Objective To explore and analysis the effect of internal heat acupuncture combined with platelet rich plasma(PRP)on T cell subsets in peripheral blood of patients with knee osteoarthritis(KOA).Methods From May 2020 to October 2023,84 cases of pa-tients with knee osteoarthritis treated at a certain hospital were selected as the research subjects for this study.According to the principle of random coin tossing,the 84 cases of patients were divided into an experimental group of 42 cases and a traditional group of 42 cases.The traditional group were treated with intra-articular injection of sodium hyaluronate,while the experimental group were treated with in-ternal heat acupuncture combined with PRP.The treatment were observed for 6 weeks.Recorded the overall efficacy,and changes of vis-ual analogue scale(VAS),serum interleukin-17(IL-17)levels,T cell subsets in peripheral blood of two groups.Results After 6 weeks of treatment,the total effective rate of the experimental group were 97.62%,while that of the traditional group were 83.33%.The experimental group showed a significant improvement compared to the traditional group(P<0.05).The pain VAS scores of the experi-mental group and the traditional group after 6 weeks of treatment were lower than before treatment(P<0.05),and the pain VAS scores of the experimental group decreased more compared to the traditional group after 6 weeks of treatment(P<0.05).After6 weeksoftreat-ment,the serum IL-17 levels in the experimental group and the traditional group were significantly lower than before treatment(P<0.05).After 6 weeks of treatment,the serum IL-17 levels in the experimental group were significantly lower than those in the tra-ditional group(P<0.05).After 6 weeks of treatment,the relative proportion of CD4+T cells in peripheral blood of the experimental group and the traditional group were lower than before treatment(P<0.05),and the relative proportion of CD8+T cells were higher than before treatment(P<0.05).After 6 weeks of treatment,the relative proportion of CD4+T cells and CD8+T cells in peripheral blood of the experimental group were also statistically significant compared to the traditional group(P<0.05).Conclusion The application of in-ternal heat acupuncture combined with PRP in patients with knee osteoarthritis can improve the treatment effect,promote pain relief,in-hibit serum IL-17 expression,regulate the proportion of T cell subsets in peripheral blood,and continuously improve the prognosis of patients.
Objective To investigate the effect of Parkin protein on aerobic glycolysis and apoptosis of breast cancer MDAMB-231 cells. Methods Western blot was firstly used to detect the expression level of Parkin in normal breast epithelial cells MCF-10A and breast cancer cells MDA-MB-231. Then MDA-MB-231 cells transfected with Parkin overexpression plasmids were used as the experimental group, and the MDA-MB-231 cells transfected with empty vectors as the control group. Glucose consumption, lactic acid, and ATP production of these two groups were detected by those related kits, respec-tively. The expression of glycolysis related proteins including GLUT1, PKM2, and LDHA was evaluated by Western blot. Cell growth, colony formation, and wound healing migration assays were used to evaluate the ability of cell proliferation and migration for these two groups. The expression of apoptosis-related proteins Bax, Bcl-2, and cleaved Caspase-3 were examined by Western blot. Results Compared with normal breast cells, the expression of Parkin protein in MDA-MB-231 cells decreased significantly(P<0.01). The results showed that high expression of Parkin led to decreased production of glucose uptake and lactic acid production and increased level of ATP(P<0.05). Compared to the control group, the expression of glycolysis-related proteins GLUT1, PKM2, and LDHA decreased in the Parkin group(P<0.05). Overexpression of Parkin in MDAMB-231 cells reduced the cell proliferation and migration. Moreover, the expression of anti-apoptotic protein Bcl-2 de-creased and the expression of cleaved Caspase-3 increased(P<0.05). Conclusion The expression of Parkin in MDA-MB-231cells is lower than that in MCF-10A cells. The high expression of Parkin leads to the inhibition of aerobic glycolysis pathway,the decrease of proliferation and migration ability, and apoptosis of MDA-MB-231 cells.
Objective To observe the effect of probiotic composition on depression with anxiety pain. Methods 97 patients with anxiety and depression were divided into the control group(48 cases) and the study group(49 cases).According to random number table, the control group was treated with paroxetine hydrochloride tablets routinely, and the study group was treated with probiotic composition. The physical and mental indexes of the patients in the two groups were compared with t-test and chi-square test 30 days later. Results TNF-α of patients in the control group 54.07±5.12 pg/mL,and that in the study group was 42.16±3.97 pg/mL, which were significantly lower than those in the control group(P<0.05). The 5-HT of patients in the control group was 256.74±22.61 ng/mL, and that in the study group was 301.65±27.05 ng/mL, which was significantly higher than that in the control group(P<0.05). SAS scores of patients in the control group were 55.28±5.41 points, and those in the study group were 47.91±4.62 points, which were significantly lower than those in the control group(P<0.05). The SDS scores of the patients in the control group were 59.83±5.62 points, and those in the study group were 51.76±4.97 points, which were significantly lower than those in the control group(P<0.05). The PSQI scores of the patients in the control group were 11.72±1.06 points, while that in the study group were 9.83±0.94 points, which were significantly lower than that in the control group(P<0.05). The incidence of adverse reactions in the control group was 20.83%, while that in the study group was 6.12%, which was significantly lower than that in the control group(P<0.05). Conclusion The probiotic composition is beneficial to reduce the level of TNF-a, promote the synthesis of 5-HT, alleviate anxiety, depression and insomnia symptoms, and reduce adverse reactions.
Objective:To compare the risk for pneumonia in antiplatelet therapy with clopidogrel and ticagrelor.Methods:The relevant databases including PubMed, Embase, Cochrane Library, CBM, CNKI, and Wanfang Med (up to March 1, 2022) were searched. The randomized controlled trials (RCTs) of clopidogrel and ticagrelor (trial group or control group to each other) in the treatment of coronary atherosclerotic heart disease were collected. The Cochrane collaboration risk of bias assessment tool was used to evaluate the methodological quality of the RCTs. RevMan 5.4 software was used for meta-analysis to compare the risk of pneumonia in patients between 2 groups. The effect sizes were the risk ratio ( RR) and its 95% confidence interval ( CI). Results:A total of 5 RCTs were enrolled in the analysis. The methodological quality evaluation results showed that 2 RCTs were with low risk of bias and the others were high. In the 5 RCTs, 36 401 patients were involved, of which 18 724 received the therapy for acute coronary syndrome, 13 842 for peripheral artery disease, 3 799 for myocardial infarct, and 36 for coma survivor with cardiac arrest. The meta-analysis results showed that the incidence of pneumonia were 1.5% (271/18 174) and 1.2% (217/18 227) in patients receiving clopidogrel and ticagrelor, respectively; the risk of pneumonia in ticagrelor-treated patients was relatively lower, and the difference was statistically significant ( RR=0.80, 95% CI: 0.67-0.95, P=0.01). Subgroup analysis showed that the risk for pneumonia was similar in patients between the 2 groups when they received short-term (several days) medication ( P=0.26), while the risk was relatively lower in ticagrelor-treated patients compared with that in clopidogrel-treated patients when the therapeutical time was more than 12 months ( RR=0.80, 95% CI: 0.67-0.96, P=0.02). And also, in the combination therapy with aspirin, the risk of pneumonia was lower in ticagrelor-treated patients than that in clopidogrel-treated patients ( RR=0.74, 95% CI: 0.56-0.98, P=0.03). Conclusion:Compared with clopidogrel, the risk for pneumonia was relatively lower in ticagrelor treatment than that in clopidogrel treatment.
Objective:To conduct a glucagon like peptide-1(GLP-1)controllability model rat by chemical genetics, and observe the impact of GLP-1 neuron excitability on appetite.Methods:Fifteen rats were evenly divided into Green fluorescent protein(GFP)group, HM3D group, and HM4D group. Various combinations of adeno-associated virus(rAAV)were injected into the nucleus tractus solitarius(NTS). rAAV-GLP-1-cre and rAAV-GFP-dio were administered in rats of GFP group. The rats of HM3D group were injected with rAAV-GLP-1-cre and rAAV-HM3D-mCherry-dio while rAAV-GLP-1-cre and rAAV-HM4D-mCherry-dio were injected in rats of HM4D group . The optimal dose of clozapine N-oxide(CNO)was selected based on feeding behavior and body weight changes of rats after intraperitoneal injection of different doses of CNO. The controllability of GLP-1 neurons was confirmed by comparing with intraperitoneal injection of saline. The number of activated GLP-1 neurons in the NTS area and the expression of POMC neurons in the hypothalamus were detected 30 minutes after CNO injection.Results:GLP-1 neurons in the NTS area of rats were successfully labeled. The rat of HM3D group revealed a decrease in food intake( P=0.021)while the rat of HM4D group showed an increase( P=0.002), when given 1 mg/kg of CNO, no changes at the dose of 0.5 mg/kg and 3.0 mg/kg. Immunofluorescence showed that the activity of GLP-1 neurons in NTS of GFP group was lower than that of HM3D group( P=0.022), and higher compared with that of the HM4D group( P=0.049). The expression of GLP-1 neurons in NTS and POMC neurons in the hypothalamus of the HM3D group after intraperitoneal injection of CNO was also higher than that in the HM4D group( P=0.003). Conclusion:Using chemical genetics technology, GLP-1 controllability model rat could be successfully established via injecting varying combinations of rAAV into the NTS area of rat. Injection of 1 mg/kg CNO can effectively activate or inhibit the neuron to regulate appetite.
