Urban forests are highly multifunctional and provide numerous ecological functions. Plant functional traits individually or jointly influence the ecological multifunctionality of tree species (TS-EMF) and can also modify TS-EMF in response to environmental changes. However, there has been limited exploration of multitrait combinations for predicting TS-EMF across seasons and of trait thresholds that enhance TS-EMF. Here, for 10 dominant tree species in urban forests of Northeast China, 14 traits were measured and four aboveground and three belowground ecological functions assessed in three seasons. Ecological functions and TS-EMF differed significantly throughout the seasons (P < 0.05). Synergistic relationships were found between carbon sequestration and oxygen release, between cooling and humidification, and between organic carbon accumulation and nutrient cycling. Notably, aboveground multifunctionality played a leading role in TS-EMF. With seasonal changes, resource allocation shifted toward traits related to resource acquisition rather than conservation to maintain TS-EMF. The combination of traits that predicted TS-EMF varied by type, accounting for up to 66.45
With continuous urbanization and climate warming, increased air temperature and elevated ozone (O3) concentration often co-occur in many urban areas, but we still lack information about the interactive effects of warming and elevated O3 on urban trees. In the present experiment, the single and combined effects of increased air temperature (IT, ambient air temperature + 2 degrees C) and elevated O3 (EO, ambient air O3 concentrations + 80 ppb) on carbon (C) fixation and allocation in Quercus mongolica and Pinus tabuliformis, which are widely used as street tree species in urban areas of China, were investigated over two consecutive growing seasons by using 13C isotope techniques. The results showed that IT increased biomass, photosynthetic gas exchange parameters and total 13C content of both tree species. Compared to ambient temperature, IT significantly increased the total 13C content labelled by 56.6 % in Q. mongolica and by 31.2 % in P. tabuliformis in 2021. Elevated O3 induced a decrease in biomass and net photosynthetic rate (Pn) in both tree species. Compared to ambient O3, elevated O3 significantly decreased Pn by 52.6 % in Q. mongolica and by 37.4 % in P. tabuliformis in 2020. The treatment EO decreased 13C allocation to roots but increased 13C content and distribution in leaves in Q. mongolica. These findings demonstrated that EO inhibited the growth and photosynthesis of the two tree species. Our results showed that Q. mongolica was more sensitive to IT and EO than P. tabuliformis, but the former has a self-repair mechanism under increased O3 stress as it is able to invest more carbon to repair leaf damage to a certain extent. Our study also found that the total biomass, relative growth rate, Pn and total 13C content remained higher under the combination of IT and EO compared to EO alone, suggesting that moderate warming may mitigate the negative effects of elevated O3 stress to some extent.
As a medicinal and edible plant, Chinese yam (CY) can promote the enrichment of intestinal probiotics. Mucilage polysaccharides, diosgenin and taxifolin are the dominant components of CY. The purpose of this study was to investigate whether the impact of Chinese yam on gut microbiome structure and metabolism is attributable to its components. In the in vitro gastrointestinal digestion and colon fermentation system, the changes in gut microbiota composition and function were determined by 16S rRNA sequencing, and the levels of bacterial metabolites including short-chain fatty acids (SCFAs) and indole-like metabolites were detected by gas chromatography and an enzyme-linked immunoassay. The results show that CY, mucilage polysaccharides, diosgenin and taxifolin could increase the microbial diversity index. Furthermore, probiotics including Lactobacillus and Bacteroides were significantly increased, while harmful bacteria such as Escherichia and Proteus declined. CY could increase the production of SCFAs including acetic acid and butyric acid. Of note, CY and diosgenin displayed similar impacts on enhancing the abundance of Clostridium and promoting the production of indole-3-lactic acid and lactic acid. These findings provide evidence supporting Chinese yam as a natural food to regulate intestinal health. Diosgenin as a component of CY contributes mostly to the impact on regulating intestinal flora.
The study aimed to investigate the effects of different levels of added sucrose on the fermentation quality and microbial community of kudzu (Pueraria lobata) silage. The three sucrose supplementation levels utilized were 0, 0.5, and 1.0%, and kudzu was silaged for 15, 30, and 60 days. Sucrose supplementation significantly decreased the pH levels, acid detergent fiber, ammonia nitrogen content, and relative abundance of Pantoea in the silages (p < 0.05). The addition of 1% sucrose to kudzu silage at 60 days had the lowest acid detergent fiber content, the highest crude protein, and the highest relative feed value. Additionally, the sucrose-supplemented silage had a lower pH than that of the control group at each time point. The dominant genera in all groups were Klebsiella, Enterobacteriaceae, Lactobacillus, and Weissella, and the relative abundance of Enterobacteriaceae was lower in the 1% sucrose-supplemented group than in the control group. These results showed that sucrose addition could improve the quality of kudzu silage and increase its beneficial microbial community.
Climate warming and surface ozone (O3) pollution are important global environmental issues today. However, the combined impacts of air warming and O3 on phenology and its functional traits of urban trees are still poorly understood. Here, an experiment was performed to explore the variations of the spring phenological and functional traits in leaves of Populus alba 'Berolinensis' and Forsythia suspensa under ambient air (15.8 °C, 35.7 ppb), increased air temperature (IT, ambient air temperature + 2 °C, 17.9 °C), elevated O3 (EO, ambient air O3 concentrations + 40 ppb, 77.4 ppb), and their combined treatments (17.7 °C, 74.5 ppb). Our results showed that: IT advanced the beginning of leaf bud expansion phase of P. alba 'Berolinensis' and F. suspensa for 6 d and 5 d, respectively, increased leaf unfolding rate, leaf area and dry weight, and enhanced photosynthesis and antioxidative enzyme activities. EO delayed the beginning of leaf bud expansion phase of P. alba 'Berolinensis' for 5 d, decreased leaf area and biomass, and inhibited photosynthesis and caused oxidative damage of plant leaves. Compared to EO, the combined treatment advanced the spring phenophase, increased growth and induced the higher level of photosynthetic rate and antioxidative enzymes activities in plant leaves, which indicated that the positive effects of increased temperature (17.7 °C) alleviated the inhibition of growth and photosynthesis induced by ozone. Our findings can provide a theoretical reference for predicting the adaptation of functional traits of the two trees blossomed early under warming and O3 pollution at spring phenological stage.
In the past ten years, the research and application of microbiome has continued to increase. The microbiome has gradually become the research focus in the fields of life science, environmental science, and medicine. Meanwhile, many countries and organizations around the world are launching their own microbiome projects and conducting a multi-faceted layout, striving to gain a strategic position in this promising field. In addition, whether it is scientific research or industrial applications, there has been a climax of research and a wave of investment and financing, accordingly, products and services related to the microbiome are constantly emerging. However, due to the rapid development of microbiome sequencing and analysis related technologies and methods, the research and application from various countries have not yet unified on the standards of technology, programs, and data. Domestic industry participants also have insufficient understanding of the microbiome. New methods, technologies, and theories have not yet been fully accepted and used. In addition, some of the existing standards and guidelines are too general with poor practicality. This not only causes obstacles in the integration of scientific research data and waste of resources, but also gives related companies unfair competition opportunity. More importantly, China still lacks national standards related to the microbiome, and the national microbiome project is still in the process of preparation. In this context, the experts and practitioners of the microbiome worked together and developed the consensus of experts. It can not only guide domestic scientific research and industrial institutions to regulate the production, learning and research of the microbiome, the application can also provide reference technical basis for the relevant national functional departments, protect the scale and standardized corporate company's interests, strengthen industry self-discipline, avoid unregulated enterprises from disrupting the market, and ultimately promote the benign development of microbiome-related industries.
Dark septate endophytes (DSEs) are one of the most studied groups of root fungal endophytes in recent years. However, the effects of DSE on host plant are still under debate, and the molecular mechanisms are poorly understood. In this study, we identified a DSE fungus of the genus Anteaglonium, named T010, from the wild blueberry. When inoculated into Vaccinium corymbosum L. plants, T010 could enhance root growth and promote shoot branching, leading to increased plant growth. By comparative transcriptome analysis, we obtained 1948 regulated differentially expressed genes (DEGs) from the V. corymbosum plants treated by T010. Further functional enrichment analysis identified a series of DEGs enriched in transcriptional regulation, material transport, phytohormone biosynthesis and flavonoid biosynthesis. Moreover, the comparative analysis of liquid chromatography-mass spectrometry verified that T010 treatment induced the changes in the contents of various phytohormones and flavonoids. This is the first report on the isolation of DSE fungi of the genus Anteaglonium from blueberry roots. Moreover, our results suggested that T010 colonization could result in a series of changes in cell metabolism, biosynthesis and signal pathways, thereby promoting plant growth. Particularly, the changes of phytohormone and flavonoid metabolism induced by T010 colonization might contribute to the promotion of blueberry growth. Our results will provide new insights into understanding of the interaction of DSE fungi and host plants, as well as the development and utilization of DSE preparations.
Atmospheric warming and increasing tropospheric ozone (O3) concentrations often co-occur in many cities of the world including China, adversely affecting the health status of urban trees. However, little information is known about the combined and interactive effects from increased air temperature (IT) and elevated O3 (EO) exposures on urban tree species. Here, Ginkgo biloba and Populus alba ‘Berolinensis’ seedlings were subjected to IT (+2 °C of ambient air temperature) and/or EO (+2-fold ambient air O3 concentrations) for one growing season by using open-top chambers. IT alone had no significant effect on physiological metabolisms at the early growing stage, but significantly increased photosynthetic parameters, antioxidative enzyme activities (P < 0.05). EO alone decreased physiological parameters except for increased oxidative stress. Compared to EO exposure alone, plants grown under IT and EO combined showed higher antioxidative and photosynthetic activity. There was a significant interactive effect between IT and EO on net photosynthetic rate, stomatal conductance, water use efficiency, the maximum quantum efficiency of PSII photochemistry, the actual quantum efficiency of PSII, enzyme activities, aboveground biomass and root/shoot ratio (P < 0.05), respectively. These results suggested that during one growing season, IT mitigated the adverse effect of EO on the tested plants. In addition, we found that G. biloba was more sensitive than P. alba ‘Berolinensis’ to both IT and EO, suggesting that G. biloba may be a good indicator species for climate warming and air pollution, particularly under environmental conditions as they co-occur in urban areas.
BACKGROUND:Homing endonuclease genes (HEGs) are widely distributed genetic elements in the mitochondrial genomes of a diversity of eukaryotes. Due to their ability to self-propagate within and between genomes, these elements can spread rapidly in populations. Whether and how such elements are controlled in genomes remains largely unknown. RESULTS:Here we report that the HEG-containing introns in the mitochondrial COX1 gene in Cryptococcus neoformans are mobile and that their spread in sexual crosses is influenced by mating type (MAT) α-specific homeodomain gene SXI1α. C. neoformans has two mating types, MATa and MATα . In typical crosses between strains of the two mating types, only a small portion (< 7%) of diploid fusants inherited the HEGs from the MATα parent. However, disruption of the SXI1α gene resulted in the majority (> 95%) of the diploid fusants inheriting the HEG-containing introns from the MATα parent, a frequency significantly higher than those of intronless mitochondrial genes. CONCLUSIONS:Our results suggest that SXI1α not only determines uniparental mitochondrial inheritance but also inhibits the spread of HEG-containing introns in the mitochondrial genome in C. neoformans.
Cecropins, originally found in insects, are a group of cationic antimicrobial peptides. Most cecropins have an amphipathic N-terminal segment and a largely hydrophobic C-terminal segment, and normally form a helix-hinge-helix structure. In this study, we developed the novel 32-residue cecropin-like peptide cecropin DH by deleting the hinge region (Alanine-Glycine-Proline) of cecropin B isolated from Chinese oak silk moth, Antheraea pernyi. Cecropin DH possesses effective antibacterial activity, particularly against Gram-negative bacteria, with very low cytotoxicity against mammalian cells. Interactions between cecropin DH and the highly anionic lipopolysaccharide (LPS) component of the Gram-negative bacterial outer membrane indicate that it is capable of dissociating LPS micelles and disrupting LPS aggregates into smaller assemblies, which may play a vital role in its antimicrobial activity. Using LPS-stimulated mouse macrophage RAW264.7 cells, we found that cecropin DH exerted higher potential anti-inflammatory activity than cecropin B, as demonstrated by the inhibition of pro-inflammatory cytokines nitric oxide production and secretion of tumor necrosis factor-α. In conclusion, cecropin DH has potential as a therapeutic agent for both antibacterial and anti-inflammatory applications.
Ground-level ozone (O3) pollution has been widely concerned in the world, particularly in the cities of Asia, including China. Elevated O3 concentrations have potentially influenced growth and nutrient cycling of trees in urban forest. The decomposition characteristics of urban tree litters under O3 exposure are still poorly known. Ginkgo biloba is commonly planted in the cities of northern China and is one of the main tree species in the urban forest of Shenyang, where concentrations of ground-level O3 are very high in summer. Here, we hypothesized that O3 exposure at high concentrations would alter the decomposition rate of urban tree litter. In open-top chambers (OTCs), 5-year-old G. biloba saplings were planted to investigate the impact of elevated O3 concentration (120 ppb) on changes in nutrient contents and decomposition rate of leaf litters. The results showed that elevated O3 concentration significantly increased K content (6.31 ± 0.29 vs 17.93 ± 0.40, P < 0.01) in leaves of G. biloba, significantly decreased the contents of total phenols (2.82 ± 0.93 vs 1.60 ± 0.44, P < 0.05) and soluble sugars (86.51 ± 19.57 vs 53.76 ± 2.40, P < 0.05), but did not significantly alter the contents of C, N, P, lignin and condensed tannins, compared with that in ambient air. Furthermore, percent mass remaining in litterbags after 150 days under ambient air and elevated O3 concentration was 56.0% and 52.8%, respectively. No significant difference between treatments was observed in mass remaining at any sampling date during decomposition. The losses of the nutrients in leaf litters of G. biloba showed significant seasonal differences regardless of O3 treatment. However, we found that elevated O3 concentration slowed down the leaf litter decomposition only at the early decomposition stage, but slightly accelerated the litter decomposition at the late stage (after 120 days). This study provides our understanding of the ecological processes regulating biogeochemical cycles from deciduous tree species in high-O3 urban area.
Two psychrotolerant facultative anaerobes, strains B7-2T and B5T, were isolated from the Zoige Wetland on the Qinghai-Tibetan Plateau. The 16S rRNA gene sequences of strains B7-2T and B5T shared high similarity (>99 %) with those of the type strains of the genus Trichococcus, while their digital DNA-DNA hybridization values with each other (49 %) and with the reference type strains (48-23 %) were lower than 70 %, which suggest that they represent two novel species of the genus Trichococcus. Cells of strains B7-2T and B5T were immotile cocci, grew in the temperature range of 4-37 °C (optimum 25 °C) and were alkaliphilic with optimum growth at pH 9.0. The major components of the cellular fatty acids were C16 : 0, anteiso-C17 : 0 and C18 : 0 for strain B7-2T, and C16 : 0, anteiso-C17 : 0, C18 : 1ω9c and C18 : 0 for strain B5T. The genomic DNA G+C contents were 46.0 and 46.7 mol% for strains B7-2T and B5T, respectively. Based on physiological and genomic characteristics, it is suggested that strains B7-2T and B5T represent two novel species within the genus Trichococcus, for which the names Trichococcus paludicola sp. nov. and Trichococcus alkaliphilus sp. nov. are proposed. The type strains are B7-2T (=DSM 104691T=KCTC 33886T) and B5T (=DSM 104692T=KCTC 33885T), respectively.
Lipopolysaccharide (LPS), the important component of the outer membrane of Gram-negative bacteria, contributes to the integrity of the outer membrane, and protects the cell against bactericidal agents. LPS, also called endotoxin synonymously, is well known as a potent inducer of the innate immune system that often causes septic shock in the intensive cares. Chemically, the amphiphilic LPS is made up of three parts, i.e., hydrophobic lipid A, hydrophilic core oligosaccharide chain, and hydrophilic O-antigenic polysaccharide side chain. Specially, the lipid A is known to be responsible for a variety of biological effects during Gram-negative sepsis. LPS can elicit a strong response from innate immune system and result in local or systemic adverse reactions. LPS can trigger massive inflammatory responses and may result in immunopathology, for which the molecular basis is mediated by the signal path- way of LPS. In recent years, a tremendous progress has been made in determining the associated proteins and receptors in the LPS signaling that leads to the disease. This review gives a summary of recent progresses of research on LPS and LPS receptors.
Lipopolysaccharide (LPS), the important component of the outer membrane of Gram-negative bacteria, contributes to the integrity of the outer membrane and protects the cell against bactericidal agents, including antimicrobial peptides. However, the mechanisms of interaction between antimicrobial peptides and LPS are not clearly understood. Halictines-2 (HAL-2), one of the novel antimicrobial peptides, was isolated from the venom of the eusocial bee Halictus sexcinctus. HAL-2 has exhibited potent antimicrobial activity against Gram-positive and Gram-negative bacteria and even against cancer cells. Here, we studied the interactions between HAL-2 and LPS to elucidate the antibacterial mechanism of HAL-2 in vitro. Our results show that HAL-2 adopts a significant degree of β-strand structure in the presence of LPS. LPS is capable of inducing HAL-2 amyloid formation, which may play a vital role in its antimicrobial activity.
ORBK (LKGCWTKSIPPKPCFK) is a cyclic cationic peptide that has potent antimicrobial properties and trypsin inhibitory activities. To explore a new approach for expressing ORBK in Escherichia coli, a sequence encoding ORBK was cloned into pET28a vector in which maltose-binding protein (MBP) was used as a fusion partner and an N-terminal 6-His as an affinity tag. Protein expression was induced with 0.5mM Isopropyl-thio-galactoside (IPTG) for 4h at 37°C. The recombinant ORBK was then purified by Ni affinity column and further digested with tobacco etch virus (TEV) enzyme. The cleaved ORBK peptide was separated from MBP fusion partner by reverse phase high performance liquid chromatography (RP-HPLC) and oxidized to obtain the cyclic form. Mass spectroscopy and nuclear magnetic resonance (NMR) spectroscopy were performed for ORBK characterization. Herein we have developed an effective and reliable method to express and purify ORBK which sets a solid foundation for future structural and functional studies.
A new flavonolignan, anthelminthicol A (1), together with four known compounds, was isolated from the EtOAc extracts of the seeds of Hydnocarpus anthelminthica. Their structures were elucidated using extensive spectroscopic techniques. Bioassay showed that compounds 3–5 could inhibit nitric oxide production in LPS-stimulated RAW 264.7 macrophage cell lines, with IC50 values of 7.81, 9.38, and 10.55 μM, respectively.
