Purpose To investigate the protective effect and character of dl-praeruptorin A (Pd-Ia) on focal cerebral ischemia in mice.Methods Transient focal cerebral ischemia in mice was induced by middle cerebral artery occlusion for 1.5 h.Pd-Ia was administered intraperitoneally either with multiple doses (1,5 and 10 mg/kg) at 0.5 h before ischemia or single dose (5 mg/kg) at 0.5 h and 1 h before ischemic,the same time of ischemia,the same time of reperfusion,or 0.5 h and 1 h after reperfusion respectively.Neurological deficit score,infarct volume,brain edema,the activities of SOD and the contents of MDA were determined.Results Pretreatment with multiple doses (5 and 10 mg/kg) of Pd-Ia at 0.5 h before ischemia or single dose (5 mg/kg) of Pd-Ia at 0.5 h before ischemia,at the same time of ischemic,at the same time of reperfusion and 0.5 h after reperfusion significantly attenuated neurological deficit score,decreased infarct volume and alleviated brain edema,and the treatment at the time of reperfusion had the most marked effect.Pd-Ia (5 or 10 mg/kg) can significantly enhance the activities of SOD and lower the contents MDA.Conclusion dl-praeruptorin A has a neuroprotective effect on the injury in the acute phase of transient focal cerebral ischemia in mice,with optimal doses of 5 mg/kg and the optimal therapeutic time point of the same time of reperfusion.
Objective To investigate synergism of celecoxib combining with photodynamic therapy(PDT) on C6 glioma cells and increasing apoptosis in vitro.Methods Cell proliferation was evaluated by cell surviving curve,electron microscopy and flow cytometry(FCM).Results Sensitivity enhancemnt ratio(SER) of 10% surviving fraction was 1.34.From electron microscopy we could see that apoptosis number in combined group were the most than the others.The combined group could increase apoptosis in vitro,up to 64.3% in comparison with PDT group(4.5%) and celecoxib group(15.3%) by FCM.Conclusions Celecoxib could enhance photodynamic effect and induce apoptosis of C6 glioma cells in vitro.
近年来,很多研究显示环氧合酶-2(COX-2)与胶质瘤密切相关.Celecoxib属非甾体类抗炎药(NSAIDs),是COX-2选择抑制剂,对于胶质瘤作用方面的研究未见报道.为观察Celecoxib对胶质瘤的抑瘤效果,本实验应用MTT法、细胞生长曲线和流式细胞仪等方法,检测Celecoxib对C6胶质瘤增殖的影响.
Objective To investigate celecoxib’s function of suppressing the growth of C6 glioma and inducing apoptosis in vivo.Methods C6 glioma mice were divided into control and celecoxib group,used HE staining、curve of tumor volume、PGE_(2) and electron microscope to observe the effect of celecoxib on C6 glioma. Results HE staining and electron microscopy showed vascular injury,cell necrosis and apoptosis in celecoxib group,PGE_(2) and tumor volume between two groups through t-test had a statistical mening,P(0.05).Conclusion Celecoxib can inhibit COX-2 ,decrease synthesize of PGE_(2),there was a inhibition of proliferation on C6 glioma by celecoxib.
Objective To investigate celecoxib’s function of inducing C 6 glioma cells apoptosis in vitro. Methods Cell apoptosis was evaluated by acridine orange(AO),electron microscope and flow cytometry(FCM). Results From AO and electron microscope we could see apoptosis number of C 6 glioma cells depending on concentration of celecoxib,FCM found apoptosis of celecoxib 50μmol/L and 12.5μmol/L group were 15.32%and 5.83%,respectively. Celecoxib could increase apoptosis in vitro,there was a dose-dependent tendency. Conclusion Celecoxib can induce apoptosis of C 6 glioma cells.
恶性纤维组织细胞瘤(Malignant fibrous histocytomaMFH)是来源于间叶组织的恶性肿瘤,常见于中老年人,多发于四肢、躯干和腹膜后等部位;发生于头颈部、颅内很少见.
目的:研究转移相关基因CD44V6在甲状腺癌组织中的表达及其临床意义.方法:应用免疫组化SP方法测定45例甲状腺癌组织中的CD44V6的表达阳性率,分析其阳性率与肿瘤转移的关系.结果:CD44V6表达阳性率在淋巴结转移组(85.0%)明显高于无淋巴结转移组(48.0%)(P<0.05);在乳头状腺癌中的表达率为75.0%,滤泡腺癌组为68.8%,在髓样癌中及未化分癌中未见阳性表达;45~60岁病人组CD44V6表达阳性率高于其它年龄段.分析CD44V6阳性表达与临床分期的关系,发现随分期增加而增加,CD44V6的表达阳性率在Ⅰ、Ⅱ、Ⅲ期分别为64.3%,77.8%,100.0%;死亡的8例中有6例CD44V6表达阳性.结论:CD44V6表达与甲状腺癌的颈淋巴结转移密切相关;与临床分期、年龄、预后及病理类型有一定关系.
目的研究转移相关基因VECF-C在甲状腺癌中的表达及其临床意义.方法应用免疫组化方法检测45例甲状腺癌组织中的VEGF-C的表达情况.结果 VECF-C的阳性率在淋巴结转移组(90%)明显高于无淋巴结转移组(80%),P<0.01.在乳头状腺癌中的表达率(91.67%)高于其它病理类型,VECF-C在良,恶性肿瘤中均有表达.Ⅰ,Ⅱ,Ⅲ期的表达率分别为82.14%,100%,100%,随分期而增加.死亡8例中有6例VEGF-C呈阳性表达.结论VEGF-C与甲状腺癌的淋巴结转移密切相关,与分期及预后有一定的关系.
目的:总结鼻腔鼻窦内翻性乳头状瘤外科治疗经验.方法:收集自1976年2月至1997年10月间行外科治疗的26例鼻腔鼻窦内翻性乳头状瘤的临床资料并进行回顾性分析.结果:26例鼻腔鼻窦乳头状瘤中8例有癌变,癌变率为30.8%.均行外科治疗.随访2~5年局部复发5例,其中3例死亡(均为乳头状瘤合并有癌变者).局部复发率为19.2%(5/26).结论:术中连同上颌骨内壁切除及筛窦整块切除是治疗该病的最佳方法,内窥镜鼻窦切除术适用于局限性病变.
本文报道自1979年12月~1994年7月,50例水平垂直喉部分切除创面修复结果.①甲状软骨膜修复11例;②甲状软骨膜及其附丽肌肉修复30例;③骨肌瓣修复9例.结果:三种修复方法发音清晰响亮者分别为4例(36.4%),22例(73.3%)和8例(88.9%).全组发音清晰响亮者计34例(68.0%),略低哑13例(26.0%),低哑3例(6.0%).平均恢复正常经口进食时间分别为17.2、13.1和10.2天.拔管率分别为81.8%(9/11)、80.0%(24/30)和77.8%(7/9).认为骨肌瓣是修复水平垂直喉部分切除创面的最佳选择.
