Objective:We observed the effects of electro-acupuncture on the expression of serum ACTH and CORT in rats exposed to chronic restraint stress (CRS),to explore the antidepressive effects and possible mechanisms of electro-acupuncture in the treatment of depression.Methods:Sixty SD rats were equally divided into three groups:control group,model group and electro-acupuncture group.Except the control group,all rats were in the condition of chronic restraint stress and separation.The EA group was stimulated at GV 20 and EX-HN 3 for 20 min before daily bound.ACTH and CORT levels were detected by ELISA respectively on the 7th,28th,35th day (the 7th day after the last intervention).Results:Effects on Serum ACTH and CORT:After 7 days,there was no difference among three groups.On the 28th and the 35th day,compared with the control group,the model group significantly increased (P <0.01;P < 0.01;P <0.01;P < 0.01);Compared with the model group,EA group significantly decreased (P <0.05;P <0.05;P <0.01;P <0.01).The electro-acupuncture had a reversal effect on the expression of serum ACTH and CORT in rats exposed to chronic restraint stress.Conclusion:The hypothalamic-pituitary-adrenal (HPA) axis hyperactivity is observed in rats exposed to chronic restraint stress,and its level of serum ACTH and CORT are significantly higher.The level of serum ACTH and CORT are reversed by EA intervention,which suggests that EA intervention can regulate HPA axis hyperactivity.The 7th day after the last intervention,the level of ACTH and CORT of EA group are still significantly lower than the model group,which suggests that EA intervention has continuous regulatory effect on the HPA axis hyperactivity,and it may be one of the persistent effect mechanisms of antidepressive effects of EA.
Objective:To investigate the antidepressant effect of electro-acupuncture(EA) on the expressions of TNF-α and IL-1β in the pubescent rats with chronic restraint stress(CRS),and to explore the relative immune mechanism.Methods:60 SD rats (male,five weeks) were randomized into the blank control group (group A,n =20),the model group (group B,n =20),and the electro-acupuncture group (group C,n =20).The rats in group A were bred in groups;CRS models were established by separate breeding combined with chronic restraint stress(immobilization for 6h daily from 9:00 to 15:00);in group C,electro-acupuncture intervention was applied on point GV20 and point GV29 for 20mins one hour before the restraint,once daily for 28 days.From the 28th day to 35th day,there was no treatment application in the three groups.Open-field test was used to assess the behavior of the CRS rats.Serum levels of TNF-α and IL-1β were detected with Bio-Plex method on 7th,28th and 35th day respectively.Results:In terms of scores of the open-filed test,there was no significant difference in the three groups before the experiment;on 28th day,the scores of group C were remarkably increased compared to those of group B (P < 0.01).In terms of serum levels of TNF-α and IL-1β on 7th day,TNF-α level increased considerably in group B compared to that in group A (P < 0.01),but there was no significant differences in IL-1β level between the two groups(P > 0.05);compared to group B,TNF-α expression was down-regulated significantly in group C (P < 0.01).On 28th day,levels of TNF-α and IL-1β increased significantly in group B compared to those in group A (P < 0.01);however,they decreased obviously in group C when compared to group B(P <0.01).On 35th day,there were no significant differences in serum levels of TNF-α and IL-1β among the three groups (P > 0.05).Conclusion:CRS can raise TNF-α level and reduce IL-1β level in pubescent rats,and electro-acupuncture can regulate these changes,which may contribute to its antidepressant effect.
Acupuncture has demonstrated the function in ameliorating depressive-like behaviors via modulating PKA/CREB signaling pathway. To further confirm the antidepressant mechanism of EA on the mitogen-activated protein kinase (MAPK) and dopaminergic synapse signaling pathways, 4 target proteins were detected based on our previous iTRAQ analysis. Rats were randomly divided into control group, model group, and electroacupuncture (EA) group. Except for the control group, all rats were subjected to 28 days of chronic restraint stress (CRS) protocols to induce depression. In the EA group, EA pretreatment at Baihui (GV20) and Yintang (GV29) was performed daily (1 mA, 2 Hz, discontinuous wave, 20 minutes) prior to restraint. The antidepressant-like effect of EA was measured by body weight and open-field test. The protein levels of DAT, Th, Mapt, and Prkc in the hippocampus were examined by using Western blot. The results showed EA could ameliorate the depression-like behaviors and regulate the expression levels of Prkc and Mapt in CRS rats. The effect of EA on DAT and Th expression was minimal. These findings implied that EA pretreatment could alleviate depression through modulating MAPK signaling pathway. The role of EA on dopaminergic synapse signaling pathways needs to be further explored.
Objective To evaluate the clinical effect of the external validity and security of electro acupuncture in the real treatment environment with the real world study method ( RWS ) . Methods Patients diagnosed with depression by the ICD-10 (n=60) were divided into two groups:(1) electro acu-puncture(EA) group(n=35);(2) western medicine group (n=25). The Hamilton Depression (HAMD) scales, CGI scales, MYMOP scales were used to assess the main curative indexes during each period of the treatment. Results There was no significant difference between these two groups at baseline level (P>0. 05). (1) HAMD:Compared with the western medicine group, the clinical control rate, markedly effective rate, cure rate, effective rate of EA group were significantly better than western medicine group (P<0. 05). In both of the two groups, there were statistically significant differences between baseline score of HAMD and the score in every treatment period(P<0. 05). From the beginning of the fourth week, the HAMD score of EA group was significantly better than that of the western medicine group(P<0. 05). And four weeks after treatment, the score of EA group was still better than the western medicine group; ( 2 ) CGI:After 8 weeks of treatment, Compare with the western medicine group, score of CGI scale showed that severity of the disease, effective evaluation and effective index of EA group was significant differences better than those of western medicine group (P<0. 05);(3) MYMOP:After 8 weeks of treatment, compare with the western medicine group, the score of MYMOP scale, of EA group on symptom 1, symptom 2, activity and general health condition has significant differences ( P<0. 05 ). Conclusion In real clinical conditions, EA treatment could improve the scores of HAMD, CGI and MYMOP significantly in patients with depression. Generally, the effect of EA treatment for depression is better than western medicine.
目的:观察电针干预对束缚应激模型大鼠行为学及血清中白细胞介素6(IL-6)与可溶性白细胞介素6受体(sIL-6R)表达的影响,探讨其相关免疫学机制.方法:将60只SD大鼠随机分为对照组、模型组与电针组,每组20只.对照组群养;模型组采用孤养结合慢性束缚应激方式,每日束缚6h(9:00-15:00);电针组在每日束缚前给予电针刺激“百会”“印堂”;束缚期间3组均禁食禁水,其余时间自由饮食饮水.采用旷场和体质量实验进行行为学评价,并在第7天、28天、35天采用Bio-Plex检测大鼠血清中IL-6、ELISA检测SIL-6R水平.结果:①行为学:与对照组相比,模型组大鼠体质量及活动性均显著降低(P<0.01),电针组大鼠体质量及活动性表达无显著差异(P>0.05).②血清IL-6表达:实验day7,与对照组相比,模型组IL-6表达显著增加(P<0.01),电针组IL-6表达无显著差异(P>0.05);与模型组相比,电针组IL-6表达显著降低(P<0.05).实验day28,与对照组相比,模型组IL-6表达显著增加(P<0.01),电针组IL-6表达无显著差异(P>0.05);与模型组相比,电针组IL-6表达显著降低(P<0.01).实验day35,3组大鼠之间IL-6表达无显著差异(P>0.05).③血清sIL-6R表达:实验day7,3组大鼠之间sIL-6R表达无显著差异(P>0.05);实验day28,与对照组相比,模型组sIL-6R表达显著增加(P<0.01),电针组sIL-6R表达无显著差异(P>0.05);与模型组相比,电针组sIL-6R表达显著降低(P<0.05).实验day35,3组大鼠之间sIL-6R表达无显著差异(P>0.05).结论:慢性束缚应激模型大鼠血清IL-6、sIL-6R表达显著增加,并随时间出现相应变化,电针干预可对其进行显著调控,与行为学症状改善一致,这可能是电针抗抑郁的作用机制之一.
The clinical effect of electroacupuncture on depression is widely recognized. However, the signal transduction pathways and target proteins involved remain unclear. In the present study, rat models of chronic restraint stress were used to explore the mechanism by which electroacupuncture alleviates depression. Rats were randomly divided into control, model, and electroacupuncture groups. Chronic restraint stress was induced in the model and electroacupuncture groups by restraining rats for 28 days. In the electroacupuncture group, electroacupuncture pretreatment at Baihui (GV20) and Yintang (GV29) acupoints was performed daily (1 mA, 2 Hz, discontinuous wave, 20 minutes) prior to restraint for 28 days. Open field tests and body weight measurements were carried out to evaluate the depressive symptoms at specific time points. On day 28, the crossing number, rearing number, and body weights of the model group were significantly lower than those in the control group. Behavior test results indicated that rat models of depressive-like symptoms were successfully established by chronic restraint stress combined with solitary raising. On day 28, an isobaric tag for a relative and absolute quantitation-based quantitative proteomic approach was performed to identify differentially expressed proteins in hippocampal samples obtained from the model and electroacupuncture groups. The potential function of these differential proteins was predicted through the use of the Cluster of Orthologous Groups of proteins (COG) database. Twenty-seven differential proteins (uncharacteristic proteins expected) were selected from the model and electroacupuncture groups. In addition to unknown protein functions, COG are mainly concentrated in general prediction function, mechanism of signal transduction, amino acid transport and metabolism groups. This suggests that electroacupuncture improved depressive-like symptoms by regulating differential proteins, and most of these related proteins exist in nerve cells.
Objective:To observe the effects of electroacupuncture ( EA) on depressive-like behavior and ser-um 5-hydroxytryptamine (5-HT) of the chronic forced swim stress model rats .Methods:40 male SD rats were evenly randomized into normal control , model, fluoxetine and EA groups .The chronic stress rat model was established by chronic forced swim .The behaviors were evaluated by body weight , open field test and su-crose intake test , the serum 5-HT was detected by ELISA .Results: Compared with the control group , the body weight , crossings and rearings in the open field test , the sucrose intake , and the serum 5-HT of model group were decreased significantly (P<0.05).Compared with the model group , the body weight, crossings and rearings in the open field test , the sucrose intake , and the serum 5-HT of fluoxetine group and EA group were all increased significantly (P<0.05).But there was no significant difference between EA group and flu-oxetine group (P>0.05).Conclusion:Electroacupuncture can improve the model rats ’ depressive-like be-haviors which are caused by chronic forced swim stress , and may increase the serum 5-HT of the model rats . Hereby , electroacupuncture may have anti -depressive effects .

Objective To analyze correlations between personality traits,life events and depressive symptoms based on structural equation model and to identify risky factors of depressive symptoms.Methods Using stratified random cluster sampling method,industrial workers recruited from Shougang Qianan corporation received questionnaire survey consisted of LES,NEO-FFI and CES-D.Maximum likelihood method was applied to calculate parameters of structural equation model,then the final model was confirmed based on modification indices.Results Depressive symptom(CES-D ≥ 16) prevalence was 41.05% among industrial workers in current study.Neuroticism,extraversion,agreeableness and negative life events were significantly predictive to depressive symptoms (r=0.356,-0.304,-0.135,0.227).Positive and negative life events,neuroticism and extraversion,extraversion and conscientiousness,neuroticism and agreeableness presented high correlation,with coefficients 0.614,-0.469,0.432 and 0.402 respectively.The regression weight of affective symptoms,somatic symptoms and interpersonal relationship in measurement model were 0.779,0.678,0.464.Conclusion The onset of depressive symptoms among industrial workers is remarkably related to neuroticism,extraversion,agreeableness and negative life events.Certain measures are supposed to be conducted for mental nursing and intervention in order to strengthen depression prevention.

Immunological reactions induced by proinflammatory cytokines have been involved in the pathogenesis of depressive disorders. Recent studies showed that Electroacupuncture (EA) was able to reduce depressive symptoms; however, the underlying mechanism and its potential targets remain unknown. In the present study, we used a 21-day chronic restraint stress rats as a model to investigate how EA could alleviate depression. Open field test was carried out to evaluate the depressive symptoms at selected time points. At the end of study, immunohistochemistry (IHC) was performed to detect the expressions of IL-1beta, IL-6, and TGF-beta in hippocampal CA3 region. We found that chronic restraint stress significantly decreased behavioral activities, whereas EA stimulation at points Baihui (GV 20) and Yintang (GV 29) showed protective effect during the test period. In addition, the IL-1beta, IL-6, and TGF-beta increased in rats exposed to chronic restraint stress, while EA downregulated the levels of IL-1beta and IL-6. These findings implied that EA pretreatment could alleviate depression through modulating IL-1beta and IL-6 expression levels in hippocampal CA3 region.
