The purpose of the study was to evaluate in vitro cytotoxicity and in vivo sub-acute toxicity of conditioned medium obtained from rat bone marrow-derived mesenchymal stem cells directed to osteogenic differentiation. Material and methods . Conditioned medium was obtained by culturing rat bone marrow-derived MSCs induced under osteogenic condition. Results and discussion . Conditioned medium from rat bone marrow-derived MSCs was shown to have no significant cytotoxic effect on Ehrlich adenocarcinoma cell culture and in vitro expanded peripheral blood mononuclear cells from healthy donors. The use of MSC-CM did not have a significant effect on the state of experimental SD rats, the total body weight and growth rate of animals. A pathomorphologic study revealed no any abnormalities associated with MSC-CM injection. Conclusion . The use of conditioned medium from rat bone marrow-derived mesenchymal stem cells stimulated under osteogenic condition was found to be safe for both in vitro studies (with respect to Ehrlich adenocarcinoma cell line and peripheral blood mononuclear cells from healthy donors) and in vivo studies (injection of MSC-CM to animals).
The aim of the study is to investigate the chronic toxicity of the oral finished dosage form of pegylated interferon α-2b, obtained with the help of electron-beam synthesis technology. Materials and methods. The preparation was orally administered to rats and rabbits at the calculated therapeutic doses and doses exceeding them. The drug effect on the general condition and body weight, hematological parameters, myelogram, blood biochemical parameters, excretory and cardiovascular system, CNS were studied. As well as pathomorphological examination was carried out. Results and discussion. The drug has no toxic and locally irritating effect under its long-term use per os.
Проведено доклиническое исследование иммунокорригирующего действия суммарных препаратов из сырья эфиромасличного растения Coluria geoides (Pall.) Ledeb. (Rosaceae) при экспериментальном иммунодефиците. Исследуемые препараты устраняли проявления иммунодефицита, развивающиеся при применении циклофосфана: угнетение синтеза антиэритроцитарных антител-агглютининов и пролиферативных процессов в селезенке. Под влиянием препаратов C. geoides абсолютное число кариоцитов и количество антителообразующих клеток селезенки достоверно превышали перечисленные показатели в группе животных с экспериментальным иммунодефицитом, достигая в отдельных случаях фоновых значений. Изучаемые препараты оказывали более выраженное стимулирующее действие на синтез специфических иммуноглобулинов и пролиферацию антителообразующих клеток селезенки, чем настойка эхинацеи. Препарат С-2 (экстракт из подземных органов и травы C. geoides , полученный методом перколяции 70 % этанолом) наиболее перспективен для углубленного исследования с целью разработки нового эффективного иммунокорректора.
In vitro experiments is was revealed dose-dependent effect of polyphenolic complexes of Calendula officinalis, Urtica dioica and Sorbus aucuparia on induced synthesis of cytokines and antibodies. Cultivation of mitogen-stimulated mononuclear cells with polyphenolic complex Calendula officinalis at a dose of 500 mcg/ml resulted in a decrease in the synthesis of IL-2, IFNγ, IgM and IgG, while at dose of 50 mcg/ml it didn't affect these indicators. A similar effect was observed with the use of polyphenolic compounds of Urtica dioica, however incubation of cells with thestudied compounds at a dose of 50 mcg/ml resulted in a decrease IgG levels. Polyphenolic compounds of Sorbus aucuparia in both concentrations had expressed suppressing effect on the secretion of IL-2, IFNγ, IgM and IgG by mitogen-activated cells. Studied polyphenolic complexes, in selected concentrations didn't influence the spontaneous and stimulated production of IL-4.
The extract of Skutellaria baicalensis use in the conditions of developing cytostatic hemodepression, induced by a single administration of cyclophosphamide in a maximum tolerant doses in CBA/CaLac mice, stimulates the regenerating processes in a greater degree of erythroid and lymphoid lineages of hemopoiesis, but pantovit use – the reparation of myeloid and lymphoid lineages. Course application of Scutellaria baicalensis grades immunosuppressive effect of cyclophosphamide by stimulation of phagocyte activity of macrophages and regeneration of antibody-producing cells contents. Pantovit administration in immunized mice on the background of cytostatic immunossupression gives rise to increasing phagocyte activity of macrophages and intensification of synthetic activity of antibody-producing cells.
The influence of Sorbus sibirica, Calendula officinalis and Althaea officinalis extracts on the humoral immune response and nonspecific resistance of mice to immunosuppression by cyclophosphan was studied. It was shown that these extracts are not inferior to Echinacea purpurea tincture in terms of stimulation of humoral immune response, phagocytic and bactericidal activity of peritoneal macrophages but exceed effect of E. purpurea on phagocytic activity of peripheral blood neutrophils.
Study results of the examination of homeostasis status in 510 patients with pubertal uterine bleedings are presented in the article. Based upon studying systemic changes in peripheral erythrone, immune system, hormonal setting and vegetative nervous system tone, the matrix of homeostasis system status is created and analyzed in several variants of the disease using correlation analysis.
Extract of Filipendula ulmaria (L.) Maxim administered intragastrically in doses 10, 50, 150 and 500 mg/kg stimulated both inductive and productive phases of the humoral immunity response in CBA/CaLac and C57BL/6 mice. The extract also exhibited pronounced antiinflammatory effect, which was manifested by a decrease in the synthesis of interleukin-2 by splenocytes and by suppression of proinflammatory cytokines production in delayed-type hypersensitivity reaction. At the same time, Filipendula ulmaria extract did not influence the functional activity of peritoneal macrophages.
This project reported results of clinical investigation of hard anemia treatment in patient with juvenile uterine bleeding. Anemia was treated by additional appointment of Anaferon detskiy, which contents antibodies to human's interferon-gamma. Evaluation of antianemic therapy efficiency was made comparing with traditional iron-drug therapy. It was proved that combined prescription «Anaferon detskiy» and iron-drag normalizes rate of humoral and cellular immunity and cytokines profile earlier than iron-drags monotherapy.
It has been studied the influence of alphaand beta-adrenoblockers on production of cytokines by splenocytes of mice-hybrids F1(СВАхС57Вl/6) at development of humoral immune response. It has been shown that dihydroergotamine has lymphocytotoxicity properties, reducing on the background of immunization of the ex vivo release a lot of lymphokines IL-2, IL-4 and IL-10. However, thus there was a stimulation of monokines production TNF-„q and IL-1„r. Introduction of obsidan to immunized mice as well as dihydroergotamine enhanced production of TNF-„q and reduced production of IL-2. Influence of beta-adrenoblocker on release of others cytokines (IFN-„s, IL-4, IL-6 and IL-10) by splenocytes of immunized mice was ambiguous.
Possibilities of correction and prophylaxis. The authors presented results of study covering workers of radiation-dangerous plants of Siberian Chemical Enterprise, who underwent long-term exposure to outer and inner radiation in "low" doses range during the occupational activity. Target-systems for radiation effects were proved to be lipid peroxidation and antioxidant defence systems, cell energy metabolism--severity of disorders in which is correlated with degree of radiation exposure.
The reactogenicity of the embryonic live recombinant variola and hepatitis B bivaccine as tablets (Revax-BT) as well as its safety and immunogenicity were evaluated in clinical trials made in volunteers who had previously immunized or not with variola vaccine. A preliminary conclusion was made on a lack of side effects and drug safety in primary vaccination and been revaccination with low and high doses. Primary immunization of volunteers and as bivaccination with high doses stimulated the most pronounced immune response to the vaccine virus versus such effect observed in immunization of volunteers with low vaccine doses. Humoral immune response to HBs was observed in 75% of volunteers of both groups after as bivaccination. Such response was most pronounced in examinees immunized with low vaccine doses versus those who received high bivaccine doses. At the same time, no protective levels of humoral immunity response to HBs Ag were observed in volunteers first vaccinated.
The process of the disease due to herpes simplex virus types 1 and 2 (HSV-1 and 2) was studied on white uninbred mice weighing 10 to 12 g. The animals were infected intracerebrally or intraperitoneally. Intraperitoneal contamination of the animals with MS strain of HSV-2 was used for the experimental model of the herpes simplex infection. The prophylactic antiherpes action of ultralow doses of the human gamma-interferon antibodies (ULD of anti-IFN-gamma) at a course of its intragastral administration was evaluated. The preparation was shown to have a significant (p < 0.05) protective effect in a dose of 10 LD50, evident from a 10-fold decrease of the HSV-2 accumulation in the brain, a lower percentage of the animal deaths and an increase of the average lifespan of the animals by 3.3 days. The study of the therapeutic action of ULD of anti-IFN-gamma at a course of its intragastral administration showed that the preparation had no significant positive effect on the disease process in the animals infected with HSV-2 in a dose of 10 LD50. However, a positive effect associated with delayed virus replication in the brain was observed in the study on the therapeutic effect of ULD of anti-IFN-gamma after its intragastral administration to the mice infected with a sublethal dose of the virus.
