Practical courses in a university are the basic courses to cultivate university students’ professional quality, practical ability and innovative thinking, as well as important carriers to achieve the goal of cultivating talents in higher education. Sticking to the student-centered teaching concept and highlighting problem-oriented instruction, the teaching team of the course of Laboratory Management Innovation Practice implemented teaching reform in terms of teaching idea, instructional design, instructional delivery, and teaching reflection. The reform aimed to guide students to think actively and use brains and hands simultaneously, developing their professional skills, promote their all-round development, and ultimately accomplish the transformation from delivering knowledge to improving abilities in classrooms. The reform practices have achieved good results.

To achieve a good stealth property and to enhance the uptake by tumor cells, polymeric micelles containing a slightly negatively charged and zwitterionic corona at pH 7.4 (i.e., blood pH) and a positively charged surface at a slightly acidic pH (i.e., tumor extracellular pH) were prepared. The amphiphilic diblock copolymer which was used to prepare the polymeric micelles contains a hydrophilic block comprised of nonionic hydrophilic groups, negatively charged groups (carboxyl groups) and pH-triggered positive charge-generation groups (morpholino groups). The zeta potential of the micelles was found to increase as the pH decreased over the pH range covering the blood and tumor tissue pH ranges, and the corona of the micelles should contain zwitterionic groups. This could be due to that, in the pH range studied, more morpholino groups become protonated as the pH decreased, whereas the carboxyl groups were almost completely deprotonated to form carboxylate anions. Furthermore, by adjusting the molar ratio of morpholino to carboxyl groups, the zeta potential of the polymeric micelles was controlled to achieve a slightly negative value at pH 7.4. Thus, the combination of a slightly negatively charged surface and a zwitterionic corona suggests that these micelles would possess good stealth property. When the pH decreased from 7.4 to 6.8 or 6.5, the zeta potential value of the micelles became positive due to increased protonation of the morpholino groups. The development of graduate employment problem has been influenced by the national economic development,enrollment expansion,the prevailing concept on graduate employment and so on.With the rapid development of domestic economy and the enrollment expansion,graduates have been made to shift their position from demand exceeding supply to employment difficulty in the job market in recent twenty years.The supply-and-demand dynamics of graduates in job market,the adjustment of national employment policies and personal attitudes toward employment have influenced the graduate employment problem to different degree.To crack this severe problem,we need to make more efforts in expanding the employment demand,reforming the higher education training system,improving individual comprehensive ability,etc.
Stearic acid-grafted and lipoic acid-grafted chitosan were prepared by dehydration condensation reaction between the amino groups of chitosan and the carboxyl groups of stearic acid and lipoic acid. The modified chitosan possessed amphiphilicity due to the incorporation of the hydrophobic groups. The pKa value of the residual amino groups of the modified chitosan increased from 6.6 to 6.8~7.1 after methylation. Nanoparticles of the modified chitosan were prepared in aqueous medium through ultrasonic dispersion. The Zeta potentials of the nanoparticles were positive in values, which indicated that the residual amino groups and their methylation derivatives were located on the surface of nanoparticles. The chitosan-based nanoparticles were reduced by dithiothreitol and oxidized with oxygen in air, the disulfide cross-linked structures were formed in the core of the final nanoparticles.
Single-hole hollow polymer nanospheres were fabricated by raspberry-like template method using “graft-from” strategy through atom transfer radical polymerization (ATRP). Nanometer-sized silica spheres were covalently attached onto the surfaces of micrometer-sized silica spheres. Crosslinked polymer shells on the nano-sized spheres outside the attached area were formed by “graft-from” strategy through ATRP. After removal of the silica cores, single-hole hollow crosslinked polymer nanospheres were obtained. In this strategy, most of ATRP monomers may be used and thus many functional groups can be easily incorporated into the single-hole hollow crosslinked polymer nanospheres.

Ibuprofen was loaded into polystyrene-based macroporous weakly basic anion exchange resins,D301R and D301T.The loading was driven by electrostatic and hydrophobic interactions.The optimized loading condition was: a suspension of the resin in 5~10% ethanol aqueous solution with dispersed ibuprofen was stirred for 12h at 60℃.The loading capacity could be up to 0.49g/g resin.The release of the loaded ibuprofen in buffer mimicking gastrointestinal pH environments(2h,4h and 18h at pH 2,7.4 and 6.7,respectively) was studied.The loaded ibuprofen was released almost linearly against time in the first 12h with release percentage of 58~60%.The release percentage in the followed 12h was quite low,being 13~17%.In simulated administrations taken at 12h interval,the loaded ibuprofen in D301R was released almost linearly against time during a 24h period after the second administration.In contrast,an initial burst release of the loaded ibuprofen in a polymethacrylate-based weakly basic anion exchange resin [n-butyl methacrylate/(N,N-dimethylamino)ethyl methacrylate/ethylene glycol dimethacrylate copolymer] at neutral pH was observed.These results showed that π-π stacking interaction between ibuprofen and the carrier played a key role in the sustained release of ibuprofen from the polystyrene-based weakly basic anion exchange resins.
Amphiphilic diblock copolymers with poly(ethylene glycol) as the hydrophilic block and a random copolymer of n-butyl methacrylate or styrene and (N,N-diethylamino)ethyl methacrylate as the hydrophobic block were prepared by atom transfer radical polymerization (ATRP). Ibuprofen, a model drug that contains a carboxylic group and hydrophobic moiety, was loaded into micelles formed from the amphiphilic diblock copolymers by a combination of ionic interaction and hydrophobic effect. The loading capacity of ibuprofen in the micelles reached 60%. Loaded ibuprofen was released in a sustained fashion into media simulating gastric fluid (pH 1.6, 2h), small intestinal fluid (pH 7.4, 4h), and colon fluid (pH 6.7, 18h). Simulating the case of oral administration at 2doses per day, loaded ibuprofen was released almost linearly against time after the second dose in media simulating human gastrointestinal tract fluids.

A 9-residue peptide, CP-1 (GLRILLLKV-NH(2)), is synthesized by solid-phase synthesis method. CP-1 is a C-terminal amidated derivative of a hydrophobic transmembrane segment (CP) of the T-cell antigen receptor (TCR) alpha-chain. CP-1 shows broad-spectrum antimicrobial activities against Gram-positive and Gram-negative bacteria with the minimal inhibitory concentration (MIC) values between 3 and 77microM. Circular dichroism (CD) spectral data shows that CP-1 adopts a well-defined beta-sheet structure in membrane-mimicking environments. CP-1 kills E. coli without lysing the cell membrane or forming transmembrane pores. However, CP-1 can penetrate the bacterial cell membranes and accumulate in the cytoplasm in both Gram-positive S. aureus and Gram-negative E. coli. Moreover CP-1 shows binding affinity for plasmid DNA. These results indicate that the killing mechanism of CP-1 likely involves the penetration into the cytoplasm and binding to intracellular components such as DNA.
