Hepatocellular carcinoma (HCC) is a typical inflammation-driven cancer and ranks sixth in the incidence rate worldwide. The role of adenylate uridylate- (AU-) rich element genes (AREGs) in HCC remains unclear. HCC-related datasets were acquired from The Cancer Genome Atlas (TCGA) database and Gene Expression Omnibus (GEO) database. Differentially expressed AREGs (DE-AREGs) between HCC samples and healthy controls were identified. The univariate Cox and LASSO analyses were performed to determine the prognostic genes. Furthermore, a signature and corresponding nomogram were configured for the clinical prediction of HCC. The potential signature-related biological significance was explored using functional and pathway enrichment analysis. Additionally, immune infiltration analysis was also performed. Finally, the expression of prognostic genes was verified using real-time quantitative polymerase chain reaction (RT-qPCR). A total of 189 DE-AREGs between normal and HCC samples were identified, wherein CENPA, TXNRD1, RABIF, UGT2B15, and SERPINE1 were selected to generate an AREG-related signature. Moreover, the prognostic accuracy of the AREG-related signature was also confirmed. Functional analysis indicated that the high-risk score was related to various functions and pathways. Inflammation and immune-related analyses indicated that the difference of T cell and B cell receptor abundance, microvascular endothelial cells (MVE), lymphatic endothelial cells (lye), pericytes, stromal cells, and the six immune checkpoints was statistically significant between the different risk groups. Similarly, RT-qPCR outcomes of these signature genes were also significant. In conclusion, an inflammation-associated signature based on five DE-AREGs was constructed, which could act as a prognostic indicator of patients with HCC.
目的:观察保元汤对慢性心力衰竭(CHF)大鼠心肌损伤的改善作用及对内质网应激(ERS)凋亡通路的影响.方法:选用90只健康SD大鼠随机分为空白组,模型组,保元汤低、中、高剂量组,卡托普利组,每组15只.利用阿霉素制备大鼠CHF模型.采用HE染色观察心肌组织病理变化;Masson染色观察心肌纤维化程度;ELISA法检测血清BNP、SOD、MDA水平;Western Blot法和RT-PCR法分别检测心肌组织中GRP78、PERK、eIF2α、CHOP蛋白和mRNA表达水平.结果:与模型组比较,各给药组大鼠心肌损伤改善,心肌纤维化改善,胶原纤维沉积明显减少;保元汤中、高剂量组BNP、MDA水平显著减少(P<0.05,P<0.01),而保元汤高剂量组SOD水平显著升高(P<0.01);保元汤高剂量组GRP78、PERK、eIF2α、CHOP蛋白及mRNA表达显著降低(P<0.05,P<0.01).结论:保元汤可通过调节氧化应激及ERS凋亡通路抑制心肌细胞凋亡,改善阿霉素诱导的心肌组织损伤,发挥防治CHF的作用.
鸢尾素(Irisin)是2012年被Bruce Spiegelman教授团队发现并报导的一种新的肌肉因子,通过"PGC-1α→FNDC5/Irisin→UCP-1通路"增加机体能量消耗、调节能量代谢.本文整理Irisin及其与骨骼肌减少症、骨质疏松症的关系的最新研究.发现Irisin可能主要通过对骨骼肌细胞线粒体的作用、与肌生成抑制素的拮抗关系以及与II型肌球蛋白重链的联系这三个途径对肌少症进行干预;另外,Irisin通过对成骨细胞分化和破骨细胞形成的调节,以及对皮质组织矿物质密度,骨膜周长,极惯性矩和分形维数等的改变,可能达到治疗骨质疏松症的目的.本文旨在探索Irisin与这两种疾病的关系,为研究和治疗骨骼肌减少症、骨质疏松症提供新途径和新视角.
Objective. To assess the effectiveness and safety of melatonin for perimenopausal and postmenopausal women with osteopenia. Methods. In this meta-analysis, data from randomized controlled trials were obtained to assess the effects of melatonin versus placebo or western medicine in perimenopausal and postmenopausal women with osteopenia. The study’s registration number is CRD42018086238. The primary outcomes included bone mineral density (BMD) and T-score. Result. From 551 articles retrieved, three trials involving 121 patients were included. Due to the high-to-substantial heterogeneity (BMD: I2=96.9%, P=0.000; T-score: I2=74.9%, and P=0.019), the statistical analysis of BMD and T-score was abandoned. A systematic review was undergone for the two outcomes. Compared with the control group, melatonin may increase osteocalcin (WMD 4.97; 95% CI 3.14, 6.79; P<0.00001). Conclusion. Based on current evidence, melatonin might be used as a safe nutritional supplement to improve bone density in perimenopausal and postmenopausal women, but its efficacy needs to be further affirmed.
目的 探讨中医脾虚证与骨质疏松之间的关系.方法 统计400例诊断为骨质疏松患者,检测所有患者的骨密度、握力、肌肉含量、体质量指数(BMI)等指标,分析握力、肌肉含量、BMI与骨密度的相关性,并根据中医证状分为脾虚组和非脾虚组,比较2组的握力、肌肉含量.结果 握力≤18 kg和握力>18 kg、肌肉含量≤40.76 kg和肌肉含量>40.76 kg、BMI≤23 kg/m2和BMI >23 kg/m2患者之间骨密度的差异均有统计学意义(P<0.05).脾虚型与非脾虚型的握力、肌肉含量差异均有统计学意义(P<0.05).结论 中医脾虚证与骨质疏松有一定的关系,骨密度与握力、肌肉含量、BMI相关,中医之脾虚证是导致骨密度含量下降的重要条件.
目的 研究人参缩心饮对腹主动脉狭窄致大鼠心肌肥厚的影响,揭示人参缩心饮对心肌肥厚的保护作用机制.方法 采用结扎腹主动脉方法建立压力负荷性心肌肥厚模型,用人参缩心饮进行干预,对各组大鼠进行血流动力学指标、左心室脏器系数、左室前壁及室间隔厚度、左心室肌蛋白含量、左心室肌胶原含量测定以及心肌间质胶原容积检测.结果 人参缩心饮中、高剂量组能改善心肌肥厚大鼠血流动力学参数,能降低心肌肥厚大鼠左室前壁及室间隔厚度、左心室胶原含量及心肌间质胶原容积分数.结论 人参缩心饮对心肌肥厚有一定疗效,且呈一定量效关系,并对腹主动脉狭窄致大鼠心肌肥厚具有一定的抑制作用.
The knowledge of osteoporosis in traditional Chinese medicine (TCM) can be traced back to Huangdi Neijing,where several concepts such as ‘bone atrophy',‘bone impediment'and ‘bone necrosis' were put forward.In TCM clinic,osteoporosis treatment often need to focus on treating kidney combined with fortifying the spleen,and activating blood circulation to dissipate blood stasis is also essential.For bone remodeling balance disorders and bone microstructural degeneration,the treatment method of activating blood circulation to dissipate blood stasis can improve its root causes with promoting new bone metaplasia and preventing old bone degeneration.Many experts generally find patients with osteoporosis have the symptoms of sublingual varicose veins,dark purple tongue with ecchymosis or petechiae,ecchymosis in skin mucous membrane,dark red of lips and gingiva,etc,which are very similar to the symptoms of blood stasis in TCM.In this study,a total of 400 cases of patients with osteoporosis in Xiyuan Hospital Affiliated to China Academy of Traditional Chinese Sciences were calculated according to the Observation Table for the Standard of Pattern Classification Based on TCM Identification in Osteoporosis Cases.The bone mineral density,blood stasis pattern,sublingual vascular stasis degree,pain degree,pulse wave velocity and fracture count were tested or scored.According to the experimental results,the correlation between bone mineral density and the above parameters is analyzed,and then the close relationship between TCM blood stasis and osteoporosis is expounded and proved.
目的:探讨中医药治疗老年痴呆的基础研究与研究新进展.方法:选取中国中医科学院西苑医院2013年8月-2015年8月收治的120例老年痴呆患者作为研究对象,对所有患者的临床资料进行回顾性分析,总结临床用药情况,研究老年痴呆患者中医治疗的方法.结果:老年痴呆患者的中医病因病机学说还存在一定的问题,缺乏数据支撑,需要进一步完善,老年痴呆患者的有效中药开发进展较为缓慢,虽然在药物研究方面呈现很多种,但是,开发出来并投放市场的中药相对较少,投放在市场以后得到的应用少之又少,在临床实践中,缺乏广泛认同的个体化辨证论治方案.结论:中医药治疗老年痴呆患者的基础研究较多,但是缺乏针对性,使得中医药个体化辨证论治方案缺乏认同感,应建立并完善相关治疗个体化方案.

Objective: This paper aims to investigate whether the Ginseng Drink have the contractile function effect on the myocardial cells of heart failure rats. Methods: Isoproterenol produced heart failure in rats and isolated and cultured cardiomyocytes. The myocardial cells were divided into: normal cardiac myocytes cell group and heart failure group. Normal cardiac myocytes cell groups were separately cultured in normal DMEM medium and containing 50nmol/L β2-AR antagonist(ICI118551) DMEM medium. The culture conditions of failure cardiomyocytes group separately were DMEM medium, containing 100nmol/L DMEM medium, containing 50nmol/L β2-AR antagonist(ICI118551) and 100nmol/L Ginseng Drink DMEM. Myocardial contraction amplitude and survival were measured,β2A protein of cardiomyocytes were measured by Western blotting(WB). Results: Compared with normal cells, cell contraction amplitude and survival of heart failure were significantly lower(P0.05).The contraction amplitude and survival of failure myocardial cells were further reduced after using β2AR antagonists(P0.05). Linear regression analysis showed β2AR protein expression and cardiac contraction amplitude was positively correlated(R2=0.9656). But after using Ginseng Drink,the contraction amplitude and survival rate of failure myocardial cells both significantly increased(P0.05), the protein expression of β2AR still increased. At the same time, linear regression analysis the showed that contraction amplitude and the protein expression of β2AR was positively correlated(R2=0.9911).Conclusion: Ginseng Drink can enhanced myocardial cell contraction amplitude and survival by acting on adrenaline β2-AR to maintain the normal function of the heart.
Objective: To explore the effects of the total l avone of ginkgo(TFG) on cardiac function, AngⅡ and ET-1 of rats with heart failure induced by isopropyl adrenaline. Methods: 40 male SPF Sprague-Dawley rats were randomly divided into 4 groups: blank group, model group, TFG group and metoprolol group. Expect for the blank group, all rats were received subcutaneous injection of isopropyl adrenaline at 300mg/kg, for twice. After the model was successfully established, the TFG group and metoprolol group were respectively given TFG extract at 200mg·kg-1·d-1 and the metoprolol at 20mg/kg, while the other groups were gavaged the same volume of normal saline, lasting for 8 weeks. After the experiment, the level of ET-1, the content of AngⅡ in serum and the pathological scores were detected, and the pathologic morphological characteristics of myocardial tissue were determined. Results: Compared with those in the model group, left ventricular ejection fraction(LVEF), interventricular septum thickness(LVET) and left ventricular posterior wall thickness(LVPWT) were signii cantly decreased and left ventricular end diastolic diameter(LVEDD) and left ventricular end systolic dimension(LVESD) were increased in TFG group and metoprolol group, with signii cant dif erences between the TFG group, metoprolol group and the model group(P0.05). It suggested that cardiac function was improved after the treatment of TFG or metoprolol. The level of ET-1, the content of AngⅡ in serum and the pathological scores in TFG group were lower than those in the model group, with signii cant dif erences between the TFG group and the model group(P0.05). Conclusion: Ginkgo l avonoids extract can improve cardiac function of rats with heart failure, the mechanism of which may be related to the levels of ET-1 and AngⅡ.
Objective: To study the influences of Buyi Qiangxin tablet on cardiac function and life quality of the chronic heart failure(CHF) patients.Method: Sixty-eight cases of patients with CHF were randomly divided into control group(34 cases) and observation group(34 cases) according to before and after entering the hospital.The patients of both groups took general treatment of cardiac failure and conventional western medical treatment according to the guide of the diagnosis and treatment of chronic heart failure.The patients in observation group took Buyi Qiangxin tablets,1.2 g / time,3 times / day,the treatment course is 4 weeks.The New York Heart Association(NYHA) cardiac heart grading and 6 minutes walk test(6 MWT) were evaluated before and after treatment,recording Lee's heart failure.Left ventricular ejection fraction(LVEF) and cardiac output(CO) were tested using echocardiography.The life quality was evaluated by Minnesota life quality of heart failure questionnaires.detecting the plasma NT-proBNP and antidiuretic hormone(ADH) level.Result: The total curative effect of Lee's heart failure of the observation group was 91.17%,which is higher than that of the control group(73.52%,P 0.05).The total curative effect of cardiac function of observation was 88.23%,superior to67.64% in the control group(P 0.05).After the treatment,the Lee's heart failure points and life quality scores of observation group decreased and were lower than the control group(P 0.01).The 6MWT of both groups increased and the increasing in observation group was more significantly(P 0.01).The LVEF and CO of both groups increased after the treatment,the observation group was higher than control group(P 0.01).The levels of plasma NT-proBNP and ADH decreased and observation group was less than the control group(P 0.01).Conclusion: Buyi Qiangxin tablets can improve cardiac function grading of CHF patients,increasing LVEF,CO,6 MWT and life quality of patients.Its mechanism may be related to decreasing NT-proBNP and ADH of patients.
Objective:To explore TCM typing and TCM treatment of patients with myocarditis.Methods:A total of 209 cases of myocarditis patients were diagnosis as"Xie qi qin xi","Qi yin liang xu",and"Xin qi xu ruo"by TCM syndrome type.The number cases of the three groups were respectively 79,64 and 66.Zhigancao decoction were given for treatment according to certain Chinese medicine syndrome.Clinical efficacy between the three groups was compared before and after treatment.Results:After symptomatic treatment,three groups of patients received better effects.The efficacy of"Xie qi qin xi"was higher than the other two groups,but the difference was not statistically significant(P > 0.05).Test results of the serum myocardial enzymes showed that the CK,CKMB and LDH all improved after treatment.The difference was statistically significant(P < 0.05).The "Xie qi qin xi"group had a better result than the other two groups(P < 0.05).Conclusion:Zhigancao decoction has a good effect on the patients with clinical myocarditis.It could effectively improve CK,CKMB and LDH of patients.It can be a very good guide and reference value on TCM symptoms and treatment of myocarditis.
目的从若干个益气中药验方和古方中筛选出能够抗辐射的药味少、疗效好的中药复方。方法将110只昆明小鼠随机分为空白组、模型组、人参汤组、芎归汤组、贞元饮组、当归补血汤组、四物汤组、经验方组、芪归升血片1组、芪归升血片2组、芪归升血片3组。除空白组外其余各组进行4.0Gy的X射线照射1次,次日空白组和模型组给予生理盐水灌胃,其余相应药物灌胃,每只0.2 ml/d,1次/d,连续7 d。第8天取小鼠外周血和脾脏,检测T淋巴细胞亚群和血清Il-2水平。结果四物汤、经验方、芪归升血片2、芪归升血片3能够提高电离辐射模型小鼠脾脏T淋巴CD+4/CD8+比例,同时能够提高电离辐射小鼠血清IL-2水平。结论四物汤、经验方、芪归升血片2、芪归升血片3可对辐射小鼠的T淋巴细胞亚群起到抗辐射的作用。
恶性肿瘤的发生发展与机体免疫功能低下有关。中医的扶正治疗与免疫调节密切相关。从实验研究到临床应用均证明,中药能够调节T淋巴细胞亚群变化而发挥抗肿瘤作用,对临床应用中药扶正治疗而发挥抗肿瘤作用具有指导意义。
目的:筛选在气血相生理论指导下,由临床验方化裁的7种方剂中,益气补血作用最显著的方剂。方法:采用环磷酰胺(CY)腹腔注射致ICR小鼠红白减少症模型,连续给药15 d,对ICR小鼠外周血中的红白细胞数量和T细胞亚群CD2、CD3、CD4等指标进行检测。结果:在临床验方中的7种方剂中,各组小鼠检测指标与模型组相比,差异显著P<0.05或P<0.01,其中益气方组在RBC、WBC、Hb和CD2、CD3、CD4的检测结果中差异最为显著(P<0.01)。结论:益气方对于提升化疗后小鼠的红白细胞数量,改善化疗后的免疫力有重要作用。
