本实验首先双酶切含有Hind3和NcoI酶切位点的PRRSV目的基因的质粒pUC-57—PRRSV N gene,利用酶切的目的基因与表达载体pET-28a,,构建重组表达载体pET-28a- PRRSV N gene,为实现目的蛋白原核表达,建立针对该病的快速、有效的检测方法研发安全高效的疫苗等针对猪群PRRSV进行后续研究的奠定基础和前提条件。

To study the influences of various environmental factors on the biofilm formation of Campylobacter jejuni,the orthogonal tests were used to synthetically analyze the influences of the air condition,type of medium and temperature on the biofilm formation with the optical microscopy and scanning electron microscopy.The results showed that the influences of three factors in descending order were the air condition,temperature,and type of medium.The biofilm-forming capacities were also different with each of these factors at different levels.The results indicate that the appropriate aerobic and oligotrophic environments were more conducive to the biofilm formation.

The US10 gene was expressed in pET-32a(+)/Rosetta(DE3) pLys system using the DNA of Duck enteritis virus(DEV) C-KCE as temple,US10 gene ORF was amplified by polymerase chain reaction(PCR).Then the recombinant protein of US10 gene induction with IPTG by SDS-PAGE analysis showed that the object protein is mainly expressed as inclusion bodies.Using the Ni-NTA Purification System,the recombinant protein immunized to rabbits was purified to make the antiserum.The titer of the antiserum tested by agar diffusion reaction was 1∶4.The titer of the antiserum tested by ELISA was 1∶102 400.The prepared antiserum has a high specificity with the western-blot analysis,and the indirect immunofluorescence(IIF) test proved that the antiserum could react with DEV US10 encoded product,which lays a foundation for further research on molecular structure and function.

To investigate the effect of sodium selenite poisoning on the immune functions in mice.The experimental animals were divided into control group(0.00 mg/kg·d),low dose(0.14 mg/kg·d),middle dose group(0.54 mg/kg·d) and high dose(2.16 mg/kg·d).Sodium selenite groups mice were killed after continuous intragastric feeding for 30 days.Phagocytosis of chicken red blood cells by macrophage and hemolytic plaque assay were used to examine the phagocytic function of mouse peritoneal macrophages and numbers of antibody producing cells.Sodium selenite poisoning significantly resuced the phagocytic function of mouse peritoneal macrophages and the number of antibody producing cells.It has dose-dependent correlation.Sodium selenite poisoning inhibits the function of nonspecific immunity in mice,which presents the apparent dose effect relationship.
Single-factor experiment was designed,and 160 natal Jilin white geese and Nongan geese of the similar body weight were chosen as experimental animal. The geese of either variety were then divided into 4 treatment groups randomly and NRC-based dietary was fed to them according to their two developmental stages,0 to 4 weeks old,and 5 to 8 weeks old.By slaughter experiments and RT-PCR(reverse transcription polymerase chain reaction) technique,the expressions of MC4R(melanocortin receptor-4)and its receptor in liver,breast and leg muscles during different developmental stages were detected to further study the influence of different genetic basis on levels of MC4R mRNA and its receptor.The results show that the meat performance of different genetic basis and the expression level of MC4R mRNA in muscle show a correlation of some extent.The expression level of MC4R mRNA in leg or in breast muscle of Jilin white geese is higher than that in Nongan geese.Live weight,leg muscle weight,breast muscle weight,daily gain weight of Jilin white geese are higher than those of Nongan geese.

为了研究更快、更加准确分离空肠弯曲杆菌的方法,试验同时采用常规生化法对由12份猪盲肠内容物中分离的可疑菌进行检测,得到了6个阳性样品,经特异性PCR检测,2个样品可定性为空肠弯曲杆菌,检测时间为7 d;而对所得菌株直接进行PCR检测,检测结果相同,检测时间为2 d。结果表明:PCR可以快速、特异性地检测空肠弯曲杆菌,与常规生化检测法相比是一种更有效的方法。

Total nitrogen(TN),total phosphorus(TP),potassium permanganate index(CODMn),transparency(SD) and chlorophyll five items were selected to evaluate the eutrophication degree of Mata Lake by using index method of comprehensive nutrition evaluation.The pollution status of Mata Lake were obtained by drawing figures to compare the annual average values of major indexes,including total nitrogen,ammonia nitrogen,chemical oxygen demand(COD),5 days biological oxygen demand(BOD) and potassium permanganate index.The reasons for the eutrophication of Mata Lake was studied based on analysis and measures to improve the eutrophication were put forward.
