Putative changes in E-cadherin and β-catenin during implantation in dogs are of interest to study, as they are relevant proteins for epithelial integrity. E-cadherin and β-catenin were immunolocalized in the canine endometrium during the estrous cycle and early pregnancy, using monoclonal antibodies. Both proteins were detected in all types of endometrial epithelia (surface epithelium [SE], superficial glandular, and deep glandular epithelia) at all stages of the estrous cycle and in early placental structures. E-cadherin depicted a gradient of intensity apparently being lowest in the SE to progressively increase toward the deepness of the endometrial glands, regardless of the stage of estrous cycle. The overall immunostaining was, however, weaker at diestrus. In pregnant samples, the trophoblast was conspicuously immunolabeled compared with the endometrial surface lining epithelium. In the latter, the cytoplasmic pattern predominated over the membrane-bound, as was also seen in the decidual cells of the placental labyrinth. In the early placenta, only trophoblast cells and lacunae retained membrane signals. β-Catenin membrane labeling appeared relatively constant throughout the cycle, although a tendency toward a decrease in intensity was detected at the secretory stages. In addition, a dislocation of the immunoreaction from membrane to the cytoplasm was observed in both the SE and the glandular epithelia at particular stages of the cycle. In early pregnancy, a loss of the membranous pattern was observed in the SE and labyrinth, but neither on trophoblast nor in lacunae. The results show the existence of a softening of the adherens junctional complex in progestagen-dominated stages favoring embryo-maternal interactions and endometrial invasion during canine implantation.
The donkey breed Asinina de Miranda, with fewer than 1000 breeding females, is in danger of extinction. The objectives of this study were to predict the progression of the breed under present management and identify determinants for survival, by means of a population viability analysis program, in order to suggest suitable management strategies. The simulation showed a high risk of extinction. The most critical factor for breed survival was the percentage of females breeding per year, but the actual percentage needed depended on the carrying capacity of the breed. Reducing female mortality and age at production of first offspring, assuring registration in the Studbook, and tracking the foals will significantly foster this donkey breed’s recovery and maintenance. The breed comprised a potentially reproductive population of 589 individuals; however, just 54.1% of the adult females registered in the Studbook ever foaled, and of these 62.7% foaled just once. The overall neonatal mortality for the first month of life was 8.92% and was lower in females (6.51%) than in males (12.0%) (P = 0.028). Neonatal mortality was unevenly distributed throughout the year, with lower mortality rates recorded in February–May and October–November, and higher mortality rates in June–September and again in December–January. The neonatal foal mortality rate was lower with females aged 5–15 years (8.06%) than those younger than 4 years (10.3%) or older than 16 years (14.1%) at foaling.
Most donkey and local horse breeds are vulnerable to extinction as mechanization of agriculture progress throughout the world. The present study analyzed the pedigree and herd records of the donkey Asinina de Miranda breed (RAM), identifying genealogical and human factors that may affect the breed genetic diversity in the future and suggesting suitable strategies to breed preservation, early on the conservation program. The breeding rate was very low, with a ratio of foaling/live animals of 0.23 (178/760). The estimated number of founders and ancestors contributing to the reference population was 128 and 121. The number of founder herds in the reference population was 64, with an effective number of founder herds for the reference population of 7.6. The mean age of herd owners was 65.50±0.884 years, with a negative association among the herd size and owner's age (P<0.001). In contrast, the size of the herd and the ownership of a male were both positively associated (P<0.001) with the herd number of in-born foals. Both the owners' age and the herd location (RAM home region v. dispersal region) were negatively associated with the foaling number (P<0.001). The main identified risk factors were: low breeding rates; low number of males and their unequal contribution to the genetic pool; unequal contribution of the herds to genetic pool; and advanced age of herd owners.
Milk somatic cell count is an indicator trait for mastitis resistance. Genetic parameters for somatic cell score in the Portuguese Holstein-Friesian population were estimated by modeling the pattern of genetic correlation over the first 3 lactations (days in milk) with a random regression model. Data records from the first 3 lactations were from the national database of the Portuguese Holstein Association herds. Heritability estimates ranged from 0.05 at the beginning of the lactation for the 3 lactations, to 0.07 at the end of the lactation period for the first and third lactations, to 0.09 for the second lactation. This increase in the heritability values was due to an increase in the genetic variance and a decrease in the residual variances. Genetic correlations evaluated for monthly time points were high (0.65 to 0.99) for all 3 lactations, whereas phenotypic correlations were much less than the genetic correlations (0.13 to 0.62).
A 81-year-old black hypertensive patient noted some respiratory distress, cough and dysphagia four days after starting lisinopril. Three days later, with worsening of symptoms, the patient went to the emergency department were angioedema was diagnosed. Shortly after, he went in respiratory and cardiac arrest and CPR was started. The airway was managed with an orotracheal tube and mechanical ventilation was started until full neurologic recover. Corticoid and antihistaminic therapy was also started. On the third ICU day went under self-extubation. Initially there was not any respiratory distress, but slowly on the next two days the patient developed a worsening thoracic pain, stridor with broncospam and subcutaneous emphysema, which led to promptly re-intubation. After a clean thoracic X-ray, a thoracic CT scan revealed anterior herniation of the left lung through 5 fractured ribs, with bilateral air dissection of the muscular plans. The patient underwent surgery, with hernia reduction and prosthesis placement on 3 of 5 broken ribs. This case report describes a rare but known side effect of a commonly used anti-hypertensive drug. The angioedema developed into respiratory arrest and required CPR. This maneuver was life saving, however led to lung herniation, a rare complication in a conscious and non ventilated patient. Traumatic intercostal hernias usually occur medial to the costochondral junction. Eighty percent of lung herniations are secondary to trauma or surgery and the remaining 20% are congenital defects. Lung herniation after CPR is a rare complication end although there are some cases reported, the real percentage is not clear. The usual management of a lung hernia is conservative. Surgery is reserved for large or incarcerated hernias, for those that are intractably and painful, or those that are cosmetically unacceptable. Surgical repair was undertaken in this case because of respiratory failure, incarceration of lung tissue, and pain localized to the defect.
Three hundred and thirteen Estrela mountain dogs were examined for hip dysplasia (HD) using the standard ventrodorsal hip extended view, and graded into five categories (A, B, C, D and E) using the Fédération Cynologique Internationale's (FCI) scoring system. The Ortolani method was performed to evaluate hip joint laxity. Pedigree information was obtained from the Portuguese Kennel Club and the genetic trend was evaluated by calculating the mean breeding values (BVs) for the last 15 years, using the threshold model. HD was found in 66% of the dogs. There was low-moderate correlation between the results of the Ortolani test and FCI hip scores (r(s)=0.386; P<0.001). Grades of hip dysplasia were equal in both males and females (P=0.14) and in the animals' right and left sides (P=0.51). The mean BVs for HD were stable in dogs born between 1991 and 2003, and showed an improvement in 2004 and 2005. The data confirm the high prevalence and severity of HD in predisposed breeds that do not have breeding programmes in place. It also confirms an initial favourable change in BVs that is a likely consequence of the voluntary radiographic hip-screening programme.
Most studies on lactation curves only consider milk yield and describe a standard lactation curve of dairy cows, showing a peak or maximum daily yield occurring between 4 and 8 weeks after calving, followed by a daily decrease in milk yield until the cow is dried off. Wood's model is a widely used lactation curve function. Wood's model was fitted to test-day records of 95,405 lactations of parities lower than 5. Milk traits were milk yield (MY), fat percentage (F%). protein percentage (P%), fat yield (FY) and protein yield (PY), and the lactation curve was individually considered as a cluster of five linked curves. Milk trait and parity influence the goodness of fit of Wood's model. In 19.3% of the lactations, the shape of the MY, FY and PY curves follows the standard lactation curve while F% and P% have the reversed standard shape. A series of in vitro experiments were completed to evaluate the potential of enzyme extracts, obtained from the white-rot fungi Trametes versicolor (TV1, TV2), Bjerkandera adusta (BA) and Fomes fomentarius (FF), to increase degradation of cell wall components of wheat straw. The studies were conducted as a completely randomized design and analysed using one-way ANOVA. Enzyme activities of the extracts, previously obtained from a liquid culture medium, were characterized in terms of laccase and peroxidase for ligninolytic activity. Carboxymethyl cellulase (CMCase) and avicell digesting cellulase (Avicelase) were used for cellulolytic enzyme assays. Wheat straw samples were incubated with enzyme extracts in a citrate buffer (pH 5.0) in a forced air oven at 25 °C for 6 days. In vitro NDF digestibility (IVNDFD), and the rate and extent of NDF fermentation, without and after incubation with the white-rot enzyme extracts, were determined using a gravimetric microbiological method and a gas production technique, respectively. Results from cell wall chemical composition showed that TV2 and BA enzyme extracts decreased NDF concentration (P<0.05) and that TV1 had higher activity (P<0.05) towards cellulose. There was an increase in IVNDFD (P<0.05), resulting from treatment of wheat straw with enzyme extracts from BA, TV1 and TV2, reaching a difference of 13% for TV2 (P<0.05), versus the non-treated straw control. Treatment with enzyme extract from TV2 caused increased gas production (P<0.05) after the first 20 h of incubation, and also increased the maximum rate of gas production, thus enhancing fermentation kinetics. This study indicates that enzyme extracts from white-rot fungi can be used to develop new approaches to overcome low digestibility of some plant cell walls. Utilization of different substrates to produce enzyme extracts can lead to production of viable ligninolytic complexes which could improve the nutritive value of fibrous feeds.
Overactive bladder syndrome (OBS) is described as urinary urgency with or without incontinence, usually with increased daytime frequency and nocturia in the absence of another identifiable pathological process. Nowadays and despite other alternative therapies, the mainstay of OBS is still the pharmacological approach, mainly with anti-muscarinic drugs. To compare the efficacy of a 30-day solifenacin succinate (5 mg OD) treatment with or without previous medication with trospium chloride, a prostective open, two-arm, parallel group study was conducted for 5 weeks in 40 patients with OBS. The primary endpoint was patient self-assessment of improvement after 30 days of medication. Secondary endpoints included the reduction of the daily number of voids and urgency or involuntary leakage episodes. Adverse reactions and therapeutic stoppage were also evaluated. To be included in the trospium chloride treatment group, patients were required to have been treated with such drug for 1 to 6 months before the present study. Evaluation and efficacy assessment were accomplished using a 3-day bladder diary and an urgency severity scale (USS). Safety assessment was done by recording all the patients' complaints after starting medication. A total of 40 patients were enrolled for this study, 19 without previous medication and 21 who had already tried trospium chloride. Two patients from the non-previous medication group were excluded. Globally, there was a statistically significant reduction for the USS (2.73-->1.73), the daily number of voids (9.5-->7.0), of urgency episodes (9.1-->4.0) and of involuntary leakage episodes (3.6-->1.0) over the 24 h. Six patients had no improvement, four from the previous trospium chloride group and two from the non-previous medication group. Three patients reported side effects, two cases of dry mouth and one case of constipation. One patient dropped out of the treatment due to an unspecified intolerance. Solifenacin succinate 5 mg seems to be effective concerning patients' self-assessment of improvement and decrease in the mean number of daily voids, urgency episodes and incontinence episodes. This was reported both in patients who have already been medicated with trospium chloride and those who have never taken any kind of medication. Regarding side effects, solifenacin is quite well-tolerated in both groups.
Genetic parameters, breeding values, and genetic trends of hip dysplasia in Estrela Mountain Dogs were estimated using a linear model (LM) and a threshold model (TM). A database with 313 animals was used. Right and left hip joints were individually scored, according to the Fédération Cynologic Internationale grading rules of the canine hip dysplasia system, as normal (1), borderline (2), slight (3), moderate (4), and severe (5 and 6). The estimate of repeatability was lower in LM (0.86) than in TM (0.90). The same tendency was verified with the heritability because its estimate in LM was 0.38 and in TM was 0.43. However, these results did not establish any statistical differences between the models. The genetic trend of canine hip dysplasia for LM and TM showed a similarity in shape, but considerable individual differences were found in the EBV ranking lists. Therefore, the selection of breeding animals would not be the same with the 2 methodologies. To select the best method for genetic evaluation of hip dysplasia, further studies using more data and other dog breeds are required.
Daily milk yield over the course of the lactation follows a curvilinear pattern, so a suitable function is required to model this curve. In this study, 7 functions (Wood, Wilmink, Ali and Schaeffer, cubic splines, and 3 Legendre polynomials) were used to model the lactation curve at the phenotypic level, using both daily observations and data from commonly used recording schemes. The number of observations per lactation varied from 4 to 11. Several criteria based on the analysis of the real error were used to compare models. The performance of models showed few discrepancies in the comparison criteria when daily or 4-weekly (with first test at days in milk 8) data by lactation were used. The performance of the Wood, Wilmink, and Ali and Schaeffer models were highly affected by the reduction of the sample dimension. The results of this work support the idea that the performance of these models depends on the sample properties but also shows considerable variation within the sampling groups.
A spline animal model was fitted to 152,103 test-day milk, fat, and protein yield records from 14,423 first-lactation cows. The models included age at calving and the herd-test-month as fixed effects. Model fitting was carried out using Restricted Maximum Likelihood with ASREML. For milk yield, the heritability at 18 d in milk was 0.19, which increased to the maximum estimated value of 0.23 at midlactation and then decreased. On average, milk, fat, and protein yield heritabilities were 0.22, 0.14, and 0.19, respectively. For milk yield, all correlations were positive and ranged from 0.54 to 0.99 for the genetic component and from 0.32 to 0.78 for the phenotypic component. Genetic correlations were higher than phenotypic ones. For fat and protein yields, all genetic correlations were positive, ranging from 0.43 to 0.99. The phenotypic correlations for fat yield had the lowest correlations of the 3 traits. Curves of estimated breeding values for milk, fat, and protein over lactation had positive deviations from mean curves for sires with high genetic merit, but there was considerable variability in the shapes of the curves for different sires. More research is needed to compare the spline function with other mathematical functions used as submodels of lactation curve.
INTRODUCTION:SAFYRE is a new readjustable and minimally invasive sling for the treatment of stress urinary incontinence (SUI). Attempts to restore the normal suburethral hammock using an anatomical approach have been made in recent years. The authors report their experience with this device, which associates the efficacy of slings with readjustability.MATERIAL AND METHODS:A total of 100 consecutive patients with clinical and urodynamic diagnosis of SUI underwent SAFYRE sling procedure. The age range was from 40 to 71, mean age 63 years. Seventy-five patients (75%) presented previous failed anti-incontinence procedures. Physical clinical examination, stress test, pad use and a urodynamic study were performed before the surgery. All the patients presented symptoms of SUI and 30% also reported mild urgency.RESULTS:The average follow up period was 14 months (12-30 months). The mean operative time was of 25 minutes. Dystopia repair was performed whenever necessary, during the same procedure. The average hospital stay was 24 hours. In 3% of the implants, bladder perforation occurred. During the postoperative period, 26 patients developed urgency symptoms. During that follow up period, 92% were found to be continent, 3% reported an improvement and 5% were dissatisfied.CONCLUSION:SAFYRE is a safe and quick procedure that allows for postoperative readjustment. This technique may be an attractive alternative if the good result obtained so far proves to be long lasting.
SAFYRE es un nuevo cabestrillo (dispositivo de suspensión) reajustable y mínimamente invasivo para el tratamiento de la incontinencia urinaria por estrés (IUE). En los últimos años se ha intentado restaurar el lecho suburetral normal utilizando una técnica anatómica. Los autores comunican su experiencia con este dispositivo, que además de poseer la eficacia de los cabestrillos tiene capacidad de reajustación. Un total de 100 pacientes consecutivas con diagnóstico de IUE desde el punto de vista clínico y urodinámico se sometieron al procedimiento de implantación del cabestrillo SAFYRE. Su rango de edad era de 40 a 71 años, con una edad media de 63 años. Previamente, 75 pacientes (75%) se habían sometido a procedimientos para la incontinencia que habían fracasado. Antes de la cirugía se realizaron una exploración física clínica, una prueba de estrés, un estudio urodinámico y un estudio sobre el uso de almohadillas. Todas las pacientes presentaban síntomas de IUE y el 30% también tenían tenesmo leve. El periodo de seguimiento medio fue de 14 meses (12 – 30 meses). El tiempo medio de la operación fue de 25 minutos. Durante el mismo procedimiento se reparó la distopia cuando se consideró necesario. La estancia hospitalaria media fue de 24 horas. En el 3% de los implantes se produjo perforación de la vejiga. Durante el periodo posoperatorio 26 pacientes presentaron síntomas de tenesmo. Durante ese periodo de seguimiento, el 92% presentaron continencia urinaria, el 3% mejoraron y el 5% no estuvieron satisfechas con el procedimiento. SAFYRE es un procedimiento seguro y rápido que permite el reajuste posoperatorio. Esta técnica puede ser una alternativa atractiva si se demuestra que los buenos resultados obtenidos hasta ahora son de larga duración.
