The type 1 abomasal ulcer (AU1) does not have specific clinical signs, so there is a need to identify some early biochemical markers for its diagnosis in cattle and buffaloes. The plasma gastrin is re-ported to reflect the gastric mucosa damage but its utility for the diagnosis of AU1 in buffaloes has not been evaluated. The objective of this study was to investigate the test performance of plasma gas-trin to distinguish between healthy buffaloes and the buffaloes with AU1. Twenty-three buffaloes with AU1 and six buffaloes without any abomasal ulcer were used. The blood samples were collected from buffaloes, slaughtered in a buffalo specific slaughterhouse for estimation of the plasma gastrin. After slaughter the abomasa were examined for the presence of AU1 and the blood samples were collected for estimation of plasma gastrin. The mean plasma gastrin concentration of the ulcer-positive buffaloes was significantly (P<0.05) higher than the ulcer-negative buffaloes. The receiver operating characteristic curve analysis suggested that the optimal value of plasma gastrin for the diag-nosis of AU1 was 106.2 pg/mL. This preliminary work suggests that plasma gastrin could be a valid diagnostic test for the detection of AU1 in the buffaloes. The sensitivity, specificity, positive predictive value and negative predictive value of the plasma gastrin to diagnose AU1 in the buffaloes were 78.3, 100, 100 and 69.9 pg/mL, respectively.
The hematological alterations of bovine reticular abscess are reported in literature, but the detailed biochemical alterations and long-term outcome of bovine reticular abscess have not been reported so far and are not available in the standard textbooks. This study aims to describe clinical features, hemato-biochemical alterations, management, productive and reproductive performance, long term follow-up of reticular abscess in bovines. This prospective study was conducted on 12 bovines diagnosed with reticular abscess. All the animals were subjected to hematological and biochemical evaluation along with a healthy control group. The diseased animals were subjected to radiography, ultrasonography, and surgical treatment. The clinical findings were chronic anorexia, dehydration, persistent or recurrent tympany, scanty feces, mushy rumen, tachypnea, gradual reduction in milk yield, distended rumen, and increased fecal particle size with undigested particles in feces. Although the presence of penetrating reticular foreign bodies seemed to be the major cause, the role of non-metallic foreign bodies cannot be ruled out. Ultrasonography was more reliable than radiography in the diagnosis of reticular abscess. The majority of the reticular abscesses were single and were located cranio-ventrally. White blood cell and neutrophil counts were significantly (p < 0.05) higher while lymphocyte count was significantly (p < 0.05) lower than the control values. The hematology findings can be used for tentative diagnosis of reticular abscess in bovines. Total bilirubin, aspartate aminotrasferase, total protein, globulin, fibrinogen, blood urea nitrogen, creatinine glucose, cholesterol, and lactate were significantly (p < 0.05) higher, and albumin, albumin-globulin ratio, fibrinogen ratio, potassium, chloride, and calcium were significantly (p < 0.05) lower than the corresponding control values. Rumen chloride concentration was higher than the reference range. Peritoneal fluid analysis in four cases revealed septic peritonitis. Reticular abscess can lead to vagal indigestion, usually posterior functional stenosis. The conservative treatment was unsuccessful. The long-time survival rate in surgically treated animals was 66.7
This prospective study describes the occurrence, morphology and localisation of type 1 abomasal ulcers (AU1) in various diseases of buffaloes and cattle. The carcasses were examined to confirm the cause of death. The abomasa were examined for AU1 and their characteristics. The AU1 were catego-rised into four subtypes, 1a, 1b, 1c and 1d, as per standard procedure. Traumatic reticuloperitoni-tis/pericarditis, reticular diaphragmatic hernia, intestinal obstruction, peritonitis, bronchopneumonia and theileriosis were the common causes for AU1. The overall prevalence of AU1 was 62.9%, which did not differ significantly with species and age. The prevalence of acute ulcers (1a and 1b) was sig-nificantly higher than that of chronic ulcers (1c and 1d). Most AU1 were located in the caudal third of abomasal body on parietal surface along the greater curvature. Most of the 1a ulcers were located in the pylorus, while 1b, 1c and 1d were located in the abomasal body. The overall prevalence of AU1 was lower (P<0.05) in the fundus than in other anatomical regions of the abomasum. Type 1b ulcers were more numerous than other subtypes. It was concluded that AU1 may be an important cause of slow recovery/poor prognosis under clinical situations and hence, the therapy protocol for such cases should include treatment for probable gastrointestinal bleeding.
Background:The clinical findings, laboratory alterations, and prognosis of primary type 3 abomasal ulcer (AU3) are poorly reported in the literature.Aims:To describe clinical findings, hemato-biochemical changes, and peritoneal fluid changes in bovines suffering from primary AU3, and to monitor responses to medical treatment and outcomes.Methods:The study included 32 bovines (20 cattle and 12 buffaloes) diagnosed with primary AU3 along with a control group.Results:Common clinical findings were depressed demeanor, anorexia, dehydration, scanty feces, melena, mushy atonic rumen, tachycardia, and tachypnea. Colic was observed in 56.3% of animals. The mean hemoglobin, hematocrit, platelet count, and lymphocyte count were lower (P≤0.05), while WBC and neutrophil count were higher than the values of the control group (P≤0.05). The levels of BHBA, NEFA, glucose, total bilirubin, AST, CK, LDH, BUN, creatinine, and lactate were higher (P≤0.05), while cholesterol, total protein, albumin, sodium, potassium, chloride, and calcium were lower than the values of the control group (P≤0.05). The rumen chloride concentration was increased. The left shift was observed in a higher percentage of nonsurvivors than survivors (P≤0.05). The nonsurvivors had higher levels of bilirubin, CK, LDH, BUN, creatinine, and rumen chloride (P≤0.05), and lower levels of total protein, albumin, and globulin (P≤0.05).Conclusion:Type 3 abomasal ulcers occurred during the various stages of lactation as well as in pregnant animals. The response to medical treatment was fair, long time survival rate was good, and there was no recurrence. There was no effect on fetal survival or milk yield in the subsequent lactation.
Background: The present study aimed to describe clinical findings, hemato-biochemical changes, response to medical treatment, and outcomes for buffaloes suffering from peritonitis. Another objective of this study was to determine the prognosis of the disease. Methods: The study was conducted on 25 buffaloes suffering from peritonitis and presented to the Large Animal Hospital of Guru Angad Dev Veterinary and Animal Science University, Ludhiana, Punjab, India. The buffaloes were subjected to detailed clinical examination. Hematological, biochemical, and peritoneal fluid analysis was carried out along with treatment. Results: Peritonitis was more prevalent in pregnant buffaloes, being septic peritonitis in 60% of cases. The important clinical findings were depression, anorexia, reduced water intake, scanty feces, dehydration, and absence of rumen motility. The hematological findings were hemoconcentration, left shift, and toxic changes in neutrophils. Biochemical analysis revealed increased total bilirubin, AST, ALP, GGT, glucose, triglycerides, BUN, creatinine, lactate, fibrinogen, and rumen chloride, whereas albumin, fibrinogen ratio, potassium, chloride, calcium, and phosphorus were decreased. Abnormal peritoneal fluid changes were altered physical parameters and the presence of degenerated neutrophils, bacteria, and gut contents. Nucleated cell count and total peritoneal fluid protein were not reliable indicators of peritonitis. The absence of rumen motility, marked left shift, toxic changes in neutrophils, higher BUN, lower potassium (≤3.6 mmol/L), and unfavorable peritoneal fluid changes were the negative prognostic signs. Treatment with broad-spectrum antibiotics and supportive therapy led to a recovery in about 50 percent of cases, with diffuse peritonitis cases being unresponsive. Long time survival rate was good, and there was no recurrence. Conclusion: The prognosis of peritonitis in buffaloes has to be precisely assessed on the basis of clinical, hemato-biochemical, and peritoneal fluid alterations. Standard classification of transudate and exudate did not apply in the majority of buffaloes with peritonitis.
Aim: Goldfish (Carassius auratus L.), a freshwater fish belonging to the family Cyprinidae, is an important ornamental fish species in the world. Herewith, we report a case of systemic mycobacteriosis along with concurrent fibroma in goldfish. Methodology: The protruding mass and internal organs exhibiting lesions were collected for histopathology and molecular diagnosis. The sections of cutaneous mass were stained with haematoxylin and eosin (H&E) and Masson trichrome, whereas sections from spleen as well as kidney were stained with H&E and Ziehl-Neelsen. For identifying the etiological agent, 16S rRNA gene fragment was amplified and sequenced, using genus-specific primers for Mycobacterium sp. Results: In histopathology, multiple granulomas could be observed in kidney and spleen sections. Importantly, typical acid-fast bacilli were demonstrated in granulomas in spleen. The sequence of 16S rRNA gene fragment amplified from spleen and kidney showed maximum similarity to Mycobacterium stomatepiae. Besides, the histopathological characteristics of the protruding mass were consistent with cutaneous fibroma. Interpretation: These findings assume significance as several Mycobacterium species, including the current one from ornamental fish are reported to be of zoonotic significance.
This prospective study was conducted on 42 buffaloes diagnosed with reticular diaphragmatic hernia (RDH). The common clinical findings were depressed demeanor, chronic inappetance or anorexia, dehydration, recurrent or persistent tympany, scanty faeces, mushy rumen with normal or increased motility, tachypnea, abdominal distension, and gradual reduction in milk yield. The presence of reticular foreign bodies seemed to be the major predisposing factor. Neutrophil and lymphocyte counts were significantly (p < 0.05) higher and lower than the control values. Total bilirubin, AST, BUN, creatinine glucose, lactate, and fibrinogen were significantly (p < 0.05) higher, and total protein, albumin, albumin-globulin ratio, triglycerides, fibrinogen ratio, sodium chloride, calcium, and phosphorus were significantly (p < 0.05) lower than the control values. The haemoglobin, packed cell volume (PCV), white blood count (WBC), alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), cholesterol, and magnesium did not differ significantly (p < 0.05) from control values. RDH can lead to vagal indigestion, and rumen motility will depend upon the stage of vagal indigestion and presence of the reticular adhesions. Out of 26 buffaloes that underwent laparorumenotomy and herniorrhaphy, 14 died within a week, three died after 3–4 weeks, nine recovered within 1–2 weeks, and survived upto full follow-up period of 2 years. The non-survivors had significantly (p < 0.05) higher age, number of calvings, WBC and neutrophil count, PCV, and GGT than the survivors. The hernial ring size ≥ 12.5 cm, with or without reticular adhesions, was associated with poor prognosis. Long time survival rate was 34.6%. It was concluded that the preoperative haematology and rumenotomy findings can be used to predict the prognosis of RDH in buffaloes.
Aims:The present study was carried out for comparative evaluation of case-based learning (CBL) aided with WhatsApp and didactic lectures (DL) while teaching a pathology topic to second-year medical students. In addition, the acceptability of WhatsApp as an aid to CBL was assessed. Material and Methods:After obtaining informed consent, 70 second-year Bachelor of Medicine and Bachelor of Surgery (MBBS) students were exposed to six sessions of CBL aided by case scenarios for one set of topics of anemia posted on WhatsApp groups. This was followed by six sessions of DL for separate set of topics in anemia. The multiple-choice questions (MCQ) test scores obtained pre and postintervention, of CBL and DL sessions, were compared to paired t-test (within the groups) and Student's t-test (between the groups). Categorical data were analyzed using Chi-square (χ2) test. Student's self-administered questionnaires and focus group discussions (FGDs) were used to collect student perceptions and analyzed quantitatively, as well as qualitatively. Results:The mean MCQ scores obtained postintervention in CBL topics were significantly higher compared to DL (22.78 ± 2.99 vs 17.78 ± 3.35; P < 0.001). Students perceived that CBL enhanced their curiosity; hence, the acquired knowledge through various resources was retained better. It enhanced their analytical skills and interest in learning pathology. In FGDs, the students appreciated the use of WhatsApp as an aid to CBL for its ease of sharing scenario-related additional information and prior discussions among themselves in chat groups at their convenience. Conclusion:CBL aided by WhatsApp helped students acquire knowledge, discuss and learn actively, score more, and retain better than DL. Using WhatsApp as a platform helped them to interact at their ease and seek guidance from their mentors without resistance and hesitation.
Bovine tropical theileriosis caused by Theileria annulata is an overwhelming haemoprotozoan tick-borne disease in taurine and cross-bred cattle in Punjab, India. However, there seems to be no report from India of cutaneous nodules associated with the disease. This report describes a five-year-old cross-bred cow presented to a university clinic with a history of fever, inappetence and malaise for the past six to seven days. Clinical examination revealed normal vital parameters, pale mucous membranes, mild enlargement of the prescapular lymph nodes and multiple subcutaneous nodular masses (2-4 cm) on the neck and abdomen. Haematology revealed mild anaemia and leucopenia with 48% neutrophils, 48% lymphocytes and 4% eosinophils. Romanowsky-stained smears of fineneedle aspiration biopsy samples from swollen lymph nodes and subcutaneous masses showed an increased number of lymphoid cells, suggesting cutaneous lymphomatosis. However, a critical examination of the smears from subcutaneous nodules showed a large number of Koch's blue bodies in macrophages and lymphoblasts, and several piroplasms were also noticed within the red blood cells in lymph node smears. A peripheral blood smear revealed mild to moderate parasitaemia. Extracted DNA from the parasitologically positive blood sample was subjected to nested polymerase chain reaction (nPCR) using T. annulata speciesspecific primers encoding the 30-kiloDalton major sporozoite surface antigen. The desired 572-base pair amplified product of the nPCR was comparable to the positive control. This seems to be a rare case of T. annulata in an adult cross-bred cow, showing cutaneous nodular involvement.
Primary nephrotic syndrome encompasses a wide range of histopathological diagnoses with varied incidence, presentations, treatment options, response and outcomes based on geographical distribution, age, sex and the histopathological diagnosis. Our aim was to study the outcomes of primary nephrotic syndrome in our patient population.
Heteropneustes fossilis (Bloch) is an important candidate species for diversification of freshwater aquaculture in India. However, high mortality rate during larval rearing is the most serious bottleneck in commercial production of this species. A proper understanding of the ontogenic development of digestive system provides the basis to understand the nutritional physiology of larvae and develop appropriate feeding strategies. In the present study, the ontogenical development of the digestive tract in H. fossilis larvae was studied from hatching until 30 day post-hatching (dph) at 29 °C. At hatching (2.8 ± 0.2 mm standard length, SL), the digestive tract was undifferentiated and attached dorsally to the yolk sac. At 1 dph (2.9 ± 0.2 mm SL), the mouth opened and oral valves were visible. At 2 dph (3.0 ± 0.3 mm SL), goblet cells were observed in the buccoparyngaeal cavity. At this age, exogenous feeding started and the intestine was differentiated into the anterior and posterior regions, and the rudimentary liver and pancreas were also seen. Small supranuclear vacuoles were observed in the enterocytes of the posterior intestine at 2 dph. Zymogen granules were observed in acinar cells of pancreas by 3 dph, and islets of Langerhans were visible at 4 dph (3.5 ± 0.1 mm SL). At the same age, most of the yolk sac reserves were consumed, whereas they were completely exhausted by 5 dph (3.9 ± 0.5 mm SL). Between 4 and 6 dph, the liver elongated in size and started to accumulate lipids in the hepatocytes. Gastric glands were detected at 4 dph, and the pyloric sphincter was completely differentiated at 9 dph (6.1 ± 0.4 mm SL) as an epithelial fold that separated stomach from the anterior intestine. By 13 dph (8.6 ± 0.2 mm SL), profuse gastric glands were visible inside longitudinal mucosal folds of the stomach. The formation of gastric glands and their development were noticed as the last events in the development of the digestive tract in H. fossilis. This indicated the end of the larval period and the commencement of the juvenile stage. Considering these observations, it is suggested that H. fossilis larvae have a morphologically complete digestive tract by 13 dph. The findings of the study on the development of the digestive system in H. fossilis may help in synchronising the larval stage of development and feeding strategies and would be helpful in improving larval rearing techniques for catfish species.
Aim: Immunostimulation, before the fish is able to mount an effective immune response, can induce tolerance. Therefore, present study was carried out to investigate the structural development of lymphoid organs, namely thymus, kidney and spleen of Indian major carp, Catla (Catla catla), which will help in deciding the appropriate age, when immunocompetence is established. Methodology: In this study, ontogeny of the lymphoid organs of catla was studied from hatching up to 54 days post hatch (dph). For this, whole fish were fixed, processed and embedded in paraffin wax. Further, serial sections were cut, stained and observed under microscope. Results: The thymus anlage appeared in the dorso-anterior part of the gill cavity at 1 dph. At 5 dph, an undifferentiated mass of stem cells was observed and by 8 dph, it appeared lymphoid. At 11 dph, clear dark and light zones of cells were observed. At 26 dph, Hassall's corpuscles were observed and no further changes were observed except increase in number of lymphoid cells till end of the sampling period. Kidney was observed as an undifferentiated mass of stem cells at 1 dph with few renal tubules. At 5 dph, undifferentiated stem cells increased in number, and by 8 dph, few erythropoietic cells were observed. At 11 dph, lymphoid cells were present and by 17 dph, renal tubules started degenerating. At 26 dph, anterior kidney completely lacked excretory tissue and appeared lymphoid. Spleen was observed at 3 dph as an undifferentiated mass of stem cells, and at 8 dph, erythrocytes were seen. At 11 dph, spleen was heavily packed with erythrocytes, whereas at 20 dph, the number of erythrocytes decreased and lymphocytes were observed. Fully developed spleen was observed at 33 dph with presence of red and white pulp. Interpretation: Thymus, head kidney and spleen were fully developed on 33 dph. The information generated in the present study would be helpful in deciding the appropriate age for vaccination
India is the second largest fish producing country in the world with production of 12.6 million tonnes (mt) in 2017–18, and Indian major carps (IMCs) contribute bulk of this fish production. Catla, Catla catla is the fastest growing species among IMCs. However, the survival rate of catla during larval rearing is normally lower than the other IMCs i.e rohu Labeo rohita, and mrigal Cirrhinus mrigala. Continuous efforts are devoted for the identification of nutritional and environmental requirements of fish larvae in order to reduce hatchery mortalities. However, very little information is available regarding physiology of the immune system, especially during the late larval and juvenile stages. Hence, understanding the ontogenetic development of immune-relevant genes in the larval stages of catla will serve as the markers for the development of immune competence and thereby, will be beneficial in developing effective immune intervention strategies. In the present study, expression profiles of some of the important innate (IL-1β, IL-10, iNOS and C3) and adaptive immune (RAG-1, Ikaros, IgM and IgZ) genes during ontogenetic developmental stages and in different tissues of catla were investigated using quantitative real-time PCR. The results revealed that immune genes IL-1β, C3, IgM and IgZ were detected in the unfertilized eggs indicating their maternal inheritance. Immune genes, IL-1β, IL-10 and iNOS were expressed significantly during initial larval developmental stages whereas C3, RAG-1, Ikaros, IgM and IgZ showed significant expression during advanced stages of larval development in catla i.e. from 23 days post hatch (dph). Study of tissue distribution pattern of the genes indicated that innate immune genes were ubiquitously expressed in different tissues with varying degree of expression, whereas adaptive immune genes were predominantly expressed in lymphoid organs of catla. The information thus generated will improve knowledge on the maturation of the immune system in catla and will aid in deciding the appropriate age for vaccination in this teleost species.
The present study was conducted on four cattle and one buffalo diagnosed with intestinal obstruction. Hematological analysis showed increase in total leukocyte and neutrophil counts, and decrease in lymphocyte count. Left shift or toxic changes in neutrophils was a consistent finding. The biochemical alterations were deranged liver function tests, increased C-reactive protein, glucose and lactate, and decreased chloride, potassium calcium and phosphorus levels. The peritoneal fluid changes were increased total cell count and number of neutrophils along with higher ALP and lactate concentrations. In conclusion, bovine intestinal obstruction causes detectable changes not only in blood but also in peritoneal fluid.

Fifteen apparently healthy male buffalo calves aged between 6 months to one year with body weight range of 70 - 140 Kg divided into 3 groups of 5 buffalo calves each were used in the present investigation. Endotoxic shock was produced by IV infusion of Escherichia coli endotoxin @ 5 μg/kg BW/hr for 3 hours followed by administration of three different treatment regimens comprising of intravenous infusion of hypertonic saline solution(HSS) @ 4 ml/Kg bw, flunixin meglumine @ 1.1 mg/Kg bw and blood @ 20 ml/Kg bw to group-I, HSS, Dextran-40 and Flunixin meglumine to group-II and HSS, Dextran-40, whole blood and Flunixin meglumine to group-III with the objectives to study the major physio-pathological changes during induced endotoxemia in buffalo calves and to compare the effects of different treatment options to find out the best treatment option out of the three combinations. All the animals were further observed up to day 2 or death whichever was earlier. Endotoxin infusion to all the animals caused symptoms of restlessness, respiratory distress, snoring, diarrhoea, profuse salivation along with the significant hypoproteinemia, hypoalbuminemia, hypoglobulinemia, hypokalemia and hypocalcemia. The treatment with HSS, flunixin meglumine and blood infused as one time infusion to these endotoxemic buffalo calves not only alleviated the above mentioned symptoms but also significantly raised the circulating albumin level at 5.5 hrs and day 2 and Fibrinogen level at day 2 of observation. A significant hypocalcaemia was observed at 4.5 hours along with an increase in Alkaline Phosphatase at 3.5, 4.5 and 6.5 hrs. All the endotoxemic buffalo calves which died were subjected to post mortem and histopathological studies. Epicardial and endocardial haemorrhages, haemorrhages on intestinal mucosa, congestion, haemorrhages, emphysema and fibrinous thrombi in microvasculature of lungs were salient histopathological findings. On comparison with observations of the other treatment regimens tried, it was found that the IV infusion of the combination of HSS, Flunixin meglumine, Dextran-40 and blood was found to be the most effective leading to full recovery of 3 out of 5 buffalo calves of group-III where as only 1 out of 5 animals recovered in group-II. None of the group-I animals recovered and all of these succumbed to shock after day-2.
Epitheliocystis, an emerging aquaculture disease, has been reported in > 90 fish species from 14 countries. In the present study, we report epitheliocystis from cultured rohu Labeo rohita, an Indian major carp, which is the major cultured species in the Indian subcontinent. The affected fish exhibited lethargy, swimming near the water surface followed by mortality. In wet mounts of gills, infestation with dactylogyrids, trichodinids and myxosporeans was observed. Subsequently, following euthanasia, the affected fish were dissected and subjected to routine bacteriological, virological and histopathological investigations. The bacteria could not be isolated from internal organs, namely kidney and liver. In addition, cytopathic effects were not observed in the Cyprinus carpio koi fin (CCKF) cell line following inoculation of filtered tissue homogenate from pooled kidney samples. In histology of gill sections, amorphous intracytoplasmic inclusions typical of epitheliocystis were observed in epithelial cells, and the presence of chlamydial DNA in gills was confirmed by amplification of 16S rRNA gene sequence of Chlamydiales. The sequencing of amplicons (774 bp) revealed that the epitheliocystis agents from rohu belong to family Candidatus Parilichlamydiaceae in the order Chlamydiales. Since the sequences of epitheliocystis agent from rohu showed < 95% similarity with any member of the genera in above family, therefore, these agents appear to be novel, and it is proposed that these may be known as Candidatus Panilichalamydia rohitae. Using in-situ hybridisation, the 16S rRNA gene amplicons labelled with digoxigenin, linked to the epitheliocysts. The present study adds to the growing range of host species affected by epitheliocystis.
Columnaris disease, caused by Flavobacterium columnare, is one of the important bacterial diseases responsible for large-scale mortalities in numerous freshwater fishes globally. This disease can cause up to 100% mortality within 24 h of infection and is considered to be a cause of concern for aquaculture industry. Despite being a serious disease, scarce information is available regarding host-pathogen interaction, particularly the modulation of different immune genes in response to F. columnare infection. Therefore, in the present study, an attempt has been made to study expression of important immune regulatory genes, namely IL-1β, iNOS, INF-γ, IL-10, TGF-β, C3, MHC-I and MHC-II in gills and kidney of Catla catla following experimental infection with F. columnare. The expression analysis of immune genes revealed that transcript levels of IL-1β, iNOS, IL-10, TGF-β, C3 and MHC-I were significantly up-regulated (p < 0.05) in both the organs of the infected catla. IFN-γ and MHC-II were up-regulated in gills of infected catla whereas, both the genes showed down-regulation in kidney. The results indicate that important immune genes of C. catla are modulated following infection with F. columnare. The knowledge thus generated will strengthen the understanding of molecular pathogenesis of F. columnare in Indian major carp C. catla.
