Objective To investigate the 6-year survival rate of patients with hypertrophiccardio-myopathy (HCM) due to gene mutations of β myosin heavy chain(MYH7) and cardiac myosin binding protein C(MYBPC3).Methods Gene mutations of MYH7 and MYBPC3 in 70 HCM patients were identified by sequencing during the prospective follow-up.Results The mean follow-up time was 5.8±1.8 years during which 14 patients died,of them,0 had MYH7 mutations and 4 had MYBPC3 mutations(P>0.05).Sudden death occurred in 7 patients with MYH7 mutations in the head while no sudden death occurred in those with MYBPC3 mutations(P<0.01).Conclusion MYH7 mutations and death occur in younger HCM patients with MYH7 mutations whose sudden death rate is high.The ventricular wall is thicker in HCM patients with MYH7 mutations in the head than in those with MYH7 mutations in the stem,which is easy to lead to heart failure.It is,thus,necessary to detect gene mutations in HCM patients.
Objective To assay zinc and copper ion content in hippocampus of senescence-acceler-ated mouse prone 8(SAMP8) and the influence of metallothionein 3(MT3) on it compared with MT1. Methods Sixty SAMP8 mice were randomly divided into dementia group, three groups of different MT3 concentrations, MT1 group and blank control group with 10 mice in each group. Ten SAMR1 mice were taken as normal control group. In the experimental groups, the adult SAMP8 mice were injected intraperitoneally with MT of different types and concentrations continuously. After 4 weeks, the mice were executed. Atomic absorption speetrophotometer was used to assay zinc and copper ion content in hippocampus and the dose-effect relationship of different MT3 concentrations was observed. Results The zinc ion content in hippocampus of SAMP8 was less than that in SAMR1 group, but there was no difference in the copper ion content. The zinc ion content increased by MT3 injection,and the medium concentration group had the best result,with statistically significant difference. There was no significant difference between the MT1 and MT3 groups. Conclusions The zinc ion content in hippocampus of SAMP8 was decreased. MT3 can transport zinc ion and increase the content of it in hippocampus of SAMP8,but when the content of zinc ion get balanced it does not rise with the increase in MT3 concentration. MT3 can keep the zinc ion content in balance and had no influence on copper ion content.
Objective To study the effect of Annexin V antibody labeledby CdTe nanocrystals in differentiation peripheral nerve fascicles.Methods Ten rats were divided into two groups randomly,5 each group.Excise sural nerve,sciatic nerve and its muscle branches after the rats killed,the excised nerve stumps were quick frozen and cross-sectioned(20 μm) with a cryostat microtome.The experimental group use Annexin V antibody labeled by CdTe nanocrystals as incubating liquid,the controlled use CdTe nanocrystals,then the specimens were incubated in 37℃ incubator.Taken out to observe under fluorescent microscop at 10 min,15 min,20 min and 30 min respectively.Results The fluorescence was not seen in controlled group.while in experimental group,different from the sensory branches of sciatic nerve and sural nerve,the muscle branches of sciatic nerve could not see the fluorescence,the fluorescence intensity at 10 min was not enough to show the nerve fascicles clearly,we could see the fascicles with red fluorescence clearly at 15 min,the fluorescence intensity at 20 min and 30 min had little difference with that at 15 min.Conclusion The Annexin V antibody labeled by CdTe nanocrystals can identify sensory nerve fascicles of rat rappidly,it is possible to provide a new method for clinical differentiation peripheral nerve nerve fascicles.
Objective To study the effect of chitosan/PLGA emulsive membrane on postoperative adhesions in chicken flexor tendon.Methods A total of 45 chickens were divided into 3 groups randomly,15 for each group.The chickens of group A enrolled as controls,the deep flexor tendon of the third and forth toes in zone Ⅱ were cut and sutured with a single 6-0 nylon stitch by a modified Kessler technique,encapsulated PLGA membrane and chitosan/PLGA emulsive membrane in groups B,C respectively.Five chickens from each group were sacrified at 2,4,6 weeks after operation.The results were evaluated grossly,histologically and biomechanically.Each operated tendon was compared to the corresponding tendon in the contralateral,nonoperated foot.This approach eliminated the effect of normal restricting mechanisms eg.vincula and mesothenon.All results were input into a computer and analyzed with multiple-range test.Significance level was 0.05.Results The gross appearance and biomechanical data of groups B,C were both superior to those of group A(P 0.05).The healing process of 3 groups did not appear any difference,but the white blood cells in group B were more apparent than groups A and C.Conclusion It is demonstrated that postoperative adhesion could be controlled by using PLGA membrane and chitosan/PLGA membrane,the inflammatory reaction of former is more significant.Thus it is an ideal approach for prevention of tendon adhesions by using chitosan/PLGA emulsive membrane.
目的 回顾分析外科治疗足跟慢性溃疡的临床资料、治疗方法及疗效. 方法 1997年5月~2006年12月,收治足跟慢性溃疡患者23例.男20例,女3例;年龄18~79岁,平均45.6岁.病程1个月~7年.良性溃疡18例,恶性溃疡5例.溃疡创面2.5 cm×1.5 cm~10.5 cm×7.0 cm.患者予以局部扩创或溃疡扩大切除后,采用足底内侧皮瓣修复7例,带足背皮神经的足背皮瓣修复1例,小腿前外侧逆行岛状皮瓣修复4例,腓肠神经营养血管皮瓣修复11例,切取皮瓣4.0 cm×2.5 cm~18.0 cm×9.0 cm.供区游离植皮覆盖.结果 术后创面均Ⅰ期愈合.20例皮瓣完全成活,1例出现静脉危象,予以蒂部缝线拆除后成活;2例皮瓣远端部分坏死,经换药后愈合.供区植皮均成活,切口Ⅰ期愈合.21例获随访3个月~2年,平均11个月.皮瓣质地及外观良好,足底内侧皮瓣及足背皮瓣感觉恢复良好,小腿前外侧逆行岛状皮瓣及腓肠神经营养血管皮瓣痛温觉部分恢复.患足可正常负重,足跟溃疡未见复发.结论 足跟慢性溃疡以预防为主,创面应早期手术行皮瓣修复,以防止溃疡复发.
我院在2006年收治在基层医院误诊的下尺桡关节半脱位2例,经手术复位内固定,术后前臂旋转功能恢复良好,现报告如下.
Aim: To explore the fast method of indentifing motor nerve and sensory nerve with micro laser raman spectrum. Methods: The experiment was carried out in the Key Laboratory of Excited States Physics in Chinese Academy of Science of Changchun between October 2004 and October 2005. Thirty-two New Zealand rabbits were killed by hemospasia, and the anterior and posterior root of spinal nerves (3-5 mm) in rabbits were exposed under the operating microscope, and then made into frozen transections with the thickness of 30 mm. After that, the frozen transection was put under the microscope to be observed with raman spectrum by focusing the laser on it. Results: 1 The reproducibility of micro laser raman spectrum in samples of the same kind was good, and the characteristic of light spectrum was basically similar; While in different kinds of samples of motor nerve and sensory nerve, the relative intensity and width of peak of spectral properties were significantly different. 2 The relative intensity of raman shaking peak of motor and sensory nerve in raman spectrum around the region of 1 088 cm -1, 1 276 cm -1, 1 439 cm -1, 1 579 cm -1 and 1 659 cm -1 were markedly different, and the peak value ratio in 1 276 cm -1 and 1 439 cm -1 indicated that there were differences in microstructure between motor nerve and sensory nerve. Ratio of 1.06 was taken as the standard, and that above 1.06 was sensory nerve, otherwise was motor nerve. 3 The predictive value, judging for the sensibility, specificity, masculine and negative of motor nerve was 87.5%, 93.8%, 93.3% and 88.2% rescpectively; And the predictive value, judging for the sensibility, specificity, masculine and negative of sensory nerve was 93.8%, 87.5%, 88.2% and 93.3% respectively. Conclusion: There are respective characteristic spectrum in the raman spectrum of motor nerve bundles and sensory nerve bundles, which are constant and reliable. The nature of myeloarchitectonics can be distinguished according to the differences of spectrum characteristic, which indicateds that micro laser raman spectrum is an effective method in identifying the nature of nerve bundles.
目的:探讨甲床上皮细胞体外培养及传代的方法。方法:实验于2004-10/2005-04在吉林大学第一医院中心实验室完成。①标本消毒后,切取甲床并剪碎。②2.5g/L胰蛋白酶+3g/L乙二胺四乙酸1∶1混合液,37℃下消化重复3次,获得细胞悬液。③离心两次,清洗细胞,加入无血清角化细胞培养液制成细胞悬液,计数后接种于25mm培养瓶中。④37℃,体积分数为0.05的CO2、饱和湿度的培养箱内培养,每2天换液1次。⑤至单层细胞铺满近培养瓶底的80%以上时进行传代。⑥第3代细胞检测项目:倒置显微镜下观察细胞生长情况并摄片;细胞计数,绘制生长曲线;鼠抗人角蛋白17抗体做免疫细胞化学。结果:①细胞生长情况:倒置显微镜观察接种后的细胞12h后,大部分贴壁,11d左右时细胞生长密度可达瓶底的70%~80%,呈铺路石样排列。②免疫细胞化学鉴定结果:培养的细胞70%以上特异性角蛋白17抗体染色阳性。③生长曲线和群体倍增时间:接种后一两天细胞数减少,自第3天起细胞数迅速增多,至9~12d达高峰。对数期细胞群体倍增时间为4.46d。结论:利用酶消化法成功培养出人甲床上皮细胞,所得细胞纯度高并能传代培养,为深入研究甲床细胞的病理生理及分子生物学奠定了基础。
周围神经中含有运动与感觉两种纤维成分,某个功能束又是以占相对优势的一种纤维为主要成分.在进行周围神经端端缝合或神经移植术时,要求术者必须了解神经干内的精细结构,特别是神经干内神经功能束的定位知识,以及清楚神经纤维的数量和比例,才有可能达到精确的选择性神经束支对位缝合,减少再生纤维迷走所致的无效再生,提高术后肢体功能的恢复率.
