目的 探讨去氢木香内酯对喉癌细胞增殖、迁移和侵袭的影响,以及对磷脂酰肌醇-3 激酶(phosphatidyl inositol 3 kinase,PI3K)/蛋白激酶 B(protein kinase B,Akt)信号通路的调节作用.方法 第 4 代对数生长期 Hep-2 细胞经不同浓度(0、10、20、30μmol/L)去氢木香内酯处理后,MTT 法检测细胞增殖抑制率、流式细胞仪检测细胞凋亡率、Transwell实验检测细胞迁移和侵袭能力,蛋白印迹法检测 p-PI3K、p-Akt、Sox-2、CD44、CD133 和 Nanog蛋白相对表达量.结果 与 0μmol/L 组比较,10、20 和 30μmol/L组细胞增殖抑制率和细胞凋亡率均升高(P<0.05),迁移和侵袭能力均减弱(P<0.05),p-PI3K、p-Akt、Sox-2、CD44、CD133 和 Nanog蛋白相对表达量均降低(P<0.05),且具有浓度依赖性.结论 去氢木香内酯可抑制喉癌细胞增殖、迁移和侵袭,其可能作用机制是通过抑制 PI3K/Akt信号通路发挥作用.
目的 研究长链非编码RNA含有WW和PDZ结构域2的膜相关鸟苷酸激酶反义RNA 3(Ln-cRNA MAGI2-AS3)在鼻咽癌组织中的表达及与患者放射治疗(放疗)敏感性的关系,并探讨LncRNA MAGI2-AS3对鼻咽癌细胞放疗抵抗的影响及作用机制.方法 采用反转录荧光定量聚合酶链反应(RT-qPCR)检测LncRNA MAGI2-AS3在鼻咽癌组织中的表达水平,分析LncRNA MAGI2-AS3与鼻咽癌患者放疗敏感性的关系.常规培养鼻咽癌细胞CNE2,转染LncRNA MAGI2-AS3小干扰RNA(siRNA),根据是否转染siRNA分为si-NC组和si-LncRNA MAGI2-AS3组.采用CCK8实验检测各组细胞的放疗敏感性;放射处理后,采用平板克隆实验、流式细胞实验、裸鼠实验分别检测各组细胞克隆形成能力、细胞周期、凋亡情况和细胞体内生长能力;采用Western blot试验检测各组细胞周期蛋白D1(cyclin D1)、细胞周期蛋白依赖性激酶4(CDK4)、凋亡蛋白活化的含半胱氨酸的天冬氨酸蛋白水解酶(caspase)3、caspase9表达.结果 RT-qPCR检测结果显示,Ln-cRNA MAGI2-AS3在鼻咽癌组织中表达上调(P<0.05).LncRNA MAGI2-AS3高表达与患者放疗抵抗有关(P<0.05).与si-NC组相比,si-LncRNA MAGI2-AS3组鼻咽癌细胞中LncRNA MAGI2-AS3表达减少、对放疗敏感性增加(P<0.05).放射处理后,si-LncRNA MAGI2-AS3组鼻咽癌细胞克隆形成能力降低(P<0.05),细胞周期阻滞在G0/G1期(P<0.05),细胞凋亡增加(P<0.05),细胞体内生长能力显著降低(P<0.05),cyclin D1、CDK4表达降低,caspase3、caspase9表达增加(P<0.05).结论 LncRNA MAGI2-AS3通过调控细胞周期和凋亡促进鼻咽癌细胞放疗抵抗,是逆转鼻咽癌放疗抵抗的潜在分子靶点.
目的:检测鼻咽癌中微小RNA-221-3p(miR-221-3p)的表达,分析其与增殖、凋亡指标的相关性.方法:选取鼻咽癌患者69例作为观察组,慢性鼻炎患者69例作为对照组.均留取组织标本,应用实时荧光定量聚合酶链反应法检测两组中miR-221-3p的表达.蛋白免疫印迹法检测鼻咽癌中BCL-2相关X蛋白(BAX)的表达.免疫组织化学法检测鼻咽癌中Ki67的表达.原位杂交法检测鼻咽癌中EB病毒的表达.结果:观察组miR-221-3p表达高于对照组,差异有统计学意义(P<0.05),miR-221-3p的表达在观察组不同组织学亚型、淋巴结转移、局部浸润程度的分组中差异有统计学意义(P<0.05).生存分析显示miR-221-3p高表达患者的生存时间短(P<0.05).Spearman相关分析显示,miR-221-3p表达与Ki67增殖指数呈正相关(r=0.56,P<0.05),与BAX呈负相关(r=-0.69,P<0.05).结论:与慢性鼻炎患者比较,鼻咽癌中miR-221-3p表达升高.MiR-221-3p高表达促进肿瘤增殖、抑制凋亡,预示病变呈进展状态及较差的预后.
Objective:To study the mechanism of abnormal expression of CDK8 on the cell proliferation, apoptosis and chemosensitivity of human nasopharyngeal carcinoma (NPC).Methods:Cultured cells were divided into three groups:the untreated group (without any intervention), the negative control group (without lentivirus transfection with CDK8 expression) and the transfected group (with lentivirus with CDK8 overexpression), with 12 Petri dishes in each group.Liposomal LipofectamineTM2000 was used to transfect NPC 5-8F cells.The transfection efficiency was verified by Western-blot.The proliferation, apoptosis and cycles of NPC 5-8F cells were analyzed by MTT and flow cytometry (CDK8).Western-blot was used to detect the expression of CDK8 related proteins.The sensitivity of CDK8 to chemotherapeutic agents was detected by MTT.Results:The expression of CDK8 in NPC 5-8F cells transfected with pcDNA3.1/CDK8 was up-regulated (P<0.05).This indicates that NPC 5-8F cells with stable expression of CDK8 gene were successfully constructed.The abnormal expression of CDK8 in NPC 5-8F cells led to slow proliferation and increase the rate of apoptosis, leading to a significant decrease in the expression of proliferating cell nuclear antigen (PCNA) protein, and the expression level of Cyclin D1 was significantly reduced (P<0.05).The apoptosis rate significantly increased (P<0.05).The expression level of multidrug resistance associated protein 1 (MRP1) significantly decreased (P<0.05).The expression level of Bcl-2 was significantly down regulated (P <0.05), and the expression level of Bax significantly increased (P<0.05).Meanwhile, NPC 5-8F cells significantly increased cisplatin sensitivity (P<0.05).Conclusion: Abnormal expression of CDK8 can inhibit the proliferation of NPC 5-8F cells and increase the chemosensitivity of cancer cells.It is presumed that the mechanism is related to the down regulating expression of Cyclin D1, PCNA, MRP1 and Bcl-2, and the up-regulate expression of Bax.
目的 探讨Epstein Barr病毒(EBV)编码的microRNA bart7调节鼻咽癌放疗敏感性的机制. 方法 筛选75例鼻咽癌患者以及同期入院体检的30例健康人群为研究对象.比较两组间鼻咽癌患者EBV/DNA比率.测定鼻咽癌患者的microRNA bart7、EBV VCA IgA、EBV EA IgA、EBV Rta IgG等EBV相关肿瘤标志物阳性状态,分析EBV/DNA与microRNA bart7间的相关性. 结果 鼻咽癌患者血浆EBV/DNA比率高于健康人群(92.00% vs 3.33%,P<0.05).放疗后鼻咽癌患者血浆EBV/DNA比率低于放疗前(30.67% vs 92.00%,P<0.05).放疗后持续缓解的鼻咽癌患者血浆EBV/DNA阳性率低于复发/转移者(12.50% vs 39.22%,P<0.05).放疗后鼻咽癌患者血浆microRNA bart7、EBV VCAIgA、EBV EAIgA、EBV Rta IgG阳性率均低于放疗前(P<0.05).放疗后,持续缓解的鼻咽癌患者血浆microRNA bart7、EBV VCA IgA、EBV EA IgA、EBV RtaIgG阳性率均低于复发/转移者(P<0.05).EBV/DNA阳性率与microRNA bart7阳性率呈正相关(P<0.05). 结论 EBV可能通过调节其编码的microRNA bart7水平对鼻咽癌患者放疗敏感性产生影响.

Background/Aims: The objective of this study was to investigate the potential role of IL-17 in the development of nasopharyngeal carcinoma (NPC) and to screen microRNAs (miRNAs) that potentially target IL-17 in NPC cells. Methods: Blood was collected from NPC patients and normal subjects, and plasma IL-17 concentration was quantified by enzyme-linked immunosorbent assay. An immortalized normal human nasopharyngeal epithelial cell line, NP69, was treated with or without human IL-17 (15 ng/mL) for various times, and expression of IL-1ß, IL-6, IL-12, and TNF-α mRNA was assessed by real-time reverse transcription PCR. The candidate miRNAs that potentially target IL-17 were predicted by a bioinformatics strategy. The selected miR-135a mimic was transfected into primary NPC cells, and cell proliferation was assessed by MTT assay. Results: The concentration of plasma IL-17 was significantly higher in the NPC patients (92.5 ± 7.3 pg/mL) than in the control subjects (56.8 ± 2.9 pg/mL). In response to IL-17 treatment, the mRNA expression of IL-1ß and IL-6 was significantly upregulated and reached a peak at 12 h, followed by a slight decrease at 24 h, while the mRNA expression of IL-12 and TNF-α was significantly upregulated at 12 h and remained high even at 48 h after exposure to IL-17. Moreover, miR-135a specifically targets IL-17 and was dramatically downregulated in NPC cells compared with NP69 cells. Transfection of exogenous miR-135a mimic resulted in significant suppression of IL-17 secretion and subsequent inhibition of NPC cell proliferation. Conclusions: Blood IL-17 was significantly higher in NPC patients compared with normal subjects. Expression of miR-135a in the cancer cells isolated from nasopharyngeal tumors was significantly lower than that in NP69 cells, and suppression of IL-17 by miR-135a mimic resulted in significant inhibition of NPC cell proliferation. These findings suggested that downregulation of miR-135a may contribute to the development of NPC via the mechanism of IL-17 stimulation of proinflammatory cytokine expression.

Objective To assess the therapy effect of long-term low-dose clarithromycin joint nasal steroid in the treatment of chro-nic rhino sinusitis. Methods Totally 112 patients with chronic rhino sinusitis who were treated in our hospital from February 2010 to Au-gust 2012 were selected and randomly divided into the treatment group(65) and the control group(47). The patients in the control group were treated by the long-term low-dose clarithromycin joint nasal steroid and nasal irrigation,while the patients in the treatment group were treated by the nasal steroid and nasal irrigation. Then the visual analogue scale, Lund Mackay and the therapeutic effect were evalua-ted. SPSS 16. 0 software was used to analyze the data. Results The Results were as follws:(1) The visual analogue scale score of rhi-nobyon, nasal mucus, headache and hyposmia in the treatment group significantly decreased(P < 0. 05),while only the visual analogue scale score of rhinobyon in the control group significantly decreased(P < 0. 05). (2) The total effective rate in the treatment group (80%) was significantly higher than the control group (51%). The difference was statistically significant(P < 0. 05). (3)About the Lund-Mackey sinus CT score, the treatment group was significantly lower than the control group,and the difference was also statistically significant( P < 0. 05). Conclusion The long-term low-dose clarithromycin joint nasal steroid is superior to nasal steroid in the total effective rate in treatments of patients with chronic rhino sinusitis. It had convenient economic costs and low incidence of adverse reactions. Therefore it has great value for chronic nasal-sinusitis therapy. We recommend it as a first-line drug combination.

Methods Selected 359 cases with allergic rhinitis from February 2013 to February 2014 in our hospital were analysis. Results Methacholine air passage provocation test positive rate was 77%, the normal control group was 16.7%,χ2=28.56, P<0.0001,had difference statistically significance. Conclusion Elderly patients with allergic rhinitis increased airway reactivity, combine with bronchial asthma of high morbidity, early intervention and actively to treat the elderly allergic rhinitis to retard and prevent the occurred of asthma.
目的:探讨同种异体羊膜在中老年鼓膜修补术的临床应用疗效观察。方法将56例中老年鼓膜穿孔患者随机分为两组,在耳内镜下分别采用同种异体羊膜和自体颞肌筋膜行鼓膜修补术,比较两组患者的临床疗效。结果术后随访3~12月,观察两组鼓膜小穿孔的Ⅰ期愈合率分别为92.8%和91.7%,大穿孔的Ⅰ期愈合率分别为92.9%和85.7%,两组患者手术后的平均听阈均有明显提高,且同种异体羊膜组更明显,差异有统计学意义( P<0.05)。结论采用同种异体羊膜行鼓膜修补具有成功率高、听力改善明显、取材经济方便、操作简单、组织相容性好、损伤小的特点。
OBJECTIVE:To observe the clinical efficacy of nasal vestibule eczema by using beclomethasone dipropionate in combination with He-Ne laser therapy.METHOD:The 200 cases of nasal vestibule eczema patients were randomly divided into treatment and control groups. The control group received the rub beclomethasone dipropionate cream treatment one time per day. The treatment group supplemented with He-Ne laser irradiation treatment on the basis of the control group on the same treatment, alsoone time per day. Then the results were analyzed.RESULT:The total effective rate was 100.0% in treatment group, while 75.0% in the control group. The difference between the two groups was statistically significant (P < 0.05).CONCLUSION:Beclomethasone dipropionate cream combined with He-Ne laser irradiation therapy on nasal vestibule eczema is significant, and easily to operate, with significant anti-inflammatory, anti-itch, analgesic effects.
