates, we found a predominance of subtype Stx1 (22/40), followed by subtype Stx1 + Stx2 (11/40), while the less prevalent group was Stx2 (7/40). Also, we found phenotypical resistance to beta-lactam antibiotics in 50% (20/40) of the strains analyzed, forming two groups, one with resistant isolates and the other with non-resistant isolates. The cytotoxicity of the isolates did not vary among the groups. In addition to having this characteristic, some multiresistant isolates produced significant amounts of toxins. This leads to the conclusion that the mechanisms of antimicrobial resistance via beta lactamases are present in sheep STEC and that the cytotoxicity of those isolates is variable regarding such resistance.
ABSTRACT The indiscriminate use of antibiotics has contributed to the emergence of multiresistant bacteria. Faced with this, the search for antibiotics from plants has proven to be a promising alternative. The objective of this work was to isolate and identify Staphylococcus sp. resistant to methicillin of the ocular surface of dogs with ophthalmopathies and to evaluate its susceptibility to alcoholic extract of the bark and hexane extract of the pulp of Caryocar brasiliense. Biological material was collected from the ocular surface of 21 dogs presenting ophthalmopathies. We isolated 64 S. pseudintermedius, among these, 4 isolates were identified as methicillin-resistant S. pseudintermedius (MRSP). The alcoholic extract of C. brasiliense peel was able to inhibit the bacterial growth of MRSP isolates at a concentration of 2.2%. Thus, the extract from the C. brasiliense peel has antimicrobial potential and represents an alternative in the control of MRSP.
Background: Pseudomonas aeruginosa is a gram-negative aerobic bacterium and non-glucose fermenting, that usually causes opportunistic infections in animals, including humans. It is rarely involved in primary disease. The antibiotic-resistant bacterial strains are mainly developed due to the inappropriate use of antibiotics, however treating P. aeruginosa infections can be difficult owing to their natural resistance to antibiotics. Furthermorer resistant microorganisms such as P. aeruginosa grow by developing biofilms. Inaccurate diagnoses and absence of adequate microbiological tests can cause difficulties in resolving cases. This report describes a case of chronic superficial infection in a bitch caused by multidrug-resistant Pseudomonas aeruginosa (MDR-PA). Case: A 6-year-old bitch Shih Tzu, initially presented with an exudative erythematous lesion in the snout region, which progressed to deep lesions, and spread to the back and limbs; furthermore, the animal always experienced a fever before new wounds emerged. Lesion samples, collected using a swab and processed at the Veterinary Microbiology Laboratory of the Federal University of Jatai (UFJ), revealed the presence of Pseudomonas aeruginosa. The isolate was multidrug-resistant and a carrier of TEM and ppyR genes. In the diffusion disk antibiogram, the isolate was found resistant to 14 different antibiotics belonging to 6 classes. Antimicrobial resistance was also tested using the minimum inhibitory concentration (MIC) test against imipenem, ceftazidime, ciprofloxacin, ticarcillin + clavulanic acid and aztreonam present in the MIC test strip. Treatment with amikacin and muporicin proved to be effective; however, owing to lesions extending to the face and palpebral involvement, the animal lost its eyeballs. Discussion: Pseudomonas aeruginosa is frequently associated with nosocomial infections mainly affecting immunosuppressed patients. Among the antibiotics tested, the group with the highest number of ineffective antibiotics was beta-lactams, where sensitivity was only observed for ticarcillin and ceftazidime. Recent studies have demonstrated that ceftazidime can reduce biofilm volume, inhibit motility, and repress the expression of genes associated with bacterial adhesion in P. aeruginosa. Therefore, the production of biofilm in P. aeruginosa is an important virulence factor as it facilitates a stable environment for the microorganism, which protects the bacteria from contact with antimicrobials. In addition, prolonged exposure to a wide variety of antimicrobials creates an environment of selective pressure between microorganisms, facilitating the emergence of multidrug-resistant strains. Furthermore, it is now well recognized that low doses of antibiotics, administered during continuous and fluctuating treatments, can stimulate biofilm establishment and are partly responsible for biofilm-specific antimicrobial tolerance. The resistance profile of P. aeruginosa isolated from dogs varies considerably, and the presence of isolates with a possible biofilm production capacity represents a challenge for the interpretation of the antimicrobial susceptibility profile. Culture and antibiogram is fundamentally important, both clinically and in environmental monitoring, in addition to the use of antibiogram data for decision making in clinical treatment.
Background: The presence of resistant and potentially virulent bacterial strains in a veterinary hospital environment is a neglected problem. Pseudomonas aeruginosa is an opportunistic microorganism present and circulating in the veterinary hospital environment, of clinical importance and zooanthroponotic transmission of P. aeruginosa has also been reported. The aim of this study was to characterize the population of P. aeruginosa present in a veterinary hospital environment by evaluating their resistance profile and biofilm production.Materials, Methods & Results: A total of 306 samples were collected from the veterinary hospital environment (swabs from consultation tables, surgical tables, door handles, hospitalization cages, stethoscopes, thermometers, and muzzles). The isolates were biochemically identified as belonging to the species Pseudomonas aeruginosa through nitrate to nitrite reduction, motility and oxidase test, growth at 42°C, pigment production, and alkalinization of acetamide. Antimicrobial resistance was tested using the minimum inhibitory concentration (MIC) test. Twenty seven isolates of P. aeruginosa were obtained, with a frequency of 8.8%. The detection of beta-lactamase production and biofilm formation genes by polymerase chain reaction (PCR). Two multidrug resistant (MDR) and 3 single-drug resistant (SDR) strains of P. aeruginosa were identified. Furthermore, it was observed that the strains carried genes related to beta-lactamase production (TEM and CTX-M group 25) and biofilm production (pelA, pslA, ppyR).Discussion: Pseudomonas aeruginosa is considered a major cause of opportunistic hospital infections, as it causes significant morbidity and mortality in immunosuppressed individuals, both in animals and in humans. Veterinary hospitals can harbor microorganisms that cause infections, as well as multiresistant agents. Normally, these environments have a large circulation of people and animals, which particularly enables a facilitated dissemination of these resistant microorganisms. Recently, the World Health Organization (WHO) listed carbapenem-resistant P. aeruginosa as one of 3 bacterial species in critical need for the development of new antibiotics to treat their infections. The data found in this work strengthen the knowledge on the antimicrobial resistance capacity that P. aeruginosa exhibits. The presence of 3 multiresistant strains further highlights the advanced stage of resistance of this bacterial species. The characterization of strains of this species in a veterinary hospital environment is crucial for the control of this population circulating in this environment, and the consequent adoption of more effective measures aimed at controlling its proliferation. The study of this bacterial species in a veterinary hospital environment has a direct impact on human health, due to the mechanisms of resistance and genetic variability that can occur between infections in different animal species and in humans. In view of that, professionals working in veterinary hospitals should be aware of the importance of controlling these microorganisms. Correct measures must be taken to sanitize the environment and utensils between animal care sessions, besides frequent hand washing by all employees and the use of protective equipment such as masks and gloves. The presence of potentially biofilm-producing MDR and SDR strains indicates the free circulation of these bacteria in the veterinary hospital environment. Thus, as a potentially pathogenic microorganism to humans and animals, containment measures must be taken to prevent this possible transmission. Keywords: bacteria, antimicrobial resistance, multidrug resistant, beta-lactamase, biofilm, veterinary care.
Due to the strong selective pressure resulting from the misuse of antibiotics, the natural process of bacterial resistance has been accelerated, leading to the increasingly constant appearance of multiresistant isolates. The high number of multiresistant bacteria is a one health problem. Enterobacteriaceae are usually commensal bacteria of the gastrointestinal tract. However, they can cause infections, and the most important resistance characteristic among them is the production of β-lactamases. This study aimed to identify ESBL-producing Enterobacteriaceae of types of TEM, SHV, and the CTX-M groups. To isolate the enterobacteria, swabs were collected by swiping objects that had contact with the patients and professionals, and the water of the hospital environment. Ten collections were carried out, yielding 306 samples, from which 118 enterobacteria were identified: Escherichia coli, Enterobacter spp., Klebsiella spp., Proteus mirabilis, Serratia spp., and Citrobacter spp. Isolates. The genes TEM and CTX-M, for the production of β lactamases, were detected in 12.7% of the 118 enterobacterial isolates. It is very important to know the bacterial population circulating in the veterinary hospital environment and its resistance to antimicrobials so that professionals can take appropriate measures to minimize the risks of transmission, especially from cages and consultation tables. In addition, the correct control of the microbiological quality of the supply water, as well as environmental cleaning procedures, are essential to prevent the transmission of these microorganisms.
