Objective:To explore the ectopic osteogenesis effect of the porous hydroxyapatite (HA) scaffolds as the artificial bone substitute materials.Methods:Cultured human placenta derived mesenchymal stem cells with porous HA scaffolds for 7 d,cell-scaffold composite were taken out to implanted in the beagle's back muscles as the experimental group,and implanted blanket porous HA scaffolds in the beagle's back muscles as the control group,respectively remove the ectopic osteogenesis organization at 12 weeks after implantation,then detected the osteogenesis effect of the ectopic ossification organizations.Results:Implanted at 12 weeks there had new blood supply in the ectopic ossification organizations,and the experimental group new bone tissue significantly greater than the control group.Conclusions:Porous HA ceramic scaffolds have good ectopic bone formation in vivo,and human placenta mesenchymal stem cells can promote the formation of new bone.
目的:通过模拟观察高压氧条件下种植体周围软组织愈合情况.方法:将12只比格犬的硬腭中份后份制备5mm的圆形组织缺损,随机分为AB两组各6只,A组给予高压氧治疗,B组不予处理.在第3天7天及14天术区取材对其进行愈合情况观察,组织学观察及CD31的免疫组化染色观察.结果:大体形态观察显示:A组创伤区逐渐愈合,14天上皮黏膜覆盖完全无组织凹陷及瘢痕的形成.B组术区愈合状态欠佳,14天时术区仍可见组织凹陷.苏木精-伊红染色(H-E染色)下观察到A组较B组在术后3天及7天时炎症细胞浸润更少.Mas-son染色后可见A组较B组在术后3天及7天时有更多的胶原纤维,更有序的排列.CD31免疫组织化学染色观察显示:A组较B组有更多的毛细血管生成,差异具有统计学意义.结论:高压氧治疗使得软组织愈合更快,愈合质量更佳,更减轻了术区的炎症反应.
