The article is a brief literature review. This article provides an overview of the Quorum Sensing system in bacterial communities, highlighting the peculiarities of the system for gram-positive and gramnegative microorganisms. Basic information about the three existing Quorum Sensing systems is presented. Information is also given about different types of autoinducers, which are signaling molecules that trigger a cascade of behavioral reactions. The importance of the Quorum Sensing system as one of the fundamental mechanisms in the formation and regulation of bacterial biofilms is described, emphasizing the significance of biofilm microorganisms for modern clinical medicine and their impact on aggravating the issue of antibiotic resistance. The main mechanisms of inhibiting bacterial quorum, including by other microorganisms, are presented. The work discusses enzymatic and non-enzymatic methods of inhibiting the Quorum Sensing system, points of application and mechanisms of action. Some microorganisms with confirmed enzymatic activity by Quorum Quenching are indicated. Also presented are registered cases of suppression of other bacteria by microorganisms through the Quorum Sensing inhibitors system.
Экспериментально изучено влияние пробиотических препаратов в составе корма на рост, развитие и поведенческие реакции молоди рыб разных видов: стерляди, гибрида белуги и стерляди, чешуйчатого карпа - в условиях установок с замкнутым водообеспечением. Корма, содержащие пробиотические добавки, способствовали интенсивному росту и высокой выживаемости рыб (до 100 %). В ходе эксперимента в группах «опыт» отмечен наибольший прирост массы по показателям абсолютного и среднесуточного прироста, что говорит об эффективности использования кормов. Кормовой коэффициент в варианте «опыт» для карпа оказался ниже по сравнению с контролем на 1.25 единицы, или на 21 %. Исследование двигательной активности молоди рыб после кормления экспериментальными кормами выявило положительные изменения в их поведении. Молодь из группы «опыт» лучше адаптировалась к новым условиям среды и проявляла более высокую устойчивость к стрессовым раздражителям. Таким образом, полученные результаты подчеркивают потенциальные преимущества использования пробиотиков в составе кормов для улучшения роста и здоровья молоди рыб.
В настоящее время происходит бурный рост и развитие отечественного кормопроизводства, появляются новые рецептуры комбинированных кормов. В связи с этим растет необходимость в качественной оценке эффективности различных добавок, входящих в состав этих кормов, и для этого необходима надежная модель для оценки состояния рыбы. Проанализировано изменение морфологических и физиологических признаков, а также состояния микробиома у стерляди, потреблявшей корма различного качества, для определения физиологических показателей, наиболее подверженных влиянию состава пищи. Стерлядь, получавшая корм ненадлежащего качества, в среднем обладала на 7 % меньшей конечной массой, кровь рыб характеризовалась значительным падением уровня триглицеридов и β-липопротеидов в конце эксперимента. Результаты анализа кишечного микробиома показали, что у стерляди, получавшей некачественный корм, в кишечнике увеличивалось количество Enterococcus и актиномицетов, а также появлялось значительное количество представителей лактозоположительных бактерий, которые отсутствовали у особей из контрольной группы. Кроме того, у рыб, получавших некачественный корм, но остававшихся в хорошем физиологическом состоянии, в кишечнике были обнаружены бактерии Bacillus , отсутствовавшие у других групп, что может говорить о потенциальной пробиотической активности бактерий рода Bacillus, выделенных из их микробиома.
The oil cakes of essential oil plants were screened for prebiotic properties for further development of prebiotic feeds for livestock animals based on these essential oil plants’ extracts. For screening, a microbiota model of the chicken cecum, which was created on the basis of an artificial intestinal medium, was used. This model renders it possible to simulate conditions close to intestinal ones. Oligofructose, inulin, β-glucan, psyllium seeds, and apple fiber at a concentration of 0.5% were used as substances with known prebiotic properties. The oil cake concentration was 0.5%, 1%, 2%, and 4%. The direct antimicrobial effect of the cakes on intestinal bacteria was also investigated. The ability of the cakes to stimulate a decrease in the pH level by Lactobacillus was studied under conditions close to intestinal ones. It was shown that the cakes of big seed false flax (Camelina sativa), brown mustard (Brassica juncea), and spicate lavender (Lavandula angustifolia) exhibit prebiotic properties in relation to the microbiome of chickens in model experiments. They enhance the acid-forming properties of lactic acid bacteria, thereby lowering the pH of the medium. This leads to a decrease in the number of Enterococcus, Escherichia coli, and lactose-positive bacteria, as well as a complete suppression of Proteus. The optimal oil cake concentrations are 1% and 2%. These oil cakes are promising sources of prebiotics for the development of prebiotic feed for agriculture.
It is believed that the main goal of aquaculture is to provide the growing humanity with nutritious food. Meanwhile, modern technologies of crop production, microbial synthesis and food biotechnology make it possible to achieve this goal in less costly ways. The quality of plant protein from a number of sources is as good as that of fish, shellfish and arthropods. Unsaturated fatty acids and the most valuable vitamins in aquaculture products are of microbial origin. Thus, the real purpose of commercial aquaculture is to satisfy people's need for gourmet food. From an economic point of view, this means that the luxury fragment of the market is the most promising.
The ecological state of Lake Sevan, the largest drinking water reservoir for the South Caucasus, formed under the influence of climatic and social changes. This study assesses the bacteriological quality of water in the rivers of the Lake Sevan basin and tetracycline-resistant bacteria isolated from fish and people living near the rivers of the Lake Sevan basin in Armenia in autumn 2019 and spring 2020. No differences have been shown for the tetracycline resistance of the investigated E. coli isolated from the human gut and the Masrik, Argichi, and Gavaraget Rivers. Horizontal gel electrophoresis revealed the same plasmid bands in most of the investigated E. coli with the same tetracycline resistance from the different sources of the Argichi River (obtained from people/fish/water sources where the fish were caught). The results also showed that most of the waters carried Edwardsiella spp., Erwinia spp., Morganella spp., and Proteus spp. in addition to E. coli; the coliform index did not exceed the standard level of 5 × 104 CFU mL-1 there. These findings highlight the importance of multidisciplinary studies of bacteria from "interacting" ecosystems, which might serve as a basis for the suggestion of microbial antibiotic resistance as another indicator of water pollution.
The poultry industry is generating a significant amount of waste from chicken droppings that are abundant in microbes as well as macro- and micronutrients suitable for manure. It has the potential to improve the microbial activity and nutrient dynamics in the soil, ultimately improving soil fertility. The present study aimed to investigate the effect of chicken droppings manure (CDM) on the diversity of the soil microbiome in the free walking chicken’s area located in Stefanidar, Rostov Region, Russia. The data obtained were compared with 16 s rRNA from control samples located not far from the chicken's free-walking area, but not in direct contact with the droppings. Effect of CDM on the physicochemical characteristics of the soil and changes in its microbial diversity were assessed by employing the metagenomic approaches and 16 s rRNA-based taxonomic assessment. The alpha and beta diversity indices revealed that the application of the CDM significantly improved the soil microbial diversity. The 16S taxonomical analysis confirmed Proteobacteria, Actinobacteria, Bacteroidetes, Firmicutes, and Planctomycetes as abundant bacterial phylum. It also revealed the increase in the total number of the individual operational taxonomic unit (OTU) species, a qualitative indicator of the rich microbial community. The alpha diversity confirmed that the significant species richness of the soil is associated with the CDM treatment. The increased OTUs represent the qualitative indicator of a community that has been studied up to the depth of 5–20 cm of the CDM treatment range. These findings suggested that CDM-mediated microbial richness are believed to confer the cycling of carbon, nitrogen, and sulfur, along with key soil enzymes such as dehydrogenases and catalase carbohydrate-active enzymes. Hence, the application of CDM could improve soil fertility by nutrient cycling caused by changes in soil microbial dynamics, and it could also be a cost-effective sustainable means of improving soil health.
Fish as model objects have found wide applications in biology and fundamental medicine and allow studies of behavioral and physiological responses to various environmental factors. Representatives of the genus Nothobranchius are one of the most convenient objects for such studies. Male fish belonging to the family Nothobranchiidae are characterized by extremely diverse coloration, which constantly changes, depending on the age of the fish, environmental factors, and social hierarchical status. These fish species are characterized by a short life cycle, which allows changes in coloration, an indicator of the ontogenesis stage, to be estimated. Existing methods of fish color assessments do not allow the intensity of coloration of particular body zones to be clearly differentiated. In the present study, we suggest a method of two-factor assessment of specific fish body zones using modified methods of photofixation and image processing software. We describe the protocol of the method and the results of its application to different-aged groups of male Nothobranchius guentheri. The coloration of selected areas (i.e., red spot on the gill cover (RSGC), black border on the caudal fin (BBCF), and white border on the dorsal fin (WBDF)) differed significantly according to the size and age of the fish (p < 0.05). The data obtained suggest that N. guentheri can be a model for studying aging by the intensity of body coloration in males.
Probiotics are known for their beneficial effects on poultry health and wellbeing. One promising strategy for discovering Bacillus probiotics is selecting strains from the microbiota of healthy chickens and subsequent screening for potential biological activity. In this study, we focused on three probiotic strains isolated from the gastrointestinal tract of chickens bred in different housing types. In addition to the previously reported poultry probiotic Bacillus subtilis KATMIRA1933, three strains with antimutagenic and antioxidant properties Bacillus subtilis KB16, Bacillus subtilis KB41, and Bacillus amyloliquefaciens KB54, were investigated. Their potential effects on broiler health, growth performance, and the immune system were evaluated in vivo . Two hundred newly hatched Cobb500 broiler chickens were randomly divided into five groups ( n = 40). Four groups received a standard diet supplemented with the studied bacilli for 42 days, and one group with no supplements was used as a control. Our data showed that all probiotics except Bacillus subtilis KATMIRA1933 colonized the intestines. Treatment with Bacillus subtilis KB54 showed a significant improvement in growth performance compared to other treated groups. When Bacillus subtilis KB41 and Bacillus amyloliquefaciens KB54 were applied, the most significant immune modulation was noticed through the promotion of IL-6 and IL-10. We concluded that Bacillus subtilis KB54 supplementation had the largest positive impact on broilers' health and growth performance.
Probiotics development for animal farming implies thorough testing of a vast variety of properties, including adhesion, toxicity, host cells signaling modulation, and immune effects. Being diverse, these properties are often tested individually and using separate biological models, with great emphasis on the host organism. Although being precise, this approach is cost-ineffective, limits the probiotics screening throughput and lacks informativeness due to the 'one model - one test - one property' principle. There is а solution coming from human-derived cells and in vitro systems, an extraordinary example of human models serving animal research. In the present review, we focus on the current outlooks of employing human-derived in vitro biological models in probiotics development for animal applications, examples of such studies and the analysis of concordance between these models and host-derived in vivo data. In our opinion, human-cells derived screening systems allow to test several probiotic properties at once with reasonable precision, great informativeness and less expenses and labor effort.
One of the main problems in the poultry industry is the search for a viable replacement for antibiotic growth promoters. This issue requires a “one health” approach because the uncontrolled use of antibiotics in poultry can lead to the development of antimicrobial resistance, which is a concern not only in animals, but for humans as well. One of the promising ways to overcome this challenge is found in probiotics due to their wide range of features and mechanisms of action for health promotion. Moreover, spore-forming probiotics are suitable for use in the poultry industry because of their unique ability, encapsulation, granting them protection from the harshest conditions and resulting in improved availability for hosts’ organisms. This review summarizes the information on gastrointestinal tract microbiota of poultry and their interaction with commensal and probiotic spore-forming bacteria. One of the most important topics of this review is the absence of uniformity in spore-forming probiotic trials in poultry. In our opinion, this problem can be solved by the creation of standards and checklists for these kinds of trials such as those used for pre-clinical and clinical trials in human medicine. Last but not least, this review covers problems and challenges related to spore-forming probiotic manufacturing.
In this study, the duckweed varieties Lemna minor, Spirodela polyrhiza, and a commercially processed duckweed food supplement were investigated as potential substrates for the propagation of two probiotic Bacillus strains, B. subtilis KATMIRA1933 and B. amyloliquefaciens B-1895. Both L. minor and S. polyrhiza were found to be suitable substrates for the propagation of both bacilli, with 8.47–9.48 Log CFU/g and 10.17–11.31 Log CFU/g after 24 and 48 h growth on the substrates, respectively. The commercial duckweed product was a less favorable substrate, with growth reaching a maximum of 7.89–8.91 CFU/g after 24 h with no further growth after 48 h. Growth and adherence of the bacilli to the three products were confirmed via electron microscopy. These strains have demonstrated health-promoting benefits for poultry and thereby have the potential to enhance duckweed as an animal feed through the process of fermentation. Duckweed has been shown to be a promising alternative resource for protein and has the opportunity to become a valuable resource in multiple industries as a potential means to increase sustainability, food security, and reduce environmental impact.
BACKGROUND Chicken manure waste has a wide range of organic substances and mineral elements. This enriched source has stimulated great scientific interest in finding cleaner and more environmentally benign nutrient recovery options. This study aimed to determine an effective and eco-friendly method (i.e. subcritical water extraction) for processing fresh poultry manure. RESULTS The high content of total organic carbon, including humic acids carbon and fulvic acids carbon, in extract was found to release under subcritical conditions. The organic compounds obtained by extraction with subcritical water correspond to humic acid in composition because of the presence in the sample of all the functional groups: polymer bonded by molecular hydrogen bond (3400 cm(-1)), the presence of CH(2)and CH(3)groups (2870 cm(-1)), the presence of carboxyl groups (1720 cm(-1)) and quinones (1640-1680 cm(-1)). The solid phase left over was characterized by a high content of organic carbon, phosphorus, potassium, and microelements. The maximum extraction of humic acid and fulvic acid carbon was found between 210 and 250 degrees C at a pressure of 50-60 atm, and the content was a maximum of 3647.2 x 10(-6)g kg(-1)at an extraction temperature of 250 degrees C. CONCLUSION Given the high content of humic acid found in the extracted medium, the proposed subcritical extraction opens up new opportunities for nutrients recovery in the poultry industry. The study aims at investigating the effect of preparations of two bacilli strains on laying hens and roosters. Preparations were based on the strains Bacillus subtilis KATMIRA1933 and Bacillus amyloliquefaciens B-1895. Several groups of roosters and hens received a preparation based on either strain, or a mixture of both, from the first day to the last day of poultry in production. These preparations improved egg production, quality of sperm production, quality/hatchery of eggs, and slowed down the reproductive aging of hens. These observations were confirmed by the mathematical model proposed herein. At the molecular level, the slowing down of aging was confirmed by a decrease in the amount of mitochondrial DNA damage. Monitoring the physiological parameters of the experimental and control groups of birds showed that live weight gain in all experimental groups was higher than in the control group, and the reproductive organs of hens were more developed. There was also an improvement in the biochemical parameters of blood, the quality of the sperm of roosters, the laying of laying hens, and the morphological and biochemical parameters of the eggs. One of the most significant results is an increase in egg fertilization and a decrease in embryo death during the first 7 days of incubation.
A promising approach for slowing down the rate of reproductive aging is the use of probiotic bacteria as a feed additive. In the current study was investigated the influence of the intake of a potential probiotic on the follicle content and expression of vitellogenin genes (vtg1, vtg2, vtg3) in aged hens. RNA was isolated from liver samples collected from 570-day-old laying hens and gene expression levels were measured using RT-PCR. Bacillus subtilis KATMIRA1933 supplementation had a positive effect on the number of formed follicles in hens and also triggered a significant increase in the relative expression levels of vtg1, vtg2, and vtg3. A Bacillus amyloliquefaciens B-1895 enriched diet or a combination of the two strains had a modest effect on both the number of follicles and the expression of vitellogenin genes. Additionally, the study demonstrates that vitellogenin mRNA expression levels can be considered as a biomarker in a convenient approach for analyzing the hen's egg-laying ability.
In the current study, we performed in vivo investigation of probiotic intake influence on nuclear and mitochondrial DNA damage of hens, using quantitative PCR techniques. The probiotic supplementation to the diet of Hisex Brown hens had no significant effect on the rate of telomere shortening. After prolonged probiotic intake (225 and 445 days), the 18–21% decrease in the mtDNA lesions was detected. Since avian mitochondrial DNA damage investigations are rare, the current study of the probiotic-enriched diet’s impact on the damage of the hen mitochondrial DNA is novel and highly important. The decrease of mtDNA damage is a beneficial property, which could positively affect the reproductive aging of hens. The positive impact of probiotic supplementation on hens’ performance traits such as hen-day egg production, egg weight and mass, and feed conversion ratio was observed.
Background Dental infections, like dental degradation and periodontal diseases, are the most widespread bacterial infections in humans.Due to non-life threatening nature and ubiquity, their significance in healthcare was minimized.Still, they burden the economics of healthcare and the patients.Material and methods The study of SkQ1 (10-(6'-plastoquinoyl) decyltriphenylphosphonium) antibacterial activity against Streptococcus mutans species and dental biofilm bacteria was conducted.Bactericidal and bacteriostatic effect of this substance in micromolar concentrations on pure cultures and in higher concentrations on dental plaque bacteria was established.The compound binding effect with high molecular components of human saliva was identified and quantitatively evaluated.Results The test results showed that acidic media or competing for cation-containing drugs weaken this effect.SkQ1 is potentially applicable for oral cavity infections treatment and its efficiency can be enhanced by the utilization of proper vehicles.Conclusions SkQ1 disturbs bacteria transmembrane potential, so it can be considered as an antibacterial agent that is effective against gram-positive bacteria and can increase gram-negative bacteria sensitivity to the compounds that they are often resistant to.
A model system based on the Escherichia coli MG1655 (pRecA-lux) Lux-biosensor was used to evaluate the ability of the fermentates of eight probiotic strains to reduce the SOS response stimulated by ciprofloxacin in bacteria and mutagenesis mediated by it. Preliminary attempts to estimate the chemical nature of active components of the fermentates were conducted.
One of the important components of the concept of aging-phenoptosis (programmed aging) is the notion of aging as an accelerator of evolution having the rank of subconcept. For many reasons, the main being the problematic experimental testing of evolutionary hypotheses, verification of the above-mentioned subconcept can be based primarily on analysis of the internal inconsistency of heuristic models and their correspondence to undisputedly observed facts. To illustrate the acceleration mechanism, and most importantly to structure the evolutionary process in communities that include naturally weakened individuals, V. P. Skulachev offered in 2003 a conceptual model that he later called a “fable about hares”. Despite its simplicity, this model has undoubted internal logic. The natural trend in the development of conceptual models is their translation into the language of mathematics. The purpose of the present work was to create a variation of the known multi-agent model “predator–prey” that would allow us to “see” how the presence in the prey population of naturally weakened (old) members stimulates the selection of individuals with traits whose adaptive potential is not devaluated with age. The model (http://homebear.ru/PD) was developed on the Java platform, version 6, NetBeans development environment 8.2. Statistical analysis and preparation of illustrative materials were carried out using environment R, version 3.4.1. The results of numerical experiments set using our model correspond in principle to the provisions of the heuristic model of Skulachev and, consequently, confirm the absence in it of logical contradictions.
The study aims at elucidating the effect of bacilli probiotic preparations on the physiology of laying hens and roosters. Probiotic formulations were prepared as soybean products fermented by Bacillus subtilis KATMIRA1933 and Bacillus amyloliquefaciens B-1895. In this study, groups of male and female chickens were used. These groups received a probiotic preparation based on either B. subtilis KATMIRA1933 or B. amyloliquefaciens B-1895, or of a mixture of strains, from the first day to the age of 39 weeks. These preparations positively affected egg production, quality of sperm production, and quality and hatchery of eggs. Considering the simplicity and cost effectiveness of the soy-based probiotic preparation, these formulations should be considered as advantageous in modern livestock production.
