肥胖是多囊卵巢综合征(PCOS)常见的临床表现和负面影响因素,PCOS肥胖女性在脂代谢方面有其独特的特点,腹围增加、HDL降低是其最显著的特征,脂质的异常代谢会引起或加重患者胰岛素抵抗(IR),与高雄激素三者之间形成恶性代谢循环,增加患者发生代谢综合征的风险.随着年龄增长,代谢异常成为影响PCOS女性健康的主要问题.此外,PCOS女性循环中炎症因子显著高于非PCOS女性,肥胖可能加重PCOS患者的慢性炎症及氧化应激反应.PCOS肥胖的发病机制尚未明确,研究发现与某些基因及其变异有关.研究证实,FTO及其相关变异、apM1基因及其多态性与PCOS肥胖存在相关性,但尚缺乏足够证据证明MC4R、TCF7L2及INSIG2基因与PCOS肥胖存在确切联系.
目的 应用膳食平衡指数评价利用辅助生殖技术助孕的高龄不孕女性的膳食质量,为今后改善高龄不孕女性营养状况,提出针对性的营养干预措施提供科学依据.方法 选取2018年9月至2019年9月在山东大学附属生殖医院因不孕需利用辅助生殖技术助孕的高龄不孕女性457人,利用食物频率法调查并分析其膳食摄入情况,应用膳食平衡指数DBI-16评价患者的膳食质量.结果 高龄不孕女性的膳食状况摄入不足和摄入过量同时存在,主要存在低度膳食失衡、低度摄入不足、部分低度摄入过量的问题,分别占73.30%、66.52%、32.60%,其中39岁及以上年龄段人群膳食摄入不足和膳食失衡的程度较低.谷薯类、畜禽肉类存在摄入过量的情况,奶类、鱼虾类、大豆类摄入不足,油盐摄入过量.结论 高龄不孕女性膳食摄入状况有待改善,需要对该人群进行营养宣教及干预,提高其营养健康水平,促进优生优育.
卵巢过度刺激综合征(ovarian hyperstimu- lation syndrome,OHSS)是人类辅助生殖技术(assisted reproductive technology,ART)过程中较严重的并发症[2],是在辅助生殖过程中人体对促排卵药物产生的一种过度反应,中重度OHSS 的发病率约为 3%~8%[3].OHSS 患者血管通透性增大引起富含蛋白的液体外渗,血容量减少甚至威胁患者生命.重度OHSS甚至需要静脉输注白蛋白,但白蛋白可能带来过敏反应、输液反应等风险,并价格较贵,造成患者的生理及经济负 担[4].合理营养和饮食,补充患者蛋白质流失,提高血浆蛋白的水平可能是一种安全、经济且较容易为患者所接受的方法.本次营养干预研究旨在证实营养支持对OHSS患者病情恢复的有效性.
The aim of the current study was to investigate the effect of excess iodine on rat aorta endothelial cells and the potential underlying mechanisms. Rat aorta endothelial cells were cultured with iodide ion (3506, 4076, 4647, 5218, 5789, 6360, 6931, and 7512 mg/L) for 48 h. Morphological changes of cells were observed with microscope after Wright-Giemsa staining and acridine orange staining. Cell proliferation was determined with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, and cell apoptosis was assessed with flow cytometry. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), endothelial nitric oxide synthase (eNOS), induced nitric oxide synthase (iNOS), and concentrations of malondialdehyde (MDA), glutathione (GSH), and protein carbonyl in culture medium were determined with colorimetric method. The expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) was detected by enzyme linked immunosorbent assay. The results showed that excess iodine induced abnormal morphologic changes of cells, inhibited cell proliferation, and increased apoptosis rate. Iodine also reduced the activity of SOD, GSH-Px, and concentrations of GSH and increased the concentrations of MDA and protein carbonyl in a dose-dependent manner. Moreover, excess iodine decreased the activity of eNOS and increased the activity of iNOS and the expression of ICAM-1 and VCAM-1 in culture medium. Our results suggested that excess iodine exposure increased oxidative stress, caused damage of vascular endothelial cells, and altered the expression of adhesion factors and the activity of NOS. These changes may explain the mechanisms underlying excess iodine-induced vascular injury.
<span id="ChDivSummary" name="ChDivSummary" class="abstract-text">目的探讨有机碘和无机碘对体外培养的人甲状腺细胞凋亡相关蛋白Fas、FasL和p53表达影响。方法取正常人甲状腺细胞进行培养,实验设对照组(培养液)和碘化钾组及有机碘组(碘浓度分别为0.1、10、1 000μmol/L),培养48h,采用免疫组织化学技术测定细胞凋亡相关蛋白表达的情况。结果 10、1 000μmol/L碘化钾组与有机碘组甲状腺细胞Fas表达量分别为(0.131 8±0.010 0)、(0.217 4±0.017 7)、(0.124 5±0.007 4)、(0.158 5±0.010 0),均高于对照组的(0.072 8±0.009 3),差异有统计学意义(P<0.01);10、1 000μmol/L碘化钾组与有机碘组甲状腺细胞FasL表达量分别为(0.108 1±0.015 5)、(0.178 4±0.010 4)、(0.093 8±0.007 0)、(1 382±0.008 0),均高于对照组的(0.060 8±0.009 6),差异有统计学意义(P<0.01),呈剂量效应关系;与相同剂量碘化钾组比较,1 000μmol/L有机碘组甲状腺细胞Fas表达量较低,10、1 000μmol/L有机碘组甲状腺细胞FasL表达量较低,差异均有统计学意义(P<0.01);各组甲状腺细胞p53表达量无明显差异。结论高碘可促进甲状腺细胞凋亡的发生,且有机碘对甲状腺细胞的影响小于碘化钾。</span>
Objective To research the influence of high iodine on cellular morphology and the expression of a gene family about apoptosis(TRX-1 and ASK-1). Methods The A7r5 cells were treated with potassium iodide(KI) at the doses of 0, 750, 1 500, 3 750,6 000 and 7 500 mg/L for 24 h. MTT method was used to detect cell viability changes and Western blot assay was used to detect the protein expression of TRX-1 and ASK-1. Results Compared with the control group,A7r5 cell survival rate of 6 000-7 500 mg/L KI treated groups were significantly decreased(P0.01),and the survival rate of cells decreased with the increase of KI exposure dose. 750-7 500 mg/L KI decreased the expression of TRX-1 protein and increased the expression of ASK-1 protein compared with control(P0.01). With the increasing of KI exposure,TRX-1 expression showed a gradual decline trend and the ASK-1 expression showed a gradual upward trend. Conclusion Higher concentration of iodine can decrease the viability of A7r5 cells and the expression of TRX-1 protein,increase the expression of ASK-1 protein,and induce apoptosis of A7r5 cells.
Objective To observe the antagonism of selenium to arsenic-induced adverse effect in the activity of SOD and GSH-Px,the content of MDA,and the expression of ICAM-1 and VCAM-1 in human umbilical vein endothelial cells(HUECV). Methods The density of HUVEC in logarithmic growth phase was adjusted to 1.0 ×105/ml,then the cells were divided into two groups and were treated as follows, one group was treated with arsenic solution at different doses(0,0.05,0.1,0.5,1.5,3,6 and 12 μmol/L),the other group was treated with the same doses of arsenic plus sodium selenite(0.1 μmol/L). The medium were collected after 48 h-culture to determine the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) and the content of malondialdehyde(MDA) by spectrophotometry. The expression of intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) were detected by enzymelinked immunosorbent assay(ELISA). Results With the increase of arsenic exposure,the arsenic group showed a gradual decline trend in the activity of SOD,GSH-Px,while an upward trend in the content of MDA and the expression of ICAM-1 and VCAM-1. Selenium showed a significant antagonism to lower level arsenic-induced adverse effect in the activity of SOD and GSH-Px,the content of MDA,and the expression of ICAM-1 and VCAM-1 in human umbilical vein endothelial cells.Conclusion Exposure to arsenic in a certain level can cause decrease of antioxidation of vascular endothelial cell(VEC) and increase of lipid peroxidation and up-regulated expression of ICAM-1 and VCAM-1. Selenium supplying may have antagonism to the adverse effect of arsenic.
Endemic fluorosis is prevalent in China, which can seriously impair the bones, teeth and cardiovascular system. The study was intended to explore antagonism of selenium (Se) on high dose fluorine (F) inducing plasma 6-Keto-prostaglandin F1α,Thromboxane B2 and Endothelin-1. Twenty male rabbits were randomly divided into 4 groups: High F group (NaF,100mg/L), Se group (Na2SeO3,1mg/L), High F + Se group (NaF,100mg/L; Na2SeO3,1mg/L) and Control group (without fluorine and selenium). The ear vein blood was collected for measurement of serum selenium and fluorine at the end of 0, 3rd and 6th months. At the end of 6th months, plasma 6-Keto-prostaglandinF1α (6-K-P), Thromboxane B2 (TXB2) and Endothelin-1 (ET-1) were detected. Serum fluorine was increased in high F group and serum Se was increased in Se group. Plasma 6-K-P was decreased, while the plasma TXB2 and ET-1 increased with the development of fluorosis. Compared with the high F group, the plasma 6-K-P was higher (p<0.01), whereas plasma TXB2 and ET-1 were lower (p<0.01) in high F + Se group. The interaction showed that there was a significant antagonistic action of Se on decreased 6-K-P and increased TXB 2 induced by high dose fluorine. Our study showed that the Se may antagonize the adverse effects of high F.
Objective To research the influence of organic iodine(DIT) and inorganic iodine(KI) on cellular morphology and the expression of a gene family about apoptosis(Bcl-2 and Bax).Methods The human thyroid epithelium cells from para-adenoma normal tissues from thyroid adenoma patients were cultured.They were divided into control group(medium),KI group(the dosages of KI were 10-7,10-5 and 10-3 mol/L respectively) and DIT group(the dosages of DIT were 10-7,10-5 and 10-3 mol/L respectively).Various dosages of DIT and KI diluted with medium were applied in the culture for 48 h.The local distribution and expression of Bcl-2 and Bax of human thyroid cell were detected by immumohistochemistry after different dosages of DIT and KI were applied.Results The expression of Bcl-2 significantly decreased in KI group(l0-5,10-3 mol/L) and DIT group(10-3 mol/L),and the expression of Bax significantly increased in KI group(l0-5,10-3 mol/L) and DIT group(l0-5,10-3 mol/L) than that in the control group(P0.05,P0.01);As the level of exposure increased,the expression of Bcl-2 decreased and the expression of Bax increased in degrees.At the same level of DIT and KI,the expression of Bcl-2 of DIT was significantly higher and the expression of Bax of DIT was significantly lower(P0.05,P0.01).Conclusion High level iodine exposure may cause morphological changes and apoptosis of thyrocytes and the effect of DIT may be less compared with KI.
With the rapid development of multimedia technical,there were more and more digital video resources on the Internet.How to retrive the information in enormous amout of data fast,effectively and accurately became the focus of attention.As a result,content-based video retrieval(CBVR) had aroused widespread concern.This paper first analyzed the characteristics of the video,extracted the color and texture histogram as the main feature information,and then used an improved adaptive sliding-window double-threshold shot detection algorithm for video shot segmentation.Finally,three kinds of algorithms were compared through experiments.The results showed that the algorithm proposed in this paper was superior to the traditional in accuracy when the complexity had no significant increase.The last part of this article used the algorithm proposed in this paper implemented a retrieval system,and the results showed that the accuracy of this algorithm was better than the traditional algorithm to some extent.
