目的 系统评价环状RNA在预测胃癌患者预后时的准确性和有效性.方法 检索PubMed、Embase、Web of Science、Cochrane Library以及中国知识基础设施工程,搜索环状RNA与胃癌患者预后相关性的研究.检索时限从建库至2019年9月1日.2名研究者独立筛选文献后,采用Stata 12.0软件进行Meta分析.结果 经筛选最终纳入7篇病例对照研究文献,共计699例胃癌患者.Meta分析结果显示:环状RNA异常表达的人群,其不良预后结局是环状RNA正常表达胃癌患者的1.92倍(HR=1.92,95%CI:[0.99,3.73],P<0.001).敏感性分析显示研究之间存在差异,结果不稳定;Begg's漏斗图及Egger回归分析显示不存在发表性偏倚.结论 胃癌患者的生存结局与差异表达的环状RNA有明显的相关性.
目的 研究幽门螺杆菌(Hp)通过Wnt/β-catenin信号通路对胃癌细胞侵袭及血管新生因子的作用.方法 收集手术切除的胃癌组织并检测Wnt1、Wnt3a、β-catenin、MMP-7、N-cadherin、E-cadherin、VEGF、bFGF、Ang-2 mRNA的相对表达量.培养胃癌细胞株SGC-7901,分为对照组、Hp组、Hp+ICG-001组.对照组用不含细菌及药物的DMEM处理,Hp组用含有Hp的DMEM处理,Hp+ICG-001组用含有Hp及Wnt/β-catenin信号通路抑制剂ICG-001的DMEM处理.采用Western blotting检测Wnt1、Wnt3a、β-catenin、MMP-7、N-cadherin、E-cadherin蛋白的相对表达量.采用Transwell检测侵袭数目.采用酶联免疫吸附试验检测VEGF、bFGF、Ang-2的含量.结果 与Hp阴性胃癌组织比较,Hp阳性胃癌组织中Wnt1、Wnt3a、β-catenin、MMP-7、N-cadherin、VEGF、bFGF、Ang-2的mRNA相对表达量均增加(P<0.05),E-cadherin的mRNA相对表达量降低(P<0.05);Hp组Wnt1、Wnt3a、β-catenin、MMP-7、N-cadherin蛋白相对表达量、侵袭数目、VEGF、bFGF、Ang-2的含量高于对照组(P<0.05),E-cadherin的相对表达量低于对照组(P<0.05);Hp+ICG-001组Wnt1、Wnt3a、β-catenin、MMP-7、N-cadherin蛋白相对表达量、侵袭数目、VEGF、bFGF、Ang-2的含量低于Hp组(P<0.05),E-cadherin的相对表达量高于Hp组(P<0.05).结论 Hp通过Wnt/β-catenin信号通路促进胃癌细胞的侵袭及血管新生因子生成.
Objective Disulfiram is commonly used for alcohol abuse; however, recent studies have revealed its potential as an anti-cancer treatment. This study investigated the effects of disulfiram on gastric cancer and its underlying mechanisms of action. Methods The gastric cancer cell lines MKN-45 and SGC-7901 were used for all experiments. Cell proliferation was investigated using cell counting kit-8, cell migration and invasion were examined using Transwell assays, the proliferation and metastasis related proteins PCNA and MMP-2, respectively, were detected by ELISA. To explore the underlying molecular mechanisms, we also examined levels of proteins involved in the Wnt and NF-κB pathways by ELISA. Results Disulfiram significantly inhibited the proliferation, migration, and invasion of gastric cancer cells and decreased PCNA and MMP-2 levels. Additionally, disulfiram-treated MKN-45 and SGC-7901 cells showed reduced expression of Wnt, β-catenin, and NF-κB. Conclusion Disulfiram regulates the Wnt and NF-κB pathways, and thus could be a potential treatment for managing gastric cancer.
