Background Phlegm-dampness constitution as one of nine constitutions in traditional Chinese medicine (TCM) has been a high risk factor for glucolipid metabolic disorders (GLMD). Based on our previous findings, Hua Tan Qu Shi recipe (HTQSR) could effectively improve metabolic indicators of GLMD by targeting on phlegm-dampness constitution. However, the proteomic mechanisms of GLMD with the treatment of HTQSR targeting on phlegm-dampness constitution remain unknown. Methods Clinical participants from phlegm-dampness constitution with the prediabetic state (T), phlegm-dampness constitution with marginally elevated blood lipids (Z), and phlegm-dampness constitution before sickness (W) were included in this study, who orally took HTQSR for 12 weeks and, respectively, marked AT, AZ, and AW. Data-independent acquisition (DIA) and parallel reaction monitoring (PRM) were performed to identify the differential proteins; then, Venn analysis was used to investigate coexpressed and coregulated proteins. In addition, ingenuity pathway analysis (IPA) software was utilized to explore the related pathways and diseases and biofunctions. Results LXR/RXR activation, acute phase response signaling, and production of nitric oxide and reactive oxygen species in macrophages were obviously activated between the T and AT groups, as well as the Z and AZ groups. In contrast, these three pathways were inhibited between the W and AW groups. Importantly, one coexpressed and coregulated differential protein, B2MG, was validated by PRM among all groups. Conclusions This work firstly reported the underlying proteomic mechanisms of GLMD with the treatment of HTQSR targeting on phlegm-dampness constitution, indicating that intervention of phlegm-dampness constitution might be a novel strategy for the preventive treatment of GLMD.
Objective: To investigate the proteomic characteristics of overweight/obesity and related abnormal glucose and lipid metabolism caused by phlegm-dampness retention to identify related biomarkers.Methods: Seventy-one subjects were enrolled in the study. We assessed blood glucose, blood lipids, body mass index(BMI), and phlegm-dampness pattern, which was confirmed by a traditional Chinese medicine clinician. Of the participants, we included healthy participants with normal weight(NW, n=23),overweight/obese participants with normal metabolism(ONM, n=19), overweight/obese participants with pre-diabetes(OPD, n=12), and overweight/obese participants with marginally-elevated blood lipids(OML, n=17). Among them, the ONM, OPD, and OML groups were diagnosed with phlegmdampness pattern. The data-independent acquisition(DIA) method was first used to analyze the plasma protein expression of each group, and the relevant differential proteins of each group were screened. The co-expressed proteins were evaluated by Venn analysis. The pathway analyses of the differential proteins were analyzed using Ingenuity Pathway Analysis(IPA) software. Parallel reaction monitoring(PRM) was used to verify the differential and common proteins in each group.Results: After comparing ONM, OPD, and OML groups with NW group, we identified the differentially expressed proteins(DEPs). Next, we determined the DEPs among OPD, OML, and ONM groups. Using Venn analysis of the DEPs in each group, 24 co-expressed proteins were screened. Two co-expressed proteins were verified by PRM. IPA analysis showed that pathways including LXR/RXR activation,acute phase response signaling, and FXR/RXR activation were common to all three groups of phlegmdamp overweight/obesity participants. However, the activation or inhibition of these pathways was different among the three groups.Conclusion: Participants with overweight/obesity have similar proteomic characteristics, though each type shows specific proteomic characteristics. Two co-expressed proteins, VTN and ORM1, are potential biomarkers for glucose and lipid metabolism diseases with overweight/obesity caused by phlegmdampness retention.
Objective The objective of this study is to explore the long non-coding RNAs (lncRNAs) and messenger RNAs (mRNAs) expression profiles of monocyte-derived dendritic cells (DCs) obtained from peripheral blood mononuclear cells (PBMCs). DCs are known to play a major role in the regulating function of allergic rhinitis (AR). Methods PBMCs were separately isolated from the human peripheral blood of patients with AR and normal person (NP). The mixed lymphocyte reaction (MLR) assay was used to evaluate the function of DCs. Flow cytometry was used to determine the immune regulatory function of immature DCs (imDCs) and mature DCs (mDCs). lncRNAs and mRNAs in the NP group (DCs isolated from NP) and the test group (DCs isolated from patients with AR) were identified via chip technology and bioinformatic analyses. Moreover, bioinformatic analyses were employed to identify the related biological functions of monocyte-derived DCs and construct the functional networks of lncRNAs and mRNAs that are differentially expressed (DE) in imDCs and mDCs. Results MLR was significantly higher in the mDCs group than that in the imDCs group. CD14 was highly expressed in imDCs, whereas HLA-DR, CD80, and CD86 were highly expressed in mDCs (p < 0.001). We identified 962 DE lncRNAs and 308 DE mRNAs in the imDCs of NP and patients with AR. Additionally, there were 601 DE lncRNAs and 168 DE mRNAs in the mDCs in the NP and test groups. Quantitative RT-qPCR was used to study the significant fold changes of lncRNAs and mRNAs. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis found 16 significant regulated pathways in imDCs and 10 significant regulated pathways in mDCs, including the phagosome, cell adhesion signaling pathway, and inflammatory mediator regulation of TRP channels pathway. Conclusion Our research studied the lncRNA and mRNA expression profiles of monocyte-derived DCs and demonstrated the functional networks that are involved in monocyte-derived DCs-mediated regulation in AR. These results provided possible molecular mechanisms of monocyte-derived DCs in the immunoregulating function and laid the foundation for the molecular therapeutic targets of AR.
Background: Based on the theory of traditional Chinese medicine(TCM) and pre-epidemiological investigation, Professor Qi Wang classified the entire human population into nine constitutions and put forward the theory of "Nine-Constitution Medicine." Among these constitutions, the main feature of the yang-deficiency constitution(YADC) is intolerance of the cold, which has been proven to reduce quality of life and confer susceptibility to specific diseases. Previous studies explored the genetic and transcriptional bases of YADC. In this experiment, we explored the potential mechanism of YADC using protein microarray, to deepen our understanding of its biological mechanism.Methods: Subjects identified with a YADC(n=12) or a balance constitution(BC; n=12) in accordance with the Classification and Determination Standards of Constitutions in Traditional Chinese Medicine were selected. Blood was collected to separate serum and protein microarray technology was used to analyze serum protein expression.Results: The clustering of subjects’ constitutions based on protein expression profiling largely coincided with the TCM classification. Based on false discovery rate correction(P <.01) and fold change ≥5 or ≤0.2, a total of 85 proteins differentially expressed in YADC compared with their status in BC were selected, including 64 upregulated and 21 downregulated ones. Enrichment analysis suggested that subjects with YADC are susceptible to endocrine and energy metabolism disorders, as well as decline in immune function.Conclusion: This study revealed that YADC exhibits systematic differences in its protein expression profile. Moreover, we can potentially explain the characteristics of YADC partly via differentially expressed proteins.
目前糖尿病、高脂血症、高血压病等慢性病的高发病率、高致残率、高病死率成为困扰人类健康的主要问题.实现慢性病防治关口前移亟需找到新途径.本研究基于国医大师王琦教授“体质土壤”学说,提出“痰湿体质是许多代谢性疾病的共同土壤,其机制可能与免疫炎症进程、胰岛素抵抗及氧化应激反应失调有关”的假说.对平和体质及痰湿体质的“未病人群”、高脂血症边缘升高人群、2型糖尿病临床前期3类人群的共同分子通路表达进行检测分析对比,并观察王琦教授“化痰祛湿方”对其表达的调控效应及机制.本研究对于揭示中医痰湿体质的现代内涵,指导实施防控代谢紊乱性疾病,实现慢性病防治关口前移,具有重要的理论和实践意义.
Th17细胞是Th细胞亚群的新成员,其分化受到多种因子调节并存在多处自稳调节机制.中医体质学是在传统中医学的理论的指导下形成的,其阴阳生克、五行制化等基本理论与“自稳”调节机制异曲同工,现代研究也初步揭露了Th17与中医体质分类和调理的相关性.对Th17细胞分化自稳调节机制进行综述,并对其与中医体质相关性进行分析,以期对体质形成和调理的微观机制研究有所指导.
Objective: In this study, we investigated the composition of the intestinal microflora associated with Yang-deficiency and the function-specific members of this microflora. Method: Subjects with normal and Yang-deficient constitutions were recruited in Beijing, with 30 subjects in each group. Illumina high-throughput sequencing was used to sequence the DNA of their fecal bacteria in the 16S rRNA V3–V4 region. The species abundance and distribution of the intestinal microflora in each specimen were determined with a statistical analysis of these sequences. We identified an underlying taxonomic trend with nonparametric PCoA and other statistical techniques. Results: (1) Subjects with Yang-deficiency displayed more uneven abundances of the taxa in their intestinal microflora than the subjects with normal Yang; (2) several differences were observed in the proportions of certain bacterial groups between the Yang-deficient and control groups; and (3) the following bacteria differed significantly between the two groups: (i) Brevundimonas, Leuconostoc, Turicibacter, Defluviitaleaceae incertae sedis, Rothia, and Butyricimonas were slightly higher in the Yang-deficient subjects, whereas (ii) Megasphaera, Ruminococcus, and Lachnospira were higher in the normal subjects than in the Yang-deficient subjects. Conclusions: The composition of the intestinal microflora is altered in Yang-deficient subjects.
Objective:To analyze literature about traditional Chinese medicine treatment of chronic urticaria published in China and the development trend.Methods:The papers relevant to traditional Chinese medicine treatment of chronic urticaria published in journals collected by CNKI database were reviewed.The 1 829 articles published from 1959 to 2015 were statistically analyzed from the aspects of publish year,number,research institutions,author,authors' institution,and research funds with bibliometrics method.Results:In addition to the individual years,the number of literature in general is on the rise,and rapid growth since 2006.Papers covers many disciplines and research direction.But there are less high levels literature,and the articles are with low reference rate.Only relatively few papers get funded.Conclusion:The study of TCM therapy for chronic urticaria increase quickly,and receive the attention of the society.However,the quality of literature is low,and lack of high level research achievements.Due to the scattered research group distribution,the core research institutions have not been formed.
体质形成机制是体质科学研究的重要方面.体质是由先天禀赋和后天获得共同形成的,体现了基因和环境的交互作用.因此,在研究体质形成机制时,也要考虑先天和后天两个方面.利用现代分子生物学的方法和技术研究体质形成机制具有重要的意义.对于先天机制的研究,一般采用全基因组表达谱与SNP技术.对于后天机制的研究,目前尚未有人涉及,笔者认为表观遗传学和microRNA、lncRNA检测是可采取的方法.如何从先天后天两个方面揭示体质形成机制,尚需大量的研究工作去探索和证实.
Allergic diseases are some of the most common and refractory diseases. Professor WANG Qi, a National Chinese Medical Science Master, treats the allergic diseases guided by the model of“differentia-tion of constitution-differentiation of disease-differentiation of syndrome” and achieves remarkable effects. The authors summarize the experiences of Master WANG on treating the allergic disease. Firstly, treat-ment should be reinforcing the healthy qi priority to eliminating pathogenic factors. Secondly, patients can be exposed to allergens properly when they are undergoing TCM treatment. Thirdly, according to the pathogenesis rules of certain allergic disease, the exact time of treatment is determined. Finally, medica-tion to adjust Constitution is simpler and more flexible than medication applied to the patients suffering the allergic diseases.
目的:分析2009年《中医体质分类与判定》标准发布后,国内中医体质研究的特征、规律、及发展趋势.方法:以中国知识基础设施工程(CNKI)数据库为资料来源,对2009年4月9日至2014年4月9日国内正式发表的2 369篇中医体质文献进行文献计量学分析.结果:中医体质文献数量近5年来呈快速增长趋势,覆盖多个学科领域和研究方向,获得多项科研基金资助,已形成核心作者,北京、广州等地区对其研究处于领先地位.结论:中医体质学已基本形成核心论坛,成为中医学研究领域的热点,在临床及基础各学科中被广泛应用,但尚存在研究深度不足的问题.应加强辨体调治以及体质基础实验的研究.
中医体质学将体质分为9种,其中痰湿体质和过敏体质都属于常见的体质类型.随着研究的不断深入,肥胖、糖尿病、代谢综合征等疾病和变态反应性疾病的关联性越来越受到重视.而痰湿体质和过敏体质正是这些疾病的"病前"状态.因此,这两种体质类型也必然存在密切的关联性.试从临床研究、微观机制研究等方面对痰湿体质和过敏体质的相互关系作一分析,以期对"体病相关"和"调体防病"的深入研究提供指导.
探讨“辨体-辨病-辨证”诊疗模式在针灸治疗过敏性疾病中的应用.提出在辨病辨证论治的基础上,调理体质方面应采用扶正法为主,兼以清透伏邪.扶正取穴可取背俞穴、原穴、任脉、督脉穴等,并多采用灸法;清透伏邪采用清热的腧穴及刺络放血、拔罐等疗法.临床针灸治疗过敏性疾病在预防和治疗缓解期时重用辨体,以治本为主,再次用辨病、辨证;在发作期时以辨病、辨证论治为主,辨体为辅,灵活运用三辨模式.
气郁质是临床上常见的九种体质之一,研究发现气郁质易患郁证、失眠、惊恐等病证.《灵枢·经脉》记载了这些病证的相关经脉病变,如“胆足少阳之脉,起于目锐眦......是动则病口苦,善太息,心胁痛不能转侧.”此外手阳明大肠经病变也会出现郁证;烦心失眠可由手心主之别络、足少阴肾经、足太阴脾经病变所致;足少阴肾经、足阳明胃经病变会出现惊恐,故可选取相应的经络穴位治疗,如治疗郁证可选取太冲、阳陵泉、膻中、血海等穴;同样,治疗失眠可选取少府、内关、足三里、三阴交等穴;选取灵道、神门、涌泉、太冲等穴可治疗惊恐.根据“体病相关”的理论,疾病通过相应的经络穴位治疗而改善,体质也会改变.
偏头痛是临床上一种病因病机未明的疾病,中医一般从调补肝脾肾,祛风化痰瘀为主论治,而王琦教授临床上发现有些偏头痛与过敏体质有相关性,针对此类疾病主以“调理过敏体质”为主配合“辨体-辨病-辨证”三辨模式,用“乌梅,蝉衣,首乌,野生赤芝”四味药改善过敏体质状态,加“土茯苓”并随证加减取得良好的效果.本文笔者从偏头痛的病因病机,偏头痛与过敏的相关性及王琦教授治疗偏头痛的思路出发选取两则验案加以分析.
Antibody class switching of translating B cell into IgE antibody from producing IgM simply was the critical mechanism of allergic diseases.This paper summarized the preliminary researches of IgE Antibody class switching from the aspects as cellular mechanism,molecular mechanism and gene mechanism.It also compared the class switch mechanism of IgE antibodies with the fundamental principles of constitution theories as' theory of the process of life','theory of natural endowment and heredity','theory of environmental constraint',and so on,in order to seek the intrinsic connection between them,and laid a theoretical foundation for studying the allerqic construction from the perspective of lgE antibody class switching.
Objective: To summary the medication rule for TCM in the treatment of urticaria and provide guidance for the treatment of urticarial. Methods: The documents were retrieved from some domestic databases, such as Medline, Cochrane Library, CNKI, VIP and CBM. By searching the terms of ‘traditional Chinese medicineu0027, ‘traditional Chinese drugu0027, ‘traditional Chinese herbu0027, ‘urticariau0027, ‘hidden rashesu0027, ‘randomu0027, we included 199 documents involving 130 formulas, 191 traditional Chinese drugs and 2 097 medicine-times. A database was established by means of Microsoft Excel 2007 in order to analyze some characteristics of the formulas and traditional Chinese drugs, such as nature, l avor, channel tropism, ei ciency, and indications of principle. Results: In medication frequency of more than 5, we found that the tonic drugs, drugs for treating exterior syndromes and heat-clearing drug mainly dominated in traditional Chinese drug; The main natures were warm, cold and neutral; The main l avors were sweet, acrid and bitter; The channel tropisms mainly distributed into liver, spleen, lung and heart. Conclusion: The TCM treatment of urticaria shows regularity according to isu0027 s pathogene, provided effective guide for clinical medication.
Objective To study the relationship between facial skin conditions and TCM constitutional types; To provide new means and methods for guidance of skin conservation and skin health improvement from the aspect of constitutional types. Methods Totally 2241 female volunteers in nine provincial capital cities and municipalities participated in this research, and were investigated by web questionnaire. Constitutional types were determined according to the“Classification and Judgment of TCM Constitutional Types”standard issued by China Association of Chinese Medicine. Skin color, glossiness, delicate degree, moistness, firmness, resilience, uniformity and other indicators were scored. Skin conditions were classified into three groups:1-3 score group, 4-6 score group, and 7-9 score group, and the constitutional differences among each group were compared. Results Yang-deficiency and mild constitutional types accounted for the largest proportion. Yellowish skin color accounted for the largest proportion, and the next were white, dark and reddish. Yang-deficiency constitutional type accounted for the largest proportion in 1-3 score group, and the second was Qi-stagnation; Yang-deficiency and mild constitutional types accounted for the largest proportion in 4-6 score group; mild constitutional type accounted for the largest proportion in 7-9 score group. Conclusion People with mild constitutional type got highest score in every skin index, while biased constitutional types influenced the score in varied degrees. The constitutional types which influenced Chinese urban females’ facial skin index more were Yang-deficiency and Qi-stagnation constitutional types.
目的 考察中国城市女性的体质状况,为个体化养生养美服务.方法 在北京、上海、广州等9个省会城市和直辖市召集女性志愿者2 241名,以网络问卷的方式对受访者进行中医体质类型调查,按照中华中医药学会颁布的《中医体质分类与判定》判定中医体质类型.按年龄段、职业、婚姻状况和地域进行分层,考察9种体质类型的频数分布.结果 中国城市女性平和质占21.95%,在不同年龄段、不同职业、不同婚姻状况和不同地域的女性偏颇体质频数分布中,气虚质、阳虚质、阴虚质频数居前3位.结论 应针对气虚质、阳虚质、阴虚质对中国城市女性开展养生养美等保健服务.
