The transcription factor ETV5 is widely expressed and regulates diverse genes including a whole string related to obesity and diabetes. ETV5 is involved in the regulation of glucose-induced insulin secretion from ?-cells and its pancreatic loss increases the severity of pancreatitis. On this account, we investigated whether ETV5 is regulated by nutritional states and whether it plays a role in proliferation and survival from cellular stress using rat INS-1E ?-cells and doxycycline-inducible shRNA knockdown of Etv5. Neither low or high glucose conditions nor serum deprivation had an effect on ETV5 expression in normal INS-1E ?-cells while streptozotocin and palmitate time- and dose-dependently induced its expression to significant levels after 24h. Etv5 knockdown did not change proliferation but significantly further enhanced streptozotocin- and palmitate-mediated loss of viability. In this context, Etv5 knockdown neither had an impact on the expression of the proliferative and anti-apoptotic proteins NUPR1 and RET nor on caspases-3 and -7 activation levels. Instead, Etv5 knockdown was associated with increased basal nitric oxide production and augmented caspase-independent cell death upon exposure to streptozotocin and palmitate. In conclusion, ETV5 seems to limit nitric oxide production and may thereby play a role in the protection of ?-cells from necrosis upon certain cellular stresses.
In several studies, a strong association of the T risk-allele of the rs 7903146 polymorphism within the TCF7L2 gene with diabetes mellitus type 2 (T2DM), and also gestational diabetes (GDM) has been demonstrated in various ethnicities. Whether this association is due to increased insulin resistance or impaired insulin secretion remains unclear.
MSC are of interest for cell therapy since they secrete tissue regenerative and immunomodulatory factors which restore the local microenvironment. Own previous results demonstrated protection of viability and p-Akt concentrations in ALX- and STZ-treated INS-1E rat insulinoma cells by cocultured human telomerase immortalized MSC (hMSC-TERT). Here we investigated the effects of hMSC-TERT on apoptosis in INS-1E.
Genome-wide association studies in humans display that obesity is associated with increased gene expression of the transcription factor E-twenty-six version 5 (ETV5). In mice, ETV5 knock out results in profoundly impaired glucose tolerance associated with reduced insulin exocytosis. This indicates that ETV5 is required for insulin secretion. Since ETV5 knock out mice also exhibit a reduced pancreatic beta cell mass, we here investigated the potential role of ETV5 for survival of beta cells during cellular stress by streptozotocin (STZ) and the free fatty acid palmitate (PA).
REG3beta is acutely upregulated in acinar cells during pancreatitis and has been shown to reduce tissue inflammation via induction of the anti-inflammatory protein NUPR1. Own previous data demonstrated that transgenic mice with beta cell-specific overexpression of NUPR1 are protected against inflammatory and obesity-related beta cell stress. Therefore, this study evaluated whether acinar cells affect viability of beta cells and whether this involves REG3beta and NUPR1.

Das Ziel dieser Studie war es, die Abgabegenauigkeiten von Patch-Pumpen (OmniPod® zweite Generation, Ypsomed, OP) mit Schlauchpumpen (MiniMed®640G, Medtronic, MM) systematisch zu vergleichen.
Introduction: Injury-activated MSC secrete tissue protective factors which contribute to re-establishment of the local microenvironment. We previously demonstrated that cocultured telomerase-immortalised human MSC (hMSC-TERT) promote survival of alloxan (ALX)- or streptozotocin-treated INS-1E beta cells via AKT signalling. hMSC-TERT further mediated proliferation via ERK1/2 signalling. Here we investigated whether ERK1/2 signalling contributes to survival during ALX-mediated beta cell stress.
Background and aim: Mesenchymal stem cells (MSCs) bare the ability to home to sites of injury where they act by secreting angiogenic- and immunomodulatory factors. Previously we demonstrated that human telomerase-immortalised MSCs (hMSC-TERT) migrate towards injured INS-1E ß-cells, where they preserve cellular viability. Here we identify macrophage migration inhibitory factor (MIF) secreted by injured ß-cells as chemotactic factor that supports migration of MSCs.
Introduction: We previously demonstrated that transgenic mice with beta cell-specific overexpression of Nupr1 (Nupr1-Tg) are resistant to multiple low-dose streptozotocin (STZ) and/or high fat diet (HFD). Preserved insulin secretion of Nupr-Tg islets in vivo was associated with reduced NF-kB activation and less lymphocyte infiltration. The pro-inflammatory cytokine IL-1beta is well known to promote beta cell apoptosis and to inhibit insulin secretion via activation of NF-kB. Therefore, we investigated the response of IL-1beta and its physiologic receptor antagonist IL-1RA in WT and Nupr1-Tg islets during diabetogenic stress in vitro.
Schwerwiegende diabetische Folgeerkrankungen sind eine große Bürde für Patienten mit autoimmunem Typ-1-Diabetes, denn auch moderne Insuline und intensivierte Insulintherapien können die feinregulierte Insulinantwort einer β-Zelle im endokrinen Pankreas nur bedingt nachahmen. β-Zell-Ersatz könnte diese Situation verbessern, jedoch sind die klinisch etablierten Verfahren der Pankreas- und Inseltransplantation aufgrund der nötigen Immunsuppression nur für sehr wenige Patienten sinnvoll. Zudem gibt es kaum geeignete Spenderorgane. Die Grundlagenforschung arbeitet daher intensiv daran, Stammzellen in vitro zu vermehren und in β-Zellen zu differenzieren. Letzteres gelingt bisher nur bedingt, sodass embryonale Stammzellen und ethisch unbedenkliche induzierte pluripotente Stammzellen nicht sicher in der Klinik eingesetzt werden können. Alternativ wurde erkannt, dass adulte mesenchymale Stammzellen bei Gewebeverletzung eine Vielzahl von angiogenen, immunmodulatorischen, antimikrobiellen und trophischen Faktoren sezernieren. Mesenchymale Stammzellen zeigen in Tiermodellen ein hohes regeneratives Potenzial. Bislang konnte aber nur die Induktion von Selbsttoleranz in die Klinik übertragen werden. Dieser Beitrag erläutert den aktuellen Stand der Forschung und die Probleme, die einem klinischen Einsatz von Stammzellen zur Therapie von Diabetes derzeit noch im Wege stehen.
Objective: We have recently reported that the insulin-biosynthesis-impairing MADD rs10501320 genetic polymorphism is associated with gestational diabetes (GDM) in a European population. Aim of our study was to investigate the effects of this polymorphism on metabolic phenotype and glucose homeostasis in the same population of women 8 – 12 years after pregnancy.
Introduction: The intracellular protein NUPR1 (alias p8) reduces tissue damage during acute pancreatitis, and its expression is enriched in pancreatic islets. Therefore, we hypothesized that NUPR1 may exert protective mechanisms in the endocrine pancreas under inflammatory diabetogenic stress. This was investigated in transgenic mice with β-cell-specific NUPR1 overexpression (Tg).
Background and aim: According to the so-called "nutrition overflow hypothesis", type 2 diabetes results from nutrition-induced local tissue inflammation leading to insulin resistance, insulin secretory dysfunction and finally pancreatic β-cell loss. Free fatty acids (FFA) acting through toll-like receptors are key mediators of this lipotoxic inflammatory response. We previously demonstrated that the intracellular protein NUPR1 (p8) exerts potent anti-inflammatory and anti-diabetogenic effects in β-cells in vivo. Here we investigated the anti-lipotoxic potential of NUPR1 ex vivo in pancreatic islets of transgenic mice with β-cell-specific Nupr1 overexpression under the control of the rat insulin 1 promoter (Nupr1-Tg).
Fragestellung: SNPs (single nucleotide polymorphisms) im MADD-Gen sind assoziiert mit defekter enzymatischer Konversion von Proinsulin zu Insulin und konsekutiv mit erhöhten präprandialen Glukosespiegeln. In einer chinesischen Population konnte gezeigt werden, dass diese mit einem erhöhten Risiko für das Auftreten eines Diabetes mellitus Typ 2 (DM 2) assoziiert sind. Ziel dieser Studie war die Untersuchung der Korrelation zwischen zwei SNPs (rs10501320 und rs10838687) im MADD Gen mit dem Auftreten von Gestationsdiabetes bei Frauen mit europäischer Abstammung.
Fragestellung: Träger von Polymorphismen im TCF7L2 Gen zeigen ein erhöhtes Risiko für die Entwicklung eines Diabetes mellitus Typ 2. Die größte Bedeutung wird derzeit dem Polymorphismus TCF7L2 rs7903146 zugeschrieben, welcher mit der Insulinsekretion und dem BMI korreliert – wichtigen Kenngrößen für die Entwicklung eines Gestationsdiabetes (GDM). Wir untersuchten deshalb die Bedeutung des TCF7L2 rs7903146 Risikoallels für die Entwicklung eines GDM in zwei unterschiedlichen Kohorten von Frauen in Deutschland und Griechenland.
Fragestellung: Humanes (h)KLF11 aus der Familie der Sp1/KLF-Transkriptionsfaktoren reguliert den humanen Insulinpromoter (hInsP). Allerdings beschrieben einige Studien KLF11 als Aktivator, während wir und andere Inhibition beobachteten. Vor diesem Hintergrund ist es interessant, dass KLF11 das humane Monoaminooxidase-B Gen in Abhängigkeit von rekrutierten Kofaktoren sowohl aktivieren als auch inhibieren kann. Daher untersuchten wir hier, ob KLF11 auch den hInsP in Abhängigkeit vom molekularen Kontext unterschiedlich regulieren kann und ob diese Regulation durch Interaktion mit dem p300-PDX1-Transaktivierungskomplex vermittelt wird.
Fragestellung: MSC migrieren in entzündete Gewebe und verringern bei systemischer Injektion den β-Zellverlust in Mäusen mit experimenteller Insulitis. Hierfür könnten immunmodulatorische Eigenschaften von MSC ursächlich sein. Eigene Vorarbeiten belegen zudem direkte und vom Immunsystem unabhängige, zelluäre Interaktion über humorale Faktoren. STZ-geschädigte β-Zellen induzieren Migration in kokultivierten MSC und aktivierte MSC wirken ihrerseits β-Zell-protektiv. Hier untersuchten wir, ob humorale Faktoren aus MSC β-Zellen vor Schädigung durch Zytokine oder Alloxan (ALX) schützen und ob diese Faktoren Proliferation in β-Zellen induzieren können.
Fragestellung: Eigene in vitro-Untersuchungen weisen p8 als glucoseabhängigen Mediator der Proliferation von β-Zellen mit anti-apoptotischen Eigenschaften aus. Unter zellulärem Stress durch die diabetogene Substanz Streptozotocin (STZ) wird p8 akut und transient hochreguliert und inhibiert die Apoptoseeffektoren Caspase 3 und 7. Zur Überprüfung der proliferativen und anti-apoptotischen Eigenschaften in vivo wurde ein Mausmodell mit β-Zell-spezifischer p8-Überexpression unter der Kontrolle des Ratteninsulinpromotors etabliert. Hier untersuchten wir die Auswirkung von Hochfettdiät (HFD) auf die β-Zellmasse und, um den zellulären Stress zu erhöhen, die Auswirkungen von HFD in Kombination mit Insulitis auf die Glucosetoleranz.
Fragestellung: MSC migrieren in entzündete Gewebe und verringern bei systemischer Injektion den ß-Zellverlust in STZ-diabetischen Mäusen. Der molekularen Mechanismen dieser MSC-vermittelten ß-Zell-protektiven Wirkung sind noch ungeklärt. Die Aufklärung wird durch die große Heterogenität primärer MSC Populationen erschwert.
