To explore the protective function on nervous system of compound Chinese herbal and active ingredient,we has concluded positive effects on nervous system of compound Chinese herbal and active ingredient by collecting relative papers reported in recent years.This provides a reference to research further the mechanism of other compound Chinese herbal and active ingredient on protection of nervous system.
OBJECTIVE:To investigate the effect of 5-hydroxymethyl furfural (5-HMF) on apoptosis and BCL-2, NF-kappaB gene expression of rat hippocampal neurons injured by hydroperoxide (H2O2).METHODS:Hippocampal neurons of newly born rat were cultured in vivo and injured by H2O2. Effect of different concentration of 5-HMF on cell viability was measured by MTT. Flow cytometer (FCM) was used to measure the apoptosis of rat hippocampal neurons pre-cultured with different concentration of 5-HMF,Western blotting was used to measure the expression of BCL-2 and NF-kappaB gene.RESULTS:It revealed that the high and medium dosage of 5-HMF could increase the activity of rat hippocampal. The high, medium and low dosage of 5-HMF also increased the expression of BCL-2 gene and decreased the expression of NF-kappaB gene.CONCLUSION:5-HMF could restrain the apoptosis of cultured hippocampal neurons injured by H2O2. The mechanism may be related to increasing in BCL-2 level and decreasing in NF-kappaB level.
Objective:To study the effect on the glucose tolerance of the diabetic kunming rat of the ethanol fraction from Platycodon grandiflorum A.DC.Methods:Diabetic kunming rat models were induced by i.p.injection of streptozotocin(STZ)assisted with high energy diet,and were divided into different groups,which were given 28d of 2,8g/kg ethanol fraction from Platycodon Grandiflorum A.DC,Acarbose or water respectively.Then the glucose tolerance,serum insulin levels(INS)and insulin sensitivity index(ISI)were tested and the pathology damage of the pancreatic were examined.Result:High doses of the ethanol fraction from Platycodon grandiflorum A.DC improved glucose tolerance,INS,ISI more evidently,and alleviated the pancreas tissue injury.Conclusion:The improvement of the the ethanol fraction from Platycodon grandiflorum A.DC on glucose tolerance related to the improvement of the ISI and the partial repair of the diabetic rats' pancreas tissue.
Objective To extract α-glucosidase inhibitors from Potentilla anserina L. Methods Water-decocting method and water-decocting ethanol-precipitation were applied to obtain water extract,sodden ointment and polysaccharides of Potentilla anserina L. The colorimetry of pNPG was used for the determination of the activity of alpha-glucosidase pure enzyme and alpha-glucosidase from isolated small intestines of natural ICR mice. Results The extracts of Potentilla anserina L. had obvious inhibitory effect on the pure enzyme of α-glucosidase. The water extract and sodden ointment also had inhibitory effect on α-glucosidase from isolated small intestine in natural ICR mice,the effect being in a dose-dependant manner. Conclusion The extracts of Potentilla anserina L. have the inhibitor-like effect on α-glucosidase,and the active position is probably in the sodden ointment.
目的:观察夏枯草水提物对α-糖苷酶类活性、麦芽糖耐量试验的影响,研究其延缓正常ICR小鼠单糖吸收作用.方法:采用水煎法得到夏枯草水提物,按Van Loon's碘法测定α-淀粉酶活性,pNPG法测定α-葡萄糖苷酶活性,葡萄糖氧化酶法测定正常ICR小鼠离体小肠α-麦芽糖酶活性;并测定夏枯草水提物对正常ICR小鼠麦芽糖耐量试验的动态血糖值的影响.结果:夏枯草水提物对α-淀粉酶、α-葡萄糖苷酶、离体小肠α-麦芽糖酶活性均有抑制作用,并呈量效关系;同时能提高其麦芽糖耐量,降低正常ICR小鼠餐后高血糖.结论:夏枯草水提物能延缓正常ICR小鼠单糖吸收,其机制可能与抑制肠道α-糖苷酶类有关.
The alkaline phosphatase(ALP) from the lead homogenate of rabbit was purified by sequential extraction with organic solvent with precooling treatment at 0 ℃,and the products were charaterizated by SDS-PAGE.In the specific activity detection of the ALP,the protein was quantified by Bradford and Folin-Lowry test and the Bradford test was found to be the more desirable one due to its high sensitivity and the ability of anti-interference.Through condition optimization of the extraction condition,molecular identification method and content determination of the protein,the practicality and applicability of the experiments were developed.
