Objective:Tostudy the effects of Bushen Qutan Formula on leptin,Ob-Rb,TNFα,INFγ,PGE2,MMP-9、MMP-13 and MMP-1 mRNA experssion levels in obesity rat of kidney-deficiency and osteoarthritis by PCR.Method:The rats were dividied into normal group,model group,Bushen Qutan Formula low-dose,middle-dose and high dose groups,glucosamine sulfate capsule suspension group.The normal group and model group wereadministered with normal saline by intravenous drip and various treatment groups were administered with corresponding doses for continuous 7 days by intravenous drip.Rats were killed after the last administering.In the bacteria-free environment,we decalcified the cartilage and studied the levels of leptin,Ob-Rb,TNFα,INFγ,PGE2,MMP-9,MMP-13 and MMP-1 mRNA experssions in rats were observed by PCR after getting the cartilage.Result:Compared with the normal group,there was no statistical significance of Bushen Qutan Formula on expressionsof Ob-Rb,TNFα,INFγ,PGE2,MMP-9 and MMP-1 mRNA.There was obvious statistical significance ofBushen Qutan Formula on leptin and MMP-13 mRNA.The levels of leptin and MMP-13 mRNA experssions in model group wereincreased (P < 0.01).Compared with the model group,the levels of leptin and MMP-13 mRNA experssion in Bushen Qutan Formula low-dose,middle-dose,high-dose groups and glucosamine sulfate capsule suspension group were decreased (P < 0.05),especially in the Bushen Qutan Formula high-dose group.Conclusion:The obesity rat model of kidney-deficiency and osteoarthritis is simple and steady,which would supply the new method of making model.Bushen Qutan Formulacan decrease the levels of leptin and MMP-13 mRNA experssion in obesity rat of kidney-deficiency and osteoarthritis,which would provide a theoretical basis for clinical study of Bushen Qutan Formula.
目的 建立蛇黄凝胶的质量标准.方法 采用薄层色谱法对方中各药进行定性鉴别,采用HPLC对方中主药蛇鳞草中triphyllinA和大黄中大黄酸进行含量测定.结果 薄层色谱中特征斑点明显、重复性好、易于区别.triphyllinA在0.006 4~0.051 2 mg/mL范围内线性关系良好(r=0.999 9),平均回收率为100.5%,RSD为1.6%(n=6);大黄酸在0.003 2~0.025 6 mg/mL范围内线性关系良好(r=0.999 9),平均回收率为99.4%,RSD为1.1%(n=6).结论 所建立的方法简便、灵敏、准确、专属性强,可作为蛇黄凝胶的质量控制方法之一.
目的 建立蛇黄散(蛇鳞草、大黄、三角草、山芝麻和独行千里)的质量标准.方法 TLC法鉴定方中蛇鳞草、大黄、三角草、山芝麻;采用HPLC法测定三羽新月蕨苷A、大黄酸以及大黄素的量,色谱柱为Boston Green ODS C18 (4.6 mm ×250 mm,5μm),流动相为乙腈-甲醇-0.1%磷酸水溶液,梯度洗脱,体积流量1 mL/min,检测波长为226 nm.结果 薄层鉴别色谱中特征斑点明显,重复性较好.三羽新月蕨苷A在0.016~0.128 mg/mL范围内线性关系良好(r =0.999 9),平均回收率为100.5%,RSD为1.6%(n=6);大黄酸在0.008~0.064 mg/mL范围内线性关系良好(r =0.999 9),平均回收率为99.4%,RSD为1.1%(n=6);大黄素在0.012~0.096 mg/mL范围内线性关系良好(r =0.9999),平均回收率为99.3%,RSD为1.9%(n=6).结论 本法采用的HPLC流动相能改善三羽新月蕨苷A色谱峰拖尾现象.
Objective To study the molding process of Shehuang Gel. Methods By experiment on percutaneous absorption in vitro,the content of percutaneous absorption in vitro of triphyllin A in Pronephrium triphyllum and rhein in rheum served as the key evaluation index,the amount of carbomer,azone and glycerin in Shehuang Gel were studied by orthogonal design. Results The content of azone had statistical significance( P 0. 05),and the content of carbomer and glycerin had no statistical significance. The optimal conditions determined as follows: 1. 7% carbomer 1% azone and 5% glycerin. Conclusion This optimized process of Shehuang Gel is simple and feasible,and the prepared gel exhibited fine consistency and moderate viscosity with stable quality.
目的:优选蛇黄凝胶的提取工艺.方法:采用L9(34)正交试验设计,以三羽新月蕨苷A、大黄素和干膏得率为评价指标,在单因素试验基础上,通过综合评分优选蛇黄凝胶的提取工艺.采用HPLC测定三羽新月蕨苷A、大黄素含量,流动相乙腈(A)-0.1%磷酸水(B)-水(C)梯度洗脱(0 ~ 40 min,10% ~ 70%A,10%B,80%~20% C),检测波长226 nm.结果:最佳提取工艺条件为加14倍量70%乙醇提取3次,每次30 min;大黄素、三羽新月蕨苷A平均提取量依次为0.37,0.76 mg·g-1.结论:优选的提取工艺稳定可行,对蛇黄凝胶的生产工艺具有一定的指导和参考意义.
目的:为忧遁草抗肿瘤作用的进一步研究提供思路.方法:查阅近40年来国内、外的相关文献,对忧遁草化学成分的研究进展进行概括与归纳.结果与结论:目前国外对忧遁草化学成分的研究相对较多,已知其含有丰富的化学成分,如三萜类、黄酮碳苷类、含硫糖苷类、植物甾醇类和脱镁叶绿素等,这些成分大部分已被现代药理研究证实对肿瘤有一定的抑制作用.临床应加强对忧遁草抗肿瘤作用机制的研究,明确其有效物质基础,充分挖掘其药用价值并合理利用其植物资源.
OBJECTIVE:To optimize the extraction conditions for triphyllin A in Pronephrium triphyllum.METHODS:The content of triphyllin A in the Pronephrium triphyllum was determined by HPLC. Concentration and volume of the alcohol,time and times of the extraction were assayed by orthogonal test to detect their influences on the extraction rate of triphyllin A in the Pronephrium triphyllum.RESULTS:Alcohol volume and extraction times had significant influence on the process (P < 0.05) while alcohol concentration and extraction time had no effect. The optimal extraction conditions were as follows:50 fold 60% alcohol, extraction for 3 times and 50 min for each time.CONCLUSION:The extration rate of triphyllin A is higher,and the process can be used for the development and production of Pronephrium triphyllum.
目的了解中药经皮给药凝胶剂的研究与应用概况。方法根据经皮给药制剂在国内外的研究概况,通过文献检索,综述了凝胶剂在中药经皮给药方面的基质与渗透促进剂选择、新型凝胶及临床应用。结果中药经皮凝胶剂不仅具有使用方便、舒适、生物相容性好等多种优点,还可避免首过效应、减轻药物的不良反应,符合中医"内病外治"的理念。结论目前,凝胶剂在中药经皮给药方面的研究尚浅,随着研究的进一步深入,将会有更多的新技术与新方法被引入,使凝胶剂在中药经皮给药领域拥有更广阔的发展空间。
Objective To study the spore of Pronephrium triphyllum(Sw.) Holtt.Methods The spore of Pronephrium triphyllum(Sw.) Holtt was investigated under scanning electron microscope.The morphobgical characteristics of this spece was described and summarized.Results and Conclusion The spore is monolete and bilaterally symmetric,ellipsoidal in polar view,and kidney-shaped or asbrounded in equatorial view.The polar axe is 37.6 μm long,and the equatorial axes is 24.5 μm.The surface of the spore is echinulate.
