目的 分析新癀片外用对小鼠镇痛模型的有效成分,为新癀片镇痛的药效成分研究提供科学依据和参考.方法 ICR小鼠50只,雌雄各半,随机分为5组:模型组?阳性对照组?给药组?全方中等剂量吲哚美辛组?全方缺吲哚美辛群药组,每组10只.连续给药3d,给药后40min醋酸造模,立即观察并记录小鼠出现扭体的潜伏期和20min内小鼠发生扭体反应的次数.雌性ICR小鼠50只,根据基础痛阈值分为5组,给药后30min?60min?120min?180min和240min测定痛阈值.结果 与模型组相比,阳性对照组?给药组和全方中等剂量吲哚美辛组的潜伏期均延长(P<0.05),扭体次数降低(P<0.05).而全方缺吲哚美辛群药组潜伏期较模型组有所延长,扭体次数有所减少,但无统计学差异(P>0.05).给药后30min到给药后240min,阳性对照组和给药组的痛阈值较模型组有所延长(P<0.05).而全方中等剂量吲哚美辛组和全方缺吲哚美辛群药组的痛阈值与模型组比较差异无统计学意义(P>0.05).结论 新癀片中的吲哚美辛和中药成分之间相互作用,相互协同,共同参与小鼠的镇痛作用.
目的 探讨新癀片外用制剂的最佳溶媒,为新癀片临床外用提供参考依据.方法 扭体实验:选择ICR小鼠60只,雌雄各半,随机分为生理盐水组、阳性对照组、溶媒水组、溶媒醋组和溶媒酒组各12只,连续经皮给药3 d,最后1次给药后40 min腹腔注射1%冰醋酸生理盐水溶液0.1 ml/10 g,比较各组小鼠扭体反应的潜伏期和扭体反应次数.舔足实验:选取ICR小鼠60只,雌性,随机分为生理盐水组、阳性对照组、溶媒水组、溶媒醋组和溶媒酒组各12只,连续经皮给药3 d,比较各组小鼠最后1次给药后30、60、120、180和240min痛阈值.结果 扭体实验:阳性对照组、溶媒水组和溶媒酒组小鼠扭体反应的潜伏期长于生理盐水组(P<0.05或P<0.01),溶媒水组和溶媒酒组小鼠扭体反应的潜伏期长于溶媒醋组(P<0.05).阳性对照组、溶媒水组、溶媒醋组和溶媒酒组扭体反应次数均少于生理盐水组(P<0.01);溶媒水组、溶媒醋组和溶媒酒组小鼠的扭体反应次数比较差异无统计学意义(P>0.05).舔足实验:溶媒水组、溶媒醋组小鼠给药30、60、120、180和240 min痛阈值高于生理盐水组(P<0.05或P<0.01);阳性对照组、溶媒酒组小鼠给药后60、120、180和240 min痛阈值高于生理盐水组(P<0.05或P<0.01).结论 新癀片外用制剂的溶媒水、溶媒酒、溶媒醋中,溶媒水的镇痛效果较佳.
Intestinal mucosal barrier dysfunction is involved in the pathogenesis of inflammatory bowel disease, including ulcerative colitis (UC). Xinhuang tablets (XHTs) have been prescribed for several kinds of inflammatory diseases, including UC, whereas its possible underlying molecular mechanisms had never been explored. Mouse model of UC was constructed by DSS treatment and followed by XHT treatment. Disease activity index, histopathological of colonic tissue, tumor necrosis factor-alpha (TNF-α), and serum amyloid A (SAA) levels in serum were further assessed. The underlying mechanism was further explored by determination of the expression of epithelial tight junction-related protein. XHT administration ameliorated dextran sulfate sodium (DSS)-induced clinical symptoms, colonic histological injury, and decreased the circulating levels of TNF-α and SAA. Moreover, XHT treatment significantly increased the protein levels of zona occludens (ZO)-1, whereas decreased the levels of phosphorylation of Elk-1. In conclusion, this study confirmed the therapeutic effects of XHT treatment on UC in a DSS-induced mouse model, and indicated that by increasing expression of epithelial tight junctions and decreasing phosphorylation of Elk-1 might be one of the underlying mechanisms of XHT treatment on UC.
目的 通过对芪枣颗粒进行薄层鉴别、定量测定等研究,建立了合理可行的质量控制方法.方法 TLC法对黄芪进行定性鉴别.HPLC-ELSD法定量测定芪枣颗粒中黄芪甲苷含量.结果 薄层色谱中,黄芪斑点清晰,专属性强.含量测定中,黄芪甲苷在0.942μg~9.42μg(r=0.9994)范围内线性关系良好,平均回收率100.20%(RSD为3%).结论 所建立的方法简便、准确,可以有效控制芪枣颗粒的质量.
目的 探讨新癀片对牛奶致家兔发热模型的解热作用机制.方法 制备牛奶致家兔发热模型,测定给药后1、4h体温,并取下丘脑测定与体温相关的白细胞介素1α (IL-1α)、-p、IL-6、前列腺素E2 (PGE2)、肿瘤坏死因子α(TNF-α)、精氨酸加压素(AVP)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、Na+,K+-ATP等指标.结果 新癀片给药1、4h后均能显著降低家兔体温,显著降低家兔下丘脑IL-1α、IL-1β、IL-6、PGE2、TNF-α、cAMP水平及cAMP/cGMP值,降低Na+,K+-ATP活力;给药后4h能显著升高AVP水平.与吲哚美辛比较,新癀片给药后4h能进一步降低家兔下丘脑IL-1β、IL-6、cAMP水平及cAMP/cGMP值,差异显著(P<0.05),进一步降低PGE2水平,给药后1h能显著降低IL-1α水平.结论 新癀片的解热作用机制与调节下丘脑中体温调控相关因子IL-1α、IL-1p、IL-6、PGE2、TNF-α、cAMP、cAMP/cGMP、Na+,K+-ATP、AVP有关;其中药成分与吲哚美辛发挥协同作用,从而延长吲哚美辛解热作用时间并且增强吲哚美辛的解热效果.
ABSTRACT:OBJECTIVE To investigate effect of Xinhuang tablets and its Chinese medicine components on high uric acid Mice Caused by hypoxanthine , and discusse its mechanism .METHODS The mice models of high uric acid caused by injecting hypoxanthine and the normal mice were adopted .After giving different doses of intragastric administration with extracts from Xinhuang tablets and its Chinese medicine components on the mice by gavage ,the uric and xanthine oxidase level of each group were determined .RESULTS After 1h of intraperitoneal injection of hypoxanthine ,positive control group and Xinhuang tablets and its Chinese medicine components dosage ( 23.75、47.5、95 mg? kg -1 ) could against high uric mice induced by hypoxanthine and reduced the activity of xanthine oxi -dase in hepar ( P<0.01 ) .CONCLUSION Xinhuang tablets and its Chinese medicine components could decrese the blood uric acid levels by inhibiting the activity of xanthine oxidase .
目的:建立芪骨胶囊的多成分指标定量方法.方法:采用高效液相色谱法同时测定芪骨胶囊中松果菊苷、朝藿定B,朝藿定C,淫羊藿苷和柚皮苷的含量,Platisil-C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-乙腈-水为流动相进行梯度洗脱,流速1 mL· min-1,检测波长270 nm,柱温30℃.结果:松果菊苷,柚皮苷,朝藿定B,朝藿定C,淫羊藿苷分别在91 ~3 094 ng(r=0.999 8),19 ~646 ng(r =0.999 7),11~374 ng(r =0.999 5),25 ~850 ng(r =0.999 5),29 ~986 ng(r=0.999 6)与峰面积呈良好的线性关系;平均回收率分别为98.9% (RSD 1.2%),99.1%(RSD 1.3%),99.1% (RSD 1.5%),99.4% (RSD 1.5%),99.3% (RSD 1.5%).结论:所建立的方法简便、准确、重复性好,可用于芪骨胶囊的质量控制.
目的 建立芪骨胶囊HPLC指纹图谱,为评价其质量提供依据.方法 Platisil-C18色谱柱(250 mm×4.6 mm,5μm);以甲醇-乙腈-水为流动相梯度洗脱;柱温30℃;体积流量1.0 mL/min;检测波长270 nm.结果 建立了15批芪骨胶囊的对照指纹图谱,确定了16个共有峰,其中15个归属到各药材,其相似度均大于0.9.通过对照品对照、UFLC-ESI-MS/MS鉴定了其中8个色谱峰的化学成分.通过聚类分析,15批芪骨胶囊可分为3类.结论 该方法操作简便、准确可靠、重复性较好,为芪骨胶囊的质量控制提供了有效手段.
新癀片由肿节风、三七、人工牛黄、猪胆汁膏、肖梵天花和吲哚美辛等组成,具有清热解毒、活血化瘀、消肿止痛功效,主治热毒瘀血所致的咽喉肿痛,牙痛,痹痛,无名肿毒等症.新癀片中药组分是在新癀片的基础上去吲哚美辛而制成.近年来,新癀片在临床上也用于治疗痛风性关节炎[1-2],但未见新癀片对痛风性关节炎疗效的药效学报道.我们采用氧嗪酸钾引发小鼠出现高尿酸,观察新癀片及新癀片中药组分对小鼠血尿酸的影响.
[Objective] To investigate the anti-inflammatory effect and mechanism of Xinhuang tablet.[Methods] The anti-inflammatory models were induced by bilaterally adrenalectomized mice,carrageenan-induced hind swelling in rats,peritonitis model were induced by glacial acetic acid,the content of COX-2,HIS,PGE2 and IL-1α,IL-1β,IL-6,IL-10 were measured.[Results] Xinhuang tablets show a marked suppressive effect on carrageenan-induced hind swelling.It's effect related to inhibition of COX-2,reduction of IL-1α,IL-1β,HIS,increasing IL-6,IL-10.[Conclusion] The anti-inflammatory effect and mechanism of Xinhuang tablets may related to reduce IL-1β and HIS,inhibit cellular and humoral immunity.
OBJECTIVE To set the quality standard of Danzhi xiaoyao Pills.METHODS Microscopy was used for the identification of Rhizoma Atractylodis Macroce,Poria,Radix Bupleuri and Cortex Moutan.TLC was used for the identification of Radix Angelicae Sinensis and Radix et Rhizoma.Glycyrrhizae HPLC was used for the determination of geniposide in Danzhi Xiaoyo Pills.RESULTS Rhizoma Atractylodis ,Poria,Radix Bupleuri and Cortex Moutan were identified by Microscopy,Radix Angelicae Sinensis and Radix et Rhizoma Glycyrrhizae were identified by TLC,the average recovery was 99.5%,RSD=1.57%(n=9).CONCLUSION The methods in the article was highly specific simple and rapid, excellent precision, which can be used as the idex of quality contrl.
