The aim of the present work was to estimate the choleretic effect of extracts from Carthamus tinctorius L., Tagetes erecta L. and Calendula officinalis L. The experiments were carried out on white male rats weighing 180-200 g. The extracts in a single dose were introduced into the duodenum of the rats at the doses 50-200 mg/kg in the form of water solution. The results of the experiments have show that the extracts from Carthamus tinctorius L. and Calendula officinalis have the marked choleretic action due to the presence of biologically active substances, predominantly flavonoids.
The role of Hsp70 and nitrogen oxide in realization of adaptogenic reactions and adaptogenic effect of the extract from Astragalus membranaceus (Fischer) Bunge containing such active substances as isoflavones (28-49 mg%) and triterpenes (52-98 mg%) has been studied on the model of acute emotional stress. The acute emotional stress was simulated by water immersion method. In the control group, the acute emotional stress caused erosive injury of the stomach mucosa, hypertrophy of the adrenal glands and involution of thymus followed by 47% increase of nitrate/nitrite concentration in the blood plasma, 15% decrease of the basal level of Hsp70 in thymus in vivo, 4.5 times increase of the Hsp70 level in leucocytes induced by heat shock in vitro (p <= 0.05). The course preventive administration of the extract from Astragalus membranaceus in experimental-therapeutic dose 50 mg/kg against the background of the acute emotional stress prevents the development of the "Selye's triad": involution of thymus (by 34%), erosive injury of the stomach mucosa (by 2.8 times) and hypertrophy of the adrenal glands (by 20%) (p <= 0.05). The increased resistance to emotional stress against the background of the course administration of the extract from Astragalus membranaceus is followed by activation of Hsp70 expression in thymus by 2.6 times (p <= 0.05) that is revealed in vivo. In leucocytes undergone the heat shock in vitro, the synthesis of Hsp70 is 4.5 times increased and against the background of the course administration of the Astragalus membranaceus extract, the induction is 66% decreased (p <= 0.05). The concentration of NO metabolites in the blood plasma of the experimental group was 44% decreased against the background of the course administration of the Astragalus membranaceus extract. The administration of the Astragalus membranaceus extract was followed by activation of NO system (by 54%) (p <= 0.05). We suppose that the activation of Hsp70 synthesis and prevention of stress-induced NO generation is the molecular-cell mechanism of adaptogenic effect of Astragalus membranaceus due to the presence of flavonoids in the extract: isoflavones - calycosin, calycosin-7-O-glycoside and formononetin capable to activate the expression of heat shock proteins, also they have the direct and indirect radical scavenging effect.
The article gives the research findings of the stress-protective activity of extracts derived from rhizomes and aerial part of Rhaponticum uniflorum L., in which phytoecdysteroids and flavonoids are the main active ingredients. The developed extracts are practically non-toxic substances. Their course administration in experimental-therapeutic dose 100 mg/kg has the stress-protective effect in acute emotional stress decreasing the manifestation of the Selye’s triad signs due to inhibition of hyperactivity of central stress-realizing bodily systems. Peripheral mechanisms of adaptogenic effect of the Rhaponticum uniflorum extracts are connected with inhibition of free radical oxidation processes.

BACKGROUNDThe relevance of this study is determined by the need for the scientifically sound substantiation of the possibility for the use of acidic mineral waters in the treatment of digestive diseases taking into consideration their widespread application in ethnomedicine, in particular in the Republic of Tuva.AIMThe objective of the present study was to elucidate the gastroprotective action of acidic mineral water from the Azhyg-Sug source (Republic of Tyva) on the experimental animals as exemplified by the model of ulcerogenesis under the conditions of immobilization stress.MATERIAL AND METHODSThe experimental studies were carried out with the use of 32 white rats of the Wistar line. Ulcerogenesis was initiated by means of immobilization stress. We evaluated the pathomorphological characteristics of the gastric mucosa and the number of destructions based on the Pauls index. The state of the lipid peroxidation system and antioxidative protection were determined from the content of malon dialdehyde and reduced glutathione, extracellular catalase and superoxide dismutase activities in erythrocytes.RESULTSThe study has demonstrated that the mineral water from the Azhyg-Sug source slows down the development of the inflammatory and destructive necrotic processes in the mucosa of the stomach of albino rats. The depth of erosion in the animals receiving mineral water was 2.3 and 3.4 times lower than in the control animals (p≤0.05). The antioxidant effect of mineral water was confirmed by the 14-20% decrease of the MDA concentration as well as by the increase of the catalase and superoxide dismutase (SOD) activities by 21-25% and 20-30% respectively in comparison with the control animals.CONCLUSIONThe Azhyg-Sug mineral water has the strong gastroprotective influence on the experimental animals having the induced neurogenic ulcer. One of the mechanisms underlying the gastroprotective action of the investigated mineral water arises from its ability to inhibit the processes of lipid peroxidation with the simultaneous enhancement of the activity of the antioxidant system of the organism.
The aim of the work was to estimate the antiulcerous effect of a new multicomponent plant extract derived from the following species of medicinal plant materials: leaves of Plantago major L., grass of Gnaphalium uliginosum L., rhizomes and roots of a Inula helenium L., flowers of Matricaria chamomilla L., roots of Glycyrrhiza glabra L., grass of Polygonum aviculare L., grass of Urtica dioica L., fruits of Sorbus aucuparia L.in the ratio 3:3:3:2:2:1:1:1. Standardization of the extract was carried out in terms of flavonoids. There was used the Pauls index (PI), the index of antiulcerous effect (AE) and morphological evaluation of the gastric mucosa. In experiments on Wistar line rats with butadion-induced injury of the stomach mucosa the marked antiulcerous activity of the multicomponent plant extract in a dose of 150 mg/kg has been established. The administration of the extract restricted the formation of ulcer defects which were more marked on the 14th and 21st days of observation. Such morphological features of activation of regenerative processes against the background of introduction of the given remedies as the new growth of vessels, cleanliness of the wound, inhibition of inflammatory reaction and active granulation were more expressed in rats treated with the extract and less distinct in rats treated with the preparations of comparison. The findings of the research confirm the expressed antiulcerous effect of the plant extract which is comparable to the effects of befunginum and ranitidine. The complex of biologically active substances contained in the extract promotes the acceleration of the ulcer healing due to its versatile influence on the main pathogenesis mechanisms of the given pathology. The findings of the research have shown that the tested extract has good prospects for creation of medicinal preparations for prophylaxis and treatment of gastric ulcer.
