To study the epidemic status,molecular characteristic and phylogenetic relationship of porcine pseudorabies virus (PRV),gD gene in 17 PRV isolates isolated in western Fujian province from 2013-2015 were sequenced and analyzed.The results suggested that nucleotide sequences and amino acid sequences of 17 PRV isolates shared high homology with 7 classic PRV isolates isolated from different provinces in China,which was 99.0% 100.0%,but shared relatively low homology with 4 foreign PRV classic isolates,which were 98.6 %-99.3% and 97.0%-99.0%,respectively.The multiple sequence alignment of nucleotide sequence revealed that the most characteristic change of gD nucleotide sequence was the insertions of 6 continuous base(CCGGCC) from 831 to 836 bp,which led to the increase of 2 amino acids (RP) of gD amino acid sequence from 275 to 276.And antigenic analysis showed that these site were also located in the antigenic epitope regions of gD protein.Phylogenetic analysis of gD gene showed that 17 PRV isolates,7 classic PRV isolates isolated from China,Korea Yangsan and American Becker isolates formed a relative independent sub-branch far from 2 PRV isolates isolated from Hungary,such as Bartha.
为了解闽西地区2013年以来猪伪狂犬病病毒(PRV)流行株的现状、分子生物学特征和遗传演化规律,收集了闽西不同地区的14株PRV分离株,并对其gC基因进行遗传变异分析.结果表明,14株PRV分离株与2012年以来国内不同省份分离的PRV变异株的核苷酸序列同源性为98.8%~99.7%,而与Bartha疫苗株核苷酸序列同源性为93.6%~93.8%.氨基酸多序列比对发现,14株PRV分离株和近年来国内分离的PRV毒株gC基因氨基酸序列同源性较高,有很多相同的特征性氨基酸替换,其中最有特征性的是aa52~aa61,aa63~aa69位和aa186~aa194位氨基酸的替换和插入.遗传进化树分析表明,14株PRV分离株与2012年以来国内不同省份分离的PRV株位于一个相对独立的大分支中,且形成了一个独立的亚分支,而与Bartha疫苗株存在很大差异,亲缘关系较远.抗原表位预测分析表明,PRV闽西分离株与Bartha疫苗株相比发生了抗原漂移,该差异是否预示闽西地区流行株正在向远离疫苗株的方向变异尚有待进一步研究.
为了解2013-2014年我国闽西地区猪伪狂犬病病毒(PRV)流行株的现状、分子生物学特征和遗传演化规律,收集闽西不同地区的15株PRV分离株,并对其gE基因进行遗传变异分析.结果表明,15株PRV分离株与2012年以来国内不同省份分离的14株PRV变异株核苷酸及氨基酸序列的同源性较高,分别为98.7%~99.9%和97.1%~99.8%;而与15株PRV经典株的核苷酸及氨基酸序列的同源性相对较低,分别为97.0%~99.6%和94.6%~99.3%.氨基酸多序列比对发现,gE氨基酸序列最具特征性的变化是第48位和第496位各有1个天冬氨酸(D)的插入,该插入特征为PRV变异毒株的重要标志.同时,抗原性分析发现这2个位点还位于gE蛋白的抗原表位区内.遗传进化树分析结果表明,15株PRV分离株与近3年国内不同省份分离的PRV株位于一个相对独立的分支中,而与经典毒株处于不同的分支中,亲缘关系相对较远.以上结果表明PRV变异毒株已成为我国闽西地区主要的流行毒株.
One local strain of pseudorabies virus(PRV)was isolated from the brain of the 10-day-old pig from Fujian Province identified by methods such as necropsy,PCR and virus isolation and culture et al.It was named as PRV Fujian-LY strain.The analysis of molecular characteristic and phylogenetic relationship of its virulence associated genes TK and gE showed that the homologies of the TK gene nucleotide sequences and amino acid sequences were from 98.5% to 99.8% and from 98.1% to 99.4%,respectively.Meanwhile,the homologies of the gE gene nucleotide sequences and amino acid sequences were from97.6%to 99.8% and from 95.5%to 99.3%,respectively.The multiple sequence alignment of amino acid sequences revealed that 2unique amino acid mutations were identified in the TK sequence of Fujian-LY isolate as compared to the reference strains.It was that Lys mutated into Arg at position 16 aa and 129 aa Ser mutated into Gly,respectively.Among them,the amino acid mutation at position 16 aa led to ATPbinding domain of TK consisted of-G **G*GK-into-G **G *GR-.The most characteristic change of gE amino acid sequence of Fujian-LY isolate were two insertions of Asp at sites of 48 aa and 496 aa,and it was an important marker for the PRV variant.Phylogenetic analysis showed that the PRV Fujian-LY strain and the PRVs isolated from different province in China since 2012 had more close relationship far from the classic PRVs,and it belongs to the PRV variants prevailing in China.
