High-fat diet (HFD)-induced obesity is a crucial risk factor for metabolic syndrome, mainly due to adipose tissue dysfunctions associated with it. However, the underlying mechanism remains unclear. This study has used genetic screening to identify an obesity-associated human lncRNA LINK-A as a critical molecule bridging the metabolic microenvironment and energy expenditure in vivo by establishing the HFD-induced obesity knock-in (KI) mouse model. Mechanistically, HFD LINK-A KI mice induce the infiltration of inflammatory factors, including IL-1β and CXCL16, through the LINK-A/HB-EGF/HIF1α feedback loop axis in a self-amplified manner, thereby promoting the adipose tissue microenvironment remodeling and adaptive thermogenesis disorder, ultimately leading to obesity and insulin resistance. Notably, LINK-A expression is positively correlated with inflammatory factor expression in individuals who are overweight. Of note, targeting LINK-A via nucleic acid drug antisense oligonucleotides (ASO) attenuate HFD-induced obesity and metabolic syndrome, pointing out LINK-A as a valuable and effective therapeutic target for treating HFD-induced obesity. Briefly, the results reveale the roles of lncRNAs (such as LINK-A) in remodeling tissue inflammatory microenvironments to promote HFD-induced obesity.

浙东锣鼓是中国传统吹打乐的代表性乐种之一,它依附于民间礼俗仪式场域,是仪式行为的音声表达.在浙东一带,凡岁时节庆、庙会社火、婚丧仪礼、祭祖祈神、开渔谢洋等民间礼俗活动皆离不开锣鼓.浙东锣鼓之所以生生不息流传至今,是因为它具有礼俗文化基质和礼俗仪式功能,并在传承中不断发展.民众在礼俗仪式音乐中,获得了传统文化的认同感和本土文化身份的归属感.
Event Detection (ED) aims to identify event trigger words from a given text and classify it into an event type. Most of current methods to ED rely heavily on training instances, and almost ignore the correlation of event types. Hence, they tend to suffer from data scarcity and fail to handle new unseen event types. To address these problems, we formulate ED as a process of event ontology population: linking event instances to pre-defined event types in event ontology, and propose a novel ED framework entitled OntoED with ontology embedding. We enrich event ontology with linkages among event types, and further induce more event-event correlations. Based on the event ontology, OntoED can leverage and propagate correlation knowledge, particularly from datarich to data-poor event types. Furthermore, OntoED can be applied to new unseen event types, by establishing linkages to existing ones. Experiments indicate that OntoED is more predominant and robust than previous approaches to ED, especially in data-scarce scenarios.
Organelles use specialized molecules to regulate their essential cellular processes. However, systematically elucidating the subcellular distribution and function of molecules such as long non-coding RNAs (lncRNAs) in cellular homeostasis and diseases has not been fully achieved. Here, we reveal the diverse and abundant subcellular distribution of organelle-associated lncRNAs from mitochondria, lysosomes and endoplasmic reticulum. Among them, we identify the mitochondrially localized lncRNA growth-arrest-specific 5 (GAS5) as a tumour suppressor in maintaining cellular energy homeostasis. Mechanistically, energy-stress-induced GAS5 modulates mitochondrial tricarboxylic acid flux by disrupting metabolic enzyme tandem association of fumarate hydratase, malate dehydrogenase and citrate synthase, the canonical members of the tricarboxylic acid cycle. GAS5 negatively correlates with levels of its associated mitochondrial metabolic enzymes in tumours and benefits overall survival in individuals with breast cancer. Together, our detailed annotation of subcellular lncRNA distribution identifies a functional role for lncRNAs in regulating cellular metabolic homeostasis, highlighting organelle-associated lncRNAs as potential clinical targets to manipulate cellular metabolism and diseases.
保障公务员考录的实质价值与工具价值均衡实现是公务员考录制度建设的基本出发点与目标追求.建立在管理思维基础上的现行公务员考录制度,在保障公务员考录价值均衡实现方面尚存在明显的体制、机制缺陷,对考试手段的科学运用尚存在明显的局限.有鉴于此,应立足治理思维对公务员考录制度进行改革,重构公务员考录主体的权利、权力及其实现机制,探索实行两轮制公务员考录机制;重构公务员考录流程、重新界定公务员考录笔试与面试功能;改革公务员考录考试手段运用机制,对考试命题机制与面试机制进行改革,实现命题管理统一性与命题组织分类化的统一以及面试效度、信度、公平性的统一.如此,有助于最大限度地提升公务员考录价值的均衡实现程度,最大限度地提升公务员考录的科学性与有效性.

Organelles entail specialized molecules to regulate their essential cellular processes. However, systematically elucidating the subcellular distribution of functional molecules such as long non-coding RNAs (lncRNAs) in tissue homeostasis and diseases has not been fully achieved. Here, we characterized the organelle-associated lncRNAs from mitochondria, lysosome, and endoplasmic reticulum (ER), respectively, and revealed the diverse and abundant distribution of lncRNAs. Among them, we identified mitochondrial lncRNA Growth-Arrest-Specific 5 (GAS5) as a tumor suppressor in maintaining cellular energy homeostasis. Mechanistically, energy stress-induced GAS5 modulated mitochondria TCA flux by declining metabolic tandem association of FH-MDH2-CS, the canonical members of the TCA cycle. Remarkably, the expression of GAS5 negatively related with levels of its associated mitochondrial metabolic enzymes and breast cancer development. Together with the detailed functional annotations, this subcellular lncRNA identification revealed the human cell’s inquisitively complex architecture, aiding in the development of new strategies for the clinical application of organelle-associated lncRNAs.
Genetic factors are considered to play a critical role in patients with early-onset Parkinson's disease (EOPD). The genetic spectrum of EOPD patients has been extensively investigated in Caucasian populations but rarely in the Chinese population. In this study, a total of 21 unrelated Chinese EOPD patients were enrolled. Multiplex ligation-dependent probe amplification assay and whole-exome sequencing were performed, followed by Sanger sequencing. Detailed clinical features were presented. Two novel likely pathogenic variants (p.Q648X in ATP13A2 and p.N521fs in PINK1) and 10 previously reported Parkin pathogenic variations (exon 2 deletion, exon 3-4 deletion, exon 4 deletion, exon 6-7 deletion, exon 7 deletion; p.G284R, p.G329 V, p.R366W, p.N428fs, p.M458 L) were identified in 9 out of 21 (42.86%) patients. The frequency (33.33%) of Parkin variations is much higher in our cohort than that in the East Asian population. The patient carrying the ATP13A2 variant showed no response to levodopa treatment. Our findings broaden the genetic spectrum and clinical features of EOPD patients.
There is some doubt that all nodules <8mm are really mainly benign and that simple follow-up is adequate in all cases. The purpose of this study is to create a predictive model for the diagnosis of benign and malignant small pulmonary nodules. This was a retrospective case-control study of patients who had undergone pulmonary nodule resection at the Zhejiang University Jinhua Hospital. Patients with pulmonary nodules of <= 10mm in size on chest high-resolution computed tomography were included. Patients' demographic characteristics, clinical features, and high-resolution computed tomography findings were collected. Logistic regression and receiver-operating characteristic analysis were used to create a predictive model for malignancy. A total of 216 patients were included: 160 with malignant and 56 with benign nodules. Nodule density (odds ratio [OR]=0.996, 95% confidence interval [CI]: 0.993-0.998, P=.001), vascular penetration sign (OR=3.49, 95% CI: 1.39-8.76, P=.008), nodule type (OR=4.27, 95% CI: 1.48-12.29, P=.007), and incisure surrounding nodules (OR=0.18, 95% CI: 0.04-0.84, P=.03) were independently associated with malignant nodules. These factors were used to create a mathematical model that had an area under the receiver-operating characteristic curve of 0.744. Using a cut-off of 0.762 resulted in 63.1% sensitivity and 75.0% specificity. This study proposes a pulmonary nodule prediction model that can estimate benign/malignant lung nodules with good sensitivity and specificity. Mixed ground-glass nodules, vascular penetration sign, density of lung nodules, and the absence of incisure signs are independently associated with malignant lung nodules.

在现代管理中重提德性伦理建设具有重要的意义,它既能为现代管理培养德才兼备的管理主体,又能为现代管理实践提供重要的人文精神支撑.中国自古以来极为重视德性管理伦理建设,传统德性管理伦理在伦理精神塑造、组织考核选拔、民间舆论控制等德性养成机制方面都具有独特优势,但由于传统德性管理伦理产生于封建社会,其重私德而轻公德、强调服从而忽略平等的价值观,与现代管理伦理的公共精神背道而驰.随着我国社会主义市场经济的发展,社会结构调整使德性伦理在现代管理中的作用逐渐消解,遭遇了价值认同危机及养成机制不足等困境,现代社会管理仍需借鉴传统德性伦理来促进管理之“善”的实现.在新的时代背景下,需要对德性管理伦理的德性内容及养成机制加以重建,在社会主义核心价值观的引领下,通过构建个人私德、职业道德和社会公德体系,重建德性自修、家教训导、组织约束、道德楷模等德性养成机制,加强对组织管理者的道德约束和道德激励,才能使德性伦理在现代管理中的积极作用得到有效发挥.
本文采用波浪订正的Ekman模型,研究分析了三种Stokes漂流近似公式(单波公式、e指数公式、Phillips谱近似公式)对海洋表层流场估算的影响.海表总流场由海表面高度(SSH)数据计算的地转流和海浪模式WAVEWATCH Ⅲ输出结果计算的非地转流组成,并采用拉格朗日浮标观测数据对计算结果进行了验证.研究表明,随着Stokes漂流近似公式精度的提高,其计算的拉格朗日流速更接近于谱积分公式的计算结果,更贴近拉格朗日浮标观测数据.与谱积分公式计算的海表拉格朗日流速相比,单波公式的平均相对偏差为0.0834,e指数公式的平均相对偏差为0.0392,Phillips谱近似公式的平均相对偏差为0.0101,说明Phillips谱近似公式在不同风速下均能对谱积分公式有良好的近似效果.在低风速条件下,由Stokes漂流近似公式精度引起的海洋表层流场估算误差可以忽略不计,但随着风速增加,由近似公式精度引起的偏差逐渐变大,此时应该选择Phillips谱近似公式计算Stokes漂流,来减小误差.
浙东儿歌童谣是流传于浙东沿海地区,具有鲜明的海洋民俗文化色彩和吴越方言特色,反映儿童生活,富有童真趣味的本土原生性民间歌谣.自古以来,在浙东民间,关于海洋环境、气象、节令、渔汛、海产、航行、捕捞、贸易、娱乐、游戏、神话、传说、信仰、习俗,具有启蒙教育意义的歌谣,源源不断地涌现出来,从精神层面滋养着浙东沿海渔民的后代.海洋文化基质是浙东儿歌童谣生成的根源性因素.
Text representations is a key task for many natural language processing applications such as document classification, ranking, sentimental analysis and so on. The goal of it is to numerically represent the unstructured text documents so that they can be computed mathematically. Most of the existing methods leverage the power of deep learning to produce a representation of text. However, these models do not consider about the problem that text itself is usually semantically ambiguous and reflects limited information. Due to this reason, it is necessary to seek help from external knowledge base to better understand text. In this paper, we propose a novel framework named Text Concept Vector which leverages both the neural network and the knowledge base to produce a high quality representation of text. Formally, a raw text is primarily conceptualized and represented by a set of concepts through a large taxonomy knowledge base. After that, a neural network is used to transform the conceptualized text into a vector form which encodes both the semantic information and the concept information of the original text. We test our framework on both the sentence level task and the document level task. The experimental results illustrate the effectiveness of our work. (C) 2017 Elsevier Ltd. All rights reserved.
