Objective To evaluate the spatial and temporal variations of fibroblast growth factor 2 (FGF2) in oxygen induced retinopathy,and to explore its action mechanism.Methods C57BL /6J pups were used and assigned to experimental group and control group,experimental group was OIR model.For P17,use the pave piece of retina with HE staining and retrobulbar injection of fluorescein isothiocyanate-dextran injection to create the OIR model; P13,P15,P17 to check the retinal FGF2 expression of time and space change by WEST BLOT.Results OIR model was constructed successful,FGF2 expression showed a rising trend in the P13,P15,P17 experimental group,significantly higher than the control group.Conclusions OIR may inhibit of the oxygen induced retinopathy by increasing FGF2 expression.
目的:观察中药联合雷珠单抗治疗视网膜分支静脉阻塞(Branch retinal vein occlusion,BRVO)的临床疗效.方法:回顾分析2014年1月至2014年12月在中山大学中山眼科中心诊断为视网膜分支静脉阻塞的患者(24只眼),其中单纯玻璃体腔注射雷珠单抗治疗12例(12只眼),中药联合雷珠单抗治疗12例(12只眼).治疗前后记录患者最佳矫正视力及光学相干断层扫描及眼底荧光血管造影情况,随访1月.结果:治疗1月后应用雷珠单抗治疗总有效率为67%,中药联合治疗总有效率为86%.治疗前最佳矫正视力为0.25±0.12,治疗1月后雷珠单抗组、中药联合组最佳矫正视力分别为0.38±0.13 (P=0.021),0.45±0.16 (P=0.021).治疗前平均黄斑中心凹厚度为465±84,治疗1月后雷珠单抗组、中药联合组平均黄斑中心凹厚度为276.10±89um (P=0.030),285±72um (P=0.010).结论:活血利水法具有一定治疗视网膜分支静脉阻塞的临床疗效.
目的 探讨凉血活血药物对视网膜新生血管的干预及其作用机理.方法 通过氧诱导C57BL/6幼鼠的方法建立视网膜新生血管的动物模型.随机分为正常对照组、模型对照组、凉血活血药物高、低剂量组.计数视网膜切片中突破血管内界膜的内皮细胞核数;免疫组织化学检测VEGF在视网膜中的表达情况.结果 氧诱导后,幼鼠血管内皮细胞核数目明显增加,VEGF明显上调,提示造模成功;各治疗组血管内皮细胞核数目减少、VEGF下调,提示凉血活血药物对视网膜新生血管有抑制作用,其机制可能与通过对VEGF的抑制有关.结论 凉血活血药物可能通过抑制VEGF的表达实现抑制视网膜新生血管的形成.
Objective To study the intervention of liangxuehuoxue drugs on retinal neovascularization and the mechanism of action.Methods Retinal neovascularization model was established in young C57BL/6 mice by hyperoxia induction.Mice were divided randomly into 4 groups:normal control group,model group,liangxuehuoxue drugs high dose group and low dose group.The number of nucleus of endothelial cells which broke through the endangium in retinal section,and the expression of HIF-1 and NF-κB in retina was detected by immunohistochemistry.Results Comapred with normal control,after hyperoxia induction,the number of nucleus of endothelial cells significantly increased,and the expression of HIF-1 and NF-κB was upregulated,and the difference was statistically significant(P<0.05),suggesting the retinal neovascularization model was established successfully.Compared with model group,in liangxuehuoxue drugs groups,the number of nucleus of endothelial cells significantly decreased,and the expression of HIF-1 and NF-κB was downregulated,and the difference was statistically significant(P<0.05).Conclusion Liangxuehuoxue can inhibit the retinal neovascularization,and the mechanism may be related to downregulating the expression of HIF-1 and NF-κB related to the angiogenesis.
Objective To study the therapeutic liangxuehuoxue effect on the supression of retinal neovascularization.Methods Retinopathy was made by subjecting postnal mice to hyperoxic conditions taken retinal angiogram with ink.Quantified the degree of hyperoxia-induced neocascularization in serial paraffin cross sections by counting the number of vascular cell nuclei on vitreal side of the internal membrane.the expression of HIF-1 and NF-кB in the oxygen-induced retina with immuhistochernistry were observed.Results After the mouse were induced by oxygen,the endothelial cell nuclei increased and HIF-1、NF-кB went up obviously,The level of HIF-1、NF-кB descended.Conculsions oxygen-induced intravitreal retinal angiogenesis by down-regulated HIF-1and NF-кB related to the angiogenesis.
Objective To study the therapeutic effect liangxuehuoxue traditional Chinese medicines on the supression of ECV304 by serum pharmacological method,and to explore the interventional target for treatment of neovessels related eye diseases.Methods ECV304cells were cultured with medicated serum with different concentrations of liangxuehuoxue medicines.The proliferation rate of endothelial cells was assayed with MTT methods,the level of VEGF was tested.Result Medicated serum with liangxuehuoxue medicines at concentration of 20% could inhibit the proliferation of ECV304 cells and the expression of VEGF.Conclusion Liangxuehuoxue traditional Chinese medicines cna inhibit the ECV304 cells proliferation and the expression of VEGF,and this may be related to inhibition of neo vessels.

目的 通过建立增生性玻璃体视网膜病变(PVR)动物模型,研究化瘀散结片对兔眼PVR增生膜中结缔组织生长因子(CTGF)表达的影响.方法 采用"玻璃体腔内注入体外培养兔视网膜色素上皮(RPE)细胞"法建立PVR动物模型,用免疫组织化学法观察化瘀散结片对增生膜中CTGF表达的影响.结果 模型组增生膜CTGF的表达及明显高于正常组(P<0.01);化瘀散结片组与模型组及阳性药物对组相比较,增生膜CTGF表达明显降低(P<0.01).结论 CTGF参与了PVR增生膜的形成;化瘀散结片可能通过降低CTGF表达来抑制PVR增生膜的形成.
眼底新生血管是目前严重威胁视力的疾病,本文从中医络脉理论的角度探讨眼底新生血管的病因病机和治疗原则。认为眼底新生血管属孙络范畴,眼新生血管性疾病多属于孙络疾病。与脏腑和经络的虚衰有关,从而采用络虚通补,活血理气及辛味通络之法。
目的 研究高度近视患者的中医证候特点.方法 167例高度近视患者根据年龄分为10~29岁组,30~49岁组,50~69岁组和70~89岁组,所有患者填写中医证候量化表及CRF表,并行统计学分析.结果 高度近视患者年龄与中医症状积分有相关性,随着年龄增加,中医症状逐渐加重.结论 高度近视患者中医证候演变具有一定规律,10~29岁组患者主要表现为气阴两虚的症状,30~49岁组,50~69岁组和70~89岁组患者随年龄增加在气阴两虚症状存在的同时肝肾亏虚的症状逐渐加重.
AIM To analyze the risk factors of age, sex, course, best corrected visual acuity (BCVA), diopter and fundus features of high myopes with progressive high myopia. METHODS A total of 167 patients with high myopes were categorized into four groups: group 1, age 10-29 years; group 2, age 30-49 years; group 3, age 50-69 years and group 4, age 70-89 years. The refractive errors of all patients were measured without cycloplegia with an autorefractometer. Data of the spherical equivalent (SE) of the refractive errors in diopters (D) and fundus examined by direct ophthalmoscope were used in statistical analysis. RESULTS The number of female was statistically larger than that of male (P<0.01), also the disease course was correlated to the age. The visual acuity of high myopes significantly decreased as they grew older including the higher incidence of lacquer cracker, submacular hemorrhage, Fuchs spots, chorioretinal atrophy. CONCLUSION Female maybe a risk factor of high myopia, advanced age is an important factor of visual acuity decreased. High myopes ought to be treated early to delay the progress of myopia and development of macular degeneration.
目的 观察结缔组织生长因子(CTGF)在实验性增生性玻璃体视网膜病变(PVR)增生膜中的表达,探讨CTGF在PVR视网膜增生膜形成过程中的作用.方法 采用Nobuyo的方法从兔视网膜中分离视网膜色素上皮(RPE)细胞并进行体外培养和传代.第3代RPE细胞制备密度为1.1×105/mL的细胞悬液.32只日本大耳白兔,取8只作为正常对照组,其余24只采用玻璃体腔内注入RPE细胞悬液的方法建立PVR动物模型.动物分别于造模后7、30、60d处死并摘除眼球,每次处死8只动物.光学显微镜下观察视网膜增生膜的组织学改变,免疫组织化学法检测PVR视网膜增生膜中CTGF的表达.结果 造模后5d检眼镜下可见视网膜增生组织开始形成,增生膜随时间的推移逐渐增厚.组织学检查显示,造模后7d兔视网膜表面可见红染的条索状和网状胶原纤维,并可见大量的增生细胞分布其中.光学显微镜下可见视网膜内界膜变厚、粗糙、断裂或结构不清,视网膜各层结构欠清.免疫组织化学法检测表明,正常对照组在玻璃体及视网膜内未见CTGF的特异性染色.造模后7d,CTGF主要表达于视网膜表面增生细胞;造模后30d,CTGF主要表达于增生的细胞及增生的胶原纤维组织中;造模后60d,CTGF主要表达于增生的胶原纤维组织中.结论 CTGF在实验性PVR动物模型中呈高表达,提示CTGF参与了PVR增生膜的形成.
Objective To investigate the expression of hepatocyte growth factor (HGF) in experimental animal model of periretinal membranes (PRM) of proliferative vitreoretinopathy (PVR). Methods The retinal pigment epithelium (RPE) cells of the cultivated rabbits were separated outside their bodies,then the PVR animal model was established by injecting these RPE cells into vitreous. Using immunobistochemical technique the expression of HGF was evaluated. Results The expression of HGF in 7 days after the modeling group was higher than normal group; in 30 days it reached highest,and in 60 days it was slightly lower than than in 30 days. Conclusion Expression of HGF in periretinal membranes of experimental animal model of PVR is significantly higher than normal groups,which indicates that HGF takes part in the formation of periretinal membranes of PVR.

目的研究化瘀散结片对实验性增生性玻璃体视网膜病变的防治作用特点,探讨其作用机理。方法采用眼穿通伤加注血法制备实验性增生性玻璃体视网膜病变(experimental proliferative vitreoretinopathy,PVR)模型,进行眼底及常规组织病理学观察,ELISA法检测玻璃体中血小板源性生长因子的浓度。结果(1)化瘀散结片高、低剂量治疗组增生膜形成较模型对照组缓慢,且增生程度较轻,增生膜面积及周长总和低于模型对照组(P<0.01)。(2)化瘀散结片高低剂量治疗组视网膜神经节细胞数量,均明显高于模型对照组(P<0.05或<0.01)。(3)化瘀散结片高低剂量治疗组玻璃体腔内PDGF浓度明显低于模型对照组(P<0.01)。结论化瘀散结片能有效防止PVR的形成与发展,其作用机理与减少玻璃体中血小板源性生长因子浓度有关。
