目的 探讨SKA3在甲状腺乳头状癌中的表达及其与预后、免疫细胞浸润的关系.方法 采用免疫组化SP法检测SKA3蛋白在21例甲状腺乳头状癌组织和16例正常组织中的表达.应用癌症基因组图谱和基因芯片公共数据库分析SKA3 mRNA在565例癌组织和111例正常组织中的表达及与临床病理特征的关系.生物信息学分析SKA3的生物学功能及与免疫细胞浸润的关系.结果 SKA3蛋白和mR-NA在癌组织中均表达上调.SKA3与甲状腺乳头状癌病理N分期、无瘤生存期和无进展生存期等有关.SKA3与B细胞、CD8+T细胞、CD4+T细胞、巨噬细胞、中性粒细胞和树突状细胞的浸润水平呈正相关.结论 SKA3在甲状腺乳头状癌组织中表达上调,与患者预后和肿瘤的免疫细胞浸润相关.
目的:探讨Child-Pugh(CTP)、ALBI、MELD、MELD-Na评分预测肝细胞癌(hepatocellular carcinoma,HCC)经肝动脉化疗栓塞(transcatheter arterial chemoembolization,TACE)治疗后并发慢性加急性肝衰竭(acute-on-chronic liver failture,ACLF)的价值.方法:回顾性分析2013月10月至2015年10月广西医科大学附属肿瘤医院接受首次TACE治疗的711例HCC患者的临床资料,采用Logistic回归分析及受试者工作特征(receiver operating characteristic,ROC)曲线评估4种评分模型预测ACLF的价值.结果:单因素、多因素分析结果均提示4种评分模型能独立预测TACE术后ACLF的发生(均P<0.05);ROC曲线分析显示,ALBI的受试者工作特征曲线下面积(area under curve,AUC)均显著高于其余三种评分的AUC(均P<0.001).不同分型ACLF按4种评分模型的最佳截断(cut-off)值分组后,Child-Pugh评分>5.5分、ALBI>-2.29分、MELD>8.08分、MELD-Na>8.08分的患者ACLF总发病率和ACLF C型发病率均高于该4种评分低于cut-off值的患者(均P<0.001),而ACLF A型、ACLF B型的发病率两组间差异均无统计学意义(均P>0.05).结论:Child-Pugh、ALBI、MELD及MELD-Na评分对TACE治疗后ACLF均有一定预测价值,ALBI预测价值最优.
In this study,the carboxymethylation of polysaccharides (LYP) extracted from longan (Dimocarpus longan) pulp was investigated using monochloroacetic acid (MCA) with polysaccharides-to-MCA ratio,reaction time and reaction temperature as independent variables.The degree of substitution (DS) was taken as response value to optimize the important reaction conditions by response surface methodology.Meanwhile,the antioxidant activity and immunoregulatory of carboxymethlated polysaccharides (CM-LYP) in vivo were measured.The results showed that the optimized conditions that provided the maximum DS of 1.053 were determined as follows:reaction time,3.2 h;MCA concentration,1.2 mol/L;and reaction temperature,73 ℃.The antioxidant activity.of CM-LYP was concentration dependent in a certain range of concentration.The percentage inhibition of hydroxyl and superoxide anion radicals,lipid peroxidation,red blood cell hemolysis induced by H2O2 were (42.35 ± 5.67)%,(51.91 ± 5.34)%,(67.91 ± 5.72)%,and (47.23 ± 3.5)% by LYP,and (84.39 ± 4.47)%,(87.91 ± 7.32)%,and (79.85 ± 2.92)%,and (54.66 ± 2.83) % by CM-LYP,respectively,implying that the antioxidant activity of LYP was improved as compared to the native polysaccharides.The immunoregulatory activity in vivo showed that CM-LYP significantly improved spleen index in immunosuppressed,increased serum hemolysin level and lysozyme activity in serum and spleen,and maintained Th1/Th2 balance,and the effect was better than that of LYP.
Objective: To improve the curcumin content extracted from Curcuma kwangsiensis with surfactant and study its effect on the apoptosis of human ovarian carcinoma SKOV3 combined with cisplatin.Methods: Moistened with appropriate quantity of surfactant,the curcumin was extracted by alcohol and assayed by HPLC for its ratio.The cells inoculated in the logarithm phase were administrated with 25,50,and 100 μmol/L of curcumin or combined medication: 25,50,and 100 μmol/L of curcumin was combined with 10 μmol/L of cisplatin,respectively,cultured for 48h and their absorbances was assayed by MTT method to calculate the inhibition rate of the cell growth.The normal controls,the 50 μmol/L of curcumin group,the 10 μmol/L of cisplatin group,and the 50 μmol/L of curcumin combined with 10 μmol/L of cisplatin was AO/EB stained to observe their apoptosis.Results: A maximum yield was obtained from each 10 g of Curcuma bulk material moistened by 10 mL 1% of Sodium dodecyl sulfate(SDS) for 30 min,extracted with double refluxed ethanol for 1.5 h per time.The MTT outcome and observation of Fluorescent staining indicated a synergistic induction to SKOV3 apoptosis by curcumin and cisplatin combination.Conclusion: SDS affects significantly to the curcumin extraction from curcuma.SKOV3 apoptosis can be synergistically inducted by curcumin in Curcuma kwangsiensis and cisplatin combination.
Objective To establish a high performance liquid chromatography(HPLC) method for the content determination of levodopa isopentyl ester hydrochloride.Methods The Ultimate C18 column(250 mm×4.6 mm,5 μm) was used and the mobile phase was acetonitrile was 0.05 mol/L potassium dihydrogen phosphate(adjusting pH 3.5 with phosphoric acid,70 ∶30),the flow rate was 1.0 mL/min.The detection wavelength was 280 nm.The column temperature was 30 ℃and the injection volume was 20 μL.Results All the related substances gained a completely chromatographic separation with the principal peak.A good linear relationship was observed in the concentration range of 16.1-322.0 μg/mL,with the correlation coefficient of 0.999 9(n=6).The average recovery rate was 98.41%and RSD was 1.85%(n=6).Conclusion The method is proved to be accurate and sensitive with high precision and strong specificity,which can be applicable for the content assay of levodopa isopentyl ester hydrochloride.
Objective To study the changes of chemical composition in volatile oil from Zingiber officinale Rosc.during processing. Methods The volatile oil was extracted from samples with the interval of 7-15 days,by water distillation method and analyzed by GC-MS. Results When the content of water in the Zingiber officinale Rosc.was 99.16%,69 chemical compositions were identified;When the content of water was 4.75%,26 chemical compositions were identified.There were 25 chemical components in the volatile oil. Conclusion With the decrease of water content in Zingiber officinale Rosc.,the yield of volatile oil declines gradually,and the chemical composition in volatile oil changes greatly.
OBJECTIVE: To observe the effects of ginger essential oil on hemorheology of rats with acute blood stasis. METHODS: The model of acute blood stasis was established with high-molecular weight dextran intravenously in rats. Rats were divided into 6 groups, i.e. normal control group(0.05% tween-80),model group (0.05% tween-80),Compound danshen tablet group(1 500 mg·kg-1) and Ginger essential oil high-dose, medium-dose and low-dose groups(80,20,5 mg·kg-1). Hemorheological parameters of rats were determined. RESULTS: Ginger high-dose, medium-dose and low-dose groups could markedly decrease the whole blood viscosity with low, middle and high shear rate, plasma viscosity, blood reduce viscosity, hematocrit and erythrocyte conglomeration index; It could also rise erythrocyte deformation index. CONCLUSION: Ginger essential oil can significantly ameliorate the hemorheology and inhibit platelet aggregation in acute blood-stasis model rats, and prevent the increase of blood viscosity.
[目的]对猫豆中营养成分进行初步研究.[方法]以乙醇回流提取法提取脂肪酸,采用气相色谱法进行不饱和脂肪酸分析;以碱提酸沉法提取蛋白质,采用氨基酸分析仪进行氨基酸分析.[结果]以峰面积归一化法计算,猫豆中不饱和脂肪酸分析结果为:油酸18.3%、亚油酸48.0%、亚麻酸7.7%,三者占不饱和脂肪酸总量的74.0%;猫豆蛋白质中常见氨基酸总量为25.8%,检测到16种氨基酸,包括7种必需氨基酸.[结论]猫豆油脂中含有一定量的不饱和脂肪酸,包括油酸、亚油酸、亚麻酸;猫豆蛋白质中氨基酸种类较齐全;猫豆具有较高的营养价值.
Objective To assess the therapeutic effect and safety of cyclosprine in the treatment of proliferative lupus nephritis.Methods The CENIRAL of Cochrane library,PuBmed,EMBASE,CBMdisc,CNKI,VIP that is to the 28th February 2010 were searched.Three reviewers assessed the quality of including studies,extracted data and Meta-analyzed the homogenous studies.Results Seven studies involving 361 participants were included,and all studies were randomized control trials(RCT).There were statistical difference in proteinuria(24 h),Anti-ds DNA,CH50,height velocity,height SDS and C3(mg/dl) between cyclosporine and CTX,but no statistical difference in other indicators;There were statistical difference in CCr(6,24,48 months),arthralgias and gastrointestinal disorders between cyclosporine A and AZA,but no statistical difference in other indicators.There were statistical difference in proteinuria(24 h) between cyclosporine and pred,but no statistical difference in other indicators.Conclusion Available evidence show:comparing with CTX or Pred,CsA can improve the clinical symptom and spare steroid,but cannot control serological activity;Comparing with AZA,CsA has more side effects,while the improving effect was similar;Larger scale clinical trials are needed to evaluate its effectiveness.
目的 通过半随机对照交叉试验,验证短期小剂量糖皮质激素可以有效地减少细胞因子的释放,从而减少肾病综合征复发.方法 纳入隔日小剂量糖皮质激素(0.3~0.8mg/kg)维持治疗的原发性肾病综合征患儿,在第一次急性上呼吸道感染时,将患儿随机分为Ⅰ或Ⅱ组,分别接受醋酸泼尼松或安慰剂治疗,当第二次急性上呼吸道感染时,治疗药物则相反.醋酸泼尼松的治疗方案为将隔日顿服改为每日服用.结果 共纳入32例年龄在1~16岁之间的糖皮质激素维持治疗的原发性肾病综合征患儿,其中26例完成了研究.服用安慰剂组的26例患儿中,有19例复发,复发率为73%;而服用醋酸泼尼松的26例患儿中,有6例复发,复发率为23%,两组复发率差异有显著性(P<0.05).结论 肾病综合征患儿急性上呼吸道感染期间,增加醋酸泼尼松的剂量可以有效预防肾病综合征的复发.
Objective To assess the efficacy and safety of mycophenolate mofeti in the treatment of proliferative lupus nephritis.Methods We searched the Cochrane library、PubMed、EMBASE、CBMdisc、CNKI、VIP up to 30 June 2009.Three reviewers assessed the quality of including studies,extracted data.Meta-analysis for the result of homogenous studies were made with RevMan 5.0.Results Fourteen studies involving 1 032 participants were included.All included studies were graded on randomization,allocation concealment and blinding.Seven studies were graded B,and the other seven studies were graded C.Meta-analysis based on included studies showed:①Complete remission rate(OR=2.16,95%CI:1.31-3.55)and total remission rate(OR=1.51,95%CI:1.10-2.07)had statistical significance in MMF compared with CTX.MMF was better than control groups;amenorrhea(OR=0.16,95%CI:0.05-0.50)infections(after sensitivity analysis)(OR=0.38,95%CI:0.23-0.61)and leucopenia(OR=0.43,95%CI:0.27-0.71)had statistical significance.The side effect in MMF was less than in control groups.But mortality、ESRD 、partial remission rate、gastrointestinal symptoms、herpes zoster had no statistical significance.②Amenorrhea(OR=0.20,95%CI:0.07-0.61),infections(OR=0.46,95%CI:0.22-0.99)and leucopenia(OR=0.12,95%CI:0.03-0.52)had statistical significance in MMF compared with CTX/AZA.The side effect of MMF was less than control groups.But mortality,ESRD,complete remission rate,partial remission rate,total remission rate,relapse,SCr double,gastrointestinal symptoms and herpes zoster had no statistical significance.Conclusions Present evidences showed the efficacy of MMF and CTX or CTX/AZA was similar,but the safety of MMF was better than CTX or CTX/AZA.
