In this paper,mapping analysis of the genes that are exsisted in the 16S rDNA V3 districts of duodena,jejuna,ilea,caeca and cola of the rabbits which have been weaned 10,20 and 30 days respectively,is carried out by using PCR-DGGE thechnology.Through the clear specificity and generality of bands cutting rubber recycling of DNA and cloning sequencing,the comparisons of the structure and the diversity between bacterial communities,which are in different parts of the intestines and belong to the rabbits of differdent days have been done.Also,their community members have been ascertained.More importantly,it analyzed the influence of the days that the rabbits had been weaned and the different parts of the intestines on the structure and diversity of the micropopulation.On the basis of the DGGE map of the bacterial community in the intestines,it could be seen obviously that different parts of the intestines,as well as the days that the rabbits had been weaned,had great effects on the structure and diversity of the bacterial community.Specifically,there were varying degrees of dynamic changes: with the increase of the days,the types of the bacterial communities in the duodena and caeca were more varied;those in the jejuna and cola had experienced a transversion of scarce to abundant to scarce,while those in the ilea was on the contrary.From the DGGE map,we could conclude that the dominant bacteria in different parts of the intestines were Firmicutes,Bacteroidetes,Clostridia,Azohydromonas sp,Lachnospiraceae and many uncultured bacteria and so on.In conclution,the research results indicated: the days that the rabbits had been weaned and the different parts of their intestines did have effects on the structure and diversity of the bacterial community.
In this study,18S rDNA gene was extracted from wild Nyctereutes procyonoides matschie's meat of ear,and amplified by polymerase chain reaction(PCR) method,then cloned and sequenced.A lengh of 1831 bp was cloned,the sequences had been sent to GenBank and registered,comparison of the sequence with that of other animals 18S rDNA gene was carried out through BLAST.The results showed that the homologies of the nucleotide sequence were above 90% with Nyctereutes procyonoides matschie,and 18S rDNA gene was conserved in evolution,which could be used as phylogenetic analysis of the important reference.
The pit-1 has been implicated in regulation of body weight. Its gene sequence and protein function exhibit high evolutionary conservation and have been studied extensively in humans, mice, pigs and chickens. Mink pit-1 is nearly identical to dog pit-1 but little is know about its function. Thus, researchers investigated whether polymorphisms in th pit-1 exon4 and exon6 were associated with mink growth traits. Five population of mink were assessed for growth traits for use in statistical correlation analysis with genomic sequence data. Two SNPs were detected in the pit-1 exon4 and exon6 of each population: 43C>T and 143 A>G. The 43C>T mutation produced three genotypes: AA, BB and AB. Statistical analysis of variance revealed that 43C>T polymorphism was associated with Body Weight (BW) and Carcass Weight (CW) in black mink; the 143A>G mutation produced three genotypes as well: CC, DD and CD; statistcal analysis of variance revealed that the 134A>G polymorphism was associated Body Weight (BW) in. black mink (p<0.05). The interaction of 43C>T and 143A>G was discussed through combination genotype analysis. The result showed that the combined genotype had significant association with Body Lenght (BL) in sapphire mink and Body Weight (BW) and Carcass Weigth (CW) in black mink (p<0.05). The findings suggest that the pit-1 gene could be a qualitative trait locus or linked to a major gene that affects growth traits in. mink.
A total of 80 1-month-old meat rabbits were randomly assigned to four groups and each group contained 20 rabbits.The dietary treatments were basal diet(contain 30% chinensis),garlic straw instead of 5%,10%,and 15% chinensis diet,respectively.The trial lasted for 7 days for adaptation and 35 days for sampling.At day 7,21 and 35,the samples were collected,and daily gain,F/G,relative weight of immune organs,total protein and serum IgG and IgA was determined.The results showed that the performance was significantly affected by treatments.Average daily gain in 5%-15% garlic straw significant increased than that in the control,and F/G significant decreased than that in the control(P0.05).The treatment plus garlic straw had the tendency of increasing relative weight of immune organs,total protein and serum IgG and IgA.Especially,at day 21 the relative weight of spleen and at day 35 the serum IgA were significant higher than those in the control(P0.05).Therefore,garlic straw also play a role in improving the immune function of growing rabbit.
Two hundred and eighty 50-weeks Green-eggshell chickens were randomly allotted to 4 treatments,each group was assigned 5 replications and 14 layers per replication.The control group was provided with corn-soybean meal basal diet,the trial groups were respectively fed the basal diet supplemented with 0.5%,1.0% and 1.5% Leonurus heterophyllus Sweet,in order to observe the influence to biochemical parameters,hormone and ovary in Green-eggshell chickens.The results were showed as follows: compared with the control group,1.0% and 1.5% Leonurus heterophyllus Sweet added to Green-eggshell chickens could significantly decrease TC,TG and LDL,apparently increase HDL,E2,FSH and ovarian relative weight.Leonurus heterophyllus Sweet could affect fat metabolism,reduce the content of blood lipid.For saving cost,the most appropriate volume was 1.0%.
This experiment was conducted to obtain the suitable ISSR-PCR reaction system of Columba domestica by orthogonal design.Columba domestica was chosen to study the affecting factors of ISSR-PCR amplification.Orthogonal design of five factors(Mg2+,dNTPs,primer,Taq DNA polymerase,template) at four levels was used to establish the suitable ISSR-PCR system by primer ISSR807 in Columba domestica,and the suitable anneal temperature was yielded from gradient PCR on temperature.The results showed that the optimized ISSR-PCR system(25 μL reaction volume) in Columba domestica was proved to be: 10×PCR Buffer 2.5 μL,2.6 mmol/L Mg2+,0.2 mmol/L dNTPs,0.4 μmol/L primer,0.5 U Taq DNA polymerase,DNA template 1.2 ng/μL;and suitable anneal temperature was 56.2 ℃.According to the optimum ISSR-PCR reaction system,the results lay a foundation for the analyses of genetic diversity,germplasm resources and genetic relationship of Columba domestica.
The incidence of trichophagy in five varieties of minks (jinzhou-black mink,coffee mink,silver blue mink,Red-eye white mink and American shorts mink) was counted in Dalian mink nursery.The incidence rate of different varieties,sex,and age implanted of exogenous melatonin or not were calculated and analyzed,respectively to explore the causes and relative factors of mink trichophagy.The results showed that variety difference significantly affected the incidence rate of trichophagy (P0.05) in these 5 kinds of minks,with the highest incidence rate of 12.61% occurred in coffee mink;sex also greatly affected the happen of disease,female mink's incidence was obviously higher than that of the male mink (P0.05);the age had no significant influence on the incidence (P0.05);the while,exogenous melatonin implanting could significantly (P0.05) reduce the incidence rate of trichophagy.
