Breast milk consumption reduces the incidence of necrotizing enterocolitis (NEC) in preterm infants compared to formula. Branched-chain fatty acids (BCFAs) are present in breast milk but not in most formulas intended for preterm infants. We aimed to determine the composition of BCFAs in the breast milk of mothers with preterm infants, and to understand the impact of gestational age at birth and stage of lactation on BCFA content. The main BCFAs in preterm breast milk were iso-14:0, iso-15:0, anteiso-15:0, iso-16:0, iso-17:0, and anteiso-17:0. Breast milk BCFAs as a percent of total fatty acids (g per 100 g, %) were significantly different across lactation stages, with the highest concentration in colostrum, followed by transitional and mature breast milk (median: 0.41, 0.31, and 0.28%, respectively, p < 0.05). Lower BCFAs in preterm breast milk compared to term breast milk may have been related to maternal intake, or the ability of the mammary gland to extract BCFA from plasma, or differences in mammary gland BCFA synthesis. BCFAs were mainly in the sn-2 position (52-65%), similar to palmitic acid. Overall, preterm and term breast milk BCFAs were similar and showed specific concentration patterns, resembling 16:0 with respect to sn-2 positional distribution. BCFAs were reduced with lactation stage, similar to highly unsaturated fatty acids.
Fatty acid (FA) is the major energy resource in breast milk, which is important for infant development. FAs profiles with sn-2 positional preference were an important part of triacylglycerols due to their better availability. This profile is still not replicated in artificial formulas. This study quantified the FAs profile of total and sn-2 position in human breast milk samples from 103 healthy volunteers during colostrum, transitional, and mature stages. Multicomponent analysis showed significant differences in FAs profiles of different lactation periods, due to that with relative percentage less than 1%. Linoleic acid (LA), mostly located at the sn-1,3 positions of TAGs, was more common in the milk of Chinese women than in western women. The majority of the breast milk did not meet the standard for the ratio of LA/α-linolenic acid for infant formula. FAs related to brain development, mainly at sn-2 in TAGs, were enriched in colostrum. Capric and lauric acids were enriched in transitional and mature breast milk, and capric acid showed sn-1,3 selectivity in TAGs. This study will aid the development of infant formula containing TAGs more similar to human breast milk.
支链脂肪酸(BCFA)是胎脂的重要成分,胎儿吞食羊水可获取胎脂中的BCFA,从而参与发挥肠道的防御作用.早产儿吞食胎脂不足,肠道缺乏BCFA,需要外源补充,羊毛脂是潜在的BCFA的天然来源.对胎脂和羊毛脂的BCFA组成进行分析.结果表明:胎脂含有复杂的脂质种类,主要是非极性脂质;胎脂总脂质和胎脂甘油三酯中的BCFA含量都比较高,依次为17.16%、16.12%;胎脂中共检测到11种BCFA,其中异构C12∶0、异构C14∶0、异构C16∶0、反异构C15∶0的含量较高;羊毛脂的脂肪酸中含有约48.53%的BCFA,包括了胎脂中大部分可测得的BCFA,因此羊毛脂可以模拟胎脂中的BCFA,具有在早产儿配方食品中应用的价值.
Objective. Ovarian cancer is one of the most serious disease in female reproductive system. Platinum is the first-line drug for the treatment of ovarian cancer, while the resistance of platinum drug in clinical hindered the relief ovarian cancer. Our previous study found that decreased FOXO3a might be a poor prognosis in human ovarian cancer. In this research, we study whether FOXO3a was involved in the mechanism of platinum drug resistance. Methods. The CCK-8 and FACS analysis were used to monitor the survival of ovarian cancer, and the FOXO3a expression was detected by western-blot. Results. We found that FOXO3a expression upregulated significantly in A2780 compared with A2780/DDP cells with the treatment of platinum. Moreover, overexpression of FOXO3a in ovarian cancer inversed the platinum resistance in ovarian cancer. Conclusion. These observations reminded that the role of FOXO3a might be one of the critical mechanisms in developing platinum drug resistance in ovarian cancer. (C) 2018 IMSS. Published by Elsevier Inc.
Objective:To study the effects of Callicarpa nudiflora capsule combined with medroxyprogesterone acetate in the treatment of intrauterine cavity deciduas residue in patients received early medical pregnancy termination.Methods:84 patients with small amount of intrauterine cavity residue after medical abortion were randomly divided into observation group and control group,each with 42 cases with equal numbers.All patients were treated with conservative medical treatment for 10 d.The observation group was given medroxyprogesterone acetate on the basis of control group.The duration of vaginal bleeding,the disappearance of abnormal echo in the uterine cavity and the cure rate were compared between the two groups.Results:Compared with the control group,the observation group stopped bleeding rate was significantly higher than the control group 10 d after treatment,the difference was statistically significant (P < 0.05).Compared with the control group,the ultrasound intrauterine abnormal echo disappeared rate of observation group after treatment was higher than that of control group,the difference was statistically significant (P < 0.05).Compared with the control group,the cure rate of observation group (95.23%) was significantly higher than the control group (21.42%),the difference was statistically significant (P < 0.05).Conclusion:The curative effect of Callicarpa nudiflora capsule combined with medroxyprogesterone acetate for the treatment of intrauterine cavity residue after medical abortion was preferable.
目的 分析无锡市妇幼保健院医学SCI收录论文发表及被引情况,为其科研管理部门、科研人员了解其科研发展动态提供量化的参考依据.方法 应用WEB OF SCIENCE数据库对2011年1月~2015年12月期间以无锡市妇幼保健院为作者单位发表的SCI收录论文及被引情况进行统计分析.结果 该院2011年1月~2015年12月年共发表SCI收录论文98篇,被引67篇,被引率为68.37%,被引频次471次,均篇被引频次4.81次;单篇最高被引26次,发表量前5名期刊中,均篇被引频次最高杂志为Medical Oncology,均篇被引9.57次,Medical Oncology,Tumor Biology及Scientific Reports三个期刊的被引率均为100%.结论 该院发表SCI收录论文被引率、均篇被引频次较高,有一定的学术价值.
Branched chain fatty acids (BCFA), as the main component of trace fatty acids in human milk, are proved to be functional compounds for infant intestinal health and cancer inhibition which make it necessary to investigate their role in the diet and in humans. However, a comprehensive existing form of BCFA in lipids has not been well defined. Therefore, the objective of the present study was to characterize BCFA content in total lipid, neutral lipid and polar lipid, with a focus on their positional distribution in triglycerides of lipids in nature including human colostrum (All experiments were approved by the Ethics Committee of Jiangnan University, China JN No. 20121203-0120, All procedures of the study were carried out in accordance with ethical standards of the committee on human experiment and with Helsinki Declaration of 1975, as revised in 2008. All these mothers were explained what this study is about and were given their written consent.), goat milk, cow milk, and fish oil. A routine quantification for positional distribution of minor components like BCFA in lipids using GC/EI-MS was therefore developed in this paper. An adequate resolution of all BCFA and other FA in cow milk fat was attained, as shown in Figure 1. BCFA content in the polar lipids (PLs) of human colostrum and their positional distribution in lipids of human colostrum, ruminant milk and fish oil were reported for the first time. BCFA in PLs of human colostrum was approximately 0.36%, much less than that in neutral lipids (NLs) (0.99%). About 68% of total BCFA in human colostrum was located in the sn-2 position, which was much more than 29.66% in cow milk, 34.20% in goat milk and 5.19%~11.89% in fish samples (Figure 2). These results indicate the source of bacterial BCFA in humans and animals may come from mesophylls, which is different than in fish. Most BCFAs in fish are transferred into PLs, whereas most of them in animals and human body may be transferred into NLs. Additionally, the diversity of BCFA distribution in triacylglycerides illustrates that the source of BCFA in human and rumen animals is different from fishes, and there is relatively more BCFA settled in sn-2 in human than in other lipids. Thus, we anticipate that the physiological significance of its distinctive distribution in lipids in human should be studied and in the future this may be taken into consideration as designing BCFA supplement food. The ability to detect fetal chromosomal abnormalities associated with recurrent miscarriage precisely and effectively is increasingly being exploited to develop and improve CNV assay. G-banded karyotyping assay is the predominant assay clinically, relying on enriched fetal cells obtained invasively. A lack of less invasive and efficient methods with the same accuracy in CNVs associated with recurrent miscarriage has long prevented in-depth understanding of etiology of recurrent miscarriage and making generalized policy in prognosis of recurrent miscarriage. Here we show a ligation and PCR based multiplex-CNVplex assay, presenting 100% sensitivity with conventional karyotyping in numerical CNVs and demonstrating its improved resolution in detecting microdeletions and microduplications based on 76 patients experiencing recurrent miscarriage. Specifically, both Karotyping and CNVplex assay detect 27 autosomal trisomies, three 45, X monosomy and three polyploidy, which accounts for 95% of CNVs associated with recurrent miscarriage. CNVplex also obtained novel structural abnormalities in 8p23.1 region which includes genes associated with cardiovascular development. Besides, the cost of CNVplex is significant lower than Karotyping and other available assay, owing to its high throughput multiplexing with four dyes probe system. Thus, the advantages of CNVplex should be of benefit in specially detecting fetal CNVs associated with recurrent miscarriage. More generally, we anticipate that the ability demonstrated here to detect predominate CNVs associated with recurrent miscarriage at a reduced cost and turnaround time will benefit the further research in understanding the genetic cause of recurrent miscarriage and offer an appealing alternative option to the emerging non-invasive prenatal screening of fetal aneuploidy using cell-free DNA.
Labor-intensive karyotyping is used as the reference standard diagnostic test to identify copy number variants (CNVs) in the fetal genome after recurrent pregnancy loss. Our aim was to present and evaluate a novel molecular assay called CNVplex that could potentially be used as an alternative method to conventional karyotyping for diagnosing fetal chromosomal abnormalities associated with recurrent pregnancy loss. Using karyotyping as the reference standard, CNVplex was performed to identify fetal chromosomal abnormalities in the chorionic villus samples from 76 women experiencing at least two pregnancy losses. Its diagnostic accuracy, sensitivity, and specificity were evaluated to detect aneuploidies associated with recurrent pregnancy loss. Turnaround time and costs of CNVplex were also measured. Diagnostic accuracy of CNVplex in aneuploidies that are associated with recurrent pregnancy loss was 1.0 (95% CI 0.94–1.0), sensitivity was 100% (95% CI 0.89–1.0), and specificity was 100% (95% CI 0.875–1.0). Diagnostic accuracy of CNVplex was similar to that of karyotyping. Both karyotyping and CNVplex assay detected 27 autosomal trisomies, three 45,X monosomies, and three polyploidies. CNVplex also detected additional novel structural abnormalities of the fetal genome. Compared with karyotyping, CNVplex significantly ( p = 0.001) reduced the waiting time by 13.98 days (95% CI 13.88–14.08) and the cost by US $241 (95% CI 234.53–247.47). CNVplex is a novel effective assay for diagnosing fetal chromosomal abnormalities associated with recurrent pregnancy loss. In the routine clinical work-up of recurrent pregnancy loss, diagnostic accuracy of CNVplex is comparable to that of conventional karyotyping but it requires less waiting time and has lower cost.
Our previous studies have demonstrated overexpression of Mcl-1 in cervical cancer tumorigenesis. However, the molecular mechanism of its overexpression remains not elucidated. MiR-320 has been reported to be down-regulated in various types of cancer, and bioinformatics prediction indicated that it may regulate the expression of Mcl-1. The aim of this study is to investigate the role of miR-320 and its target gene Mcl-1 in cervical cancer progression and to assess their clinical significance. miR-320 and Mcl-1 expressions in human cervical cancer tissues were investigated by qRT-PCR, in situ hybridization, and immunohistochemical staining, respectively. The clinicopathological implications of these molecules were analyzed. Bioinformatic prediction and luciferase assays were employed to identify the predicted microRNA (miRNA) which regulates Mcl-1. The apoptosis, proliferation, migration, and invasion assays were performed to investigate the effect of miR-320 on the cervical cancer cells. MiR-320 expression is significantly down-regulated versus Mcl-1 expression is up-regulated in cervical cancer tissues compared with normal controls with a negative correlation between them. Luciferase assay showed that miR-320 negatively regulates Mcl-1 expression. In addition, miR-320 induces apoptosis via down-regulation of Mcl-1 and activation of caspase-3 but inhibits cell proliferation, migration, invasion, and tumorigenesis in cervical cancer cells. Our studies show that miR-320 expression is decreased in cervical cancer, and its expression is negatively correlated with Mcl-1 expression in cervical cancer. In addition, miR-320 inhibits cervical cancer progression by down-regulation of Mcl-1. These results indicate that miR-320 may be an important biomarker and target for diagnosis and treatment of cervical cancer patient.
P27kip1 is a negative cell cycle regulator that plays an important role in tumor suppression. Deregulation of p27kip1 is commonly observed in many human cancers. Numerous studies about p27kip1 are reported in clinical patients despite variable data for the prognostic of p27kip1 expression. Here we report a meta-analysis of the association of p27kip1 expression with the survival of ovarian cancer.
Milk fat is widely accepted to be the major nutrient in human milk. Commercial infant formulas are usually based on mammalian milk such as bovine or caprine milk, but the differences in milk fat globules (MFGs) between human, bovine and caprine milk remain unclear. We showed that saturated fatty acid content was higher in bovine and caprine MFGs (>60%) than in human MFG (<40%), but that content of the unsaturated fatty acids C18:2 in human MFG was >7 times higher than in bovine and caprine MFGs. The cholesterol content of human milk was ∼20% higher than that of bovine and caprine milk. Triacylglycerol molecular species and polar lipids also differed between bovine, caprine and human MFGs. Confocal laser scanning microscopy images of MFGs revealed that the shape of the liquid-ordered domains varied by species.

目的:研究宫腔镜子宫纵隔切除术(TCRS)术后不同辅助治疗方法对妊娠结局的影响。方法对74例子宫纵隔患者行TCRS,术后随机分为A、B、C组:A组24例,术后辅助宫内节育器(IUD)治疗;B组25例,术后辅助人工周期治疗;C组25例,术后无辅助治疗。随访2~25个月,分析比较妊娠结局。结果3组术后随访总数70例,妊娠率为91.4%。3组足月活产比例分别为A组70.0%,B组69.6%,C组71.4%。51例分娩者中经阴道分娩占37.3%,剖宫产占62.7%,发生胎盘粘连19例,未发生胎盘植入、产后出血。结论TCRS显著改善纵隔子宫妊娠结局,但不同辅助治疗结果无明显差异。
Branched chain fatty acids (BCFA) are components of common food fats but little is known about their positional distribution on triacylglycerols (TAG). Human milk fat, mature milk of cows and goats, and several fish oil products available in China were evaluated for BCFA content in the sn-2 position. Compared to the sn-1/sn-3 (α) positions, BCFA are enriched in sn-2 (β) position of human milk (68% of BCFA), but randomly distributed among the sn-2 and sn-1/sn-3 positions of cow and goat milk. Fish oil TAG sn-2 BCFA were consistently depleted compared to the sn-1/sn-3 positions (5.2–11.9% of BCFA). Human intestinal epithelial cells were incubated with human milk sn-2 MAG, or fatty acids liberated from human milk sn-2 MAG. Cells incorporated more BCFA as sn-2 MAG than as FFA. The stereochemical positioning of BCFA in human milk fat may be unique compared to other BCFA sources, and mediate selective absorption of BCFA.
The injury of α-linolenic acid on HepG2 cell was investigated by MTT colorimetric method, inverted phase contrast microscope was used to observe morphology of HepG2 cell and the reactive oxygen species(ROS) level, superoxide dismutase(SOD) activity and malonadialdehyde(MDA) content were determined to discuss the effect of α-linolenic acid on the oxidative stress of HepG2 cell. The results showed that the proliferation of HepG2 cell was inhibited by α-linolenic acid in a dosage-dependent (50-250 μmol/L) and time-dependent manner, which indicated that α-linolenic acid could induce the injury of HepG2 cell. The morphology of HepG2 cell changed after incubated with 200 μmol/L α-linolenic acid for 24 h and showed typical apoptotic morphology changes in high concentration. After treatment with α-linolenic acid,the contents of ROS and MDA in HepG2 cell increased,while the activi-ty of SOD decreased, indicating that α-linolenic acid affected the oxidative stress of HepG2 cell to in-duce the injury of HepG2 cell.
Background The increasingly recognized importance of vitamin D has been discussed and vitamin D status among young children has attracted widespread attention in recent years. However, study on vitamin D status in young children aged 1–3 y is limited in China. Objective To evaluate the nutritional vitamin D status of young children aged 1–3 y in Wuxi, southeastern China. Methods A large cohort of 5,571 young children aged 1–3 y were recruited in this study who visited the child health clinics at the Wuxi Maternity and Child Health Hospital (latitude 31.57°N) during January 2014 to January 2015. Wuxi was located in southeastern China at a latitude of 31.57°N. Finger-stick blood sampling was conducted in all the subjects and serum 25-Hydroxyvitamin D (25(OH)D) levels were measured to evaluate their vitamin D status. Results In this study, serum 25(OH)D levels of young children at the age of 1–3 years ranged from 20.6–132.9 nmol/L (Median: 71.5 nmol/L). 16.1% of the population had vitamin D deficiency (<50 nmol/L), while 38.8% of the subjects had a sufficient (50–74.9 nmol/L) vitamin D level. An optimal vitamin D status (≥75 nmol/L) was found in 45.1% of the young children. The prevalence of vitamin D deficiency was higher in autumn (19.5%) than in summer (12.1%). There was no significant difference in vitamin D status between genders. The binary logistic regression analysis revealed that child age was strongly associated with vitamin D deficiency (adjusted OR: 1.173; 95%CI: 1.053–1.308; P = 0.004). Conclusions The prevalence of vitamin D deficiency was 16.1% among young children aged 1–3 y in Wuxi. Season and child age were associated with their vitamin D status. It is implied that young children should receive adequate amounts of vitamin D supplementation and spend more time outdoors to prolong the sunlight exposure when they grow older.

Objective:To investigate the relationship between the uterine artery blood flow and endome-trial blood flow measured and the outcome of pregnancy in invitro fertilization and embryo transfer(IVF-ET) fresh transfer cycles.Methods:A total of 61 women undergoing IVF-ET were included in the study.Trans-vaginal color doppler sonography (TV-CDS)was used to monitor the blood flow of uterine artery resistance index(RI),pulsation index(PI)and endometrial(sub endometrial )blood flow which include the day of human chorionic gonadotropin(HCG)injection and the day of embryos transfer during IVF-ET cycles.Ac-cording to the outcome of pregnancy,patients were divided into pregnancy group and non-pregnancy group, compared the parameters between the two groups.According to the situation of the endometrial (sub endom-etrial )blood flow,patients were divided into three groups:type A,type B and type C group,compared the parameters in the three groups.Results:There was no difference in the uterine artery blood status between the pregnancy group and non-pregnancy group (P >0.05).There was also no difference in the rate of preg-nancy during the day of HCG injection between type A,B and C group (χ2 =0.02,P >0.05).While there was significant difference in the rate of pregnancy during the day of embryos transfered between type A,B and C group (χ2 =6.16,P <0.05).Conclusion:Endometrial perforating blood flow in the day of embryos trans-fered could be used as a factor for forecasting the outcomes of IVF-ET.
采集无锡地区150例人乳样品,提取乳中脂肪后,采用气相色谱法对人乳脂肪中总脂肪酸及sn-2位脂肪酸进行测定.结果表明:人乳脂肪总脂肪酸中饱和脂肪酸含量较低,主要为棕榈酸,不饱和脂肪酸含量较高,主要为油酸和亚油酸.随着泌乳期的延长,中碳链饱和脂肪酸(C8:0、C10:0、C12:0)及单不饱和脂肪酸(C16:1)含量逐渐增加,而部分长碳链饱和脂肪酸及长碳链多不饱和脂肪酸如二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)含量明显下降.人乳脂肪中sn-2位脂肪酸组成中饱和脂肪酸含量约为70%,主要为棕榈酸.
