Background. Orostachys spinosa (L.) Sweet. – a perennial plant of a wide habitat and contains various metabolites (amino acids, flavonoids, polysaccharides, etc.). Extracts from the aerial part of the plant are used in traditional medicine as an anticonvulsant and sedative.The aim of the work. To study the neuroprotective effect of O. spinosa in cholinergic deficiency.Materials and methods. The studies were carried out on 52 Wistar rats. The animals were administered scopolamine (1 mg/kg) daily for 21 days, followed by O. spinosa dry extract per os at a dose of 100 mg/kg for 14 days. On the day 32, the animals developed a conditioned passive avoidance reflex (CPAR), the integrity of which was checked after 1, 24 and 72 hours; on the day 35 they were tested in an “open field”. On the day 36, biochemical and histological studies of the brain were carried out.Results. It has been established that O. spinosa, against the background of scopolamine intoxication, reduces the anxiety of animals, stimulates exploratory activity in the open field test, improves the production and preservation of the CPAR, and also reduces the number of functionally inactive neurons (pyknotic and shadow cells) in the cerebral cortex. The extract reduces the lactate/pyruvate ratio by 47 %, intensifies the activity of mitochondrial complexes I and II by 54–64 %, and increases the concentration of adenosine triphosphate by 1.6 times compared to the control. O. spinosa exhibits antioxidant properties by reducing malondialdehyde and increasing the activity of catalase, glutathione peroxidase and glutathione reductase in the brain.Conclusion. O. spinosa dry extract has a neuroprotective effect in cholinergic deficiency. The studied extract exhibits antioxidant properties and stimulates energy processes in the brain
Аннотация.Необходимость разработки гепатопротективных средств определяется распространением повреждений печени вирусной этиологии, увеличением частоты токсических медикаментозных и алкогольных повреждений печени, отсутствием достаточно эффективных средств лечения.Растительное сырье -траву копеечника альпийского -заготавливали в период цветения в Байкальском регионе в 2020-2021 гг.Собранное сырье соответствовало ВФС 42-1998-85 «Трава копеечника».Экстракт сухой из данного сырья получали спиртоводной экстракцией.В полученном экстракте в значительных количествах содержались флавоноиды, ксантоны, полисахариды и другие биологически активные вещества.Эксперименты проведены на белых крысах-самцах линии Wistar с массой 170-200 г.В экспериментах на белых крысах установлена выраженная желчегонная активность экстракта сухого hedysarum alpinum L. в дозах 50-300 мг/кг.Наряду с этим полученный экстракт копеечника оказывает благоприятное влияние на биохимический состав желчи
Проведено исследование гастропротективного действия экстракта сухого Ferulopsis hystrix (Bunge) Pimenov на моделях эрозивно-язвенных повреждений желудка у крыс линии Вистар. Экстракт сухой F. hystrix в дозах 100 и 200 мг/кг и препарат сравнения плантаглюцид в дозе 300 мг/кг вводили животным внутрижелудочно в течение 7 дней, последнее введение осуществляли за 1 ч до ульцерогенного воздействия. Установлено, что экстракт сухой F. hystrix в дозе 200 мг/кг оказывает гастропротективное действие, превосходящее таковое для препарата сравнения плантаглюцида. На фоне введения животным экстракта F. hystrix в дозе 200 мг/кг в слизистой оболочке желудка не развиваются эрозии и полосовидные язвы, тогда как у крыс контрольной группы они выявляются у 100 и 50 % животных, соответственно. Индекс Паулса для точечных кровоизлияний в 2,7 раза ниже показателя контрольных животных. На фоне индометациновой гастропатии F. hystrix в дозе 200 мг/кг уменьшает среднее число эрозий и полосовидных язв в 2 и 5 раз (p ≤ 0,05), а индекс Паулса для них соответственно в 2 и 20 раз ниже по сравнению с контролем (p ≤ 0,05). Установлено, что экстракт F. hystrix в дозах 100 и 200 мг/кг усиливает темп желудочной секреции на 32 и 52 % (p ≤ 0,05), общую кислотность на 39 и 67 % (p ≤ 0,05). Полученные результаты позволяют рекомендовать экстракт сухой F. hystrix для дальнейших исследований в качестве перспективного растительного гастропротективного средства.
В статье представлены результаты исследования стресспротективной активности экстрактов, полученных из корневищ и надземной части растения Rhaponticum uniflorumL., основными действующими веществами которого являются фитоэкдистероиды и флавоноиды. Разработанные экстракты - практически нетоксичные вещества. Их курсовое введение в экспериментально-терапевтической дозе 100 мг/кг оказывает стресспротективное действие при остром эмоциональном стрессе, уменьшая выраженность признаков триады Селье благодаря ограничению гиперактивации центральных стрессреализующих систем организма. Периферические механизмы адаптогенного действия экстрактов R. uniflorum связаны с ингибированием процессов свободнорадикального окисления. The article gives the research findings of the stress-protective activity of extracts derived from rhizomes and aerial part of Rhaponticum uniflorum L., in which phytoecdysteroids and flavonoids are the main active ingredients. The developed extracts are practically non-toxic substances. Their course administration in experimental-therapeutic dose 100 mg/kg has the stress-protective effect in acute emotional stress decreasing the manifestation of the Selye’s triad signs due to inhibition of hyperactivity of central stress-realizing bodily systems. Peripheral mechanisms of adaptogenic effect of the Rhaponticum uniflorum extracts are connected with inhibition of free radical oxidation processes.
The antioxidant activity of the dry extract from the complex plant remedy has been studied in model systems in vitro. The complex plant remedy includes the following species of medicinal plants: Serratula centauroides (L.), Bergenia crassifolia (L.) Fritsch, Rosa davurica Pall., inula helenium L., Echinacea purpurea (L.) Moench. It has been established that the plant remedy under study inhibits the oxidation of the biological substrate preventing from the destruction of beta -carotene (IC50=24.3 mu g/ml)and yolk lipoproteids (IC50=65.8 mu g/ml) and having membrane stabilizing effect in peroxide (IC50=0.97 mu g/ml) and osmotic hemolysis (IC50 =0.11 mu g/ml) of erythrocytes. The complex remedy manifests the radical binding activity for 2,2-dipheny1-1- picrylhydrazyl (DPPH') (IC50 =61.4 mu g/ml), superoxide anion-radical (IC50 =28.6 mu g/ml), nitrogen oxide (IC50 =55.3 mu g/ml), also for Fe2+ (IC50 =639.3 mu g/ml). The marked antioxidant activity of the tested remedy is due to the complex of biologically active substances (flavonoids, tannins, polyphenolic compounds, phenol carbonic acids, ecdysteroids, etc.) contained in its components.
A polyextract consisting of the extracts of the five title plants shows a marked prostate-protective effect in experimental chronic prostatitis. The pharmacotherapeutic efficacy of the phytoextract gives a basis to recommend its use in complex treatment and prophylaxis of prostatitis.
A chromatography–spectrophotometry method is developed for determining total ecdysteroids in terms of 20-hydroxyecdysone in Serratula centauroides (L.) and Fornicium unflorum (L.) raw materials, based on the purification of extracts from the plant raw materials with aluminum oxide and subsequent spectrophotometric determination relative to a 20-hydroxyecdysone reference sample. Ecdysteroids were determined in plants by HPLC; rhapontisterone and turkesterone are found in F. uniflorum for the first time.
The aim of the work was to estimate a wound-healing effect of soft drug products from the Pentaphylloides fruticosa L. shoots on the model of planar skin-fascial wound. The liniment was developed with the use of the NanoDeBee 30 device and the ointment was developed with the use of pharmaceutical technology. Experiments were carried out on white male Wistar rats. The skin injury was simulated by inflicting planar wounds 2.0×2.0 cm in size. The 5%-liniment and 5% ointment from P. fruticosa were put on the wounds of the first and second experimental groups respectively. In the third group the wounds were treated with glycerin-vaseline ointment. Bioptate of the wound surface was taken on the 7th, 14th and 21st day. Histological preparations were studied using standard methods. Cell elements of the first group animals were displayed as fibroblasts and fibrocytes; it was noted high level of collagen organization that testified the effective staging of the wound process in this case. The amount of fibrocytes and fibroblasts and ratio between juvenile and mature forms of collagen-producing cells vary in the first and second groups. Biopsy material obtained from the animals of the third group only on the 21st day corresponded to the picture observed in animals of the 1st group on the 14th day. The findings of the study verify that the use of the product obtained on NanoDeBee device provides the earlier forming of the scar as compared with the ointment developed using pharmaceutical technology.
The aim of the work was to evaluate the influence of the liniment from Pentaphylloides fruticosa L. on lipid peroxidation in rats with experimental diabetes mellitus accompanied by wound process.The liniment was developed with the use of NanoDeBee30 device. Experiments were carried out on white male Wistar rats. Experimental diabetes mellitus was simulated by a single intraperitoneal introduction of alloxan in a dose of 100mg/kg. The skin injury was simulated by inflicting planar wounds 2.0×2.0cm in size. 5mg of the 5%-liniment from Pentaphylloides fruticosa was put on the wounds of the first experimental group once a day for 14 days. The sec-ond experimental group was treated by the preparation of comparison – balsamic liniment. The wounds of the control group were treated with glycerin-lanolin ointment. On the 7th, 14th and 21st day the concentration of TBA-active products in the blood serum was determined to estimate the intensity of lipid peroxidation processes; the contents of reduced glutathione and catalase activity were determined to evaluate the antioxidant status. The results of the experiments verify the marked curative effect of the liniment from Pentaphylloides fruticosa L. on the healing of the wounds on the background of experimental diabetes mellitus in rats
This work is devoted to the research of pharmacotherapeutic efficacy of multi-component herbal medication “Polinoophit” which was developed on the basis of Tibetan formulation for patients suffering from mild cognitive impairment with early stages of chronic circulatory insufficiency. In our research we observed patients with diseases of central nervous system of vascular and mixed genesis; with neurosis-like and soft affective mnestic disorders. The medication under investigation consists of the roots of Baikal skullcap (ScutellariabaicalensisGeorgi), anomalous peony (Paeoniaanomala L.), garden burnet (Sanguisorbaofficinalis L.), herb of waterwort (Gnaphaliumuliginosum L.) and other plants which are used in Tibetan Medicine. The therapeutic efficacy of the phyto-remedy was estimated according to the data of patients’ physical examination with the usage of the neurological status investigation, the haemodynamic parameters, the estimation of patients’ symptoms, the dynamics of cognitive, behavioral and emotional-affective deteriorations. The examination of these indices revealed the decrease of all symptoms except the behavioral ones. The received yields approved the theoretical expectations that this agent has a nootropic efficacy.
The experiments were carried out on white Wistar rats. The probable embryotoxic, teratogenic and fetotoxical influences of the plant remedy "Pancreophyt" at the doses of300 (1/10 DL50) and 600 mg/kg (1/5 DL50) were studied. The findings have shown that all indices of embryo lethality (the number of implantation sites, the number of alive fetus and the number of resorptions) in female white rats treated with the "Pancreophyt" do not differ significantly from the indices of the control group. The mass and craniocaudal dimensions of the fetus in the experimental group corresponded to the indices in the control group. In the second set of experiments the influence of the "Pancreophyt" (300 mg/kg) on the postnatal development of white rat offspring was studied. It was revealed that the death of infant rats was noted only on the 1st day of observation and was 4,7%; the liveweight gain and linear size of infant rats do not differ from the indices of those in the control group.
