Проведено исследование гепатопротекторных свойств экстракта сухого Calendula officinalis L. на экспериментальной модели острого токсического гепатита, вызванного введением четыреххлористого углерода. Показано, что экстракт C. officinalis при внутрижелудочном введении в дозе 100 мг/кг в течение 13 дней снижает выраженность окислительного стресса, повышая активность каталазы, супероксиддисмутазы и содержание восстановленного глутатиона. Исследуемое фитосредство способствует увеличению активности глутатионпероксидазы на 15 % (p ≤ 0,05) и глутатионредуктазы на 39 % в ткани печени крыс относительно контроля (p ≤ 0,05), а также снижает содержание малонового диальдегида на 28 % (p ≤ 0,05). Установлено, что экстракт C. officinalis положительно влияет на энергетический обмен в гепатоцитах, что отражается на повышении содержания АТФ на 28 % (p ≤ 0,05), снижении содержания лактата на 31 % (p ≤ 0,05) и нормализации соотношения лактат/пируват в ткани печени крыс с экпериментальным токсическим гепатитом. Полученные результаты позволяют рекомендовать экстракт сухой C. officinalis для дальнейших исследований в качестве перспективного растительного гепатопротекторного средства.
The influence of the complex plant remedy (Urtica dioica L., Polygonum aviculare L., Achillea millefolium L., Zingiber officinalis L., Cinnamomum cassia L.) on the morphofunctional state of the liver in rats with ethanol-induced hepatitis was studied. The liver injury was simulated with 40% ethanol introduction to rats in dose of 10.0 ml/kg for 7 days. The extract of the complex remedy was administered to animals in dose of 200 ml/kg for 14 days. The findings showed that the tested remedy inhibited the development of microcirculatory impairments, decreased the intensity of inflammatory infiltration and diminished the amount of hepatocytes subjected to adipose and cloudy degeneration as well as to necrotic changes. The increased amount of hypertrophied and binuclear hepatocytes in the area of the portal tract and central vein testifies reparation processes in the liver of the rats treated with the tested plant extract.
To study the probable negative influence of the plant remedy “Aterophyt” on the reproductive function of the laboratory animals the following tests have been carried out, namely the probable occurrence of embryo-toxicity, teratogenicity, fetotoxicity and its influence on the postnatal development of the laboratory animals progeny. The experiments have been carried out on the white Wistar rats. It has been established that the “Aterophyt” at the volumes of 5 and 10 ml/kg has no embryotoxic, teratogenic and fetotoxic effect. The introduction of the “Aterophyt” at the volume of 10 mg/kg since 6th to 19th day of pregnancy does not influence on the postnatal development of white rats progeny.
Hypoxia, as universal pathological process accompanying or determining the development of various pathological conditions, activates lipid peroxidation followed by disturbance of cell energy-producing system. The paper deals with the influence of phytoremedy «Tireoton» on free radical processes and energy metabolism in the brain of rats subjected to hypobaric hypoxia/reoxygenation. Experiments were conducted on 44 Wistar rats. It has been found that «Tireoton» at the dose of 50 mg/kg increases catalase activity and the content of reduced glutathione on the average by 20% as well as decreases the content of TBA-active products in 1,5 times in rat brain against the background of acute hypobaric hypoxia. «Tireoton» administration improves cell energy status at the early post-hypoxia period: a 2,8-fold increase in ATP content and 1,5-fold decrease in lactate/pyruvate ratio compared to the control animals were noted. The results obtained permit «Tireoton» to be considered as promising herbal remedy to correct free radical and energetic processes in the brain under hypoxia/reoxygenation.
В опытах на экспериментальных животных изучено влияние превентивного курсового введения препарата растительного происхождения «кардекаим» на основные проявления стресса. В опытах in vitro изучены механизмы антистрессорного эффекта тестируемого препарата. Курсовое превентивное введение кардекаима в дозе 100 мг/кг 7 дней перед острым иммобилизационным стрессом оказывает выраженное стресс-протекторное действие, препятствуя развитию признаков «триады Селье» на 15 – 63 % (p < 0,05). Преобладающим молекулярно-клеточным механизмом стресс-протекторного действия кардекаима является ингибирование процессов перекисного окисления липидов, что обусловлено наличием соединений фенольной природы, обладающих прямым антирадикальным действием и повышающих мощность эндогенной антиоксидантной системы.
The paper deals with the influence of Phlojodicarpus sibiricus extract at the dose of 50 mg/kg body weight on ATP content, pyruvate kinase (PK) and Н+-АТPase activity in brain cells after ischemia-reperfusion. In total, 36 albino Wistar rats were taken into experiments. Bilateral occlusion of both common carotid arteries for 5 min. followed by reperfusion for 60 min. resulted in the disturbance of energetic processes in brain cells. It has been found that the extract in question increases the ATP concentration by 50 % and leads to a 1.5- and 1.7-fold induction of Н+-АТPase and pyruvate kinase activities respectively compared with the control animals.
The aim of the work: to determine the neuroprotective properties of Humulus lupulus dry extract in chronic emotional stress. The studies were carried out on 48 white Wistar rats. Neuroprotective properties of the plant remedy were studied according to the “Manual on the experimental (pre-clinical) study of new pharmacological substances” (2005). It has been established that the Humulus lupulus dry extract increases the number of outputs and staying period in the open branch of the lifted crosswise labyrinth and stimulates the development of conditioned reflex of passive avoiding in animals and causes the safety of a memorable trace in the remote period after training in the passive avoidance reaction (PAR) test. The property of Humulus lupulus extract to restore the emotional state and cognitive functions of the animal in stress conditions is due to the weakening of neurotransmitter systems disturbances.
Stress-protective and antioxidant effects of Humulus lupulus dry extract were studied in experiments on white Wistar rats at prolonged emotional stress. It has been established that the H. lupulus dry extract had stress-protective effect, reducing the manifestation of degenerative catabolic changes in the internal organs of white rats in chronic emotional stress. The antistress activity of Humulus lupulus dry extract is caused by the pronounced antioxidant activity. On the background of the test extract introduction the MDA content decreased and the activity of catalase, SOD and the GSH content in the blood increased.
The aim of the work: determination of pharmacological properties of the herbal remedy « Pankreophyt». The studies were carried out on 78 white Wistar rats. Pharmacological properties of the herbal remedy were studied according to the “Manual on the experimental (pre-clinical) study of new pharmacological substances” (2005). It has been established that «Pankreophyt» in the dose of 300 mg/kg has a marked anti-inflammatory activity in aseptic peritonitis, reducing the volume of peritoneal fluid and degranulation level of mast cells; hypoglycemic activity, reducing the level of glucose in the blood serum in alimentary glycemia. Besides, the tested herbal remedy has analgesic effect, reducing the amount of «acetic cramps».
Pharmacotherapeutic efficiency of the dry extract from Lomatogonium carinthiacum in toxic hepatitis was studied. Experimental hepatitis was caused by the introduction of tetracycline hydrochloride to Wistar white rats in the dose of 1.0 g/kg of the animal weight once a day for 7 days. The extract from L. carinthiacum was administered per os in the dose of 100 mg/kg for 26 days. 1t has been established that the extract from L. carinthiacum has a marked hepatoprotective effect in the case of tetracycline hepatitis in white rats that is characterized by inhibition of the disturbances in choleretic and cholate-synthetic functions of the liver: the increase of bile secretion rate, preservation of cholate concentration in the bile, the decrease in the content of cholesterol triacylglycerides and alkaline phosphatase activity in the animal blood serum during the whole period of experiments. The tested remedy diminishes dystrophic and necrotic processes in hepatocytes, decreases the intensity of inflammatory infiltration in the liver and stimulates regeneration of liver cells in tetracycline hepatitis. The decrease of MDA content in the liver homogenate and increase in catalase activity against the same indices in control animals testify the marked antioxidant effect of L. carinthiacum.
There has been conducted the study of the antioxidant activity of the vegetable remedy “Pankreophyt” and its components in conditions in vitro ; there were defined an antiradical activity (DPPHmethod), the ability to bind of superoxide radical anion, nitric oxide binding molecules and chelating ions Fe2+. It has been established that vegetative remedy is characterized by antiradical action (IC50 = 29,4 mkg /ml) due to the presence in it a composition of tannins and phenologlykosides; has the ability to inactivate reactive oxygen species and other initiators of the process of lipid peroxidation O2NO(IC50=78,7 mkg/ml, IC50=721,3 mkg/ml), due to the presence in it a composition of P. fruticosa, V. myrtillus, I. helenium and H. perforatum. Fe2+chelating activity of herbal remedy (ICFe2+ = 750,2 mkg/ml) is stipulated by the presence of50polysaccharides in its chemical composition. The results of these studies allow to consider vegetative remedy “Pankreophyt”as a potentially promising antioxidant in combined therapy and prevention of pancreatitis.
The effect of Lomatogonium carinthiacum ( Wulf.) Reichenb. dry extract on the energetic status and antioxidant system of rats with experimental tetracycline hepatitis was investigated. It was established that the using of L. carinthiacum dry extract result to inhibition of lipid peroxidation, increasing the activity of endogenic antioxidant system, normalising of energetic status of hepatocytes and restrict the development of distrophic processes in liver.
