Currently, the performance of sea ice thickness (SIT) data retrieved from multisource satellite products in the Arctic seasonal ice zones remains unclear. This study presented the spatiotemporal intercomparison and evaluation of satellite data, including the latest versions of Soil Moisture and Ocean Salinity (SMOS), CryoSat-2, combined CryoSat-2 and SMOS (CS2SMOS), and Ice, Cloud, and Land Elevation Satellite-2 (ICESat-2), specifically focusing on area with mean SIT below 0.5 m. Five evaluation datasets were used. During 2010–2023, SMOS had the smallest mean SIT, with CryoSat-2 showing the largest mean SIT. During 2018–2023, with the inclusion of ICESat-2, SMOS still showed the smallest mean SIT. CryoSat-2 exhibited the largest mean SIT, followed by ICESat-2, CS2SMOS ranked third. Evaluation results indicated that four satellite products generally underestimated SIT. In two periods, SMOS consistently exhibited the weakest performance, which showed a large gap from what was expected in previous studies. In contrast, CS2SMOS demonstrated the highest alignment with five evaluation datasets during 2010–2023, indicating the best overall performance. During 2018–2023, ICESat-2 exhibited the best overall performance with two evaluation datasets. This study refreshes previous knowledge about SMOS in the seasonal ice zones and contributes to further improvements in SIT retrieval.
The Canadian Arctic Archipelago (CAA) includes the critical region of the Northwest Passage (NWP) and is one of the areas with the most severe sea ice conditions in the Arctic. Currently, studies on sea ice projections focusing on the CAA are limited. Furthermore, the prediction results for the CAA from different models based on the Coupled Model Intercomparison Project Phase 6 (CMIP6) exhibit uncertainty due to the inter-model spread. This study evaluated the projected data for sea ice concentration (SIC) and thickness (SIT) within the CAA from 14 CMIP6 models for the period 2015-2022 under the SSP2-4.5 scenario based on the satellite and reanalysis data. Although most models can capture the major characteristics of spatiotemporal variations in SIC and SIT within the CAA, there are considerable numerical differences compared to observations and reanalysis. Additionally, there is a notable spread among 14 CMIP6 models. The assessment of SIC indicates that CESM2, GFDL-CM4, IPSL-CM6A-LR, and UKESM1-0-LL exhibit better performance, with a relatively low bias (less than 7%), a root mean square error (RMSE) below 22%, and a relatively high correlation coefficient (CC) exceeding 0.75. In the evaluation of SIT, the four best-performing models are GFDL-CM4, MPI-ESM1-2-HR, MPI-ESM1-2-LR, and MRI-ESM2-0. The multi-model ensemble of the best performance group (MMMBPG) projects a declining trend in both sea ice area (SIA) and SIT for the CAA from 2025 to 2100, with respective trends of -0.21 x 105 km2 per decade and -0.06 m per decade under the SSP2-4.5 scenario, and -0.56 x 105 km2 per decade and -0.11 m per decade under the SSP5-8.5 scenario. Under both scenarios, the MMMBPG predicts a more notable reduction of sea ice in the NWP compared to the multi-model ensemble of all 14 models. Navigable conditions for the northern and southern routes are defined by SIA and SIT respectively, with each route's SIA being less than 5% of its total area and the mean SIT being below 0.15 m. Based on the SIA threshold, under the SSP2-4.5 and SSP5-8.5 scenarios, MMMBPG projects that continuous annual navigability in the NWP, with at least three navigable months per year, will be achieved starting in 2060 and 2054, respectively. According to the SIT threshold, the MMMBPG projects that it will reach continuous annual navigability with at least one navigable month per year under both scenarios, starting in 2038 and 2036, respectively. This study enhances the understanding of CMIP6 model performance in projecting sea ice within the CAA and provides insights into future sea ice conditions in the region.
Currently, Arctic sea ice thickness (SIT) data with extensive spatiotemporal coverage primarily comes from satellite observations, including CryoSat-2, Soil Moisture and Ocean Salinity (SMOS), and the Ice, Cloud, and Land Elevation Satellite-2 (ICESat-2). The studies of the intercomparison and evaluation of multi-source satellite products in recent years are limited. In this study, three latest version products of ICESat-2, CryoSat-2, and CS2SMOS (a merged product of CryoSat-2 and SMOS) were examined from October to April, between 2018 and 2022. Three types of observation including airborne data from the Operation IceBridge (OIB) and IceBird, and in situ data from Beaufort Gyre Exploration Project (BGEP) are selected as the reference in the evaluation. The intercomparison results show that the mean SIT is generally largest in ICESat-2, second largest in CryoSat-2, and smallest in CS2SMOS. The SIT in CryoSat-2 is closer to the SIT in ICESat-2. The thickness displayed by the three satellite products starts to increase at different freezing months, varying between October and November. The three satellite products demonstrated the strongest agreements in SIT in the Beaufort Sea and Central Arctic regions, and exhibited the most distinct differences in the Barents Sea. In the evaluation with OIB data, three satellite-derived SIT were generally underestimated and CS2SMOS demonstrates the closest match. The evaluation using IceBird data indicates an underestimation for all satellites, with CryoSat-2 showing the best agreement. In the assessment with BGEP data, ICESat-2 displayed a more pronounced degree of overestimation or underestimation compared to the other two satellites, and CS2SMOS exhibited the optimal agreement. Based on the comprehensive consideration, CS2SMOS demonstrated the best performance with the airborne and in situ observational data, followed by CryoSat-2 and ICESat-2. The intercomparison and evaluation results of satellite products can contribute to a further understanding of the accuracies and uncertainties of the latest version SIT retrieval and the appropriate selection and utilization of satellite products.
Objective Glucocorticoid-induced osteoporosis (GIOP) represents a major complication arising from the long-term use of glucocorticoids, which are widely prescribed for various inflammatory and autoimmune conditions. Despite its prevalence, the current therapeutic options for GIOP are limited in terms of efficacy, safety profiles, and patient compliance. The Modified Danggui Buxue Decoction (DGBXD), a traditional Chinese herbal formulation, has shown promise in preliminary studies for its potential osteoprotective effects. The present study aimed to explore the mechanistic underpinnings of DGBXD’s action on GIOP using network pharmacology and molecular docking approaches, bridging traditional medicine with modern pharmacological insights. Method Network pharmacology is applied to screen drug-active compounds and potential core target proteins for disease treatment and to explore the drugs’ therapeutic mechanisms. Result Altogether, 78 DGBXD active compounds and 223 DGBXD-related, 146 component-disease common, and 2,168 GIOP-associated target genes were obtained. The PPI network had 43 nodes and 462 edges, and a total of 10 core target genes, including TP53, JUN and MAPK3, were identified. The results of the GO enrichment analysis implied that DGBXD might participate in biological activities, including responses to oxidative stress and nutrient levels. The outcomes of the KEGG pathway enrichment analysis showed that DGBXD may treat GIOP through TNF, IL-17, and phosphatidylinositol 3-kinase (PI3K)-Akt signaling pathways. Based on to the molecular docking results, biologically active compounds (beta-carotene, formononetin, luteolin, and isorhamnetin) exhibited good binding to AKT1 and ESR1. Conclusion DGBXD may aid in GIOP treatment by modulating multiple therapeutic targets and signaling pathways.
The accuracy and reliability of the latest version of multisource satellite derived Arctic sea ice thickness (SIT) in thinner ice regions are currently uncertain. This study integrated a comprehensive comparison and assessment of Arctic SIT derived from CryoSat-2, Soil Moisture and Ocean Salinity (SMOS), fusions of CryoSat-2 and SMOS (CS2SMOS), and Ice, Cloud, and Land Elevation Satellite-2 (ICESat-2) during 2011–2022. The focus was on the region with mean SIT less than 1 m. The five datasets from the Operation IceBridge (OIB) L4 and Quick Look, IceBird campaign, CryoSat Validation Experiment (CryoVEx), and the Canadian Arctic Archipelago (CAA) stations were utilized as the reference data in the assessment. The four satellite products could capture similar major spatiotemporal variations in SIT. During 2011–2022, CryoSat-2 generally derived the largest multi-year mean SIT, followed by CS2SMOS, and SMOS exhibited the smallest mean. During 2018–2022, ICESat-2 recorded the largest mean SIT and the rankings of the other three satellite products remained consistent. The comparison and assessment results indicated that all four satellite products generally exhibited some underestimations of SIT. During 2011–2022, the comprehensive results highlighted CryoSat-2 as the best overall performance product, exhibiting optimal agreement with all five reference datasets. During 2018–2022, CryoSat-2 consistently demonstrated the best overall performance. CS2SMOS exhibited a performance similar to CryoSat-2 in the two selected periods. This study contributes to further understandings of reliabilities and potential disparities among the latest versions of multisource satellite products in the thinner ice region.
Bone marrow mesenchymal stem cell (BMSC) chondrogenic differentiation contributes to the treatment of osteoarthritis (OA). Numerous studies have indicated that microRNAs (miRNAs) regulate the pathogenesis and development of multiple disorders, including OA. Nevertheless, the role of miR-20a-5p in OA remains obscure. Forty male C57BL/6 mice were divided into four groups and were surgically induced OA or underwent sham surgery in the presence or absence of miR-20a-5p. Flow cytometry was implemented to detect surface markers of BMSCs. Reverse transcription quantitative polymerase chain reaction revealed the upregulation of miR-20a-5p during BMSC chondrogenic differentiation. Western blotting displayed that miR-20a-5p inhibition decreased protein levels of cartilage matrix markers but enhanced those of catabolic and hypertrophic chondrocyte markers in BMSCs. Alcian blue staining, hematoxylin‑eosin staining and micro-CT revealed that miR-20a-5p inhibition restrained chondrogenic differentiation and miR-20a-5p overexpression promoted the repair of damaged cartilage and subchondral bone, respectively. Luciferase reporter assay identified that mitogen activated protein kinase kinase kinase 2 (Map3k2) was a target of miR-20a-5p in BMSCs. Moreover, the expression of miR-20a-5p and Map3k2 was negatively correlated in murine cartilage tissues. Knocking down Map3k2 could rescue the suppressive influence of miR-20a-5p inhibition on chondrogenic differentiation of BMSCs. In conclusion, miR-20a-5p facilitates BMSC chondrogenic differentiation and contributes to cartilage repair in OA by suppressing Map3k2.
Background: Young adult men who have sex with men (YMSM) are at increased risk for HIV, especially minority YMSM. Pre-exposure prophylaxis (PrEP) is a breakthrough daily pill to prevent HIV. Consistent adherence is key to PrEP effectiveness, which is why the CDC recommends adherence support be provided to all PrEP patients. Mobile health can overcome barriers to the delivery of adherence support, particularly for YMSM who may be most in need of it due, at least in part, to their young age. Methods: We created a culturally-and developmentally-sensitive PrEP adherence mobile app (called "Dot") that was tailored for culturally-diverse young adult MSM. After formative research and usability testing, we conducted a 6-week pre-post study to evaluate the impact of Dot on self-reported PrEP adherence, PrEP treatment self-efficacy, PrEP knowledge, and intention to practice safe sex among culturally-diverse YMSM, age 20-29. Results: At 6 weeks, there were significant improvements in PrEP adherence, PrEP self-efficacy, and intention to practice safe sex. PrEP knowledge scores did not significantly change. Conclusions: The Dot app proved feasible and effective at improving PrEP adherence among culturally -diverse YMSM. Moreover, the app had a high-degree of user appeal, which is foundational to success of an mhealth intervention.
BACKGROUND AND OBJECTIVESexually transmitted diseases (STDs) are increasing among older adults concomitant with a rise in divorce after the age of 50 years. The objective of this study was to examine the effectiveness of a web-based human immunodeficiency virus (HIV)/STD risk reduction intervention for divorced and separated women aged more than 50 years.RESEARCH DESIGN AND METHODSTwo hundred nineteen divorced or separated women, aged 50 years and older, participated in 60-day randomized pre-post control group study. Recruitment occurred via health agencies in Boston and Columbia, SC, and Craigslist advertisements placed in Boston, Columbia, Charleston, New York City, Washington DC, Baltimore, Chicago, Atlanta, Orlando, and Miami.RESULTSIntervention group reported greater intention to practice safe sex compared to the control group (B = .55, p = .03). Intention to practice safe sex differed by perceived stress (B = .15, p = .005), with no difference between control and intervention groups for those with low levels of stress. For high levels of stress, intervention group reported greater intention to practice safe sex compared to controls. Sexual risk was reduced by 6.10 points (SD: 1.10), and self-efficacy for sexual discussion was increased by 2.65 points (SD: 0.56) in the intervention group.DISCUSSION AND IMPLICATIONSA web-based intervention represents a promising tool to reduce HIV/STD risk among older women. Offering HIV/STD education in the context of other topics of interest to at-risk older women, such as divorce, may solve the problem of at-risk older women not seeking out prevention information due to lack of awareness of their heightened risk.

ObjectiveThis study aims to address gaps in health conditions and unmet needs in daily activities between people with dementia (PWD) and without dementia in a developing country and to identify the variables associated with unmet needs among PWD to guide practitioners and policymakers in dealing with an increased burden of dementia.MethodsNationally representative data on 4655 Chileans age 60 and over were used to compare health conditions and unmet needs in daily life activities between individuals with (N = 455, 9.6%) and without dementia. Regression analysis was conducted to identify the variables associated with unmet needs among PWD.ResultsOverall, PWD had worse health and needed greater assistance in performing daily activities than people without dementia. Among PWD, being male was associated with more unmet needs, in both activities of daily living (ADL) and instrumental ADL. Lower educational level and fewer caregivers were associated with more unmet needs for ADL, while inferior functional ability was associated with more unmet needs for instrumental ADL.ConclusionsThe results from this study call for action by practitioners and policymakers to foster caregiver training, increase supportive services, and advance care planning for PWD.
Background: China is facing a continuously rising numbers of people with cognitive impairment (CI). Objectives: To investigate the prevalence and risk factors of CI among elderly people living in rural and urban communities. Methods: We conducted a face-to-face survey of CI on 7,900 individuals aged 50 years or older meeting inclusion criteria in the Malu (rural community, n = 4,429) and Wuliqiao (urban community, n = 3,471) communities of Shanghai. The Mini-Mental State Examination (MMSE) was used to evaluate the cognitive function. Information on demographic features and potential risk factors for CI was collected during the interview. Multivariate logistic regression was performed to identify risk factors associated with CI. Results: Based on the education modified MMSE score, we identified 329 CI cases in rural community and 227 in urban community. The prevalence of CI was 7.43% in rural population and 6.54% in urban population (p = 0.13). In the urban population, risk of having CI was associated with age (OR = 1.04; 95% CI: 1.01–1.08), lack of physical activities (OR = 2.25; 95% CI: 1.11–4.57), presence of diabetes mellitus (OR = 1.79; 95% CI: 1.04–3.07), and having three or more children (OR = 2.39; 95% CI: 1.27–4.50). In contrast, factors associated with rural populations included female gender (OR = 2.03; 95% CI: 1.08–3.82), age (OR = 1.06; 95% CI: 1.03–1.08), exposure to pesticides (OR = 4.68; 95% CI: 1.27–17.21), history of encephalitis or meningitis (OR = 6.02; 95% CI: 1.92–18.85) and head trauma (OR = 1.89; 95% CI: 1.10–3.24). Conclusions: Urban rural and populations showed different risk factors for CI, suggesting that different preventive strategies in these areas should be performed.
MicroRNA-146a is upregulated in the brains of patients with Alzheimer's disease (AD). Here, we show that the rho-associated, coiled-coil containing protein kinase 1 (ROCK1) is a target of microRNA-146a in neural cells. Knockdown of ROCK1 mimicked the effects of microRNA-146a overexpression and induced abnormal tau phosphorylation, which was associated with inhibition of phosphorylation of the phosphatase and tensin homolog (PTEN). The ROCK1/PTEN pathway has been implicated in the neuronal hyperphosphorylation of tau that occurs in AD. To determine the function of ROCK1 in AD, brain tissue from 17 donors with low, intermediate or high probability of AD pathology were obtained and analyzed. Data showed that ROCK1 protein levels were reduced and ROCK1 colocalised with hyperphosphorylated tau in early neurofibrillary tangles. Intra-hippocampal delivery of a microRNA-146a specific inhibitor (antagomir) into 5xFAD mice showed enhanced hippocampal levels of ROCK1 protein and repressed tau hyperphosphorylation, partly restoring memory function in the 5xFAD mice. Our in vitro and in vivo results confirm that dysregulation of microRNA-146a biogenesis contributes to tau hyperphosphorylation and AD pathogenesis, and inhibition of this microRNA could be a viable novel in vivo therapy for AD.
BACKGROUND:Our study was aimed to validate a modified RBD (REM sleep behavior disorder) single question (RBD1Q-C), study the prevalence of probable RBD (pRBD) in a rural community based on RBD1Q-C and investigate the association between pRBD and Parkinson's disease (PD).METHODS:The validation study of RBD1Q-C included 32 Chinese participants (14 idiopathic RBD patients and 18 controls). All participants underwent a polysomnogram (PSG). We then conducted a door-to-door survey to estimate the prevalence of pRBD assessed by RBD1Q-C, and its association with PD among 19614 residents who lived in Malu community of Shanghai, China.RESULTS:RBD1Q-C demonstrated a high sensitivity of 100%, a moderate specificity of 55.6%. The agreement between RBD1Q-C and PSG-based RBD diagnosis was good (k = 0.552). PPV of the RBD1Q-C was 63.6% and NPV was 100%. The prevalence of pRBD in Malu community was 4.9%. In people over 50 years old, presence of pRBD was significantly associated with increased risk of having PD (odds ratio = 2.61, 95% CI: 1.56-4.39).CONCLUSION:RBD1Q-C was shown to be a useful screening tool. Based on the RBD1Q-C, we found that pRBD was not rare in Chinese rural population and associated with odds of PD, calling for more attention from patients, caregivers and physicians.
Prevalence of atopic dermatitis (AD) is increasing worldwide. Up to date, there has been no face-to-face nation-wide study in China. We aim to explore the prevalence of clinical diagnosed AD in children aged 1–7 ys in China. Twelve metropolises were chosen from different areas of China. In each region, we selected 4–10 kindergartens and 2–5 vaccination clinics randomly. A complete history-taking and skin examination were performed by dermatologists. The definite diagnosis of AD and the severity were determined by two or three dermatologists. All criteria concerned in UK diagnosis criteria, characteristic presentation of AD and atypical manifestations were recorded in detail. A total of 13998 children from 84 kindergartens and 40 vaccination clinics were included. The prevalence of AD was 12.94% by clinical diagnosis of dermatologists overall, with 74.6% of mild AD. Comparatively, prevalence of AD based on UK diagnostic criteria was 4.76%. This is the first face-to-face nation-wide study in Chinese children aged 1–7 ys, revealing that the prevalence of AD in children is closer to that of wealthier nations.
Objective: Few studies regarding the epidemiology of essential tremor (ET) and its non-motor symptoms (NMS) have been conducted to date. We thus conducted a door-to-door survey to investigate the ET and its NMS and their clinical manifestations in a rural area of Shanghai, China.Methods: We recruited 19,614 residents (>= 50 years old), living in Malu town, Shanghai. Investigations were divided into two steps. In first step, trained physicians conducted door-to-door surveys and reported probable ET subjects to movement disorder specialists. In the second step, participants with suspected ET had their diagnosis confirmed by movement disorder specialists. We randomly selected 123 subjects from those adults who screened negative for neurological conditions as healthy controls for the study of NMS.Results: The prevalence of ET in individuals (>= 50 years old) was 0.306% (PR = 3.06 per 1000). The mini-mental state examination (MMSE) scores were lower in ET patients than in controls (25.81 +/- 4.20 vs. 26.63 +/- 3.14, P = 0.024). Complaints of restless legs reported by ET patients were significantly higher than those in controls (P < 0.05). In contrast, we did not observe a significant difference for other NMS between ET patients and controls, including olfactory function (P = 0.82), depression (P = 0.94), and rapid eye movement sleep behavior disorder (RBD) (P = 0.71).Conclusions: The prevalence of ET in people >= 50 years old in a rural area of Shanghai China is low. A series of investigations have been performed regarding microRNA (miRNA, miR) of Alzheimer's disease (AD) patients. However, most of these used microarray with neither validation by PCR nor any follow-up on the biological mechanism implicated by findings. Further, there were rarely any analyses linking clinical phenotype of de novo, drug-naive patients to cellular pathogenic mechanism(s) to date. Microarray screening followed by validation via quantitative PCR (Q-PCR) assays and the relationship between miRNAs and phenotypic indices were evaluated. Additionally, the cellular mechanism of miRNAs through effects of β-site amyloid precursor protein (APP) cleaving enzyme (BACE1) was assessed. We identified 2 specific differentially expressed (DE) miRNAs (miR-339 and miR-425) as potential diagnostic biomarkers for AD and revealed that these DE miRNAs could be involved in modulating the pathogenesis of AD via BACE1 protein inhibition. The findings presented here reveal a detailed snapshot of the profile of peripheral blood mononuclear cells (PBMC) miRNA changes in AD patients, association with clinical phenotype, and potential roles in cellular pathogenesis.
We explored the potential differences in cognitive status, lipid and glucose metabolism, ApoEε4 alleles and imaging between diabetic and non-diabetic subjects. 83 subjects with normal cognitive function and 114 mild cognitive impaired patients were divided into four groups by history of diabetes. General demographics was collected from all participants followed by MRI scan, biochemical examinations and a series of neuropsychological tests. Student's t test, multiple regressions and one-way ANOVA were applied to investigate the differences between groups. Comparing diabetic patients with non-diabetic subjects in the mild cognitive impaired group, we found several decreased items in recall of three words in MMSE (p=0.020), AVLT and SCWT (p<0.050). The multiple linear regression revealed that two-hour glucose level (B= -0.255, p<0.001) and fasting C-peptide (B= -0.466, p=0.001) had negative effects on the score of MMSE. In addition, diabetic patients treated with insulin and other diabetes medication performed better in part of the AVLT (p<0.050) compared to patients with insulin treatment or oral antidiabetic medication only. Patients with metformin medication had a better memory outcome compared to patients with sulphonylurea medication in the AVLT long delay free recall (p =0.010). These findings show that patients of mild cognitive impairment with diabetes mellitus have a worse outcome in attention, information processing speed and memory compared to non-diabetic patients. Higher two-hour glucose level and C-peptide level may be risk factors for severe cognitive impairment in type-2 diabetes mellitus patients. The results of this study also suggest that medication may have effects on cognitive function.
Epidermal growth factor (EGF) is a neurotrophic factor that plays an important role in Parkinson's disease (PD). We measured plasma EGF level in PD, essential tremor (ET) and normal controls to investigate whether it changes in PD and whether it is associated with motor and non-motor symptoms of PD. 100 patients with PD, 40 patients with ET as disease control and 76 healthy persons were enrolled in the present study. Motor and non-motor symptoms were assessed by different scales. Plasma EGF levels of three groups were measured by enzyme-linked immunosorbent assay kit. Spearman test and linear logistics regression model were used to test the correlation of EGF with motor and non-motor symptoms of PD. Plasma EGF level was significantly decreased in early PD patients compared with normal control, but not in advanced PD patients. Interestingly, plasma EGF level was significantly increased in advanced PD and total PD patients compared with ET patients, but not in early PD patients. In addition, plasma EGF level was correlated with UPDRS-III scores in PD. Also plasma EGF level was correlated with UPDRS-III scores and NMS scores in early PD. Our results suggested that plasma EGF decreased in the early stage of PD and increased later on in the PD disease course. Also, plasma EGF level was increased significantly in PD compared with ET patients and correlated with motor and non-motor symptoms in early PD.
Abstract Objective: We conducted a case–control study to investigate whether TREM2 polymorphism (rs75932628-T) was associated with late onset Alzheimer’s disease in Chinese Southern Han population. Methods: PCR-restriction fragment length polymorphism assay was performed to genotype rs75932628 in 279 cases with late onset Alzheimer’s diseases patients and 346 control subjects in Shanghai and Nanjing. Results: There was no rs75932628-T variant detected in our sample. However, APOEϵ4 was shown closely associated with the risk of Alzheimer’s disease (Chi-square = 60·288, P = 0·000). Conclusion: Our study suggested that TREM2 (rs75932628-T) was rare in Chinese Han population. Further association studies with large samples are needed to further study the association of TREM2 with late-onset Alzheimer’s disease.
