本文研究不同浓度盐碱环境下羊蹄中3种药用成分大黄素、大黄酚和大黄素甲醚的含量变化,并探讨盐碱胁迫对3种药用成分的影响.采用超声波辅助法制备供试品溶液,应用华谱UnitaryC18色谱柱(250mm×4.6mm,5μm),以甲醇(A)-0.1%磷酸水溶液(B)为流动相;梯度洗脱,流速为1.0mL/min,柱温设置35℃;检测波长254nm,进样量20μL;建立羊蹄指纹图谱,测定3种药用成分的含量,分析盐碱胁迫对其3种药用成分的影响和中药指纹的整体变化.结果显示,在盐胁迫下,羊蹄根部大黄素、大黄酚和大黄素甲醚都在盐浓度为200mmol/L时含量达到最大,且显著高于对照;羊蹄茎叶大黄素含量在盐浓度为40mmol/L时相对较高,但低于对照处理,大黄酚和大黄素甲醚含量在盐浓度为160mmol/L时最大,且高于对照处理.在碱胁迫下,羊蹄根部大黄素、大黄酚和大黄素甲醚在碱浓度为20mmol/L时含量最大,且高于对照;茎叶大黄素含量在碱浓度为10mmol/L时含量最高,但显著低于对照处理,大黄酚含量在碱浓度为20 mmol/L时显著高于其他处理,大黄素甲醚在碱浓度为10 mmol/L时显著高于其他处理.盐碱胁也影响大黄素、大黄酚和大黄素甲醚在羊蹄根部和茎叶当中的分布,盐胁迫对茎叶的影响比较明显,碱胁迫对根部的影响更为显著.中药指纹相似度分析和聚类分析结果显示,盐碱胁迫同样会影响羊蹄次生代谢产物的形成,碱胁迫影响大于盐胁迫.从而得知盐碱胁迫下药用植物羊蹄不仅能够存活,而且在适宜的盐碱浓度下3种有效成分的含量也会提高.
采用超声波法提取北五味子果实中的果胶成份,咔唑-浓硫酸显色法测果胶得率,并应用正交试验设计对其提取工艺参数进行了优化.试验结果表明:各因素对北五味子果胶提取的影响顺序依次为超声功率>提取温度>料液比>提取时间,最佳工艺参数为超声功率200W,提取温度为65℃,料液比为1∶30,提取时间为50 min,果胶提取率为6.32%.
生物信息学方法预测出miRNA-375可能的靶基因为DIAPH2和QKI,实时荧光定量法检测了靶基因在1日龄、1~7月龄军牧1号白猪睾丸组织中的表达量.结果表明,3月龄前猪睾丸组织中miRNA-375表达量极低,4~7月龄表达水平逐渐上调,差异显著.预测靶基因DIAPH2在1~7月龄呈显著下调趋势,OKI基因在1~7月龄睾丸组织中表达水平呈显著上调趋势.经SPSS13.0分析可知,不同时期睾丸组织中miRNA-375与2个预测靶基因DIAPH2和QKI的相关系数r分别是-0.396(P<0.05),0.411(P<0.05),2个预测靶基因DIAPH2和QKI之间的相关系数是0.151(P>0.05),靶基因DIAPH2和QKI无明显联系,初步证明DIAPH2可能是miR-NA-375的靶基因,为探索猪睾丸组织发育提供了一定的研究基础.
Epimedii Folium is a traditional Chinese medicine,which is adopted in Chinese Pharmacopoeia.It has initially been recorded in Sheng Nong's Herbal Classic.The Epimedii Folium have the effects of warming kidney,strengthening muscles and eliminating rheumatism.Modern pharmacology has proven that Epimedium herb has the ability of anti-aging,enhancing immunity,anti-arrhythmia,anti-inflammatory and antipathogenic microorganism.Currently,the analysis of Epimedium is mainly focused on the chemical constituents at home and abroad.Five species of the Epimedium,which are adopted in Chinese Pharmacopoeia,are taken as representatives to overview the analysis methods of the constituents in the Epimedium in recent years.These methods provide a foundation for the study of the bioactive constituents.
To compare the detection of the blood physiological and biochemical indexes in lactation and mature stages for pigs from three different genetic backgrounds,104 healthy pigs in lactation(at one-month old) and mature stages(at seven-month old) were selected under the same environment,including 41 Chinese experimental miniature pigs,38 Junmu No.1 white pigs,and 25 Duroc,and then anterior vena cava or ear vein blood samples were collected to analyze 28 blood indexes.A general linear model analysis of variance was used to analyze the measured data using SAS9.2 statistical software.The results showed that most of the blood physiological and biochemical indexes were affected by developmental stages,breeds,and the interactions of them.The developmental stages had the greatest impact on blood physiological and biochemical indexes,followed by the interactions of breeds and developmental stages,breeds and gender.The differences between breeds on the levels of red blood cell(RBC) count,hemoglobin(Hb)content,and hematocrit(HCT) might be related to the behavioral characteristics of pigs,and creatinine(CREA),triglyceride(TG),and total serum cholesterol(CHOL) were related to the content of pig lean meat and fat percentage.
MicroRNAs(miRNAs) are short(-22 nucleotides) non-coding RNA molecules that play important roles by repressing translation or cleaving RNA transcripts.Some reports showed miRNA-34b is an important regulator in germ cell differentiation process of male humans and mice.We detected the different expressions of miR-34b of testicular tissue by real time quantitative PCR(RT-PCR) in Junmu No.1 white pig among the whole developmental stages.The results showed the expressions of miRNA-34b keep very low level before 3-month-old,rising dramatically from 5-month-old,reaching a peak at 7-month-old,and then descend apparently.Overview the expression trend of miRNA-34b in the whole sexual development processes,it runs a rule that first rising and then descending.The conclusion is anticipated that miR-34b keep a high possibility correlation with sexual development of male pig.
目的 通过实验筛选出马勃中促大鼠成纤维细胞增殖的有效成分.方法 利用极性不同的溶剂分离提取马勃的主要组分,作用于体外培养的成纤维细胞,观测各组分对成纤维细胞生长的影响,筛选出作用最强的组分,并利用质谱等方法确定有效成分.结果 80%乙醇提取物对成纤维细胞增殖作用明显,并筛选出对成纤维细胞增殖作用较强的有效成分.结论 分析得到其有效成分可能为萜类化合物.
The interactions of icariin and icarisideⅠ with bovine serum albumin(BSA) under simulated physiologic conditions was investigated by fluorescence spectroscopy.The binding constants(KA),number of binding sites(n) of icariin / icarisideⅠ with BSA,and other thermodynamic parameters(△G,△H and △S) were calculated.The experimental results showed that both icariin and icarisideⅠ significantly quenched the intrinsic fluorescence of BSA through a single static process by forming a ground state complex.The values of KA and n at different temperatures(17 ℃,27 ℃,37 ℃) indicated that icarisideⅠ bound BSA stronger than icariin could.The thermodynamic measures suggested that the forces for icariin to bind BSA were mainly the hydrogen bonding and van der Waals force,whereas the hydrophobic and electrostatic interactions were the leading forces to form icarisideⅠ and BSA complex.In addition,conformational changes of BSA were observed from the synchronous fluorescence spectra upon the binding of icariin and icariside I.
The EDNRB complementary deoxyribonucleic acid was molecularly cloned from the normal silver-black fox lung tissue by reverse transeription-polymerase chain reaction. Sequence analyses indicated that the cloned cDNA of EDNRB was 1 636 bp in length which contained the entire open reading frame of 1 332 bp encoding 443 amino acid residues. Alignment analysis indicated that the nucleotide sequence of the coding region showed a high homology with the EDNRB sequences of dog(99.0%), pig(90. 7~//oo), human(89.5%), cattle(89. 1 ~//0), rabbit (86.9%o) and mouse(82.8%), respectively. The deduced amino acid resides contained a signal peptide and seven putative trans- membrane domains which showed highly conserved amino-acid resides with that of other species above mentioned. Results of the tissue expression of EDNRB gene in silver-black fox indicated that EDNRB gene expressed in six of seven tissues. It expressed with the higher abundance in brain, liver and lung, but it was a little present in heart, spleen and muscle. No expression in stomach tissue was detected in this experiment. The data provide a valuable foundation for studying on polymorphisms of EDNRB gene and their association with different phenotypes of coat color in the silver-black fox.
Pork quality is a quantity trait,which is controlled by several major genes and many candicate genes.In order to detect the polymorphism of meat quality major gene of Junmu No.1 white pig,the halothane gene(RYR1),Rendement napole(RN),and heart fatty acid binding protein gene(H-FABP) were checked by PCR-RFLP among the 400 Junmu No.1 white pig.The result indicated that,the frequency of N type RYR1 gene was 0.908 3,n was 0.091 7;in H-FABP gene 5 'region HinfⅠ,the frequency of H was 0.688 8,h was 0.311 2;in H-FABP intron 2 of the Hae Ⅲ locus,the frequency of D was 0.478 8,d was 0.521 2;no polymorphism evidence was found in intron 2 of H-FABP MSPⅠ sites and Rendement Napole gene.
目的:制备时间分辨荧光免疫分析双功能螯合剂4,7-二(氯磺酸基苯基)-1,10-菲咯啉-2,9-二羧酸(BCPDA).方法:合成物BCPDA的结构由熔点、元素分析、红外光谱和核磁共振谱确证;采用标记曲线法测定标记物中蛋白质的含量;应用荧光分光光度计测定Eu3+-BCPDA的荧光强度;采用放免法测定标记蛋白质的活性.结果:BCPDA与Eu3+结合后可测量到的最低浓度为1×10-15mol*L-1 标记蛋白质的相对生物活性高于80%.结论:成功地制备时间分辨荧光免疫分析双功能螯合剂BCPDA,并建立Eu3+-BCPDA-蛋白质标记物中蛋白质浓度的测定方法.
