A signed graph is one that features two types of edges: positive and negative. Balanced signed graphs are those in which all cycles contain an even number of negative edges. In the adjacency matrix of a signed graph, entries can be 0, -1, or 1, depending on whether ij represents no edge, a negative edge, or a positive edge, respectively. The index of the adjacency matrix of a signed graph G(center dot) is less or equal to the index of the adjacency matrix of its underlying graph G, i.e., lambda(1)( G(center dot))
In epistatic fitness landscapes, the fitness effect of a mutation depends on the genetic background and may even switch between deleterious and beneficial depending on the presence of another mutation. Epistatic interactions may cause both mutations to change the sign of each other's fitness effects (reciprocal sign epistasis) or only one mutation to do so (simple sign epistasis). Both these forms of epistasis influence evolutionary trajectories. While reciprocal sign epistasis has been associated with multi-peaked landscapes and their ruggedness, the role and relative frequency of simple sign epistasis in fitness landscapes have not been systematically investigated. Here, we prove that the presence of simple sign epistasis is associated with evolutionary detours, i.e., indirect, longer fitness-increasing paths to fitness peaks that include back-mutations. We also show that in experimentally resolved, weakly epistatic landscapes, simple sign epistasis occurs much more frequently than reciprocal sign epistasis. This result is consistent with the theoretical predictions we derive for most landscape models, with the exception of the block model and of landscapes dominated by pairwise allelic incompatibilities, such as RNA stability landscapes. Our results suggest that detours represent a general feature of evolutionary trajectories in weakly epistatic landscapes.
Abstract Structural network representations of metastatic dissemination typically focus on static topology without resolving transport dynamics, relaxation timescales, or steady-state behaviour. Here, we formulate a discrete Markovian transport model on a directed higher-order network with transition rates derived from qualitative clinical affinity classes. By constructing a non-Hermitian row-stochastic transfer operator, we characterise the relaxation dynamics through its spectral decomposition. The system exhibits a fast-mixing regime characterised by a spectral gap of γ ≈ 0.67, corresponding to a characteristic relaxation timescale of τ ≈ 1.49 discrete steps, with the influence of the primary tumour origin progressively attenuated during dissemination. Convergence towards a non-equilibrium steady state (NESS) is accompanied by a reduction in Shannon entropy, concentrating probability mass within specific topological sinks. This spectral relaxation delineates two distinct dynamical regimes: early transient dissemination ( n < τ ), dominated by local organ-specific transition probabilities (organotropism), and the asymptotic regime ( n > τ ), determined increasingly by the global transport architecture of the network. Comparison with independent clinical and autopsy observations across 21 primary tumours and 23 target organs indicates that the predicted stationary distribution is consistent with the observed hierarchy of metastatic organ involvement.
