目的:研究莪芪抗瘤方对二乙基亚硝胺(DEN)诱导肝癌模型鼠肿瘤微环境的调控机制.方法:建立DEN诱导大鼠肝癌模型,造模4周后,70只大鼠随机分成7组,空白组,模型组,顺铂治疗组,单纯祛邪中药组,莪芪抗瘤方组随机分为高、中、低剂量组.每组10只.空白组正常喂养,模型组每日12.5 mg/kg体重维持DEN摄入量喂养,莪芪抗瘤方中剂量组、单纯祛邪中药组、顺铂治疗组,分别以8 mL/kg体重剂量灌药;莪芪抗瘤方高、低剂量组分别以12 mL/kg体重、4mL/kg体重剂量灌药.16周实验结束,测定大鼠血清白细胞介素-1β (IL-1β)、白细胞介素-2 (IL-2)、肿瘤坏死因子-α(TNF-α)的表达.结果:莪芪抗瘤方组IL-1β与单纯祛邪中药组、空白组比较,差异有统计学意义(P<0.05),与顺铂治疗组比较,差异无统计学意义(P>0.05):莪芪抗瘤方组IL-2与单纯祛邪中药组比较,差异有统计学意义(P<0.05);莪芪抗瘤方组IL-2、TNF-α与空白组、顺铂治疗组比较,差异无统计学意义(P>0.05).结论:莪芪抗瘤方能抑制肝癌大鼠模型血清中IL-1β表达,抑制肿瘤组织血管的形成和浸润,促进IL-2的分泌,增强免疫力,抑制肿瘤的形成,在肝癌形成过程中具有一定的延缓和保护作用.

Objective:To study the influence of the Ewi-anticarcinogen Prescription on associated gene protein in process of rat hepatocarcinoma induced by diethylnitrosamine ( DEN ) .Methods:Rats hepatocarcinoma model was induced by DEN,while the Eqi-anticarcinogen Prescription was used during the stage.At the end of 12th week and 16th week,the influence of the Eqi-anticarcinogen Prescription on positive expression of PCNA and p53 protein were examined by immuneohistochemical method.Results:The positive expression of p53 protein was occurred in precancerous hyperp lastic nodules;The Eqi-anticarcinogen Prescription can significantly inhibit the overexpression of p53 (P<0.05),and strikingly reduce PCNA (P<0.01) at the end of 12th week and 16th week.Conclusion:p53 gene mutation could occur in precancerous stage in DEN-induced liver cancer model and may relate to the initiation of hepartocarcinoma.The Eqianticarcinogen Prescription can inhibit the proliferative character of hepatocytes and the mutateon of p53 gene,as well as slow down the process of hepatocarcinoma induced in rats.
