Salt-tolerant germplasm of sweet potato is screened by evaluating the salt tolerance of sweet potato tissue culture seedlings. In this study, seven sweet potato varieties were used as test materials, and the stem segments of the test materials were inoculated into Murashige and Skoog(MS) media containing 0, 2, 4, 6, 8and 10 g/L NaCl, followed by the measurement of vine length, germination rate, rooting rate, root number and salt damage index after 30 days. The comprehensive evaluation of salt tolerance for sweet potato materials was carried out by using Statistic Package for Social Science(SPSS) cluster analysis and membership function. The results showed that‘Funingzi No.4’and‘Funingzi No.5’were sensitive materials. The root growth was completely inhibited and vine growth was inhibited by 90% in MS medium containing 2 g/L NaCl.‘Funingzi No.3’,‘Funingzi No.6’,‘Funingshu No.12’,‘Funingshu No. 20’and‘Funingshu No.23’showed certain resistance to salt damage in the appropriate concentration range of salinity. All the indicators indicated that,the seven sweet potato varieties were ordered in terms of salt tolerance strength as follows:‘Funingshu No.12’>‘Funingzi No.3’>‘Funingzi No.6’>‘Funingshu No. 20’>‘Funingshu No. 23’>‘Funingzi No.4’>‘Funingzi No.5’.
为选育高产优质紫甘薯新品种,以福薯13号为母本,采用集团杂交育成的甘薯新品种福宁紫4号,于2012-2013年参加福建省区域试验.结果表明,福宁紫4号2 a平均鲜薯亩产量2 888.72 kg,比对照品种金山57(CK1)增产7.38%;薯干亩产量672.78 kg,比CK1增产1.84%;并于2015年通过福建省品种审定,2019年通过农业农村部品种登记.福宁紫4号具有高产、抗病、较优质、适应性广等特点,早、晚薯均可种植,适宜在福建省大面积推广应用.
2019-2021年连续3年对软枣猕猴桃'奇异莓6号'在福建寿宁地区种植的植物学特征、生物学特性、适应性及果实主要经济性状等进行了系统观测与研究.结果表明,'奇异莓6号'植株生长势较强、枝梢生长健壮、适应性、抗逆性强,在田间未发现溃疡病等病害.种植第2年开始结果,第4年平均株产160个果,平均单果质量18.81 g,平均单株产量3008 g,可溶性固形物含量16.8%~18.7%,最高可达22%,风味甜香,且果实成熟期较早,可作为早熟品种在福建适宜生态区种植.
通过对国内关于软枣猕猴桃组织培养技术的相关文献进行归纳和总结,综述了国内软枣猕猴桃组织培养中外植体、基本培养基、激素选择等方面的内容,以期为该领域今后的研究提供参考.
为防止紫甘薯新品种福宁紫4号和5号田间种植感染病毒导致优良种性退化,运用茎尖分生组织培养获得脱毒苗.试验结果表明,福宁紫4号和5号茎尖诱导成苗培养基为MS+6-BA 0.1 mg/L+NAA 0.01 mg/L,成苗率达70.8%,继代增殖培养基为1/2MS基本培养基,福宁紫4号月均繁殖系数达2.83倍,福宁紫5号月均繁殖系数达2.41倍;福宁紫4号脱毒种薯产量比对照原品种提高11.60%,达极显著水平,5号脱毒种薯产量比对照原品种提高5.27%,达显著水平.
为可持续开发霍山石斛资源,采用响应面法优化霍山石斛原球茎增殖培养基.在单因素试验基础上,根据Box-Behnken试验设计原理,采用三因素三水平的响应面分析法,以增殖率为响应值进行回归分析.结果 表明,萘乙酸(简称NAA)及6-苄氨基嘌呤(简称6-BA)浓度对增殖率影响最为显著,6-BA与马铃薯两者间交互作用显著,适宜的NAA、6-BA及马铃薯浓度分别为0.05 mg·L-1、0.70 mg·L-1和185 g·L-1,此时增殖率为2 989.82%±205.55%,与模型预测值基本符合.响应面法优化霍山石斛原球茎增殖培养基切实可行.
目的 建立金线莲工厂化高效育苗技术.方法 以金线莲种子在培养基上萌发获得原球茎、不定芽,通过正交试验对愈伤组织的诱导和分化先行筛选,然后根据响应面分析法中心组合试验,以愈伤组织分化率为响应值,对6-BA、NAA、芋泥3因素进行优化.结果 正交试验分析结果表明:影响愈伤组织诱导和分化的关键因子为外植体类型,不定芽类型最易诱导产生愈伤组织,诱导率较高,达到27.64%,筛选出的最优培养基为1/2MS+6-BA3.0mg/L+ NAA0.5mg/L+芋泥200g/L,在该培养基上继代培养,愈伤组织月增殖系数达50倍.利用响应面优化得到的愈伤组织分化培养基为1/2MS+6-BA1.0mg/L+ NAA0.46mg/L+芋泥200g/L,预测分化率为为83.16%,验证值为81.4%.生根壮苗培养1/2MS+ NAA0.5 mg/L+80g/L香蕉+7g/L芋泥.结论 因此利用响应面方法可有效优化金线莲愈伤组织分化培养条件,可应用于实际生产.
为调整福建省芋栽培品种结构,加快优良品种推广应用,以宁糯芋1号田间生长的块茎顶芽或侧芽为外植体,运用茎尖分生组织培养进行了不同TDZ浓度处理试验获得脱毒苗,结果发现,与6-BA相比,TDZ更适于芋茎尖诱导成芽,在MS培养基中附加0.1~0.2 mg/L的TDZ能显著促进芋茎尖的萌动,产生丛生芽.茎尖初代培养基为MS+TDZ 0.2 mg/L时成苗率达78%,继代增殖培养基MS+TDZ 0.3 mg/L繁殖系数达7.88,生根壮苗培养基1/2MS+NAA 0.5 mg/L+活性炭0.2 mg/L.脱毒芋种产量比对照原品种提高29.7%,达到极显著差异水平.
为丰富福建省优质芋种源,加快优良品种的推广应用,以‘宁糯芋1号’块茎顶芽和侧芽为外植体,运用茎尖分生组织培养获得了不定芽.在正交试验基础上,根据响应面分析法中心组合试验,以不定芽增殖系数为响应值,对TDZ、温度2个因素进行优化.结果 发现,与6-BA相比,TDZ更适于芋茎尖诱导丛芽,在MS培养基中附加0.1~0.2 mg/L的TDZ能显著促进芋茎尖的萌动,成苗率达76.7%~80.0%.经芋花叶病毒(DMV)检测试剂盒的ELISA检测表明,诱导芽的脱毒率为90%;响应面优化结果表明,‘宁糯芋1号’不定芽继代增殖的最佳培养条件为MS+TDZ 0.31 mg/L,培养温度30.32℃,增殖系数预测值为9.08,验证得到的实际值为8.86.利用响应面方法可有效优化‘宁糯芋1号’不定芽继代增殖培养条件.
目的 通过研究水浴热烫炮制对栀子中栀子苷(Geniposide)含量的影响,建立最佳的水浴热烫炮制方法.方法 将霜降前和霜降后采收的栀子经过不同温度和不同时间的水浴热烫处理后,用HPLC检测其中栀子苷含量.结果 栀子的最佳采收期为霜降后,即1/3青2/3黄时采收为宜;水浴热烫炮制最佳方法为90℃下处理10min,Geniposide的含量可达7.12%.结论 霜降后采收的栀子中Geniposide含量高于霜降前采收的;水浴热烫炮制法可提高栀子中Geniposide含量.
通过对近年来国内外关于铁皮石斛栽培技术的相关文献进行归纳和总结,发现适合铁皮石斛的种植基质多种多样,这些基质有共性,也有特性.通过对其栽培环境因子的研究,为合理开发和种植铁皮石斛提供理论依据.
为防止紫甘薯新品种福宁紫3号经田间无性繁殖感染病毒导致优良种性退化,运用茎尖分生组织培养获得脱毒苗.试验结果表明,福宁紫3号茎尖诱导成芽培养基为MS+6-BA(0~0.1 mg/L)+NAA (0~0.5 mg/L),成苗率达50%~66.7%,继代增殖培养基为MS基本培养基,繁殖系数达5~6倍;福宁紫3号脱毒种产量比对照原品种提高5.13%,差异达到显著水平.
从金线莲生物学特性、成分分析、生理活性及分子生物学等方面的研究结果进行综述,并讨论目前金线莲生产及应用方面存在的主要问题.
A pot experiment was carried out to study the effect of Brevibacillus brevis FJAT-0809-GLX on the growth of tomato plants.The results showed that the leaf count was significantly increased when the plants were sprayed with the bacteria,the fermentation supernatant or the fermented liquid.Both the length and diameter of the stems were also significantly greater than those of control.The stems of the tomato plants treated with the bacteria were the longest and the numbers of leaves greatest among all treatments.B.brevis FJAT-0809-GLX significantly enhanced the chlorophyll and carotenoids contents,reaching the highest when the plants were sprayed with the fermentation supernatant.As compared to control,the chlorophyll a,b and carotenoids contents increased by111.3%,136.0% and 102.7%,respectively.B.brevis FJAT-0809-GLX appeared to induce significant effect on promoting the growth of the tomato plants in the laboratory.The result prompted an inquiry for further test in the field.
In order to identify the causal agent of black spot disease on wax apple(Syzygium samarangense Merr.et Perry)in its taxonomic status,the causal fungi FJAT-9860 was isolated and characterized with pathogenicity test,morphology and culture feature observation,rDNA-ITS sequences assay and biological characteristics test.The results indicated that the causal agent belong to Lasiodiplodia theobromae.The colony of strain FJAT-9860 on PDA was white at first,then gradually became as grey to black.The monadic conidia were elliptic to round,and colourless.The rDNA-ITS sequence of the strain shared 99%identity with that of Lasiodiplodia theobromae strain.The results showed that the optimum temperature for mycelium growth of this pathogen was 25℃and the optimum pH was 8.0.The mycelial growth was affected by different carbon sources and the best one was mannitol.
The volatile compounds produced strains of Bacillus spp.were antagonistic to the loquat anthracnose pathogen(Colletotrichum acutatum)in two-compartment plastic plates.The results showed that the inhibition rates of FJAT-4748 and FJAT-10011 were 90.06%and 54.99%,which were better than the other tested strains.Colony morphology and 16 SrDNA sequence assays showed that the tested eight bacterial strains of Bacillus spp.were Lysinibacillus xylanilyticus、Brevibacillus borstelensis,Brevibacillus brevis,Brevibacillus agri,Bacillus aryabhattai,Brevibacillus formosus and Bacillus simplex,respectively.Our results suggested that these volatile compounds produced Bacillus strains could be promising biocontrol strains for anthracnose in harvested loquat fruit.
