Objective:To investigate the effects of serum medicated withWenyang Bushen Formula (温阳补肾方) on osteoclast apoptosis rate and RANK protein.Methods:Differentiation in vitro was made from the osteoclast precursor cells RAW264.7 to osteoclast,which was interfered respectively with low,medium and high dose ofWenyang Bushen Formula and normal saline serum.The flow cytometry was used to detect the apop-tosis rate of each group,and the Western Blot was used to detect the level of RANK protein in each group.Results:Compared with the blank control group,serum medicated withWenyang Bushen Formula could induce apoptosis of osteoclasts(P = 0.000),and reduce the expression of osteoclast RANK protein(P = 0.000).Conclusion:Serum medicated withWenyang Bushen Formula can induce apoptosis of osteoclasts,high dose of which is the best.
Objective: To explore the effects of serum containing Wenyang Bushen Formula on expression of bone resorption in osteoclasts.Methods: The preosteoclasts of RAW264.7 cell were induced to mature osteoclast by RANKL and M-CSF.Then cultured the mature osteoclast and the Wenyang Bushen Formula,low,medium and high dose group,and normal saline(NS) contained serum were respectively added.Vigour of serum TRACP were measured by biochem istry for cell culture fluid.The expression of Cts K and MMP-9 mRNA was measured by real time QPCR.Results: Compared with control group,Wenyang Bushen Formula group can significantly inhibited the TRACP activities(P0.01).The Expression of Cts K and MMP-9 in osteoclast of Wenyang Bushen group was significantly lower than NS group(P0.01).Conclusion: Wenyang Bushen Formula can effectively inhibit the expression of bone resorption in osteoclasts,and the effects of Wenyang Bushen Formula medium dose group is the best.
ABSTRACT:OBJECTIVE To investigate effect of Xinhuang tablets and its Chinese medicine components on high uric acid Mice Caused by hypoxanthine , and discusse its mechanism .METHODS The mice models of high uric acid caused by injecting hypoxanthine and the normal mice were adopted .After giving different doses of intragastric administration with extracts from Xinhuang tablets and its Chinese medicine components on the mice by gavage ,the uric and xanthine oxidase level of each group were determined .RESULTS After 1h of intraperitoneal injection of hypoxanthine ,positive control group and Xinhuang tablets and its Chinese medicine components dosage ( 23.75、47.5、95 mg? kg -1 ) could against high uric mice induced by hypoxanthine and reduced the activity of xanthine oxi -dase in hepar ( P<0.01 ) .CONCLUSION Xinhuang tablets and its Chinese medicine components could decrese the blood uric acid levels by inhibiting the activity of xanthine oxidase .
目的:观察灌胃凤凰衣水解物对膝骨关节炎大鼠膝关节的影响,进而考察凤凰衣用于防治OA的合理性及可行性.方法:灌胃凤凰衣水解物及奥泰灵胶囊干预膝骨关节炎大鼠,观察软骨形态及软骨中Collagen-Ⅱ、MMP-3及蛋白多糖的指标变化,以及血清和关节液中IL-6,IL-1β,TNF-α的指标变化.结果:1.各用药组对降低膝骨关节炎大鼠血清IL-1β、IL-6、TNF-α表达,关节液中TNF-α的表达以及软骨中MMP-3表达,升高Collagen-Ⅱ的表达看,两用药组未见明显差异(P>0.05),但均优于模型组(P<0.01);2.在提高蛋白多糖含量及降低关节液中IL-6的表达方面,灌胃凤凰衣组优于灌胃奥泰灵组(P<0.01);3.在降低关节液中IL-1β的表达方面,灌胃奥泰灵组优于灌胃凤凰衣组(P<0.01).结论:1.灌胃凤凰衣水解物能降低膝骨关节炎大鼠的炎症因子表达,提高关节软骨中蛋白多糖和Collagen-Ⅱ含量;2.凤凰衣中含有丰富的可以合成软骨基质中蛋白多糖的前体物质,是其改善OA指标的机制之一.
目的:探讨龟鹿二仙胶不同提取部位对豚鼠软骨细胞活性的影响.方法:采用溶剂极性依次递增分离法分别制备龟鹿二仙胶的三氯甲烷提取物、正丁醇提取物和水提取物.然后分别以三氯甲烷提取物(烷提组)、正丁醇提取物(醇提组)和水提取物(水提组)及生理盐水(对照组)对4组Wistar大鼠进行灌胃,制备含药血清,再分别以这些含药血清对体外培养的豚鼠软骨细胞进行干预.干预结束后分别采用免疫组织化学技术、实时荧光定量聚合酶链式反应法及酶联免疫吸附法测定经4种含药血清干预后的豚鼠软骨细胞中Ⅱ型胶原蛋白、Bcl-2和Bax基因及MMP-3水平.结果:①Ⅱ型胶原蛋白水平.4组细胞Ⅱ型胶原蛋白水平比较,差异有统计学意义[(0.178±0.002),(0.188±0.003),(0.177±0.003),(0.173 ±0.001),F=32.905,P=0.000];对照组Ⅱ型胶原蛋白水平低于其余3组(P=0.003,P=0.000,P=0.023);醇提组高于烷提组和水提组(P=0.000,P=0.000);烷提组和水提组比较,差异无统计学意义(P =0.367).②Bcl-2基因和Bax基因水平.4组细胞Bcl-2基因水平比较,差异有统计学意义[(0.569±0.052),(l.000±0.000),(0.636±0.072),(0.387±0.061),F=136.767,P=0.000];对照组低于其余3组(P =0.000,P=0.000,P=0.000);醇提组高于烷提组和水提组(P=0.000,P=0.000);烷提组低于水提组(P=0.045).4组细胞Bax基因水平比较,差异有统计学意义[(1.789±0.240),(1.000 ±0.000),(1.374±0.226),(2.251±0.196),F=47.445,P=0.000];对照组高于其余3组(P=0.000,P=0.000,P=0.000);醇提组低于烷提组和水提组(P=0.000,P=0.003);烷提组高于水提组(P=0.000).③基质金属蛋白酶3水平.4组细胞基质金属蛋白酶3水平比较,差异有统计学意义[(11.828±0.363)ng·mL-1,(10.685 ±0.317)ng·mL-1,(11.397±0.314) ng·mL-1,(13.136±0.379) ng·mL-1,F=71.401,P=0.000];对照组高于其余3组(P =0.000,P=0.000,P=0.000);醇提组低于烷提组和水提组(P=0.000,P=0.000);烷提组高于水提组(P=0.019).结论:龟鹿二仙胶的三氯甲烷、正丁醇和水3种提取部位均能提高软骨细胞中Ⅱ型胶原和Bcl-2基因水平,降低Bax基因和基质金属蛋白酶3水平,其中以正丁醇提取部位的作用最强.
新癀片由肿节风、三七、人工牛黄、猪胆汁膏、肖梵天花和吲哚美辛等组成,具有清热解毒、活血化瘀、消肿止痛功效,主治热毒瘀血所致的咽喉肿痛,牙痛,痹痛,无名肿毒等症.新癀片中药组分是在新癀片的基础上去吲哚美辛而制成.近年来,新癀片在临床上也用于治疗痛风性关节炎[1-2],但未见新癀片对痛风性关节炎疗效的药效学报道.我们采用氧嗪酸钾引发小鼠出现高尿酸,观察新癀片及新癀片中药组分对小鼠血尿酸的影响.
