Cryopreservation of Asian elephant semen remains problematic with respect to maintenance of both adequate semen quality and fertility post-thaw. The present study compared the effects of the penetrating cryoprotectants glycerol and dimethyl sulfoxide (DMSO) in the freezing extender, in conjunction with the surfactant Equex STM paste, on Asian elephant spermatozoa during freezing and thawing. Eight ejaculates from three mature bulls were used for this study. Semen was diluted with Tris egg yolk-based extender which contained either glycerol or DMSO, either with or without Equex, and frozen over liquid nitrogen in straws. Sperm motility, membrane, acrosome, and DNA parameters were assessed over 2 h incubation post-thaw to evaluate the cryoprotective capabilities of each freezing extender. Spermatozoa frozen with glycerol resulted in higher total (P < 0.001) and progressive (P < 0.05) motility compared to spermatozoa diluted with DMSO, regardless of Equex inclusion in the freezing extender. When in the presence of Equex, Asian elephant spermatozoa demonstrated higher proportions of viability (P < 0.001) and intact acrosomes (P < 0.001) post-thaw, regardless of the type of cryoprotectant. A large variance between individual bulls was noticed when assessing sperm DNA integrity, nevertheless freezing extender with glycerol plus Equex showed the highest proportion of spermatozoa with intact DNA after 2 h post-thaw (P < 0.05). Altogether, these results indicate a Tris egg yolk-based freezing extender containing cryoprotectants glycerol and Equex should be integrated into current Asian elephant semen cryopreservation protocols to yield the best sperm quality post-thaw and thus improve fertilising potential.
Context. The removal or supplementation of ejaculates with seminal plasma (SP) can affect cryotolerance and post-thaw survival of spermatozoa in many species. In the Asian elephant (Elephas maximus), elucidation of the SP proteome and investigation of how it affects spermatozoa may enable improvement of cryopreservation protocols. Aims. Herein, we characterise the Asian elephant SP proteome and investigate the impacts of SP on sperm cryotolerance in the presence of conspecific or heterospecific SP. Methods. Proteomic analysis of Asian elephant SP was performed using mass spectrometry on nine samples from three individuals. In a separate study, SP was removed from six ejaculates and spermatozoa were resuspended in Tris extender supplemented with: no seminal plasma (NOSP), conspecific SP from ejaculates exhibiting 'good' (GSP, >60%) or mixed sperm total motility (MSP), or horse SP (HSP). Samples underwent cryopreservation, and sperm parameters were compared prior to cryopreservation and after thawing (0 and 2 h). Key results. Mass spectrometry identified 155 proteins from an array of families. Significant differences were observed in post-thaw sperm quality between SP treatments: high concentrations of MSP (25%, v/v) displayed greater average path and straight-line velocity immediately after thawing (P < 0.05) and greater sperm motility index and beat cross frequency than NOSP after 2 h post-thaw incubation (P < 0.05). The addition of HSP improved sperm kinematic parameters compared to NOSP and GSP treatments (P < 0.05). Conclusions and implications. These preliminary findings suggest the potential of SP to enhance the cryosurvival of Asian elephant spermatozoa, with HSP showing particularly promising results compared to conspecific SP (GSP). Further research into the specific effects of Asian elephant SP proteins is warranted.
Seminal plasma proteins have recently been reported to play a significant role as valuable materials for understanding male reproductive biology, identifying causes of fertility problems, and developing reproductive biomarkers. Proteomic analysis of seminal plasma holds promise in advancing the understanding of male Asian elephant reproductive biology. This study aims to explore seminal plasma proteins of Asian elephants and their probable functions to provide fundamental information about male reproduction in this species. The protein solution from pooled seminal plasma from 10 bulls (a total of 33 ejaculates) was digested into peptides and identified using LC-MS/MS. Out of 986 proteins, 597 were mapped and matched with 58 species in UniProt databases, including Elephas maximus. These mapped proteins were mostly involved in binding function, catalytic activity, cellular process, and metabolic process. Only 29 mapped proteins were recognized to be related in reproductive process, mainly associated in spermatogenesis and sperm capacitation. Additionally, several seminal plasma proteins related to fertility or semen quality in other mammals were also found in Asian elephant semen, such as keratin type I, aldose reductase, thrombospondon-1, fibronectin 1, platelet-activating factor acetyl hydrolase, mannosidase, and semenogelin-2. This discovery clearly reveals the beneficial protein profile in seminal plasma of the Asian elephant and serves as a crucial step in investigating infertility and poor semen quality in this valuable species.
Although artificial insemination (AI) technology is widely used in buffalo breeding in Thailand, AI in buffaloes has a lower conception rate compared to AI in beef cattle. It is crucial to understand the development of the ovary and reproductive system of buffalo heifers and cows. The objective of the present study was to evaluate the reproductive tract characteristic using ultrasound measurements during the luteal and follicular phases in Thai swamp buffalo heifers and cows. The studies of reproductive tract were conducted in buffalo cows (n=8; BCS=3.38) and heifers (n=8; BCS=3.25). The reproductive tract was evaluated by measuring the size of the ovary, dominant follicle, CL, uterine horn, cervix, and vulva. The buffalo cows had larger diameter of the cervix and vulvar width (P<0.05) compared with those of heifers. In the follicular phase, buffalo cows had a mean ovarian diameter (ipsilateral POF) of 2.35 cm, and heifers had a mean ovarian diameter of 2.24 cm (P>0.05). The average preovulatory follicle diameter was 1.29 cm in buffalo cows and 1.18 cm in heifers (P>0.05). In the luteal phase, the mean ovarian diameter (ipsilateral CL) of buffalo cows was significantly larger than heifers (P<0.05; 2.46 vs. 2.09 cm, respectively). The diameters of the CL in buffalo heifers and cows were 1.27 and 1.47 cm, respectively (P>0.05). Buffalo cows and heifers have no differences in ovarian components in the follicular and luteal phases, but there are differences in the size of the reproductive tract, which reproductive tract of buffalo cows is larger than heifers.
To evaluate the effect of P. mirifica supplementation on ovarian activity in crossbred Thai native does, a study was carried out for three consecutive estrus cycle periods: 1) D0-D21(pre-treatment), 2) D21-D42 (treatment) and 3) D42-D63 (post-treatment); D0, D21 and D42 were the days of expected estrus. P. mirifica was orally fed at 1000 mg/kg BW/d for 21 days in supplemented groups. Twenty-one does were equally divided into three groups: 1) CON – control, no supplementation and 2) supplemented groups included 2.1) FEC – start feeding at early-cycle (D21-D42), and 2.2) FMC – start feeding at mid-cycle (D32-D53). Estrus behavior was determined by observation, ovarian activity was monitored by transrectal ultrasonography and fecal progesterone concentration (FPC) was analyzed by EIA. The results showed that CON had lower (P<0.05) receptive behavior and shorter (P<0.05) estrous duration than those of FEC and FMC at treatment and post-treatment periods. FEC had a lesser number of developed follicles (P<0.05) and a greater number of undeveloped follicles than others at treatment period. FMC had a higher (P<0.05) number of undeveloped follicles at post-treatment period. Moreover, FEC and FMC had a higher (P<0.05) number of anovulatory follicles than that of CON, while only FMC had a lower (P<0.05) number of CLs along with a greater (P<0.05) number of early CL regression at the post-treatment period. FPC of FEC and FMC was lower than that of CON during administration. In conclusion, the P. mirifica dosage used in this study enhanced estrus behavior but suppressed ovarian activity in the consequent cycle when fed either at early or mid-cycle. Feeding over ovulation might have more adverse effects concerning CL function.
This study aimed to compare the follicular dynamics in goats during naturally infected with foot-and-mouth disease virus (FMD) subjected to induced ovulations, and after disease recovery, crossbred Thai native does were synchronized with CIDR for 14 days, and then PGF2α and PMSG were administered on the day following CIDR removal. The ovarian activity was determined by transrectal ultrasonography. Clinical signs (fever, anorexia, lameness, and foot lesion) were observed on day 12 post-estrus (day 0, day of expected estrus). The study was carried out for 2 periods: FMD outbreak (day 0–day 21) and FMD recovery (day 63–day 84). Infected does were classified into two groups: (I) does without (n = 5) and (II) does with clinical signs (n = 5). The results showed that during FMD outbreak, the number of follicles/waves and number of follicles > 5 mm in ovulatory follicle wave of group II were lower than those of group I and those of its own group after FMD recovery (P<0.05). Higher in follicular regression rate were found in group II compared to group I in the does with 3 follicular waves during FMD outbreak (P<0.05). Moreover, during FMD outbreak, the does had lower number of follicles > 5 mm and longer day of emergence and day of largest follicles in ovulatory follicle wave than of those after FMD recovery. This observation demonstrated that FMD impaired folliculogenesis in goats, and the ovarian activity could be restored about 1 month after disease recovery.
The dilution effect and effect of restoring seminal plasma (SP) proportion in diluted semen were determined in chilled Asian elephant sperm. Semen was collected from eight males, and samples with ≥30% motile sperm were used in the study. Tris-glucose-egg yolk extender (TE) was used for cooled storage at 4°C for 48 hr. In experiment 1 (n = 18), semen was diluted to 1:1, 1:3, 1:7 and 1:15 with TE (volume per volume). There were no significant changes in sperm viability and sperm with normal acrosome integrity among dilutions, but sperm motility and motility velocities were greater (p < .05) in the 1:1 dilution than those of the 1:7 and 1:15 dilutions at 48 hr of storage. In experiment 2, supplemented SP was derived from elephants and stallions. In experiment 2.1, diluted semen (1:7 dilution) was restored with SP to obtain a 1:2 proportion (n = 8). Sperm motility, viability and sperm with normal acrosome integrity were similar among treatments, but motility velocities were greater (p < .05) with stallion SP at 48 hr of storage. In experiment 2.2, diluted semen (1:15 dilution) was restored with SP to obtain a 1:3 proportion (n = 10). Sperm viability and sperm with normal acrosome integrity were similar among treatments at 48 hr of storage. However, sperm motility and motility velocities were greater (p < .05) with stallion SP than those of others. In conclusion, elephant sperm motility was affected by a dilution effect and restoration of SP proportion with stallion SP, but not with elephant SP, could improve motility in chilled highly diluted sperm.
The objectives of this study were to evaluate the effects of higher parity and litter size (LS) on piglet birth weight (BW) and to compare the BW and survival rate of piglets born in different birth orders (BO). The study was conducted at a commercial pig farm in Thailand. Data from 230 sows and 3,500 piglets were analyzed including parity, litter size (LS), average birth weight (AvBW), coefficient of variation of piglets' BW (% CV), standard deviation of piglets' BW (SD), percentage of small piglets (% Small), individual piglet's BW, piglet birth order (BO; 1-10, 11-16, 17-25), sex and characteristics at birth (mummified fetuses (MM), stillborn piglets (SB) and born alive piglets (BAL)). Results revealed that the reduction in AvBW was prominent when LS increased (p < 0.001) and the expansion of BW variability (% CV, SD, % Small) was significant in the higher parity sows with large LS (p < 0.001). A difference in piglet BW was found among different BO; the later born piglets in BO 17-25 were lighter than the piglets born in BO 1-10 and 11-16 (p < 0.001). The percentage of BAL steadily decreased concomitantly with the rising of BO (p < 0.001), whereas the born dead piglets including MM and SB increased significantly (p < 0.001). This study demonstrated that higher parity and LS had negative effects on piglet BW including the reduction in mean BW with a greater variability. Moreover, the later born piglets in large litters had a lower survival rate.
ObjectiveTo establish baseline normal scotopic electroretinograpic (ERG) parameters for two wild cat species: fishing cats (FC) and leopard cats (LC).Animal studiedTwelve normal, FC and eight LC kept in the Chiang Mai Night Safari Zoo, Thailand. The mean ages of FC and LC were 7.08 and 5.00 years, respectively.ProcedureAll animals were studied using a standard scotopic protocol of a portable, handheld, multi-species electroretinography (HMsERG).ResultsThere were significant differences in the means of ERG b-wave amplitude of the rod response (Rod, 0.01 cd.s/m(2)), a- and b-wave amplitudes of standard light intensity of rod and cone response (Std R&C, 3 cd.s/m(2)) and b-wave amplitude of high light intensity of rod and cone response (Hi-int R&C, 10 cd.s/m(2)) with LC having higher amplitudes than FC. There was no significant difference in a- and b- wave implicit time except for the b-wave of Hi-int (P=0.03). No significant differences were observed in b/a amplitude ratios.ConclusionsData from this report provides reference values for scotopic ERG measurements in these two wild cat species. It showed that the normal scotopic ERG responses have some differences between the two species which might be due to the skull conformation, eye size or physiology of the retina.
The aim of this study was to investigate the effect of altrenogest (ALT) treatment on follicular development and ovulation in sows. Sows (parity 6-9; D0: weaning day) were assigned to four treatments as follows: control (no ALT treatment; n = 20), ALT-4-0 (ALT 20 mg/day, 5 days; D-4-D0; n = 20), ALT-4-2 (ALT 20 mg/day, 7 days; D-4-D2; n = 23), and 2ALT-4-2 (ALT 20 mg twice a day [40 mg/day], 7 days; D-4-D2; n = 24). The sows in each group were further divided into three groups based on slaughter periods: day 1 (DF1) and day 4 (DF4) of the follicular phase, and after the initiation of estrus. The number of follicles was reduced on DF4 compared to DF1 (P < 0.001). On DF1, follicular size (FS) of the control was larger than that of ALT-4-0, ALT-4-2, and 2ALT-4-2, but on DF4, FS of ALT-4-2 and 2ALT-4-2 was larger than that of the control and ALT-4-0 (P < 0.001). The coefficient of variation of FS on DF4 was smaller in ALT-4-2 compared to the control and 2ALT-4-2 (P < 0.05). Ovulation rate was similar between groups (P >= 0.05). ALT improved follicular growth and the homogeneity of FS without affecting ovulation.
The effect of altrenogest (ALT) feeding combined with induced ovulation by human chorionic gonadotropin (hCG) in sows was evaluated on piglet birth weight (BW) variation and pre-weaning performance. Sows were divided into four groups: the control (no ALT; without hCG induction; artificial inseminated (AI) at 12 and 36 h after estrus; n = 40), ALT + hCG72 (ALT 20 mg/d, D-4–D2 (D0: weaning day); hCG 750 IU at 72 h post ALT; AI at 24 and 40 h after hCG; n = 41), ALT + hCG96 (ALT 20 mg/d, D-4–D2; hCG 750 IU at 96 h post ALT; AI at 24 and 40 h after hCG; n = 41) and ALT + no hCG (20 mg/d, D-4–D2; without hCG induction; AI at 12 and 36 h after estrus; n = 41). The results revealed that piglet BW was not different among the groups (p > 0.05). However, the standard deviation of piglet BW (SDBW) was lower in ALT + hCG72 (0.32 ± 0.02 kg; p = 0.032), compared to ALT + hCG96 (0.40 ± 0.02 kg) and ALT + no hCG (0.40 ± 0.02 kg), except for the control (0.39 ± 0.02 kg). In addition, the pre-weaning mortality rate (%PWM) due to underweight elimination at weaning (below 3.50 kg) was decreased in ALT + hCG72 (8.33%) compared to the control (32.50%; p = 0.007) but similar to ALT + hCG96 (10.71%) and ALT + no hCG (24.05%). Therefore, ALT + hCG72 treatment in sows could reduce piglet BW variation and the number of piglets eliminated at weaning.
The duration of ejaculatory suppression of alpha adrenoceptor antagonist (tamsulosin, TAM) was investigated in goats. Five males were injected intramuscularly with TAM 80 μg/kg (179.8 nmol/kg). Semen was collected using an artificial vagina with estrous female goats and the semen quality and libido score were evaluated at 1, 3, 6 and 9 h post TAM injection. Two replicates were carried out in the study. Heart rates were also measured before the injection and at 30 min before semen collection. The results showed that the libido score was not affected by TAM. At 1 h post injection, the highest percentage of suppressed ejaculation (80%) was recorded and this effect was maintained for up to 3 h post injection (60%). Thereafter the percentage of suppressed ejaculation was significantly decreased (p < 0.05) at 6 h (20%) and ejaculation was completely recovered at 9 h post injection. The semen characteristics of the collectable semen, semen volume and number of spermatozoa were lowest at 1 h and 3 h, thereafter they suddenly increased at 6 h and the significantly highest peak was noted at 9 h post injection. TAM injection suppressed goat ejaculation within 1 h and the duration was maintained up to 3 h post injection.
There is little information on the endocrinology of fishing cats (Prionailurus viverinus), an endangered species in Southeast Asia, especially that pertaining to adrenal function. This study characterized faecal glucocorticoid metabolites in female fishing cats housed at Chiang Mai Night Safari to investigate seasonal and age relationships in hormone patterns. Faecal samples were collected 3 days/week for 1 year from seven females ranging in age from 4.5 to 9.6 years. A corticosterone enzyme immunoassay was validated for fishing cats by showing increases (∼60%) in faecal glucocorticoid immunoactivity above pre-treatment baseline levels within 1-2 days after an adrenocorticotrophic hormone injection. Faecal glucocorticoid metabolite concentrations were not related to age (P > 0.05), but there was a seasonal effect, with concentrations being higher (P < 0.05) during the winter (1.54 ± 0.04 µg/g) and rainy season (1.43 ± 0.04 µg/g) compared with the summer (1.22 ± 0.05 µg/g). Significant relationships were found between faecal glucocorticoids and rainfall (positive) and day length (negative), but not a temperature-humidity index. This is the first study to assess adrenal steroidogenic activity in female fishing cats, and we found that glucocorticoid metabolite production was influenced by seasonal factors, but not by age. We conclude that weather patterns should be taken into consideration in future studies of glucocorticoid activity in this endangered species, especially those studies aimed at improving captive management to create self-sustaining and healthy populations.
The effects of seminal plasma (SP), derived from autologous, homologous and heterologous species (stallion, boar and dog) on chilled Asian elephant sperm quality, were determined. Semen was collected from eight males and samples with ≥30% motile spermatozoa were used in the study. Semen was diluted with Tris-glucose-egg yolk extender, supplemented with different SP types and preserved at 4°C for 48 hr. Experiment 1 (n = 31), showed that the presence of SP (autologous) helped to preserve sperm quality in terms of sperm motility and acrosome integrity (p < .05). Homologous SP did not result in better sperm quality than autologous SP. Heterologous SP from stallion provided higher sperm motility and velocities compared to autologous SP (p < .05). Experiment 2 (n = 14) determined the effect of different SP from four stallions. All stallion SP gave higher (p < .05) results for motile spermatozoa and sperm velocities than autologous SP. In conclusion, the presence of SP helps preserve Asian elephant sperm quality and stallion SP supports the motility of Asian elephant spermatozoa during cold storage.
The aims of this study were to determine the relation of glucose transporter 3 (GLUT3) expression on the spermatozoa membrane with the quality of semen in Asian elephants. Fresh semen samples were collected from 10 Asian elephants and classified according to the percentage of motile sperm into Group 1 (<= 20%; n = 4 males), Group 2 (>20% to 60%; n = 3 males), and Group 3 (>60%; n = 3 males). The immunocytochemistry results showed the presence of GLUT3 at the principal and end piece of the sperm tail. GLUT3 expressions in each group were significantly different (P < 0.05). The group with good sperm motility (Group 3) significantly (P < 0.05, R = 0.960) expressed higher GLUT3 (99.40 +/- 0.69%) than Group 1 (21.46 +/- 10.25%) and Group 2 (84.37 +/- 8.70%). Moreover, the percentage of live sperm of this group was also significantly highest (P < 0.001, R = 0.938) at 82.00 +/- 4.00% compared to Group 1 (20.75 +/- 3.30%) and Group 2 (53.00 +/- 8.02%). Therefore, the motility of Asian elephant spermatozoa and percentage of live sperm may be influenced by GLUT3 expression, and GLUT3 expression may be involved in energy production via the glycolytic pathway.
Fecal samples of 14 fishing cats from Chiang Mai Night Safari were examined by simple floatation and simple sedimentation methods. These results show 42.85% was found for fluke egg and 7.14% for roundworm egg. The cause of infection in fishing cat is being unknown. Therefore, this data can be utilized for control and prevention of internal parasite infestation with fishing cats. Furthermore, these data can be utilized for management, for example: sanitation and water drainage to reduce the contamination in natural water and outbreak to the other animals.
The effect of homologous seminal plasma (SP) on chilled sperm kept at 4 C for 48 h was determined in Asian elephant. The homologous SP were obtained from semen with high (60 – 70%; SP1) and low (0 – 10%; SP2) sperm motility. Total of 18 semen samples from 7 males were studied. Each sample was divided into 3 groups depended on SP using – 1) autologous SP 2) SP1 and 3) SP2. The results found that there were no differences in the percentages of live sperm, sperm with normal acrosomal and with normal plasma membrane integrities among groups. But the percentages of motile sperm and motility velocities in Group 3 were lower (p<0.05) than those of Group 1 and 2, and no differences between Group 1 and 2. This study indicated that in Asian elephant, homologous SP could not give better results of semen quality than autologous SP during cold storage. Katila et al., 2004) สุกร (Caballero et al., 2004; Hernández et al., 2007) และแกะ (Rickard et al., 2014) จึงนาสนใจวา จะพบ ลักษณะดังกลาวในชางเอเชียหรือไม การศึกษานี้ จึงมีจุดประสงคเพื่อศึกษาวา SP จากชางเชือกอื่น สามารถชวย รักษาคุณภาพของอสุจิไดดีขึ้นหรือไม ในการเก็บรักษาน้ําเชื อ้แบบแชเย็น อุปกรณและวิธีการ ชาง ชางเอเชียตัวผู 7 เชือก อายปุระมาณ 20 – 35 ป เลี้ยงอยูที่ ศูนยอนุรักษชางไทย สถาบันคชบาลแหงชาต ิ องคการอุตสาหกรรมปาไม จ. ลําปาง ทําการรีดเก็บน้ําเชื้อระหวางเดือนตุลาคม 2555 ถึงเดือนกรกฏาคม 2556 โดยรีดเก็บเดือนละ 1 ครั้ง การรีดเก็บน้ําเชื้อและการประเมินคุณภาพน้ําเชื้อ การรีดเก็บน้ําเชื้อทําโดยวิธีนวดสวน ampullae ผานทางทวารหนัก (Schmitt and Hildebrandt, 1998) น้ําเชื้อที่ปนเปอนปสสาวะ จะไมนํามาศึกษา ทําการประเมินคุณภาพน้ําเชื้อ โดยวัดปริมาตร (มล.) รอยละของ อสุจิที่เคลื่อนที่ ความเปนกรด-ดาง (pH indicator stripes; Universalindikator, Merck, Germany) ความ เขมขนของอสุจิ (Neubauer hemocytometer; Freshman, 2002) รอยละของอสุจิที่มีรูปรางปกติและรอยละของ อสุจิที่มีชีวิต (eosin – nigrosin; Bjorndahl et al., 2003) รอยละของอสุจิที่มีเยื่อหุมเซลลปกติ (hypo-osmotic swelling test; Matson et al., 2009) และรอยละของอสุจิที่มีสวนอะโครโซมปกติ (Coomassie blue; Larson and Miller, 1999) โดยการตรวจอสุจิ ประเมินจากอสุจิจํานวน 200 ตัว การเตรียม SP จากชางเชือกอื่นสําหรับใชทดแทน SP เดมิ การเตรียม SP จะเตรียมจากน้ําเชื้อที่ไมมีการปนเปอนของปสสาวะ ตรวจดูรอยละของอสุจิที่มีการ เคลื่อนที่ แลวทําการปนเหวี่ยงน้ําเชื้อที่ 1600xg เปนเวลา 20 นาที และดูดเฉพาะสวน supernatant ซึ่งเปนสวน SP เก็บไวสําหรับใชในการทดลอง โดย SP ที่ไดนั้นแบงเปน 2 กลุมคือ SP จากน้ําเชื้อที่มีรอยละของอสุจิที่ เคลื่อนที่เทากับ 60 – 70 (SP1) และ SP จากน้ําเชื้อที่มีรอยละของอสุจิที่เคลื่อนที่เทากับ 0 – 10 (SP2) ทําการ เก็บรักษา SP ดังกลาวแบบแชแข็งที่ – 20 องศาเซลเซียส เปนเวลา 1 – 6 เดือน จนกวาจะนํามาใช การเก็บรักษาอสุจิแบบแชเย็น แตละตัวอยางน้ําเชื้อ แบงเปน 3 กลุม ดังนี้ กลุมที่ 1 น้ําเชื้อ 0.5 มล. ปนเหวี่ยงที่ 125xg เปนเวลา 10 นาที + สารเจือจางน้ําเชื้อ Tris-glucose-egg yolk (T; Michael et al., 2007) 0.5 มล. กลุมที่ 2 น้ําเชื้อ 0.5 มล. ปนเหวี่ยงที่ 125xg เปนเวลา 10 นาที และดูดสวน supernatant (SP เดิม) ออก + T 0.5 มล. + SP1 0.5 มล. และ กลุมที่ 3 น้ําเชื้อ 0.5 มล. ปนเหวี่ยงและดูดสวน SP เดิม ออก + T 0.5 มล. + SP2 0.5 มล. ทั้งนี้ในกลุมที่ 2 และ 3 ไมมีการเติม SP ของตัวเอง หลังจากนั้นเก็บรักษาแบบแชเย็นในตูควบคุมอุณหภูมิที่ 4 องศาเซลเซียส เปน เวลา 48 ชั่วโมง โดยทําการตรวจคุณภาพอสุจิที่ 24 และ 48 ชั่วโมงของการแชเย็น การประเมินการเคลื่อนที่ของอสุจ ิ การเคลื่อนที่ของอสุจิ ในสวนของน้ําเชื้อสดทั้งหมด และน้ําเชื้อที่ผานการแชเย็น 24 ชั่วโมงบางสวน จะ ประเมินโดยใชกลองจุลทรรศนแบบใชแสง (Light microscope) ที่กําลังขยาย 100 – 400 เทา ซึ่งทําการประเมิน ในพื้นที่ที่ปฏิบัติงาน น้ําเชื้อที่ผานการแชเย็น 24 ชั่วโมงบางสวนและที่ผานการแชเย็น 48 ชั่วโมงทั้งหมด จะ ประเมินในหองปฏิบัติการโดยใช Computer Assisted Sperm Analysis (CASA) (Hamilton Thorne Biosciences IVOS, Version 12 TOX IVOS, Beverly, USA) ซึ่งแสดงคารอยละของอสุจิที่เคลื่อนที่ (motility) ความเร็วของการเคลื่อนที่ในลักษณะ average path velocity (VAP; μm/s), straight line velocity (VSL; μm/s) และ curvilinear velocity (VCL; μm/s) โดยตั้งคาสําหรับการตรวจอสุจิของชางตามการศึกษากอนหนานี้ (สิทธิเดช และคณะ 2557) การวิเคราะหขอมูลทางสถิต ิ เปรียบเทียบคุณภาพของอสุจิระหวางกลุม ใชวิธี One-way repeated measures ANOVA และ Friedman Test กําหนดระดับนัยสําคัญที่ p < 0.05 ผลและวิจารณผลการทดลอง คุณภาพน้ําเชื้อสด คุณภาพน้ําเชื้อของชางตัวผูแตละเชือกแสดงใน Table 1 จากชางทั้งหมด 7 เชือก ไดตัวอยางน้ําเชื้อที่ไม มีการปนเปอนปสสาวะ จํานวน 18 ตัวอยาง โดยไดจากชางแตละเชือกตั้งแต 1 ถึง 6 ตัวอยาง คุณภาพน้ําเชื้อที่เก็บรักษาแบบแชเย็นที่ 24 ชั่วโมง หลังจากเก็บรักษาน้ําเชื้อที่ 4 องศาเซลเซียส เปนเวลา 24 ชั่วโมง พบวา ไมมีความแตกตางของคุณภาพ อสุจิในทุกคาที่ทําการประเมิน (Table 2) ขอมูลรายตัวซึ่งสามารถวิเคราะหไดทางสถิติ ไดแก E2, E7 และ E10 พบวา ไมมีความแตกตางของคุณภาพอสุจิในทุกคาที่ทําการประเมินเชนกัน (ขอมูลไมไดแสดง) คุณภาพน้ําเชื้อที่เก็บรักษาแบบแชเย็นที่ 48 ชั่วโมง ที่ 48 ชั่วโมงของการเก็บรักษาน้ําเชื้อแบบแชเย็น พบวา รอยละของอสุจิมีชีวิต อสุจิที่มี acrosome ปกติ และอสุจิที่มีเยื่อหุมเซลลปกตขิองทุกกลุมไม มีความแตกตางกัน แต รอยละของอสุจิที่เคลื่อนที่และความเร็วในการ เคลื่อนที่ของอสุจิแบบตางๆ ของกลุมที่ 1 และ 2 สูงกวาของกลุมที่ 3 อยางมีนัยสําคัญ (p <0.05) (Table 3) การวิเคราะหขอมูลรายตัวของ E2, E7 และ E10 พบวาใน E2 กลุมที่ 1 มีคารอยละของอสุจิที่เคลื่อนที่ สูงกวากลุมที่ 2 และ 3 อยางมีนัยสําคัญ (p = 0.022;
