介绍林下栽培大球盖菇技术,包括高产培养料配方(木屑30%、玉米芯30%、稻壳30%、虫粪沙10%),一行双拱的菇床铺设方式,二次播种,覆土打孔,以及菌丝期、菇蕾期、幼菇期的温度、湿度、光照管控技术要点,采用该技术模式栽培,每潮菇出菇时间均可保持在一周左右,每亩每潮产量约4 000 kg.
为筛选出适合平顶山地区保护地种植的优质、高产小果型新品种西瓜,引进了9个小果型西瓜品种进行品种筛选试验.结果筛选出早红蜜、超越梦想、京颖、小兰等几个综合表现较好的适合平顶山地区保护地种植的品种.

With ethanol reflux extraction leek seed active ingredient, product yield and adenosine content as the index, to optimize the extraction technology research, the results show that the material liquid than 1∶6, 4 h, 70% ethanol concentration, extraction time, pH value of 6.0, in this condition, the product yield is around 14%, adenosine content is above 400 mg/kg.

In order to study on the corresponding relationship between ISSR markers and heterosis of leek ,this paper calculated the heterosis of tiller number and other traits ,and compared the of heterosis of tiller number ,plant weight and sheath length with ISSR genetic distance of the parents based on the investigation of the traits of 27 hybrid combinations of leek and their parents .The inquiry elicited the strong heterosis was mostly concentrated in the position of genetic distance over 5 .0 .

The seed oil of Allium tuberosum is extracted by supercritical CO2 fluid extraction, and the result shows that granule sizes 50~60 mesh, extraction pressure 22~25 MPa, extraction temperature 45℃, extraction time 2.5 h and CO2 flow rate on the production 3.0 L/h, after investigating the ifluences of above-mentioned three factors on oil yield, which reached above 17%under the feasible operating conditions.

In order to provide theoretical information for leek breeding,this study was to probe genetic diversity of the leek germplasms.96 leek samples were used to detect the intra-and inter-specific genetic variation by inter-simple sequence repeat(ISSR) analysis.The result showed that 20 ISSR primers were screened from 77 primers based on 96 leek samples.The first leek DNA fingerprint library was constructed by PCR amplification,PAGE fingerprint detection,and fingerprint data collection of Gel imaging.The ISSR amplification diversity reached 94.7%,which was an effective molecular marker.The genetic similarity(GS) among 96 leek samples ranged from 0.58 to 0.88,which could fully show that they have rich genetic diversity.The result of cluster analysis showed that the cluster analysis correlated with sample origin and was consistent with species distribution.The breeding and spreading cultivars with relatively low diversity clustered more together,which means the new leek breeding innovation should introduce germplasms from home and abroad base.

A total of 174 accessions of leeks germplasm collected from Germplasm Repository were used in this study.According to the clustering result of 15 phenotypic traits,all the accessions were divided into 14 categories when the genetic distance was 5.3.Based on combination of simple ratio method and artificial,we established a primary core collection containing 69 accessions of leeks germplasm,which accounted for 40% of the whole collection.Compared with the whole collection,the genetic diversity index of primary core collection was higher than that of initial accessions except for plant sheath length and leaf width.Between core collection and initial collection,the mean difference percentage was 0,the variation coefficient rate was 106.14%,and coincidence rate for range was 94.43%.The significant correlation was found between the leaf thickness and other traits,and the leaf number and other traits were respectively missed two traits in the primary core repository.The other traits correlations were consistent with the traits correlations in total repository.The results showed that the genetic variation and genetic structure of the accessions can be well preserved in this primary core collection.

研究了基于ISSR-PCR扩增的DNA指纹检测韭菜种子纯度的方法。从发芽种子的嫩芽提取基因组DNA,筛选6种ISSR引物进行ISSR-PCR扩增。聚丙烯酰胺凝胶电泳检测表明,用ISSR-64引物检测平丰8号,韭菜种纯度约92%。该技术具有准确性高、时效性好、检测成本较低等特点。
