建立了当归芍药散的HPLC-DAD指纹图谱分析方法,确定了部分特征峰的归属.采用高效液相色谱法,色谱柱为Welchrom C18柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.05%磷酸水溶液(梯度洗脱),检测波长为230 nm,流速为l mL/min.从建立的当归芍药散指纹图谱中,对照出当归芍药散中的21个特征峰8个来自当归,11个来自白芍,13个来自川芎,4个来自白术,2个来自茯苓,3个来自泽泻,同时鉴定出7个峰的化学结构.当归芍药散指纹图谱的结果为鉴别当归芍药散的质量提供了依据,也为阐明各部位的药效物质基础提供了参考.
建立人血浆中盐酸马尼地平浓度测定的LC/MS/MS法,进行人体药动学研究。方法:通过24名健康志愿者口服受试制剂,采用单次空腹(20 mg(20 mg/片))和高脂餐后(20 mg(20 mg/片))两周期双交叉试验及单次空腹10 mg (10 mg/片)和连续多次给药10 mg(10 mg/片)试验设计,测定人血浆中盐酸马尼地平的浓度。结果:主要药动学参数分别为:单次空腹口服10 mg,20 mg盐酸马尼地平后:Cmax3.52±1.023 ng · mL-1,Tmax1.264±0.372 h,AUC0-36h13.399±3.451 ng/ml · h;Cmax5.59±1.863 ng · mL-1,Tmax1.417±0.359h,AUC0-36h23.697±10.179 ng/ml · h;单次餐后口服20 mg盐酸马尼地平后:Cmax9.307±2.288 ng · mL-1,Tmax2.292±0.782h,AUC0-36h39.674±7.362 ng/ml · h;连续多次给药10 mg盐酸马尼地平后Cmax3.454±1.316 ng · mL-1,Tmax1.288±0.38 h,AUC0-36h16.427±4.554 ng/ml · h。结论:本方法灵敏度高,结果准确,盐酸马尼地平在大部分人体内的过程符合二室开放模型,从而指导临床合理用药。
Objective To qualitatively analyze the main chemical constituents in Danggui Shaoyao San,which is a famous traditional Chinese prescription mainly composed of Angelica sinensis and peony,and recorded in Synopsis of Golden Chamber.Methods High performance liquid chromatography-diode array detector(HPLC-DAD) was used,with Welchrom XB-C18(250 mm×4.6 mm,5 μm) as the column,and acetonitrile-0.05% phosphoric acid solution(gradient elution) as the mobile phase.The scan range of DAD ranged from 200 to 400 nm.Results By comparing the experimental results to related literatures,the three-dimensional atlas could show the main chemical constituents in Danggui Shaoyao San,including ligustrazine,albiforin,paeoniflorin,ferulic acid,ligustilide,atractylenolide Ⅰ,and alisol B monoacetate.Conclusion HPLC-DAD can qualitatively detect the main chemical constituents in Danggui Shaoyao San,and provide an experimental basis for its pharmacodynamics studies.
Objective:To establish a method for determination of isosorbide-5-mononitrate in human plasma by UPLC-TOF-MS.Methods:ACQUITY UPLC Hss T3(1.8 μm,2.1 mm×50 mm) column was adopted with the mobile phase consisted of acetonitrile-H2O(containing 0.1% formic acid) with gradient elution at flow rate of 0.50 mL·min-1.The Waters TOF-MS equipped with an atmospheric pressure ESI source was used as a detector and was operated in negative ion mode by full scaned.Results:Chromatograms showed no endogenous interference peaks with the respective blank human plasma samples.Each analysis was completed within 3.0 min.The calibration was linear in the concentration ranges of 0.02-1.00 μg·mL-1.The extraction recoveries were 98.6%-109.0%.The intra-day and inter-day RSDs were less than 8.7% and 10.6%,respectively.Conclusions:The method is suitable for clinical investigation of pharmacokinetics and therapeutic dose for isosorbide-5-mononitrate,which offers advantages of sensitivity,quickness and repeatability.
Objective To observe the effects of Danggui Shaoyao Powders(DSP),a traditional Chinese medicine composed of angelica,peony,chuanxiong,poria cocos,alisma rhizoma,and atractylodes macrocephala,in the treatment of dysmenorrhea,and to explore its effects on the levels of arginine vasopressin(AVP).Methods The effects of DSP on twisting counts were observed in rats with dysmenorrhea induced by intraperitoneal injection of oxytocin.The contents of prostaglandin F2α(PGF2α) in the uterus,and AVP in the plasma,the hypothalamus and the uterus tissues were assayed by enzyme linked immunosorbent assay.Results DSP significantly reduced the contents of PGF2α in the uterus,and increased the contents of AVP in the hypothalamus,while reduced the contents of AVP in the rat uterine and plasma.Conclusion One of the mechanisms of DSP in treating dysmenorrhea is related with its intervening the AVP system in rats.
Objective: To study the pharmacokinetic characteristics of Sodium Ferulate Capsules and Tablets in healthy volunteers and to evaluate the bioequivalence of them.Methods: In a randomized,crossover and self-control study,20 healthy volunteers were orally administrated Sodium Ferulate Capsules and Tablets(100 mg respectively).The plasma concentrations were determined by the HPLC-UV.Pharmacokinetic parmeters were calculated and the bioequivalences were evaluated by DAS2.1 software.Results: The main pharmacokinetic parameters of Sodium Ferulate were as follows: Cmax(1.021±0.286) μg/ml and(1.066±0.397) μg/ml,tmax(0.388±0.078) h and(0.371±0.083) h,t1/2(0.887±0.431) h and(0.824±0.404) h,AUC(0-t)(0.613±0.172) μg·h/ml and(0.614±0.235) μg·h/ml.Conclusion: The test and reference sodium ferulate are bioequivalent.
目的:研究当归芍药散对慢性应激抑郁模型大鼠行为及中枢单胺类递质的影响。方法:应用分养和长期不可预见性温和应激造成大鼠抑郁模型,并观察当归芍药散对该模型大鼠的治疗作用。定期测定各组大鼠糖水嗜好程度;敞箱实验方法测定大鼠水平及垂直活动;RP-HPLC-荧光法测定动物大脑额叶皮质中神经递质的含量。结果:与正常对照组比较,模型组大鼠糖水消耗、垂直活动、水平运动显著减少,大脑额叶皮质中NE,DA,5-HT,5-HIAA的含量均明显降低;当归芍药散高剂量可明显升高DA,5-HT,5-HIAA的含量(P<0.01),低剂量仅对5-HIAA含量有明显增高(P<0.01),对NE含量无明显改变。结论:初步表明当归芍药散具有抗抑郁作用,机制可能与调节脑内单胺类神经递质DA,5-HT,5-HIAA的含量有关。
Objective:To compare diffirent extraction mathods for the content paeoniflorin and ferulic acid of dangguishaoyaosan. Methods:Using RP HPLC,compared with water extraction、water extraction by precipitation with ethanol and extracted by ethanol reflux three methods on paeoniflorin and ferulic acid content of impact.Results:extracted by ethanol reflux﹥water extraction﹥extraction by precipitation with ethanol.Conclusion:The results showed that the extracted by ethanol reflux on the content of paenflorin and ferulic acid is the highest in the three kinds of extraction method of dangguishaoyaosan.
